A New Method for Sclerotial Isolation of Two Species of Sclerotium from Infested Soils

  • White rot on Allium crops recently had a high incidence with incrensed cultivating areas of tropical garlic types in Korea. Two types of Sclerotium have known as causal agents that produce different size and shapes of sclerotia in infested fields. Therefore, we developed a new method for isolation of sclerotia from infested field soils that can be used for ecological study of Sclerotium spp. and establishment of control strategy. Soil samples collected from heavily infested fields were evenly mixed and placed on a automatic sieve shaker connected with tap water, After 10 min of shaking, residues on 0.5 mm and 0.25 mm sieves were separately collected and suspended with 70% sugar solution, which method floats sclerotia in aqueous layer, Then, floated fraction was carefully separated and mixed with a same volume of 1% sodium hypochlorite solution to differentiate with organic materials. This method provides a direct count of sclerotia under a dissecting microscopy.

Difference of Physiochemical Characteristics Between Citrus Bacterial Canker Pathotypes and Identification of Korean Isolates with Repetitive Sequence PCRs

  • The difference of carbon source utilization and fatty acid composition between the pathotypes of Xanthomonas strains, which causing citrus bacterial canker was compared, and the physiochemical characteristics were used to analyze relationship of the strains for the first time. The pattern of several carbon sources utilization and fatty acids composition reliably discriminated the pathotypes of Xanthomonas strains. The dendrogram which was constructed by 95 carbon source utilization profiles differentiated X. axonopodis pv. citri A, $A^*$ and $A^w$ from the other pathotypes. When the dendrogram was drawn by combined analysis of carbon source utilization pattern and fatty acid composition, X. axonopodis pv. aurantifolii B, C and X. axonopodis pv. citrumelo formed a distinct cluster. The difference of carbon source utilization and fatty acid composition could be used effectively for the identification of pathotypes of citrus bacterial canker. The physiochemical characteristics strongly indicated that the strains isolated in Korea belong to X. axonopodis pv. citri A type. The cluster analysis by the band patterns of ERIC-, BOX- and REP-PCR allowed the discrimination of the pathotypes isolated from Korea. However, the rep-PCRs could not differentiate X. axonopodis pv. citri A types from $A^*$ and $A^w$ types. The overall results of metabolic profiles and rep-PCRs strongly indicated that the Korean isolates are X. axonopodis pv. citri A type.

Planting Structure and Ecological Characteristics of Green Spaces at the Dangjin Steam Power Plant (당진화력발전소 녹지공간의 식재현황 및 생태적 특성 분석)

  • This study was conducted to analyze the planting structure and ecological characteristics of greon spaces on the grounds of e Dangjin steam power plant. To achieve these goals, we surveyed existing vegetation, plant community structure, Plant volume and growth rate. Based on e vegetation analysis, existing vegetation was classified into six types: herbaceous species $(70.54\%)$, evergreen coniferous trees $(21.17\%)$, deciduous broad-leaved trees $(5.10\%)$, deciduous coniferous trees $(1.47\%)$, shrubs $(0.12\%)$, and other types $(1.59\%)$. The coal storage, office, and playground areas were community is the natural forest area where a Pinus thunbergii / Pinus densiflora community is distributed in terms of vegetation structure, species diversity, plant volume, and growth rate. The artificial green spaces(near the coal storage, office, playground areas) had a single-layer structure. Species diversity indices of the artificial green areas were $0.1655\~0.4807$ compared to 0.8628 in the natural forest, which presented a good growth environment. Also, the plant volume in the artificial green space was lower than that of the natural green space. Therefore, it would be desirable to develope a multi-layer structure similar to that of the vegetation in the natural green space in order to improve the amount of plant volume. The plant-damage ratio of Pinus thunbergii was $52.48\%$ in the coal storage area, and $8.48\~ 11.52\%$, in the other survey areas. Also, the vitality of Pinus thunbergii was $15.45k{\Omega}$ in the coal storage areas, which indicates bad growing conditions. This suggests that soil characteristics and dust have a bad impact on growth. The investigation into deciduous tres' growth status showed that appropriate plants would be Albizzia julibrissin, Acer palmatum var. sanguineum, Acer palmatum, Malus spp., Prunus sargentii.

