Pollen morphology and character evolution in the subtribe Neoguillauminiinae (Euphorbiaceae)

  • A pollen morphological study was conducted using light and scanning electron microscopy involving six species belonging to the subtribe Neoguillauminiinae. Pollen samples from the six species are tricolporate, and the colpi are surrounded by broad margo, with the widest width in the equator, narrower toward the pole, and rounded at the end. Based on the pollen morphology, pollen of the species in the subtribe Neoguillauminiinae were divided into four types: the Neoguillauminia type (T1), the C. collinus type (T2), the C. casuarinoides type (T3) and the C. paucifolius type (T4). The generic divergence between Neoguillauminia and Calycopeplus was supported by the pollen characters of the size, amb and lumina shape. In particular, the traits of rounded shape in the outline of the polar view and circular lumina, which appear only in the pollen grains of N. cleopatra, support the recognition of Neoguillauminia as a monotypic genus. Calycopeplus oligandrus and C. paucifolius had the same reticulate pattern of pollen grains, supporting Forster's hypothesis that these two species are closely related. On the other hand, the close relationship between the morphologically similar C. collinus and C. casuarinoides was not supported by the pollen characters. Within the subtribe there are two equally parsimonious hypotheses regarding the evolution of exine characters. The first consists of two changes of microreticulate through parallel evolution from the primitive reticulate exine, and the second is that the microreticulate pattern is differentiated from the reticulate state and then reversed to reticulate pollen grains.

평택지구 토탄의 화분분석 (A Pollen Analysis in the Peat Sediments from Pyung Taek County Korea)

  • Pollen analysis was made on peat deposits in the area of Pyung Taek County, Central Korea. Arboreal pollen(AP) of Alnus, Abies, Ligustrum, Viburnum and Carpinus composed 70-80% of the total pollen detected in this sample area. Judging from the large amount of arboreal pollens, one can assume that those species were dominant in this district when the pollen was deposited. Among the arboreal pollens, Alnus pollen occurred more often than any other trees. Pollens of Betula, Tilia and Ulmus occurred rarely in each layers of peat deposit. The pollen diagram showed that Pinus and Abies markedly increased from the bottom to the top of the peat, while Ligustrum, Quercus and Tilia decreased proportionately. Non-arboreal pollens (NAP) and spores composed about 30 percent at the lower layer, but decreased considerably in the upper layer.

Pollen Morphology of Genus Sedum in Korea

  • Pollens of 20 species of Sedum were investigated with a scanning electron microscope. The pollen morphology of Sedum was rather variable, within particular species or even within a single inflorescence. Differences occurred in the number and shape of apertures and surface sculpture. Besides 3-colporate, various aperture types including 2-syncolporate, 3-syncolporate, 40stephanocolporate, 5-stephanocolporate, zonate, and irregular types were found in a single specimen. Also, striate-rugulose and psilate sculpture were found in S. viviparum. No correlation was found between the pollen morphology and the floral formula. Pollen characters appeared to be not useful for infrageneric classification of Korean Sedum.

Comparison of Microbial Community Structure in Kiwifruit Pollens

  • Flowers of kiwifruit are morphologically hermaphroditic and survivable binucleate pollen is produced by the male flowers. In this study, we investigated microbial diversity in kiwifruit pollens by analyzing amplicon sequences of 16S rRNA. Four pollen samples were collected: 'NZ' was imported from New Zealand, 'CN' from China in year of 2014, respectively. 'KR13' and 'KR14' were collected in 2013' and 2014' in South Korea. Most of the identified bacterial phyla in the four different pollens were Proteobacteria, Actinobacteria and Firmicutes. However, the imported and the domestic pollen samples showed different aspects of microbial community structures. The domestic pollens had more diverse in diversity than the imported samples. Among top 20 OTUs, Pseudomonas spp. was the most dominant specie. Interestingly, a bacterial pathogen of kiwifruit canker, Pseudomonas syringae pv. actinidiae was detected in 'NZ' by the specific PCR. This study provides insights microbial distribution and community structure information in kiwifruit pollen.

