This study determined the relationships between the total anthocyanin content in apple skin and the total polyphenols, flavonoids, proanthocyanidins, and soluble solids contents in the flesh of cold-stored Fuji apples. Total anthocyanin content in apple skin ranged from 0.130±0.005 mg CE/g fw to 0.262±0.028 mg CE/g fw, and the overall average was 0.200±0.008 mg CE/g fw. The total polyphenolic compounds in the flesh was ranged from a 4.283±0.141 mM GAE/g fw to 8.207±0.234 mM GAE/g fw, and the average content was 6.275±0.177 mM GAE/g fw. The total flavonoid content ranged from 4.510±0.080 mM QE/g fw to 2.467±0.458 mM QE/g fw, and the average total flavonoid content was about 3.586 mM QE/g fw. The total proanthocyanidin content was relatively high, ranging from 3.475±0.577 mM EE/g fw to 6.816±0.277 mM EE/g fw, and the soluble solid in the flesh was about 12 °Brix to 14 °Brix. The DPPH radical scavenging activity of extracts from apple flesh ranged from 66.36% to 94.99%, and the ascorbate equivalent concentration was 0.482 mM. The ABTS radical scavenging activity was 99.12% to 99.9%, indicating a higher inhibitory activity than the DPPH inhibitory activity, and the ascorbate equivalent concentration was 0.486 mM. The correlation between the total anthocyanin and total polyphenolic compounds was y = 15.192x + 3.2169 (R2=0.2748), but the concentration of total polyphenolic compounds increased when the total anthocyanin content was increased. The correlation equation of total anthocyanin with total flavonoids was y = 15.18x + 0.5555 (R2=0.6226), with total proanthocyanin was y = 14.918x + 2.3422 (R2=0.3372), and with soluble solid was y = 10.558x + 11.126 (R2=0.1925), indicating that the correlation of total anthocyanin with total flavonoid was higher than that with soluble solid.

The purpose of this study was to determine the capsaicinoid and antioxidant compounds and to evaluate the biological activity of 40 different pepper (Capsicum annuum L.) breeding lines. Capsaicinoid and ascorbic acid content were measured with HPLC. Total polyphenolic and flavonoid contents and antioxidant activity were measured with spectrophotometric methods. The antiproliferative qualities of the samples against human breast tumor cells (MCF7) were assessed with a MTT assay. The nitric oxide content in the culture media was measured to evaluate the anti-inflammatory qualities of the samples using Raw264.7 macrophages. The capsaicinoid, ascorbic acid, polyphenolic and flavonoid content of the 40 pepper breeding lines were 0.1~204.2 mg/100 g (dry weight basis, DWB), 279.1~1695.5 mg/g (DWB), 2.6~10.2 mg/g (DWB), and 1.4~5.7 mg/g (DWB), respectively. The highest antioxidant, antiproliferative, and anti-inflammatory qualities were found in breeding line numbers 2500, 3201, and 3232, respectively. This study provides basic information useful to plant breeders and biotechnologists who are planning to breed pepper genotypes with high phytochemical content.
This study was carried out to investigate the total polyphenols, antioxidant activities, and melanin synthesis inhibition of several pear cultivars (Pyruspyrifolia). The total polypenolic content of five pear cultivars was high in the unripe developmental stage. Total polyphenolic content of the Chuwhangbae cultivar extract was higher than that of other pear cultivars regardless of the developmental stage. However, the total flavonoid content did not differ between cultivars or developmental stages. The phenolic compound, arbutin has an inhibitory effect on melanogenesis. Arbutin levels in pear cultivars declined as the fruit matured. The free radical scavenging activity of the extract also decreased as the fruit ripened. In B16F10 mouse melanoma cells, most of the cultivar extracts inhibited melanin synthesis by about 50% at a $100{\mu}g{\cdot}mL^{-1}$ concentration, except in the Gamcheonbae extract until 90 days after full bloom. We have confirmed that the extract of pear cultivars have antioxidant activity and skin-whitening effects.
This study aimed to investigate the biological activities, including polyphenolic content, flavonoid content, and antioxidant activities, of various cultivars of Korean perilla leaves. The results indicated that among nine cultivars (Namcheon dlggae, Saedora, Nulbora, Donggel 1, Donggell 2, Soim, Sangyeop, Somirang, and Saebom) of perilla leaves, the total polyphenolic content (gallic acid equivalent mg/g, GAE) was the highest in "Nulbora," while it was lowest in Namcheon dlggae. Moreover, flavonoid content in the extracts of nine cultivars leaves was in the range of 132.93~268.50 mg catechin equivalent/g sample. The antioxidant effects of the perilla leaves were determined using two different in vitro bioassays measuring DPPH and ABTS radical-scavenging activities. The results revealed that antioxidant activity was also higher in "Nulbora" compared with other cultivars. Xanthin-oxidase-inhibition activity ranged from 65.65% to 80.58%, with "Nulbora" exhibiting the highest activity, although the difference with other cultivars was not significant. "Nulbora" extracts reduced the expression of pro-inflammatory genes and several cytokines, including IL-6 activation induced by LPS in macrophages in the range of 100-50 ㎍/mL. These results suggest that extracts from perilla leaves can be used as bioactive and functional materials that could be important in industrial applications in the future.

