Comparison of Antioxidant Activities by Different Extraction Temperatures of Some Commercially Available Cultivars of Rice Bran in Korea (추출온도에 따른 국내 벼 품종 미강의 항산화 활성 비교)

  • This study was conducted to determine antioxidant compounds and antioxidant activities of rice bran at temperatures of $10^{\circ}C$, $25^{\circ}C$, and $60^{\circ}C$ using 70% ethanol solution. DPPH, ABTS radical scavenging activities and reducing power have been used to investigate the relative antioxidant activities of extracts. The contents of total polyphenol and total flavonoids in the 70% ethanol extracts were measured by spectrophotometer. The total polyphenol content in the rice bran extracts were 790.39~930.07, 782.71~1,039.57, and 848.22~1,052.96 mg gallic acid eq./100 g in $10^{\circ}C$, $25^{\circ}C$ and $60^{\circ}C$, respectively. Antioxidant activities, such as DPPH, ABTS radical scavenging activity and reducing power, in all cultivars and temperatures was the highest in Cheonghaejinmi rice bran extracted at $25^{\circ}C$. The results obtained in the in vitro models suggest that extracts from rice bran, especially Cheonghaejinmi, has significant health-promoting effects, as it exhibits antioxidant activities.

Chemical compositions and functional characteristics of Korean and imported pomegranate (Punica granatum L.) (국내산과 수입산 석류의 화학적 성분과 기능적 특성)

  • In recent years, polyphenol-rich herbs, fruits and processed foods, which are made of plant origin, have attracted much attention due to their potential health benefits. Pomegranate (Punica granatum L.) is an important source of bioactive compounds and has been used to treat diseases because of its medicinal properties. This research was focused on characterizing Korea's national cultivar and a similar product from California, USA. To evaluate their bioactive compounds and pharmacological activities, their anti-oxidation and cancer inhibition properties, as well as their organic acid and free sugar contents, were investigated. The national cultivar had low total sugar and high organic acid contents, contrary to the imported product. The results showed that the peel of national cultivar had high polyphenol and ellagic acid contents compared to imported product. The free radical scavenging capacity was evaluated via 2,2-diphenyl-1-picrylhydrazyl (DPPH) and its positive correlation with the total polyphenol contents was found. The anti-cancer activity of methanol extracts revealed growth inhibition against the prostate cancer cell. These results signify that while pomegranate, national cultivar, is more sour than the imported product, its health benefits could be excellent. Also, the polyphenol compound content of the non-edible part (such as the peel and the seed) was higher than that of the juice. Thus, it is suggested that the byproduct of the juice extraction could be potentially used in other fields such as medicine or dietary agents.

Antioxidant Effects of the Methanol Extracts Obtained from Aerial Part and Rhizomes of Ferns Native to Korea (자생 양치식물 지상부와 근경 메탄올 추출물의 항산화 효과)

  • Phenolic compound contents and scavenging activity on DPPH and ABTS radicals in twelve fern species were studied by analysing ultrasonification extraction of dried aerial parts and rhizomes using methanol solvent. Total polyphenol content ranged from 2.97 to 140.1 mg per 1 g dried sample and 0.80 to $49.58\;mg{\cdot}g^{-1}$ fresh sample. Highest polyphenol content was obtained with aerial part of Davallia mariesii and Polystichum lepidocaulon. Total flavonoid content of dried sample was $2.56{\sim}34.91\;mg{\cdot}g^{-1}$ and fresh sample $0.71{\sim}11.49\;mg{\cdot}g^{-1}$. Higher flavonoids were obtained with dried aerial part of Dryopteris crassirhizoma, but with fresh aerial part of P. lepidocaulon. In general, total polyphenol content was higher in rhizomes, except in case of total flavonoid. Scavenging effects on DPPH and ABTS radicals was higher with rhizome extracts compared to aerial parts. The rhizome of Polystichum polyblepharum showed highest effects on both radicals. However, the amount of fresh sample for $RC_{50}$ with consideration of water content and extraction yield. P. lepidocaulon aerial part was more advantageous than P. polyblepharum rhizome. All the fern extracts except for the Coniogramme japonica extract demonstrated superior scavenging effects on ABTS radicals, being similar activity of ascorbic acid and BHT.

