Utilization of Pollen Grain from Liluim cv. Casablanca as a Transient Gene Expression Host (유전자의 일시발현 분석용 숙주개발을 위한 카사블랑카백합(Lilium cv. Casablanca) 화분립의 이용)

  • Lilium cv. Casablanca pollen grains stored at $-70^{\circ}C$ were grown in pollen germination medium with Agrobacterium tumefaciens LBA4404 cells harboring pBI121 for 18 hr at $27^{\circ}C$. Following this, cefotaxime (250 mg/L) was treated for 6 hr to eradicate the bacterial cells. Histochemical GUS analysis revealed that the transgenic pollen displayed deep blue color mostly from 12 hr after the co-cultivation. Presence of $200\;{\mu}M$ acetosyringone was determined not to be more effective for GUS transformation than its absence. GUS DNA integration in the transgenic pollen genomic DNA was clearly demonstrated by Southern blot analysis.

  • Three-dimensional (3D) refractive-index (RI) imaging and quantitative analyses of angiosperm pollen grains are presented. Using optical diffraction tomography, the 3D RI structures of individual angiosperm pollen grains were measured without using labeling or other preparation techniques. Various physical quantities including volume, surface area, exine volume, and sphericity were determined from the measured RI tomograms of pollen grains. Exine skeletons, the distinct internal structures of angiosperm pollen grains, were identified and systematically analyzed.

  • I tried to specify the taxa of Fagaceae in Korea by the character of their pollen grains. The light microscope(LM) and the Scanning electron microscope(SEM) were used to examine the pollen grains of 19 taxa, 5 genera. The result are as follows. 1. The pollen of Fagaceae in Korea could be grouped into four types and 4 subtypes. 1) Fagus type 2) Castanea type 3) Lithocarpus, Castanopsis type 4) Quercus type (1) Cyclobalanopsis subtype (2) Prinus subtype (3) Dentatae subtype (4) Cerris subtype. 2. The morphology of the granula on the pollen of Quercus was closely related to the differantiation of the shape of the cup scales. 1) The uniformity of branching granula on the pollen grain surface corresponds to the morphological features of the concentric arrangement of cup scales. 2) The morphological features of the pollen grain surface intermingled with large or small granula, simple-granula and tuber granula which have small points of circular prominence, corresponded to those of short cup scales. 3) The morphological features of the polllen grain surface intermingled with large or small granula, simple-granula and tuber granula with an apex of amoeba type corresponded to those of Q. dentata Thunb, with thin, fine and long cup scales. 4) The morphological features of the pollen grain surface intermingled with large of small granula of with only simple-granula, corresponded to those of Q. acutissima carr. with thick, fine and long cup scales. 3. The result of cluster analysis by coding the sculpture pattern of the pollen grain surface, the existence and nonexistence of surface perforate, the grain size and granula type were coincident with the system of classification of plants and showed an intimated relationship even under th level of species.

  • A method of collection and long-term storage of viable lily (Lilium longiflorum) pollen grains were developed for their in vitro growth and transformation in consistency. Petroleum ether, n-heptane, cyclohexane and benzene, as pollen collection medium, were determined less toxic to pollen growth in vitro than others tested. Pollen grains, however, lost their growth activity if stored in these solvents more than a week, So, a serial performance, that is, pollen grain collection in these solvents, air-drying and immediate transfer to low temperature condition was determined desirable for keeping the viability much longer. Pollen grains from this storage showed a successful transformation in vitro with a cDNA encoding tissue plasminogen activator (TPA) protein using Agrobacterium via vacuum infiltration according to western blotting analysis.