Morphological characteristics of major airborne pollen in Korea peninsula

  • Although airborne pollen is invisible to the eye, it has been known as a major source to respiratory allergic reactions. For this reason, airborne pollen is monitoring in many countries to predict pollen concentration based on locality and season. However, the morphological characteristics of airborne pollen and their potential tendency as an allergen are still obscure. In the present study, we selected 52 airborne pollen samples based on previously reported data and investigated their detail pollen characteristics using LM and SEM. Major airborne pollen in Korea has sorted in 19 families (most angiosperms except four gymnosperm families), and all pollen grains are small to medium in size ($P=17.34-49.86{\mu}m$) apart from the bisaccate pollen grains of Pinaceae ($P=46.49-106.20{\mu}m$). The aperture number and shape vary from sulcate to polyporate. While the inaperture pollen has found only in gymnosperm (Cupressaceae and Taxaceae), triporate or polyporate is common pollen type in angiosperm. The sexine ornamentations could divide into several types, but the most sculpturing types are inconspicuous like psilate, rugulate and granulate. Reticulate pollen grains as a semitectum have occurred the species of genera Platanus and Fraxinus only. To estimate the possible relationships between pollen features and allergen, the results are discussed in botanical context.

Characteristics of Heavy Metals Uptake by Plants: Based on Plant Species, Types of Heavy Metals, and Initial Metal Concentration in Soil (식물정화공법에서 다양한 중금속의 식물체로의 흡수 및 축적 특성 비교: 식물체 종류, 중금속 종류, 토양 내 중금속 농도를 중심으로)

  • Phytoextraction, one type of phytoremediation processes, has been widely used in the removal of heavy metals from polluted soil. This paper reviewed literature on metal uptake by plants and characterized the metal uptake by types of metals (Zn, Cu, Pb, Cd, and As), plant species, initial metal concentrations in soil and the distribution of metals in different parts of plants. The potential of metal accumulation and transport by plants was closely related to plants species, types of metals, and initial metal concentrations in soil. The plants belonging to Brassicaceae, Solanaceae, Poaceae, and Convolvulaceae families have shown the high potential capacity of Cd accumulation. The Gentianaceae, Euphorbiaceae, and Polygonaceae families have exhibited relatively high Pb uptake potential while the Pteridaceae and Cyperaceae families have shown relatively high Zn uptake potential. The Pteridaceae family could uptake a remarkably high amount of As compared with other plant families. The potential metal accumulation per plant biomass has increased with increasing initial metal concentration in soil up to a certain level and then decreased for Cd and Zn. For As, only Pteris vittata had a linear relationship between initial concentration in soil and potential of metal uptake. However, a meaningful relationship for Pb was not found in this study. Generally, the plants having high metal uptake potential for Cd or Pb mainly accumulated the metal in their roots. However, the Euphorbiaceae family has accumulated more than 80% of Pb in shoot. Zn has evenly accumulated in roots and stems except for the plants belonging to the Polygonaceae and Rosaceae families which accumulated Zn in their leaves. The Pteridaceae family has accumulated a higher amount of As in leaves than roots. The types of metals, plant species, and initial metal concentration in soil influence the metal uptake by plants. It is important to select site-specific plant species for effective removal of metals in soil. Therefore, this study may provide useful and beneficial information on metal accumulation by plants for the in situ phytoremediation.

Antimicrobial Effect of Natural Plant Extracts against Periodontopathic Bacteria (치주염 원인균에 대한 천연 식물 추출물의 항균효과)

  • In this study, we examined the antimicroboal effect against Actinobacillus actinomycetemcomitans and Prevotella intermedia which were the bacteria causing the Periodontopathic by using 34 types of natural plant extracts. Therefore, this study measures growth inhibition activity and Minimum Inhibition Concentration (MIC) of a sample extract with the use of organic solvent extracts in order to analyze the antibacterial effect of natural plant extracts on periodontopathic bacteria. Each of the 34 types of natural plant extracts were extracted by using the ethanol, and subsequently, the size of growth inhibition zone(clear zone, ㎜) of respective extracts were measured through the disk diffusion method. As a result, it was found that the growth inhibitory activity was found for A. actinomycetemcomitans, which is the bacteria causing the Periodontitis, in 13 types of natural plant extracts such as Raphanus sativus, Akebia quinata, Paeonia lactiflora, Belamcanda chinensis, Inula britannics, Houttuynia cordata, Forsythia saxatilis, Gentiana macrophylla, Melia azedarach, Scutellaria baicalensis, Coptis chinensis, Phellodendron amurense, Kalopanax Pictus, etc. In the case of P. intermedia, the growth inhibitory activity was found in 13 types of natural plant extracts such as Raphanus sativus, Angelica acutiloba, Akebia quinata, Belamcanda chinensis, Inula britannics, Houttuynia cordata, Cinnamomum cassia, Aster tataricus, Melia azedarach, Scutellaria baicalensis, Coptis chinensis, Phellodendron amurense, Kalopanax Pictus etc. For A. actinomycetemcomitans, anti-bacterial effect was exhibited in Belamcanda chinensis, Cinnamomum cassia, Kalopanax Pictus, Phellodendron amurense, Coptis chinensis. The Coptis chinensis showed the most excellent growth inhibitory activity in all organic solvent fragment, while P. intermedia showed the growth inhibitory activity in Belamcanda chinensis, Cinnamomum cassia, Meliaazedarach, Phellodendron amurense, and Coptis chinensis.