Curcumin Induces Apoptosis in SGC-7901 Gastric Adenocarcinoma Cells via Regulation of Mitochondrial Signaling Pathways

  • Curcumin, a polyphenol compound derived from the rhizome of the plant Curcuma longa L. has been verified as an anticancer compound against several types of cancer. However, understanding of the molecular mechanisms by which it induces apoptosis is limited. In this study, the anticancer efficacy of curcumin was investigated in human gastric adenocarcinoma SGC-7901 cells. The results demonstrated that curcumin induced morphological changes and decreased cell viability. Apoptosis triggered by curcumin was visualized using Annexin V-FITC/7-AAD staining. Curcumin-induced apoptosis of SGC-7901 cells was associated with the dissipation of mitochondrial membrane potential (MMP) and the release of cytochrome c into the cytosol. Furthermore, the down-regulation of Bcl-2 and up-regulation of Bax that led to the cleavage of caspase-3 and increased cleaved PARP was observed in SGC-7901 cells treated with curcumin. Therefore, curcumin-induced apoptosis of SGC-7901 cells might be mediated through the mitochondria pathway, which gives the rationale for in vivo studies on the utilization of curcumin as a potential cancer therapeutic compound.

Study of Mori Fructus and Dried Mori Fructus Extracts on the Antioxidant Effect and the Inhibitory Effect on Adipocyte Differentiation (상심자와 건조상심자 추출물의 항산화 효과 및 전지방세포 분화억제 효과에 관한 연구)

  • Objectives This study was to investigate the antioxidative capacity, antiobesity effect and anti-diabetes effects of Mori Fructus and dried Mori Fructus in Raw 264.7 cells and 3T3-L1 cells. Methods 3 different types of Mori Fructus extracts (water 100%, ethanol 30%, ethanol 100%) were used in this study. And 3 different types of dried Mori Fructus extracts (water 100%, ethanol 30%, ethanol 100%) were used in this study. Total polyphenol compund, total favonoid compound, DPPH radical scavenging, ROS activity, NO, cell proliferation were measured in the experiment. Expressions of adipogenic transcription factors including $C/EBP-{\alpha}$, $C/EBP-{\beta}$, $PPAR-{\alpha}$, $PPAR-{\gamma}$, $AMPK-{\alpha}$ were analyzed by Real time PCR. Results Mori Fructus extracts measurements are higher than dried Mori Fructus extracts measurements at Total flavonoid compound and total flavonoid compound. Mori Fructus extracts measurements are lower than dried Mori Fructus extracts measurements at DPPH radical scavenging, ROS activity, NO. In RT-PCR analysis, there is a tendency that dried Mori Fructus extracts inhibit the expression of $C/EBP-{\alpha}$, $C/EBP-{\beta}$ genes. In RT-PCR analysis, there is a tendency that dried Mori Fructus extracts promote the expression of $PPAR-{\alpha}$, $PPAR-{\gamma}$, $AMPK-{\alpha}$ genes. Conclusions Mori Fructus is effective on inhibiting the oxidation and dried Mori Fructus is effective on inhibiting the obesity and diabetes.

Effectiveness of Scirpi rhizoma Ethanol Extract on Skin Whitening Using in vitro Test (삼릉 에탄올추출물의 in vitro 피부 미백 유효성)

  • The purpose of this study is to evaluate the effectiveness of Scirpi rhizoma ethanol extract (SREE) on skin whitening using in vitro test. In the antioxidative activities, it was found that SREE contains 38.9 mg/g of polyphenol and 74.5 mg/g of flavonoid in total. In the electron donating ability, SREE showed a dose-dependent response, showing a high antioxidative capacity of 86.1% at 1000 ppm. It was found that the maximum permissible level of SREE to Melan-a cells was over 200 ppm, showing a quite low toxicity of SREE against Melan-a cells. Both in the inhibitory measurement for tyrosinase activity and melanogenesis using Melan-a cells, SREE presented a dose-dependent response with excellent efficacy.

