Effect of Ginseng and Herbal Plant Mixtures on Anti-Obesity in Obese SD Rat Induced by High Fat Diet (홍삼 및 천연물 혼합소재의 비만 유도 흰쥐에 대한 항비만 효과에 관한 연구)

  • Anti-obesity effects of ginseng and herbal plant mixtures were investigated to develop natural materials for anti-obesity. After inducing obesity with high fat diet for 8 weeks in male SD rats, ginseng and herbal plant mixtures DM1 (ginseng, puer tea, opuntia) and DM2 (ginseng, puer tea) were administrated orally to rats for another 8 weeks. During administration, food efficacy ratio and body weight of rat were measured twice weekly. After administration, body weight, body fat contents, and serum lipid level were estimated for anti-obesity effect and hematological analysis blood level of ALP and ASP was checked for safety. Body weight in rats fed high fat diet was significantly increased. Body weight in obese rats induced by high fat diet was significantly decreased by DM1 and DM2 feedings. The amount of body fat (epididymal, perirenal and visceral fat, brown adipose tissue) was significantly reduced by DM1 and DM2 treatments. The amount of TG, the concentration of leptin in blood plasma, and the concentration of insulin in blood plasma were significantly diminished by DM1 and DM2. Lipid accumulation on liver was reduced in DM2. There were no side effects among all groups according to blood analysis, hematological findings, and body weight. The findings of this study suggest that DM1 and DM2 may be effective materials for anti-obesity through reducing plasma triglyceride and body fats, and also decreasing body weight without side effects.

Association Between the Polymorphism on Intron 5 of the Lipoprotein Lipase Gene and Carcass Traits in Hanwoo (Korean cattle) (한우 Lipoprotein Lipase 유전자 Intron 5번의 Polymorphism과 경제 형질과의 관련성 분석)

  • The primary role of lipoprotein lipase(LPL) is the hydrolysis of triglycerides(TG) from the core of triglyceride-rich lipoproteins such as chylomicrons and very low density lipoproteins in plasma. Fatty acids liberated by LPL on capillary endothelial surfaces are available for tissues as energy sources especially in muscles or for storage in the form of TG in adipose tissues. Therefore, as the candidate gene related to the carcass traits of the beef cattle, we have directly sequenced the exon 5${\sim}$exon 6 region in the bovine LPL gene for discovery of single nucleotide polymorphism(SNP) with 24 unrelated Hanwoo(Korean cattle). Novel eight sequence variants were detected: three loci on exon 5, three on intron 5 and two on exon 6. All SNPs identified were strongly linked each other, and one hundred twenty eight Hanwoo samples were genotyped one SNP on intron 5 using PCR-restriction fragment length polymorphism method by digestion with Hae III restriction enzyme. The allele frequency of the polymorphism was 0.76 and 0.24. The effects of this polymorphism on the breeding values of the carcass weight, loin muscle area, back fat thickness and marbling score were analyzed using least square methods of SAS GLM. The marbling score of BB genotype was significantly higher than those of AA and AB genotypes(P<0.05). This result indicates that this polymorphism may be associated with the variation of marbling score. Further study is warranted to investigate the phenotypic association in Hanwoo.

Effect of Dietary Supplementation with Bitter Melon on Lipids and Hepatic Enzyme Levels in Streptozotocin Induced Diabetic Rats (여주열매 첨가식이가 당뇨 흰쥐의 지질과 항산화효소 수준에 미치는 영향)

  • This study examined the effect of Momordica charantia L. (bitter melon: BM) on lipid and hepatic antioxidative enzyme levels in diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats by injection of streptozotocin (STZ), and rats were fed for 4 weeks with experimental groups divided into four groups: a normal control group, STZ-control and STZ-BM 5% & STZ-BM 10% treated groups. Levels of free fatty acids (FFA), high-density lipoprotein cholesterol (HDL-chol), triglycerides (TG) in plasma and malondialdehyde (MDA) & protein in liver, catalase (CAT), superoxide dismutase (SOD), glutathione-S-transferase (GST), and xanthine oxidase (XOD) were measured in liver cytosol. Level of HDL-chol significantly increased in the STZ-BM 5% diabetic group. TG & FFA levels were significantly higher in all diabetic groups compared to the control group. MDA and protein levels were significantly higher in the STZ-BM 5% group compared to all other experimental group. CAT level was higher in the supplementary group with BM compared to the STZ-control group, although the difference was not significantly different. SOD level was not significant in any experimental groups. GST level was significantly higher in the BM-treated groups compared to the STZ-control group. XOD level was significantly lower in the BM 5% group and significantly decreased in all experimental groups. These results show that supplementation of BM fruit powder may have beneficial effects on diabetic complications and damage caused by oxidative stress.