Gynodioecy and floral dimorphism of Glechoma longituba (Nakai) Kuprian. (Lamiaceae) in Korea

  • The present study was focused on the careful investigation of Glechoma longituba flowers collected from four populations in Korea to clarify their precise sexual systems. All floral characters of this taxon were examined by using a stereo microscopy. The nutlet size and production were compared between two distinct morphs (i.e., hermaphrodites and females), and pollen viability test of hermaphrodite and intermediate phenotype (i.e., individuals with a mixture of females and perfect flowers) was also observed by light microscopy (LM). All of the investigated floral characters of G. longituba are significantly different from one sexual morph to another. Hermaphroditic plants were larger than the female ones in corolla width and length, calyx length, filament length, anther length, and style length (all P < 0.0001, respectively). However, the nutlet width of the female plants was larger than those of the hermaphrodites (P = 0.013). Nutlet production of females was higher than hermaphrodites in Na-Ju population [H (Hermaphrodite): ca. 17.76%, F (Female): ca. 37.45%], but in Mun-San population was no significant difference between these sexual morphs (H: ca. 57.52%, F: ca. 53.16%). We have found significant differences between the fertile pollen grain of hermaphrodite and sterile pollen grain of intermediate phenotype based on pollen size and viability. The results of the present study suggest that compensation (flower size dimorphism, seed set, and pollen viability) is one of the main mechanisms in order to maintain female plants in gynodioecious G. longituba.

Cone Sex Ratio and Pollen Traits in Pinus densiflora (Pinaceae)

  • The pattern of variations in cone sex ratio and pollen traits such as pollen number and size was examined for two years in populations of red pines In Korea. We evaluated; (1) the relationship between tree size and cone sex ratio, (2) the relationship between year and cone sex ratio, and (3) the relationship between cone sex ratio and pollen traits. Larger trees in height and circumference at breast height produced more male as well as female cones. However, cone sex ratio was independent of these plant sizes. Across the two populations, both female cone numbers per tree and male cone numbers per new shoot increased 2.2 and 1.2 times, respectively, in 1999 compared to 1998, while the yearly pattern of male cone-bearing shoots per tree differed significantly between populations. Thus, cone sex ratio varied significantly between years in one of the two populations. Pollen traits such as the number of pollen grains and diameter did not vary significantly among populations. Pollen diameter and grain numbers per male cone were significantly negatively correlated with the cone sex ratio in two populations and one population, respectively. Trees which stopped female cone production in the first year and those which produced female cones in both years differed in the cone sex ratio and pollen size in the second year. The long duration of one reproductive episode and the potential of masting in red pines emphasize the need to conduct long-term studies on the dynamics of cone production, cone sex ratio, and simultaneous changes in pollen traits in red pines.

Recent Advances in the Studies of Self-Incompatibility of plants (식물의 자가불화합성, 최근의 진보)

  • Many flowering plants possess genetically controlled self -incompatibility (SI) system that prevents inbreeding and promotes outcrosses. SI is usually controlled by a single, multiallelic S-locus. In gametophytically controlled system, SI results when the S-allele of the pollen is matched by one of the two S-alleles in the style, while in the sporophytic system self-incompatible reaction occurs by the interaction between the pistil genotype and genotype of, not the pollen, but the pollen parent In the former system the self-incompatible phenotype of pollen is determined by the haploid genome of the pollen itself but in the latter the pollen phenotype is governed by the genotype of the pollen parent along with the occurrence of either to-dominant or dominant/recessive allelic interactions. In the sporophytic type the inhibition reaction occurs within minutes following pollen-stigma contact, the incompatible pollen grains usually failing to germinate, whereas in gametophytic system pollen tube inhibition takes place during growth in the transmitting tissue of the style. Recognition and rejection of self pollen are the result of interaction between the S-locus protein in the pistil and the pollen protein. In the gametophytic SI the S-associated glycoprotein which is similar to the fungal ribonuclease in structure and function are localized at the intercellular matrix in the transmitting tissue of the style, with the highest concentration in the collar of the stigma, while in the sporophytic SI deposit of abundant S-locus specific glycoprotein (SLSG).is detected in the cell wall of stigmatic papillae of the open flowers. In the gametophytic system S-gene is expressed mostly at the stigmatic collar the upper third of the style length and in the pollen after meiosis. On the other hand, in the sporophytic SI S-glycoprotein gene is expressed in the papillar cells of the stigma as well as in e sporophytic tape is cells of anther wall. Recognition and rejection of self pollen in the gametophytic type is the reaction between the ribonuclease in the transmitting tissue of the style and the protein in the cytoplasm of pollen tube, whereas in the sporophytic system the inhibition of selfed pollen is caused by the interaction between the Sycoprotein in the wall of stigmatic papillar cell and the tapetum-origin protein deposited on the outer wall of the pollen grain. The claim that the S-allele-associated proteins are involved in recognition and rejection of self pollen has been made merely based on indirect evidence. Recently it has been verified that inhibition of synthesis of S$_3$ protein in Petunia inflata plants of S$_2$S$_3$ genotype by the antisense S$_3$ gene resulted in failure of the transgenic plant to reject S$_3$ pollen and that expression of the transgenic encoding S$_3$ protein in the S$_1$S$_2$ genotype confers on the transgenic plant the ability to reject S$_3$ pollen. These finding Provide direct evidence that S-proteins control the s elf-incompatibility behavior of the pistil.

