Nanofibrous membranes of poly(D,L-lactic acid)/chitin blend were prepared by electro spinning for a barrier of guided tissue regeneration. A miscible solution was obtained by the blending chitin-salt complex into 1-methyl-2-pyrrolidone solution of poly(D,L-lactic acid). The properties of the blend were examined for nanofibrous fabrication. The viscosity of the blend solution was increased significantly due to chain entanglement despite the low ratio of chitin to poly(D,L-lactic acid). An interaction between two polymeric compositions was confirmed by Fourier transform infrared spectroscopy. X-ray diffraction detected an appreciably ordered microstructure in the nanofiber of the blend. A membrane of thinner nanofibers was fabricated by electro spinning the chitin blend. The permeability of the membranes was examined using bioactive model compounds.
Poly(D,L-lactic acid) (PLA) microspheres containing alprazolam(APZ) were prepared by a solvent-emulsion evaporation method and their release patterns were investigated in vitro. Various batches of microspheres with different size and drug content were obtained by changing the ratio of APZ to PLA, PLA concentration in the dispersed phase and stirring rate. Rod-like APZ crystals on microsphere surface, which were released rapidly and could act as a loading dose, were observed with increasing drug content. The release rate was increased with increase in drug contents and decrease in the molecular weight of PLA. The release rate of APZ for long-acting injectable delivery system in vitro, which would aid in predicting in vitro release profile, could be controlled by properly optimizing various factors affecting characteristics of microspheres.
Electrospinning was performed using an eco-friendly solvent composed of acetic acid, ethyl acetate and distilled water to investigate the effect of gelatin concentration on mechanical properties and cytotoxicity of absorbable poly(D,L-lactic acid) (PDLLA)/gelatin blend membrane. The tensile stress, strain at break, and WUC of the PDLLA/gelatin (97/3) scaffold at 26 wt% concentration were determined to be 3.9 ± 0.7 MPa, 37 ± 1.3 %, and 273 ± 33 %, respectively. FT-IR results revealed that PDLLA and gelatin were bound only by van der Waals interactions. The cell viability of PDLLA/gelatin membranes containing 0 %, 1 %, 2 %, 3 %, and 4 % gelatin were more than 100 %, which makes all membranes highly suitable as a barrier membrane for absorbable periodontal tissue regeneration due to their marketed physical properties and biocompatibility.
본 연구에서는 폴리유산 나노복합재의 열탄성 거동을 예측하기 위해 분자동역학 전산모사를 수행하고 그 결과를 열탄성 미시역학 모델 예측해와 비교하였다. 폴리유산의 두 이성질체인 D유산(Poly D-lactide)와 L유산(Poly L-lactide)을 혼합한 스테레오 콤플렉스를 모델링하였고 이들을 기지로 사용한 탄소나노튜브 나노복합재를 구성하였다. 유산의 분해 유무에 따른 유리전이온도와 탄성계수 그리고 열팽창계수를 앙상블 전산모사를 통해 예측하였다. 미시역학 모델에서는 계면의 완전 결합을 가정한 이중입자 모델을 적용하여 탄성계수와 열팽창계수를 동일한 조성에서 예측하였다. 그 결과 열적 안정성에 있어 스테레오 콤플렉스에 탄소나노튜브가 첨가될 경우 유산의 뛰어난 계면 흡착과 이에 따른 열적 안정성 향상을 보였다. 순수한 유산과 나노복합재 모두 가수 분해에 따른 열적 특성 변화는 관찰되지 않았다. 또한, 스테레오 콤플렉스와 나노튜브 간 계면은 약한 불완전 결합상태 임을 알 수 있었다.
There has been a growing attention on PDLA (poly D-lactic acid) since stereocomplex PLA, a kind of polymer alloy between PLLA and PDLA was known much thermally stable compared PLLA. Superior characteristics of stereocomplex PLA result in the elevated demand for D-lactic acid. Although many research works have been reported for L-lactic acid production especially food industry, however there are relatively few research works for D-lactic acid production since D-lactic acid cannot find any applications in food industry. Most imminent issue for D-lactic acid is the economic production process that requires low cost medium, efficient lactic acid producing microorganism and finally large scale-up design. In this review, current status of D-lactic acid production process will be summarized and discussed for the further improvement of D-lactic acid production process.