Hepatotoxicity Reducing Effect of Ethanol Extracts from Fermented Youngia denticulata Houtt Kitamura in Ethanol-treated Rats (발효 이고들빼기(Youngia denticulata Houtt Kitamura) 에탄올 추출물이 알코올 투여로 손상시킨 흰쥐의 간독성 해소 효과)

  • This study was performed to investigate the hepatotoxicity reducing effect of ethanol extracts from Youngia denticulata Houtt Kitamura (YD) fermented by Aspergillus oryzae, which is well known as a Korean traditional meju microbe. Experimental rats were divided into 4 diets groups (six rats/group) (NC: normal control group, EC: ethanol supplemented control group, ENY: ethanol extracts 0.5% supplemented diets of the non-fermented YD group, EFY: ethanol extracts 0.5% supplemented diets of the fermented YD group). After feeding them for four weeks, the activities GOT and GPT in serum, contents of neutral lipid, total cholesterol, LPO and GSH, and activities of XO (O type) enzyme, SOD and GST in liver tissue were observed. Compared to the NC group, the GOT and GPT activities in the EC group were markedly increased by 87.91% and 57.02%, and the contents of neutral lipid, total cholesterol and LPO were increased by 77.51%, 18.56% and 193.86%, respectively. On the other hand, the GSH content was decreased by 29.52%. The activities of SOD and GST which are anti-oxidation-related enzymes, were decreased by 42.78% and 0.94%, respectively, compared to the NC group, but the XO (O type) enzyme activity was significantly increased by 70.83%. These results confirmed that the liver tissue and anti-oxidation system of the EC group had been damaged. Compared to the EC group, the GOT and GPT activities of the ENY and EFY groups were significantly decreased by 24.73~41.99% and 22.91~30.11%, respectively. The contents of neutral lipid, total cholesterol and LPO were decreased by 16.65~33.33%, 22.44~23.11% and 63.34~66.13%, respectively, but the GSH content was significantly increased by 24.61~35.60%. The GST activity was not significantly affected, but the SOD activity was significantly increased by 214.94~261.19%. The XO (O type) enzyme activity was significantly decreased by 40.34~51.20%. Compared to the ENY group, the GOT and GPT activities of the EFY group were significantly decreased by 22.93% and 9.34%, respectively. The plasma contents of neutral lipid, total cholesterol and LPO were decreased by 20.01%, 0.86% and 7.59%, respectively. The SOD activity was significantly increased by 21.52%, and the XO (O type) enzyme activity was decreased by 18.19%. Given decreasde TG, total cholesterol, LPO contents of liver tissue, GOT, GPT and XO activity, and increased SOD activity, these study results that ethanol extracts from fermented Youngia denticulata Houtt Kitamura may be effective in reducing hepatotoxicity.

Green Tea Intake and Exercise Reduce Some Biochemical Markers of Obese Adolescents (녹차섭취와 운동에 의한 비만 청소년의 혈중 biochemical marker 함량 감소)

  • The effects of green tea consumption and/or exercise for 12weeks on body weight and select biochemical markers in plasma were evaluated in obese adolescents with a fat ratio of greater than 25%. The subjects were randomly divided into a control group (n=9), green tea consumption group (n=9), exercise group (n=9) and green tea consumption with exercise group (n=9). Subjects in both green tea consumption group and green tea consumption with exercise group were given more than five cups of green tea extract in hot water ($90^{\circ}C$) daily, equivalent to 1.5-1.7 g dry green tea, for 12 weeks. Subjects in both the exercise group and green tea consumption with exercise group participated in a training program (HRmax 60-70%, 60 min/day) for 12 weeks. Control subjects were only given water equal to the quantity of green tea being given to the subjects of green tea consumption. No significant changes body weights were seen in any of these treatments, but the waist to hip ratio was reduced with treatments of both green tea and exercise. The control group showed no significant changes in TNF-$\alpha$, IL-6 and leptin levels. Green tea consumption reduced leptin (p<0.05), TNF-$\alpha$, and leptin levels. Exercise lowered TNF-$\alpha$ (p<0.05), IL-6 (p<0.01), and leptin (p<0.05) concentrations. Meanwhile, a combination of green tea consumption and exercise lowered TNF-$\alpha$, IL-6 (p<0.05) and leptin (p<0.05) levels. These results indicate that green tea consumption and exercise both had a positive effect on the reduction of inflammatory cytokines, TNF-$\alpha$, IL-6 and leptin, in obese adolescents, but no synergistic effect on the reduction of these cytokines.

