Here, we designed to examine the anti-inflammatory effects on RAW264.7 cells and the immunosuppressive effects by evaluating interleukin-2 (IL-2) production in Jurkat T cells using a MeOH extract of Panax notoginseng roots. The results showed that the MeOH extract inhibited the synthesis of nitric oxide (NO) in a dose-dependent manner (IC50 value of 7.08 ㎍/mL) and displayed effects on T cell activation at a concentration of 400 ㎍/mL. In efforts to identify the potent compounds, bioactivity-guided fractionation of the MeOH extract and chemical investigation of its active CH2Cl2-, EtOAc-, and butanol-soluble fractions led to the successful isolation and identification of eleven compounds, including two polyacetylenes (1, 2), a steroid saponin (3), seven dammarane-type ginsenosides (4 - 10), and an oleanane-type ginsenoside (11). Among them, compound 11 was isolated from this plant for the first time. Compound 2 exhibited potent inhibitory effects on NO synthesis and an immunosuppressive effect with IC50 values of 2.28 and 65.57 μM, respectively.
Background: Panax ginseng Meyer (Araliaceae) is a highly valued medicinal plant in Asian regions, especially in Korea, China, and Japan. Chemical and biological studies on P. ginseng have focused primarily on its roots, whereas the seeds remain poorly understood. This study explores the phytochemical and biological properties of compounds from P. ginseng seeds. Methods: P. ginseng seeds were extracted with methanol, and 16 compounds were isolated using various chromatographic methods. The chemical structures of the isolates were determined by spectroscopic data. Antiinflammatory activities were evaluated for triterpene and steroidal saponins using lipopolysaccharide-stimulated RAW264.7 macrophages and THP-1 monocyte leukemia cells. Results: Phytochemical investigation of P. ginseng seeds led to the isolation of a novel triterpene saponin, pseudoginsenoside RT8, along with 15 known compounds. Pseudoginsenoside RT8 exhibited more potent antiinflammatory activity than the other saponins, attenuating lipopolysaccharide-mediated induction of proinflammatory genes such as interleukin-1β, interleukin-6, inducible nitric oxide synthase, cyclooxygenase-2, and matrix metalloproteinase-9, and suppressed reactive oxygen species and nitric oxide generation in a dose-dependent manner. Conclusion: These findings indicate that pseudoginsenoside RT8 has a pharmaceutical potential as an antiinflammatory agent and that P. ginseng seeds are a good natural source for discovering novel bioactive molecules.
Background: The roots of Panax ginseng contain two types of tetracyclic triterpenoid saponins, namely, protopanaxadiol (PPD)-type saponins and protopanaxatiol (PPT)-type saponins. In P. ginseng, the protopanaxadiol 6-hydroxylase (PPT synthase) enzyme catalyses protopanaxatriol (PPT) production from protopanaxadiol (PPD). In this study, we constructed homozygous mutant lines of ginseng by CRISPR/Cas9-mediated mutagenesis of the PPT synthase gene and obtained the mutant ginseng root lines having complete depletion of the PPT-type ginsenosides. Methods: Two sgRNAs (single guide RNAs) were designed for target mutations in the exon sequences of the two PPT synthase genes (both PPTa and PPTg sequences) with the CRISPR/Cas9 system. Transgenic ginseng roots were generated through Agrobacterium-mediated transformation. The mutant lines were screened by ginsenoside analysis and DNA sequencing. Result: Ginsenoside analysis revealed the complete depletion of PPT-type ginsenosides in three putative mutant lines (Cr4, Cr7, and Cr14). The reduction of PPT-type ginsenosides in mutant lines led to increased accumulation of PPD-type ginsenosides. The gene editing in the selected mutant lines was confirmed by targeted deep sequencing. Conclusion: We have established the genome editing protocol by CRISPR/Cas9 system in P. ginseng and demonstrated the mutated roots producing only PPD-type ginsenosides by depleting PPT-type ginsenosides. Because the pharmacological activity of PPD-group ginsenosides is significantly different from that of PPT-group ginsenosides, the new type of ginseng mutant producing only PPD-group ginsenosides may have new pharmacological characteristics compared to wild-type ginseng. This is the first report to generate target-induced mutations for the modification of saponin biosynthesis in Panax species using CRISPR-Cas9 system.