Diurnal and Nocturnal Behaviour of Airborne Cryptomeria japonica Pollen Grains and the Allergenic Species in Urban Atmosphere of Saitama, Japan

  • Japanese cedar (Cryptomeria japonica) pollinosis is the most popular pollinosis in Japan. It has been reported that Cryptomeria japonica pollen allergenic species are suspended as fine particles in the urban atmosphere. These allergenic fine particles are responsible for inducing asthma by breaking into the lower respiratory tract. It has also been found that pollinosis symptoms on the sufferers appear mainly at night-time by the results from epidemiological studies. However, the exact reason for these phenomena is not yet clarified. In this study, the diurnal and nocturnal behaviours of Cryptomeria japonica pollen grains and their allergenic species in the urban area of Saitama city of Kanto Plain were investigated. Airborne pollen grains and allergenic Cry j 1 concentrations in total suspended particulate matter (TSP) were investigated at two sampling sites, a heavy traffic road (roadside site) and at the balcony of the $10^{th}$ floor of the Building of Research and Project of Saitama University (general urban site). The latter sampling site where located about 300 m away from the roadside site was used as a general urban site unaffected by automobile traffic. The airborne pollen counts were measured with a real-time pollen monitor. Cry j 1 particles were collected with two high volume air samplers, and these concentrations were measured by surface plasmon resonance method with a Biacore J system. The diurnal variation of the airborne pollen counts was similar to the trends of temperature and wind speed during the day-time; whereas its tendency with wind speed trend was not observed during the night-time. Airborne pollen counts were lower with northern wind than with southern wind because the pollen comes from the mountainous areas, and the mountains in the south are closer, about half the distance to the northern mountains. It is suggested that the peaks of airborne pollen counts during night-time in the sampling site occurred by transport of pollen grains released during day-time in the mountainous forest areas, located c.a. 100 km away from the sampling site. On the roadside site the allergenic Cry j 1 concentrations were higher than at the general urban site, nevertheless pollen grains counts were lower. These results suggested that worsening of pollinosis symptoms during night-time in urban area was caused by transport of pollen grains during day-time in the mountainous forest areas. Moreover, pollen allergenic species become different morphology from pollen grain at roadside site, and the subsequent pollen grains re-suspension by automobile traffic.

Changes of RNA synthesis in Anther Wall of Brassica napus during Male Gametogenesis

  • The distribution of RNA in the anther wall of Brassica napus during male gametogenesis was followed by 3H-uridine autoradiography. Silver grain(SG) density was not above background in the anther wall just after microspore was released from tetrad callose wall. Significant accumulation of SGs occurred in tapetum, endothecium, and epidermis before microspore vacuolation. Accumulation of RNA in the tapetal cells was peak before the vaculation occurred in the microspore. With the onset of tapetal senescence at the partially vacuolated microspore stage, SGs decreased and they completely disappeared in the tapetum at the bicelled pollen stage. Accumulation of RNA in the endothecial cells was peak after the microspore mitosis and continued just after the generative cell mitosis. Appreciable SGs also occurred in cells of epidermis from nonvacuolate microspore stage to bicelled pollen stage. During this period, SG density was almost same and was not high as compared with tapetum or endothecium. At tricelled mature pollen stage, no incorporation occurred in anther wall. SGs were found mostly over the nucleouls and chromation of the cell nuclei.
