Population Genetics of Plasmodium vivax in Four High Malaria Endemic Areas in Thailand

  • Recent trends of malaria in Thailand illustrate an increasing proportion of Plasmodium vivax, indicating the importance of P. vivax as a major causative agent of malaria. P. vivax malaria is usually considered a benign disease so the knowledge of this parasite has been limited, especially the genetic diversity and genetic structure of isolates from different endemic areas. The aim of this study was to examine the population genetics and structure of P. vivax isolates from 4 provinces with different malaria endemic settings in Thailand using 6 microsatellite markers. Total 234 blood samples from P. vivax mono-infected patients were collected. Strong genetic diversity was observed across all study sites; the expected heterozygosity values ranged from 0.5871 to 0.9033. Genetic variability in this study divided P. vivax population into 3 clusters; first was P. vivax isolates from Mae Hong Son and Kanchanaburi Provinces located on the western part of Thailand; second, Yala isolates from the south; and third, Chanthaburi isolates from the east. P. vivax isolates from patients having parasite clearance time (PCT) longer than 24 hr after the first dose of chloroquine treatment had higher diversity when compared with those having PCT within 24 hr. This study revealed a clear evidence of different population structure of P. vivax from different malaria endemic areas of Thailand. The findings provide beneficial information to malaria control programme as it is a useful tool to track the source of infections and current malaria control efforts.

Fluctuations in the Abundance of Common Squid, Todarodes pacificus and Environmental Conditions in the Far East Regions during 52 Years

  • Environmental variables, fishing and biological data of the common squid, Todarodes pacificus were used to describe changes in structure, migration and abundance of the squid population in relation to ocean climate shifts. It was possible to consider the main groups of the squid (autumn and winter-spawned groups) as a single population to aid conservation in the waters around Korea and Japan (TWC and KOC regions). The patterns of yearly fluctuations in abundance of the squid population in the two regions were the same during 52 years of $1952{\sim}2003$. The abundance of the squid began to decrease in both regions in the early 1970s, remained low in the 1980s and the main squid groups synchronously increased in the 1990s coincident with favorable changes of thermal conditions and plankton production in those ecosystems. The mechanisms of changes in the structure, distribution and abundance of common squid population in relation to current-mediated migration circuits are explained on the basis of phenological variables responding to climate shifts.

가막산 참굴의 집단 구조 분석 (The Population Genetic Structure of the Oyster Crassostera gigas (Bivalvia:Ostreidae) from Gamak Bay in Korea)

  • 본 연구는 2007년 3월 가막만에서 양식하고 있는 참굴 34마리를 대상으로 미토콘드리아COI gene 염기서열분석을 통하여 유전자 집단을 조사했다. PCR 증폭에 사용된 primer는 HCO2918과 LCO1491로 product는 658 bp였다. 유전자 배열 결과 가막만 참굴 집단에서 CR1, CR2, CR3, CR4 4개의 haplotype이 나타났다. 그 중 CR3 haplotype이 73% 발생되어 가장 높은 빈도율을 보였다. 계층구조에서도 가막산 참굴은 남해안산과 유전적 유연관계를 보이나, 유전적 거리는 남해안 및 서해안과도 마이너스 값을 보이며 통계적으로도 유전적으로 하나의 집단을 형성하고 있음을 알 수 있다.

사회네트워크 분석을 이용한 광주 전남지역의 공간 구조 변화 및 중심지 분석 (Analysis of Spatial Structures and Central Places of Gwangju and Jeonnam Region using Social Network Analysis)

  • When an age of low growth and population decline, population migration plays an important role in spatial structure of region. There have been many researches on migration and regional spatial structure. The purpose of this study is to examine the changes of Gwangju and Jeonnam region's spatial structure and central area using social network analysis methods. For analysis it was used that population and migration data and passenger OD(Origin and Destination) travel data released by Statistics Korea and Korea Transport Database(KTDB). Using Gephi 0.8.2, migration and passenger OD networks were visualized, and this describe network flow and density. The results of the network centrality analysis show that the most populated village is not always network center though population mass is an important factor of central places. The average eigenvector centrality of 2010 migration is the lowest during 2005-2015, and it means few regions have high centralities. When comparing migration and travel networks, travel data is more effective than migration data in determining the central location considering spatial functions.

  • Korea is one of the most advanced countries in kelp aquaculture. The brown algae, Undaria pinnatifida and Saccharina japonica are major aquaculture species and have been principally utilized for human food and abalone feed in Korea. This review discusses the diversity, population structure and genomics of kelps. In addition, we have introduced new cultivar development efforts considering climate change, and potential carbon sequestration of kelp aquaculture in Korea. U. pinnatifida showed high diversity within the natural populations but reduced genetic diversity in cultivars. However, very few studies of S. japonica have been conducted in terms of population structure. Since studies on cultivar development began in early 2000s, five U. pinnatifida and one S. japonica varieties have been registered to the International Union for the Protection of New Varieties of Plants (UPOV). To meet the demands for seaweed biomass in various industries, more cultivars should be developed with specific traits to meet application demands. Additionally, cultivation technologies should be diversified, such as integrated multi-trophic aquaculture (IMTA) and offshore aquaculture, to achieve environmental and economic sustainability. These kelps are anticipated to be important sources of blue carbon in Korea.