Complete Genome Sequence of the RNAs 3 and 4 Segments of Rice stripe virus Isolates in Korea and their Phylogenetic Relationships with Japan and China Isolates

  • The complete genome sequences of RNA3 and RNA4 of the 13 different Rice stripe virus (RSV) isolates were determined and characterized in this study to address the possible causes of the recent re-emergence of RSV that affected many rice fields in Korea. The genome size of each RNA segment varied among isolates and significant differences were observed in the intergenic region. There was up to 4% average divergence in the RNA4 nucleotide sequence among 13 Korean isolates and only 1.4% in the RNA3. Phylogenetic relationships among different Korean isolates revealed that there were at least 2 types of RNA3 and 4 distinct types of RNA4 genomes present in Korea. However, Korean isolates with one type of RNA3 predominate over the other while the occurrences of the RSV Korean isolates with the 4 types of RNA4 genome were not correlated to specific geographical areas. Results further indicate that RNA4 had diverged more than RNA3 and these differences in accumulation of mutations in the individual RNA segments indicate that genetic reassortment were likely to contribute to the genetic divergence in the 13 Korean isolates. All of the Korean-RNA3 sequences except for one isolate grouped with Chinese isolates (JY and Z). In contrast, the RNA 4 sequences segregated together with either Chinese (JY and Z) and Japanese (M and T) isolates but genetic relationships of Korean isolates- RNAs 3 and 4 segments to Chinese-Y isolate were low. Altogether, these results suggest that the occurrence of mixtures of RNAs 3 and 4 genotypes in the natural population of RSV may have contributed to the sudden outbreak in Korea.

Conceptual Models of Violation Error in a Nuclear Power Plant (원자력 산업의 위반오류 발생 메커니즘 개발 및 유형 분류)

  • Although many studies have been conducted to find solutions to deal with human errors effectively, violations have been rarely studied in depth. The violation is a type of human error when an employee takes an action with intention but does not intend harmful outcomes. Violations have characteristics similar to other types of human errors but it is difficult to understand the intention of an employee from accident reports. The objective of this study is to develop a conceptual model of violation errors for preventing accidents/failures in a nuclear power plant from violation errors. Based on the previous studies, the characteristics of violations were collected in 9 categories and 136 items. They were classified into three-kinds of characteristics (human-related, task-related, organization-related characteristics) to construct conceptual models of routine/situational violations. The representative cases of accidents/failures in a nuclear power plant were analyzed to derive the specific types of routine/situational violation occurrence. Three types of conceptual model for each violation were derived according to whether the basic, human-related, and task-related characteristics are included or not. The conceptual models can be utilized to develop guidelines to support employees preventing routine/situational violations and to develop supportive system in nuclear power plant.

Sensitivity of Lavender to Proton, Electron, and Gamma Radiation

  • While ion beams are widely used in plant breeding, little is known about the sensitivity of Lavandula angustifolia (lavender) to ionizing radiation. To compare the biological effects of different types of ionizing radiation on the germination and survival rates of lavender, we exposed lavender seeds to gamma rays, 3 MeV electron beams, and 1.89 MeV proton ion beams. We observed that the seed germination rate decreased with increasing dosages of all three types of ionizing radiation. The malformation rate of lavender seedlings exposed to electron beams and gamma rays increased with increasing radiation dosage. By contrast, the effect of the accelerated proton beams on the malformation rate was negatively correlated with the dosage used. The survival rate of lavender seedlings exposed to the three types of ionizing radiation decreased in a dose-dependent manner. In addition, the survival rate of seedlings irradiated with proton and electron beams decreased more slowly than did that of seedlings irradiated with gamma rays. The half-lethal dose of gamma rays, electron beams, and proton beams was determined to be 48.1 Gy, 134.3 Gy, and 277.8 Gy, respectively, and the most suitable proton-ion energy for lavender seeds in terms of penetration depth was determined to be 5 MeV. These findings provide valuable information for the breeding of lavender by radiation mutation.

Opal Phytolith Morphology in Rice (벼의 규소체 형태)

  • The opal phytolith types in Oryza sativa L. were examined with light and scanning electron microscope, and we suggested the standard tenninoligies based on original cells of the phytoliths in situ. The distinctive fonus of opal phytoliths were observed in all plant parts such as leaf blade, leaf sheath, culm, root, glume, lemma, and palea. The phytolith fonus originated from long cell, short cell, and dennal appendages were different from part to part. The opal phytoliths derived from the long cells showed the most various form: twelve types were identified based on side wall shape. The results showed that the leaf blade did include almost all types of opal phytolith observed in the other parts of rice.f rice.