Objective : The study's objective is to assess the components of Dendropanax morbifera (DM) utilizing UPLC-MS/MS and assess their antioxidant properties in order to establish fundamental information for quality control of herbal formulations. Methods : The DM leaves were ground into powder and extracted with water at 80℃. The extract was subsequently concentrated and subjected to freeze-drying for subsequent analysis. The LC-MS/MS analysis was performed using a 1260 series HPLC system and a 3200 QTrap tandem mass system in positive ion mode, with detection conducted at 280 nm. The Folin-Ciocalteu method was employed to measure the phenolic content, while a colorimetric method using aluminum chloride was used to determine the flavonoid content, with gallic acid and quercetin as standards, respectively. The evaluation of antioxidant activity was conducted through the measurement of DPPH radical scavenging activity, by adding the DPPH solution to the extract and recording the absorbance at 517 nm. Results : The UPLC-MS/MS analysis identified five polyphenolic compounds in the DM extract, specifically syringin, 6-hydroxyluteolin 7-O-laminaribioside, shaftoside, rutin, and kaempferol-3-O-rutinoside. The extract was found to contain a total phenolic content of 83.106 ± 0.21 mg GAE/g and a total flavonoid content of 87.963 ± 1.014 mg QE/g. The DM extract demonstrated substantial antioxidant properties, resulting in a reduction of DPPH radicals that was evident at concentrations as low as 40 ㎍/㎖. Conclusions : The study determined important polyphenolic compounds in DM and established its considerable antioxidant efficacy. These findings provide evidence for the efficacy of DM in disease prevention related to oxidative stress and establish a foundation for ensuring quality control in herbal preparations.

This study compared the physicochemical properties of root hair of white ginseng (WG), root hair of tissue cultured mountain ginseng (MG), root hair of red ginseng (RG) and extruded ginseng samples. The comparison of crude ash and total sugar resulted insignificant differences between extruded and raw samples. MG had a higher content of crude ash, crude protein, amino acids and polyphenolic compound than WG and RG; the total sugar and reducing sugar were highest in RG. Crude fat and acidic polysaccharide in RG and WG were similar to and higher than MG. Crude saponin of treated samples WG1 (moisture content 25%, barrel temperature $110^{\circ}C$) and WG3 (moisture content 35%, barrel temperature $110^{\circ}C$) were 9.80% and 9.73%, respectively, which were the highest among ginseng samples. In conclusion, the extrusion process can be applied to red ginseng manufacturing, and some characteristics of MG were higher than in RG and WG.

