Effect of Polyamines on Indigo Biosynthesis in Hairy Root Cultures of Polygonum tinctorium Lour. (Polyamine이 쪽 모상근배양에서 인디고 생합성에 미치는 영향)

  • We herein studied the effect of polyamines (putrescine, spermidine and spermine) on growth and indigo biosynthesis in hairy root cultures of Polygonum tinctorium Lour. Our results revealed that polyamine treatment increased hairy root growth and indigo biosynthesisat all tested concentrations, with the highest growth rate (4.4 g/ flask) and indigo yield (216 ug/g) induced by 70 mg/L putrescine. These results show far the first time that the growth rates and indigo biosynthesis of Polygonum tinctorium hairy roots may be improved by addition of polyamines to the liquid culture medium.

Influence of Eco-Friendly Underwears on Atopic Dermatitis (친환경 속옷이 아토피 피부염에 미치는 영향)

  • The present study was undertaken to evaluate effects of eco-friendly underwears(three kinds of natural dying underwears) in patients with atopic dermatitis. We recruited 45 patients with atopic dermatitis admitted to Dongshin Oriental Hospital from August to September 2013. These patients were divided into three groups (A, B and C). Each group include 15 patients and three kinds of natural underwears dyed with polygonum tinctorium, loess, and non-dyed general cotton were given, respectively. We also used SCORAD index and digital infrared thermal imaging to investigate the effects of each eco-friendly underwear in patients with atopic dermatitis. In SCORAD index, Group A after wearing loess-dyed underwears for 4 weeks showed statistically significant reduction to 27.96 (P<0.05) compared to 34.20 before wearing loess-dyed underwears. In Group B after wearing polygonum tinctorium-dyed underwears for 4 weeks, SCORAD index and objective severity assessment were markedly reduced into 22.97 (P<0.01) and 18.17 (P<0.01) compared to 35.95 and 27.89 before wearing polygonum tinctorium.-dyed underwears, respectively. In Group C after wearing general cotton (non-dyed) underwears for 4 weeks, there was statistically no difference in SCORAD index, objective severity assessment and body temperature between before and after wearing general cotton (non-dyed) underwears. Taken together, polygonum tinctorium-dyed underwears used to patients with atopic dermatitis shows significant amelioration in atopic symptoms than loess-dyed underwears as well as general cotton (non-dyed) underwears.

Dyeability and Antimicrobial Activity of Silks Dyed Singly with Sophora japonica L., Curcuma longa L., Combination-Dyed with Polygonum tinctorium L. and Sophora japonica L., and with Polygonum tinctorium L. and Curcuma longa L. (회화 및 울금에 의한 단독 염색 견직물 그리고 쪽과 회화 및 쪽과 울금에 의한 복합 염색 견직물의 염색성 및 항균성)

  • We examined the dyeability and antimicrobial activity of silks dyed singly with Sophora japonica L., Curcuma longa L., combination-dyed with Polygonum tinctorium L. and Sophora japonica L., and with Polygonum tinctorium L. and Curcuma longa L.. The color of silk dyed singly with Sophora japonica L., Curcuma longa L. was Yellow with Hue of 5.0Y and 0.6Y, respectively. Two types of combination-dyed silk were Blue Green with 2.6BG and 1.5BG, respectively. In the case of dry Cleaning fastness and rub fastness, all four types of dyed silk was good, achieving grade 4~5. The antimicrobial activity of Staphylococcus aureus of silk dyed with Sophora japonica L. extract showed excellent antibacterial activity of 99.5% and the other three types of dyed silk of 99.9%. In addition, in antibacterial activity against Klebsiella pneumonia, silk dyed with Sophora japonica L. extract showed 99.6%, and the other three types of dyed silk exhibited excellent antibacterial properties of 99.9%.

Dyeability of Saxon Blue Derived from Polygonum (Tinctorium Polygonum Tinctorium을 이용한 Saxon Blue 염색)

  • 본 논문에서는 쪽의 발효염색이 잿물과 같이 강 알칼리 매체 하에서 사용해야 하고 또한 자연 발효의 경우 온도조절이 제한되어 있어 단백질 섬유 특히 양모에는 적합하지가 않은 점 등을 해결하기 위해 우리나라에서 주로 현재 재배되고 있는 polygonum tinctorium을 사용하여 Saxon blue 법으로 단백질 섬유를 염색하여 염색성을 살펴보았다. 그 결과 Saxon blue로 염색할 때의 등온흡착곡선은 Langmiur형임을 보였으며 색상은 주로 B 영역에서 나타났다. Saxon Blue로 염색한 견 직물과 양모 직물 모두 우수한 항균성을 나타내었으며 드라이 크리닝과 마찰에 대해서는 좋은 견뢰도를 보여 주었다.

Improved Optimization of Indirubin Production from Bioreactor Culture of Polygonum tinctorium

  • Effect of the two-stage operation and cell concentration on indirubin production was investigated using bioreactor culture of Polygonum tinctorium. Two-stage culture was operated successfully for 110 days without any adverse effects on continuous indirubin production. Maximum indirubin concentration was found to be at 80 mg/bioreactor. Initial cell concentration significantly affected indirubin production. The indirubin production at 29.2% PCV was improved by 845%, compared to that at 5% PCV. For high-density bioreactor culture of P. tinctorium, a maximum production rate of 10.2 mg indirubin/L day was obtained. Indirubin recovery for bioreactor operation was also examined using XAD-2, XAD-4, XAD-7, and solid silicon. XAD-4 was 1.6-fold more effective than that for solid silicon in indirubin recovery.

  • To examine the expression of foreign protein in Polygonum tinctorium cells, plasmid pCAMBIA1302 encoding Green Fluorescent Protein(GFP) was used to transform the cells and the expression was confirmed using Western blot analysis. When the effect of sodium buryrate on the formation of GFP was examined, cell growth was retarded at the addition of 10 mM and was stalled at more than 15 mM. The amount of GFP production was increased by 15% when 5 mM of sodium butyrate was added at three-days after inoculation as compared to at 0-day. Moreover, when sodium butyrate was added at three-days after inoculation, the amount of GFP was increased by 50% at the addition of 5 mM of sodium butyrate as compared to 10 mM.

  • This experiment was carried out to analyze the effect of indole on the synthesis of indirubin in suspension culture of Polygonum tinctorium. Adding indole and L-tryptophan into culture media was re­vealed that indirubin was synthesized in callus grown on solid medium containing indole and proper concentration of indole for indirubin production was decided as 200mg/1. Indirubin content in suspension culture was higher than in solid medium with considerable amount of indirubin secresed into media in suspension culture and highest quantity of indirubin was obtained when indole was added into medium after 20 days suspension culture.