  • Lactic acid bacteria (LAB) are used as fermentation starters in vegetable and dairy products and influence the pH and flavors of foods. For many centuries, LAB have been used to manufacture fermented foods; therefore, they are generally regarded as safe. LAB produce various substances, such as lactic acid, ${\beta}$-glucosidase, and ${\beta}$-galactosidase, making them useful as fermentation starters. Existing functional substances have been assessed as fermentation substrates for better component bioavailability or other functions. Representative materials that were bioconverted using LAB have been reported and include minor ginsenosides, ${\gamma}$-aminobutyric acid, equol, aglycones, bioactive isoflavones, genistein, and daidzein, among others. Fermentation mainly involves polyphenol and polysaccharide substrates and is conducted using bacterial strains such as Streptococcus thermophilus, Lactobacillus plantarum, and Bifidobacterium sp. In this review, we summarize recent studies of bioconversion using LAB and discuss future directions for this field.

  • In Peucedanum japonicum and Aster tataricus L. chlorogenic acid was identified by methods of P.P.C. and T.L.C. $L-Phenylalanine-U-^{14}C\;and\;sodium\;acetate-2-^{14}C$ were administered to Peucedanum japonicum, $L-Tyrosine-U-^{14}C$ to Aster tataricus and $caffeic\;acid-carboxyl-^{14}C\;and\;L-tyrosine-U-^{14}C$ to Fagopyrum esculentum $M_{OENCH}$. The incorporation of each compound into chlorogenic acid was compared. $L-Phenylalanine-U-^{14}C$ showed higher incorporation to chlorogenic acid than sodium $acetate-2-^{14}C$ in Peucedanum japonicum. $Caffeic{\;}acid-carboxyl-^{14}C$ was higher to chlorogenic acid than $L-tyrosine-U-^{14}C$ in Fagopyrum esculentum. $L-Tyrosine-U-^{14}C$ was comparatively low in Aster tataricus.

  • Development of chocolate products to improve the quality and sensory properties with Saengmaeg-san powder (SP) was attempted. Effects of SP on color, viscosity, total phenol compound content, radical scavening activity and sensory characteristics. In color, lightness value was increased than milk chocolate without SP Also viscosity was significantly higher than chocolate without SP.Total polyphenol content of Saengmaeg-san chocolate was 191.52 mg/1 OOg and Dpph scavenging activity was 87.06%. Results of sensory evaluation, significant different (p < 0.05) were shown in softness, sweet taste, sour taste, bitter taste, texture and overall acceptability depending on the addition of SP.

  • Chemoprevention entails the use of synthetic agents or naturally occurring dietary phytochemicals to prevent cancer development and progression. One promising chemopreventive agent, procyanidin, is a naturally occurring polyphenol that exhibits beneficial health effects including anti-inflammatory, antiproliferative, and antitumor activities. Currently, many preclinical reports suggest procyanidin as a promising lead compound for cancer prevention and treatment. As a potential anticancer agent, procyanidin has been shown to inhibit the proliferation of various cancer cells in "in vitro and in vivo". Procyanidin has numerous targets, many of which are components of intracellular signaling pathways, including proinflammatory mediators, regulators of cell survival and apoptosis, and angiogenic and metastatic mediators, and modulates a set of upstream kinases, transcription factors, and their regulators. Although remarkable progress characterizing the molecular mechanisms and targets underlying the anticancer properties of procyanidin has been made in the past decade, the chemopreventive targets or biomarkers of procyanidin action have not been completely elucidated. This review focuses on the apoptosis and tumor inhibitory effects of procyanidin with respect to its bioavailability.