The seeds from two mulberry fruits [Morus alba (MA) and Cudrania tricuspidata (CT)] were examined for their oil content, and fatty acid, phytosterol and tocopherol compositions and contents. Moreover, polyphenolic compounds and biological activity of the two defatted seed residue extracts were also evaluated. Oil contents of MA and CT seeds were 29.36% and 16.69%, respectively, while MeOH extracts of the defatted MA and CT seed residues were 5.10% and 6.22%, respectively. The two seed oils were composed of 81.4 and 74.37% linoleic, 5.75 and 11.39% oleic, 8.40 and 10.18% palmitic acid, and 3.52 and 3.0% stearic acids, and two other minor fatty acids, such as linolenic and arachidic acids. MA seed had higher contents of phytosterols (507.59 mg/100 g of oil), tocopherols (99.64 mg/100 g of oil), and total flavonoid (106.50 mg/100 g of seed) than CT seed, whereas CT seed had higher levels of total polyphenol than MA seed. The MeOH extract of MA seed residue showed higher antioxidant, anti-diabetic, and anti-melanogenic activity than that of CT seed residue. trans-Resveratrol (9.62 mg/100 g), quercetin (54.83 mg/100 g), and 4-prenylmoracin (48.70 mg/100 g), were found to be the main polyphenolic components in the MeOH extract of MA seed residue. These results indicate that MA seeds are good sources of essential dietary phytochemicals with antioxidant, anti-diabetic and anti-melanogenic activity.
Objectives : In this study, we compared the anti-oxidant and anti-inflammatory activities of Curcumae longae Radix (CLRa) and Curcumae longae Rhizoma (CLRh). Methods : We performed 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and 2,2-azinobis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) cation scavenging assays, and determined total polyphenolic content to examine the anti-oxidant effects of CLRa and CLRh. We also evaluated the anti-oxidant effects of CLRa and CLRh against hydrogen peroxide ($H_2O_2$)-induced toxicity in PC12 cells using thiazolyl blue tetrazolium bromide (MTT) and reactive oxygen species (ROS) assays. Next, to compare the anti-inflammatory effects of CLRa and CLRh against lipopolysaccharide (LPS)-induced inflammation in microglia BV2 cells, we measured nitric oxide (NO) assay and inducible nitrite synthase (iNOS) using Western blotting analysis. Results : CLRa showed higher activity in DPPH and ABTS assays and lower total polyphenolic contents compared with CLRh. In PC12 cells, CLRa and CLRh showed no difference in H2O2-induced cell toxicity and ROS overproduction. In BV2 cells, CLRa showed higher effect than CLRh in NO and iNOS production induced by LPS. Conclusions : These results demonstrate that CLRa has higher radical scavenging activities and anti-inflammatory effect in BV2 cells comparing CLRh. However, CLRa and CLRh have no effect and no difference in $H_2O_2$-induced toxicity.
The antioxidant activities of the extracts of butterbur (Petasites japonicus) derived from different extraction methods were investigated. SOD (superoxide dismutase)-like activity differed according to the extraction solvents, showing a greater antioxidant effect with ethanol solvent than that of water. Ethanol extracts of butterbur leaves showed higher SOD-like activity of 96.7% than those of water extracts. The contents of polyphenolic compounds were higher in water extracts than those in ethanol extracts. The highest content of polyphenolic compounds was 223mg/g dry weight for butterbur leaves. EDA of butterbur roots was 61.5% in the water extract and EDA in butterbur leaves was 34.9% in the ethanol extract. The anticancer effects with the extracts of butterbur were experimented by fractionations with different solvents. 41.9% of the growth of stomach cancer cells, SNU-719, were inhibited and also 72.7% of the growth of liver cancer cells, Hep3B, were inhibited by the butanol fractions of butterbur, while not affecting the growth of normal cell, DC 2.4.

Ecklonia cava is popular in Korea as a marine functional materials. E. cava is generally collected and used on the coast of Jeju Island. However, the continuous use of collected natural E. cava may be limited because difficult to secure throughout the year and may be exposed to environmental pollution. Jeju magma seawater (MSW) was known to be significant advantages such as safety, cleanness, stability, and functional improvement. Attempts have been reported on application of MSW to the culturing of macro- and microalgae and showed improved results. Thus, the objective of the present study was to explore the anti-melanogenesis activity of brown seaweed E. cava (E. cava cultured with MSW [MSWE]) extract cultured in tanks with MSW of Jeju Island to evaluate the possibility of cosmeceutical industrial application. MSWE extract showed the higher polyphenolic and dieckol contents than natural E. cava (NE) extract. Anti-melanogenesis activity of MSWE extract and NE extract are tested and compared using tyrosinase and dihydroxyphenylalanine (DOPA) oxidation inhibition assay. MSWE extracts evidenced more effective tyrosinase and DOPA oxidation inhibition activity than that of the NE extracts and the commercial whitening agent, arbutin. MSWE extracts also markedly inhibited melanin synthesis and decreased the expression of melanogenesis-related protein in ${\alpha}$-melanocyte stimulating hormone-stimulated B16F10 melanoma cells without cytotoxicity. These results suggest that MSW cultivation process would be more effective in releasing bioactive compounds with whitening effect from seaweed such as E. cava at an industrial scale.