Study of Gene-gene Interaction within GNB3, ACE, ADRB3, ADRB2 among Korean Female Subject (한국인 비만 여성의 GNB3, ACE, ADRB3, ADRB2 유전자 다형성간의 상호관계에 관한 연구)

  • There have been several reports on the relationship between G protein β3 subunit gene (GNB3), angiotensin converting enzyme gene (ACE), β3-adrenergic receptor gene (ADRB3), and β2-adrenergic receptor gene (ADRB2) genotype and obesity or obesity related disease. The objective of this study was to examine the relationship between the combinations of these four genes' polymorphism and probability of obesity related disease in Korean female subjects. The experimental group was consisted of 85 obese Korean female subjects (body mass index, BMI≥27㎏/㎡). To determine the polymorphism, genomic DNA was isolated, and PCR was performed. Serological examinations (fasting plasma glucose, FPG; aspartate aminotranferase, AST; alanine aminotransferase, ALT; total cholesterol, TC; triglyceride, TG; high density lipoprotein-cholesterol, HDL; low density lipoprotein-choles terol, LDL) were carried by an autoanalyzer and serological methods. BMI, waist circumference (WC), hip circumference and waist hip ratio (WHR) were measured. Consequencely in the analysis with grouping of general genotyping and variant allele carrier/non-carrier, the result was not significantly different within all gene combinations and polymorphic pairings except higher waist circumference in Arg16Arg group of ADRB2 codon16 (P=0.024). And there was no significantly contrast result about age, height, weight, AST and ALT that are index feature of liver and gall bladder disease in polymorphic pairings of gene combinations. However, the statistical analysis of waist-hip ratio and waist circumference that could be recognized as the physical type of obesity showed T-Arg16 pairing carrier in GNB3-ADRB2 codon16 combination had increased WHR and WC significantly (P=0.046 and P=0.015 respectively). Futhermore, the levels of total cholesterol (TC) and low density lipoprotein choresteral (LDL) were significantly lower in C-I pairing of GNB3-ACE combination (P=0.032 and P=0.005). These results suggest that the T-Arg16 pairing carrier in GNB3-ADRB2 codon16 gene might have increased waist circumference and C-I pairing carrier in GNB3-ACE combination have lower possibility of contraction of cardiovascular disease related cholesterol and LDL despite of obese state.

Effects of Belly Dancing and Nutritional Education on Body Composition and Serum Lipids Profiles of Obese Women in a Study, 'Obesity Clinic Projects at Community Healthcenter' ('보건소 비만클리닉 시범사업'으로 실시한 Belly Dance와 영양교육이 비만여성의 신체조성 및 혈청지질 농도에 미치는 영향)

  • The effects of a 12-week weight reduction program applied to obese women at a community health center were studied. Middle aged women participated in the beginning of the program, but 34 persons finished the study (85% completion). Subjects practiced belly dancing three times a week for 60 minutes (60~85% HR max) per session, and nutritional education was carried out four times during the 12 week program. The weight, %body fat, and BMI significantly decreased by 5.6%, 9.5%, and 5.6%, respectively. The concentrations of serum triglycerides, total cholesterol, and LDL-C were also significantly reduced by 13.5%, 9.5%, and 10.4%, respectively. Insulin level was reduced by 19.8%, thus lowering the HOMA-IR index. Leptin level significantly was reduced by 32.35%, adiponectin concentration. Total ROS concentration increased while total antioxidant capacity in serum remained unchanged. In conclusion, a 12-week 'Obesity Clinic Program' belly dancing and nutritional education held at a Community Center seemed to have health-promoting effects through reducing body fats, plasma lipids, and improving insulin resistance.

Thyroid Hormone-like Activity of Alachlor as R Endocrine Disruptor in Rats and HeLaTRE Cell Culture (랫드와 HeLaTRE Cell에서의 Alachlor에 의한 갑상선 호르몬성 영향 연구)

  • This study was designed to investigate the suitability of the pubertal assay and the enhanced TG 407 as methods for detection of endocrine-mediated effects, especially thyroid function. Male and female Sprague-Dawley rats were gavaged daily with 0, 12.5, 25, 50 mg/kg alachlor in corn oil during 30 days. The effects of alachlor on thyroid gland, the genital organs and thyroid hormone were measured in male and female rats. Dose of alachlor 25, 50 mg/kg/day increased relative weight of testis and thyroid gland in exposed male rats and decreased relative weight of vagina in exposed female rats. Relative weight of thyroid gland was decreased in alachlor 25 mg/kg/day exposed female rats. Dose of alachlor 25, 50 mg/kg/day decreased plasma T4 and testosterone in female rats. Another purpose of this study was to investigate the effects of endocrine disruptors as like thyroid hormone in vitro. Luciferase activity was measured to dectect reaction of test chemicals and thyroid hormone response elements in HeLaTRE cell. Dose of alachlor 1 nM-1000 nM increased 100-134% luciferase activity compared with control.