비바이러스성 유전자 전달체의 주요 단점인 낮은 transfection 효율에 기인한 반복투여 등을 극복하기 위하여 poly (D,L-lactide-co-glycolide)를 이용하여 DNA가 봉입된 미립구를 제조하였다. pDNA 그 자체 또는 여러 비율의 키토산/pDNA 복합체를 사용하여 봉입하였고, 그 결과 44%(pDNA 그 자체), 5%(0.7:1 미토산/pDNA 복합체), 그리고 8%(1:1 키토산/pDNA 복합체)의 봉입효율을 나타내었다. 주사전자현미경(SEM)을 통해 본 표면구조에서는 미립구 제조 직후에서는 매우 매끈한 구형을 보이다가 제조 후 41일 경에는 찌그러진 다공성의 구조를 보였는데 이는 미립구 제조에 사용한 poly(D,L-lactic-co-glycolic acid)(PLGA) 고분자의 분해에 의한 것으로 생각된다. 시험관내 방출실험에서는 0.7:1 키토산/pDNA 복합체를 사용한 미립구에서 47%의 pDNA가 26일만에 방출된데 반해, pDNA 그 자체 혹은 1:1 키토산/pDNA 복합체를 사용한 미립구에서는 각각 15% 혹은 32%의 pDNA 방출을 나타내었다.
폴리락트산의 열적 안정성과 기계적 물성을 개선하고자 측쇄에 벤젠고리를 함유한 천연 $\alpha$-히드록시산인 만델린산(mandelic acid)과 L-락트산을 직접 용액 축중합법으로 공중합체를 합성하고 그 열적, 물리적 성질 등을 조사하였다. 합성된 공중합체는 무정형이었으며, 만델린산의 함량이 증가할수록 유리전이온도와 분해개시온도가 상승함을 열분석을 통해 알 수 있었다. 또한 만델린산을 5, 10 wt% 함유한 공중합체의 필름의 경우 그 기계적 성질이 다소 향상된 결과를 얻었다.

Lactic acid and its derivatives are widely used in the food, pharmaceutical, and cosmetic industries. It is also a major raw material for the production of poly-lactic acid (PLA), a biodegradable and environmentally friendly polymer and a possible alternative to synthetic plastics derived from petroleum. For PLA production by new strains of lactic acid bacteria (LAB), we screened LAB isolates from shellfish. A total of 51 LAB were isolated from 7 types of shellfishes. Lactic acid production of individual isolates was examined using high-performance liquid chromatography using a Chiralpak MA column and an ultraviolet detector. Lactobacillus plantarum T-3 was selected as the most stress-resistant strain, with minimal inhibition concentrations of 1.2 M NaCl, 15% ethanol, and 0.0020% hydrogen peroxide. In a 1 L fermentation experiment, $\small{D}$-lactic acid production of 19.91 g/L fermentation broth was achieved after 9 h cultivation, whereas the maximum production of total lactic acid was 41.37 g/L at 24 h.
Lactic acid is an important product arising from the anaerobic fermentation by lactic acid bacteria (LAB). It is used in the pharmaceutical, cosmetic, chemical, and food industries as well as for biodegradable polymer and green solvent production. The poly lactic acid (PLA) is an important material for bio-plastic manufacturing process. For PLA production by new LAB, we screened LAB isolates from shellfish. A total of 28 LAB were isolated from various shellfishes. They were all Gram positive, oxidase and catalase negative. Based on API 50CHL kit, 7 strains among the 28 isolates were identified as Lactobacillus plantarum, 6 strains as Lactobacillus delbrueckii, 5 strains as Leuconostoc mesenteroides, 3 strains as Lactobacillus brevis, 2 strains as Lactococcus lactis, 1 strain as Lactobacillus salivarius, 1 strain as Lactobacillus paracasei, 1 strain as Lactobacillus pentosus, 1 strain as Lactobacillus fermentum and 1 strain as Pediococcus pentosaceu. Also, we examined the amount of total lactic acid produced by these new strains by HPLC analysis with Chiralpak MA column. One strain E-3 from Mytilus edulis was indentified as Lactobacillus plantarum and found to produce 20.0 g/L of D-form lactic acid from 20 g/L of dextrose. Further studies are underway to increase the D-lactic acid production by E-3.