  • In recent years, the population structure has been changed by the progresses of 6th-industrialization and transportation in farming areas. This study aims to suggest a regional development plan of farming areas in accordance with the demographic changes. Population structure indicators were derived from previous studies and literature review in order to identify the types of farming areas. Demographic indicators separated to depopulation and population growth in farming areas through a standardized scoring method. This research found that the division of economy is not only the most important division in any other regional development divisions but also necessary to develop new sources of non-farm income through traditional culture, natural environment. In the social division, it is necessary to secure the facilities for the formation of a sense of community to multicultural families and existing residents in farming areas. In the environmental division, it is desirable to improve the quality and satisfaction of life for residents such as the sidewalk and park that utilize ecological environment, culture, history for both depopulation and population region. In the physical division, there is a need for improvement of the facilities of basic living infrastructure service such as roads, water supply and sewerage systems. In the institution division, sustainable financial support of the central government policy for farming areas is crucial for the improvement of residential environment in the farming areas of depopulation and population region.

Small-scale spatial genetic structure of Asarum sieboldii metapopulation in a valley

  • Background: Asarum sieboldii Miq., a species of forest understory vegetation, is an herbaceous perennial belonging to the family Aristolochiaceae. The metapopulation of A. sieboldii is distributed sparsely and has a short seed dispersal distance by ants as their seed distributor. It is known that many flowers of A. sieboldii depend on self-fertilization. Because these characteristics can affect negatively in genetic structure, investigating habitat structure and assessment of genetic structure is needed. A total of 27 individuals in a valley were sampled for measuring genetic diversity, genetic distance, and genetic differentiation by RAPDPCR. Results: The habitat areas of A. sieboldii metapopulation were relatively small (3.78~33.60 m2) and population density was very low (five to seven individuals in 20×20 m quadrat). The habitat of A. sieboldii was a very shady (relative light intensity = 0.9%) and mature forest with a high evenness value (J = 0.81~0.99) and a low dominance value (D = 0.19~0.28). The total genetic diversity of A. sieboldii was quite high (h = 0.338, I = 0.506). A total of 33 band loci were observed in five selected primers, and 31 band loci (94%) were polymorphic. However, genetic differentiation along the valley was highly progressed (Gst = 0.548, Nm = 0.412). The average genetic distance between subpopulations was 0.387. The results of AMOVA showed 52.77% of variance occurs among populations, which is evidence of population structuring. Conclusions: It is expected that a small-scale founder effect had occurred, an individual spread far from the original subpopulation formed a new subpopulation. However, geographical distance between individuals would have been far and genetic flow occurred only within each subpopulation because of the low density of population. This made significant genetic distance between the original and new population by distance. Although genetic diversity of A. sieboldii metapopulation is not as low as concerned, the subpopulation of A. sieboldii can disappear by stochastic events due to small subpopulation size and low density of population. To prevent genetic isolation and to enhance the stable population size, conservative efforts such as increasing the size of each subpopulation or the connection between subpopulations are needed.

The utility of sperm DNA damage assay using toluidine blue and aniline blue staining in routine semen analysis

  • Objective: The aim of the present study was to examine the relationship among male age, strict morphology, and sperm chromatin structure and condensation. Methods: Sperm samples from a total of 100 men underwent semen analysis, and sperm chromatin structure and condensation were assessed with toluidine blue (TB) and aniline blue (AB) tests. Results: Prevalence of strict morphology of less than 4%, and abnormal sperm chromatin structure and condensation did not show any statistically significant differences according to male age (p=0.605, p=0.235, and p=0.080). No significant correlation was demonstrated among age of male partners, strict morphology, and abnormal sperm chromatin structure using TB and AB tests. However, abnormal sperm chromatin condensation was positively associated with sperm chromatin structure (r=0.594, p=0.000) and showed negative correlation with strict morphology (r=-0.219, p=0.029). Conclusion: The tests for sperm chromatin condensation showed a significant association with strict morphology. Further study is needed to elucidate the relationship between clinical outcome and sperm chromatin tests.

EST-SSR Based Genetic Diversity and Population Structure among Korean Landraces of Foxtail Millet (Setaria italica L.)

  • Understanding the genetic variation among landrace collections is important for crop improvement and utilization of valuable genetic resources. The present study was carried out to analyse the genetic diversity and associated population structure of 621 foxtail millet accessions of Korean landraces using 22 EST-SSR markers. A total of 121 alleles were detected from all accessions with an average of 5.5 alleles per microsatellite locus. The average values of gene diversity, polymorphism information content, and expected heterozygosity were 0.518, 0.594, and 0.034, respectively. Following the unweighted neighbor-joining method with arithmetic mean based clustering using binary data of polymorphic markers, the genotypes were grouped into 3 clusters, and population structure analysis also separated into 3 populations. Principal coordinate analysis (PCoA) explained a variation of 13.88% and 10.99% by first and second coordinates, respectively. However, in PCoA analysis, clear population-level clusters could not be found. This pattern of distribution might be the result of gene flow via germplasm exchanges in nearby regions. The results indicate that these Korean landraces of foxtail millet exhibit a moderate level of diversity. This study demonstrated that molecular marker strategies could contribute to a better understanding of the genetic structure in foxtail millet germplasm, and provides potentially useful information for developing conservation and breeding strategies.

  • A two-strain epidemic model with an age structure mutation and varying population is studied. By means of the spectrum theory of bounded linear operator in functional analysis, the reproductive numbers according to the strains, which associates with the growth rate ${\lambda}^*$ of total population size are obtained. The asymptotic stability of the steady states are obtained under some sufficient conditions.