Evaluation of the Risk of Metabolic Syndrome for the Young Adults in Korean Students of a University (한국인 대학생군 대상의 청.장년층 대사이상증후군 위험성 평가)

  • Metabolic syndrome, defined as the clustering of several metabolic disorders including obesity (waist circumference ${\geq}90$ if male or ${\geq}80$ if female, cm), dyslipidemia ($TG{\geq}150$ or HDL-C<40 if male or <50 if female, mg/dl), hypertension ($BP{\geq}130/85mmHg$) and hyperglycemia (fasting plasma $glucose{\geq}110mg/dl$), increases the cardiovascular risk of the general population. Recently, risk of this syndrome arises in young adults world widely. Therefore, we randomly selected and evaluated the risk of metabolic syndrome of total 43 people (group I-22, group II-21) for 2 years. Group I was 22 peoples (15 males, 7 females) with age of 22 thru 35 year old (average 28 year old) and group II was 21 people (19 male, 2 female) with age of 22 thur 32 years old (average 24 year old) in Cheongju area from March 1st thru 30th of 2008 in Cheongju area from September 1st thru 30th of 2007 in order to find out how serious this phenomenon is in young adult of Korea. 13.95% (n=7) of total people has a metabolic syndrome by NCEP/ATPIII definition among this group (group I-6, group II-1). Those of 6 have 3 or over risk factor for metabolic syndrome such as obesity, hypertension, fasting blood glucose and hypetriglyceridemia at the same time (group I-5, group II-1). Group I have more risk factor because of more higher age than group II. Therefore we need aggressively to monitor and provide them for early diagnosis, educational programs and assistance for lifestyle changes in order to prevent metabolic syndrome among young adults.

The Effect of Dansamtongmek-tang and Dansamsengmek-san on Hyperlipidemia and Brain & Cell Damage by Hypoxia (단삼통맥탕(丹蔘通脈湯)과 단삼생맥산(丹蔘生脈散)이 고지혈증 및 Hypoxia로 유발된 뇌손상과 세포손상에 미치는 영향)

  • Background and Aims: Dansamtongmek-tang (DSTMT) and Dansamsengmek-san (DSSMS) have been used for many years as therapeutic agents for the acute stage of cerebrovascular disease, hypertension and hyperlipidemia in Oriental medicine, but the effects of DSTMT and DSSMS on hyperlipidemia and safety for cell damage are not yet well-known. This study was done to investigate the effects of DSTMT and DSSMS on hyperlipidemia. Methods: In vivo test: after administering DSTMT and DSSMS to SHR and ICR occurred hyperlipidemia for 3 weeks, we analyzed body weight, cholesterol levels. TG, HDL-chol, LDL-chol, LDH in plasma, brain, liver and kidney tissue, and DNA by RT-PCR. In vitro test: after administering DSTMT and DSSMS to human hepatocellular carcinoma in hypoxia, we observed cell cohesion by light microscope, analyzed the inflow of Ca2+ by confocal laser scanning microscope and DNA by RT-PCR. Results: DSTMT significantly decreased the levels of triglyceride and increased the levels of HDL-cholesterol in SHR, and significantly decreased the levels of LDL-cholesterol and body weight and increased the levels of HDL-cholesterol in ICR. DSSMS significantly decreased body weight, total cholesterol levels, LDL-cholesterol, LDH and cardiac risk factor (CRE) in SHR and significantly decreased the levels of total cholesterol, triglyceride, LDL-cholesterol, LDH and CRF in ICR. DSTMT had an effect on protecting cells from damage by inhibiting production of p53 mRNA, and in DSSMS, by inhibiting production of p53 mRNA and p21 mRNA after hypoxia. DSTMT effectively blocked off Ca2+ at low density, but DSSMS effectively blocked off Ca2+ at high density. Both DSTMT and DSSMS had an effect on inhibiting lipid metabolism by blocking off production of apo B mRNA. Conclusions: These results suggest that DSTMT and DSSMS might be usefully applied for treatment of hyperlipidemia and suppression of brain damage.