  • In the present experiment as a basic study for establishing a hybrid seed orchard, an observation was made on the survivability of grafts using Pinus rigida as the stock and its hybrids as the sciion, and on the shape and germination of pollen collected from grafted plants. The results are summarized as follows: 1. The hybrid scion of P. rigida.taeda or $P. rigida{\times}(P. rigida.taeda)$ grafted into P. rigida stock showed lower survivability than the homoplastic graftings of P. rigida. 2. Five among the eight eleven-year old scions homoplastically grafted into two-year old stock for the staminate strobile one year after their grafting. 3. There was no difference in the size and germination ability between the pollen collected from homoplastically grafted plant and from the normal one. 4. The pollen collected from the grafted plant is thought to be usable for the control pollination as the germination of such pollen was considerably good.

Pollen-Mediated Gene Flow between Glufosinate Ammonium-Tolerant GM and Non-GM Rice

  • To assess the risk of genetically modified (GM) rice on the agricultural ecosystem, agronomic characteristics, pollen longevity and outcrossing rate between GM (Iksan 483 and Milyang 204) and non-GM (their wild types and female parents) varieties were investigated using the bar gene as a tracer marker in paddy field. The agronomic characteristics of two GM rice were similar to their female-parents (non-GM rice) except heading date and 1,000 grain weight of Iksan 483, and they did not show a difference by the introgression of the bar gene as the genetic traits of rice varieties. Pollen viability was more than 90% just after shedding, and it was rapidly decreased below 50% at 5 minutes after shedding both GM and non-GM varieties. The Pollen longevity was lost after 30 minutes of anthesis. When the distance of gene flow from GM to non-GM rice detected to 6 m from the edge of GM rice plant, the maximum distance of pollen dispersal was 4.5m and 3.9m in Iksan 483 and Milyang 204, respectively, and that was increased in order of west, south, east, and north to the dominant wind direction, west-south. Mean outcrossing rate was very low as 0.003 and 0.001% within 1.5 m from the edge of Iksan 483 and Milyang 204, and the GM hybrids by the pollen dispersal did not detected over 4.5 m from the edge of GM rice plant. The results may help to establish the strategy which reduce the risk of pollen-mediated gene flow between GM and non-GM rice.

Isolation and Characterization of a Pollen-specific cDNA Clone from Easter Lily

  • A pollen-specific cDNA clone, LMP50, was isolated from the mature pollen cDNA library of the Easter lily. The LMP50 transcript was highly abundant in mture pollen grains but not detectable in other organs. The LMP50 cDNA clone contains 1383 nucleotides and two open reading frames. The first codes for a peptide of 15 amino acid residues. The role of this peptide is nuclear. The second encodes a protein containing 329 amino acid residues. This protein exhibited a significant homology to human tartrate-resistant acid phosphatase and porcine uteroferrin. Both of these enzymes have been suggested to play a role in iron transport. Therefore, LMP50 may act as an iron carrier protein in mature pollen grains.

자외선 형광장치현미경으로 본 화분관생장과 석탄 및 aureomycin의 상호작용에 대하여 (On the synergistic action of calcium and antibiotics in pollen growth as observed with ultraviolet fluorescence microscopy)

  • Observations were made on Crinum and Catharanthus pollen growth in artificial media by an ultraviolet transmission fluorescence microscope showing synergistic effect on pollen growth with calcium (Ca) and aureomycin. Bright yellow fluorescence of aureomycin enabling to trace out at tissue or cellular level did reveal that the greater accumulation of fluorescence occurred in the pollen tube wall if Ca was supplemented to the media than when aureomycin alone was present. The promotive pollen growth the media of Ca alone was further enhanced by the addtion of aureomycin. It was assumed that the promoted pollen growth with aureomycin in the Ca media was probably brought about by a supporting role of aureomycin in the Ca action.

자당과 붕산이 구기자 화분 발아에 미치는 영향 (Effect of Sucrose and Boric Acid on In Vitro Pollen Germination of Boxthorn (Lycium chinense M.))

  • This study was carried out to investigate the effect of sucrose, boric acid, and calcium nitrate in optimum pH and relative humidity on pollen germination and pollen tube growth of boxthorn (Lycium chinense M.). Medium containing 100mg/L of $H_3BO_3$ showed maximum pollen germination and pollen tube elongation, 64.8% and $920{\mu}m$. Additionally supplied calcium nitrate to the medium lowered pollen germination rate and pollen tube elongation. Here we report the optimum conditions for the pollen germination and pollen tube development was observed at 10% sucrose, 1.0% agar, pH 6.0, and 100% relative humidity.