This review deals briefly with the various medicinal components(mainly saikosaponins), their biological activities and the variation of their contents by different cultivation environment and plant parts in Bupleurum species. Bupleuri radix, a crude drug, is the root of Bupleurum falcatum L. (Korea, Japan), B. chinense(China), and their related species (Umbelliferae). There are over 120 species in Bupleurum genus throughout world, mainly Asian area, and over 5 species in Korea, investigated up to now. These plants contain many physiological active compounds and the principal components are saikosaponins. Major activities of this crude drug and saikosaponins are the anti-inflammatory and antihepatotoxic activities. Saikosaponins and their derivatives in Bupleurum spp. have been chemically studied, isolated and identified over 70 compounds in over 50 species. Other components, physiologically active ones, also have been investigated, which are the groups of lignan, flavonoid, essential oil, polyacetylene, polysaccharide, etc. Saikosaponins belong to the group of triterpenoid saponin chemotaxonomically and occur the accumulation and turnover in plant tissues through secondary metabolism, mevalonic acid pathway. The contents and kinds of saikosaponins and other components in Bupleurum spp. plants are various due to different species and growing environments, as the plant growth characters and yield are various. Most of medicinal plants as well as Bupleurum species are very useful as agricultural products and traditional medicines, and also are very valuable as genetic resources and natural products. So we need to collect, evaluate, preserve, and utilize various medicinal plants, and also to under-stand secondary metabolism and improve the breeding and cultivation techniques for the safe production of crude drugs with high quality and yielding.
Ocimum gratissimum is a common plant in the tropics and has been used in food and medicine. Its usage in food and medicine could be attributed to its phtyochemical and antimicrobial properties. In this study we investigated the proximate, phytochemical, and antimicrobial attributes of air dried leaves of O. gratissimum. The aqueous extract was found to contain phtyochemicals with alkaloid and saponin present in appreciable amounts. The proximate analysis (crude protein and crude fibre content were 15.075% and 17.365%, respectively) showed that the leaf could be a good source of protein and fibre. The aqueous ethanolic extract of the leaf exhibited activity against a wider range of organisms when compared to the aqueous extract at the investigated concentrations. Aqueous ethanolic extracts of O. gratissimum leaf was active against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, and Bacillus cereus and the aqueous extract of the leaf was active against P. aeruginosa.
The pharmacognostical and pharmacological studies of 'Manryong' were carried out, which has been widely used in Chun-Ra Province, Korea and the following results were obtained. 1) The original plant of the bulb MANRYONG is Erythronium japonicum DECNE. (Liliaceae). 2) The internal structure consists of mostly parenchyma containing numerous starch grains, vascular bundles and substituted fibers. 3) The external surface occurs as a yellowish white and slightly bented ovoidal shape. 4) 'Manryong; contains 11 kinds of free amino acids, such as asparagin, tryptophan, cystine, glutamic acid, threonine, glycine, leucine, proline, histidine, methionine, and alanine. 5) Hemolytic action of the crude saponin isolated from 'Manryong' is weaker than that of digitonin. 6) Effects of 'Manryong' extracts on ceruloplasmin were studied and an antidotal activity of the extracts was found in liver intoxicated with carbon tetrachloride.
이 연구는 도라지의 유기재배를 위하여 유기질 비료 시용이 도라지 뿌리의 생육특성 및 사포닌, 항산화 활성에 미치는 영향을 구명하고자 실시되었다. 3년근 도라지를 이식하기 전 기비로 화학비료, 유기질비료, 균배양체, 유박 및 퇴비를 시용한 결과, 뿌리의 길이는 화학비료와 유기질 비료 시용구에서 25.3 cm와 24.0 cm로 가장 길었으며, 직경에서도 화학비료 시용구에서 가장 굵은 26.6 mm 나타났다. 지근의 수는 유기질퇴비와 화학비료 시용구에서 가장 많은 20.0개와 17.0개로 조사되었으며, 수량과 관련성이 있는 생체중은 유기질비료 시용구에서 55.7 g으로 가장 높게 나타났다. Platycodin D의 함량은 327.4~373.8 mg/100 g의 범위로 유기질퇴비 시용구에서 높은 값이 나타났으며, 도라지의 총 polyphenol 및 flavonoid 함량은 유기질비료 시용구에서 각각 15.5, 15.3 mg/g 가장 높게 나타났다. 따라서 도라지의 유기재배시 유기질비료 시용구에서 수량성을 물론, 약리성 증대를 위한 사포닌 함량이 많은 지근의 수와 항산화 성분이 높은 것으로 나타났다.
인삼의 답전윤환재배에 있어서의 문제점을 파악하는 한편 답전윤환 재배지 토양의 이화학적 특성과 인삼의 생육 및 수량을 조사하며 논에서의 인삼연작재배의 안전성 기작을 구명하여 인삼의 답전윤환재배를 위한 기초를 확립하고자 본 실험을 수행하였던 바 그 결과를 요약하면 다음과 같다. 1. 금산지방에서의 답전윤환 인삼재배시 가장 심각한 문제점은 벼의 재배후에 토양에 잔류하는 무기염류의 과다였다. 2. 밭재배지에 비하여 답전윤환 인삼재배지에서 유기물함량과 CEC 그리고 가리를 위시한 각종 양이온의 함량이 높은 경향이었다. 3. 답전윤환 초작지와 재작지에서의 3년생 및 4년생 인삼의 경엽생육은 밭초작지 인삼의 생육과 뚜렷한 차이가 없었으나 이들에 비해 밭재작지 인삼의 경엽생육은 현저하게 떨어졌다. 또한 답전윤환 재배지에서는 황병의 발생이 많았으며 땅강아지가 우점해충이었다. 4. 답전윤환 초작지와 재작지에서의 4년생 인삼근의 생육 및 간당 근수량은 밭초작지 인삼의 생육 및 수확량과는 뚜렷한 차이가 없었으나 이들에 비해 밭재작지 인삼근의 수량은 현저한 감소를 나타내었다. 5. 답전윤환재배시 인삼의 재배후에 4년정도 벼를 재배한 다음 다시 인삼을 재배하여도 인삼에 연각장해현상이 뚜렷하게 나타나지 않았으며 또한 근부병의 발생도 심하지 않아 비교적 안전한 재작이 가능하였다. 6. 답전윤환지에서 재배된 4년생 인삼의 조-Saponin 및 개별 Saponin함량은 관행의 방법으로 재배된 인삼과 전혀 차이가 없었다. 7. 정상답에 비하여 인삼재배답 토양의 비옥도가 대체로 약간 저조한 경향이었으나 수도의 생육 및 수량에는 전혀 영향을 미치지 않았다.
이 연구는 도라지의 유기재배를 위하여 피복물 처리에 따른 도라지 뿌리의 생육특성 및 사포닌, 항산화 활성에 미치는 영향을 구명하고자 실시되었다. 3년근 도라지를 이식 후, 흑색 비닐, 톱밥, 쌀겨 및 무피복구의 뿌리의 길이는 비닐, 톱밥, 무피복구에서 24.0~27.5 cm로 길었으며, 직경에서는 비닐 피복구에서 가장 굵은 30.0 mm로 나타났다. 지근의 수도 비닐 피복구에서 가장 많은 36.0개로 조사되었으며, 수량과 관련성이 있는 생체중은 지근의 수와 직경이 가장 굵었던 비닐 피복구에서 130 g으로 가장 높게 나타났으며 무피복구에서 56 g으로 가장 낮게 나타났다. plantycodin D3와 deapioplatycodin D 함량은 톱밥 피복구에서 가장 높게 나타났으며, 도라지의 총 polyphenol 및 flavonoid 함량은 비닐 피복구에서 각각 11.0, 8.6 mg/g으로 가장 높게 나타났다. Polygalacin D와 ABTS와의 상관관계는 $-0.591^*$로 부의 상관관계를 보였으나, 나머지 성분에서는 상관관계가 없는 것으로 나타났다. 따라서 도라지 유기 재배 시 피복재료에 따른 뿌리의 특성, 사포닌함량과 항산화성분 등의 유의적인 차이가 나타나는 것을 확인할 수 있었다.
홍삼(紅蔘)과 백삼(白蔘)을 실온(室溫)에서 장기보존(長期保存)하였을 때 인삼(人蔘)의 유효약리성분(有效藥理成分)으로 밝혀진 사포닌의 보존년수(保存年數)에 따른 함량변화(含量變化) 및 triol계(系)사포닌과 diol계(系)사포논의 비(比) 즉 PT/PD ratio 등을 조사(調査)하여 다음과 같은 결과(結果)를 얻었다. (1) 보존년수(保存年數)에 따라 총질소(總窒素), 조단백질(粗蛋白質) 및 조지방(粗脂肪)의 함량(含量)은 서서히 감소(減少)하였으나 홍삼(紅蔘)과 백삼(白蔘)사이에는 큰 유의성(有意性)을 인정(認定)할 수 없었다. (2) 총(總)사포닌 함량(含量)은 보존년수(保存年數)에 따라 상당한 감소현상(減少現象)을 나타내었는데 그 정도는 홍삼(紅蔘)보다 백삼(白蔘)이 현저하였다. 3년(年)동안 보존(保存)했을 때 홍삼(紅蔘)은 12%만 감소(減少)한 반면 백삼(白蔘)은 27%나 감소(減少)하였다. (3) HPLC로 정량(定量)한 주성분(主性分)사포닌의 함량(含量)도 보존년수(保存年數)에 따라 감소현상(減少現象)이 뚜렷하였는데 보존기간(保存期間) 3년(年)이 경과(經過)했을 때 홍삼(紅蔘)은 16.8%만이 감소(減少)한 반면 백삼(白蔘)은 같은 기간(期間)에 무려 42.2%가 감소(減少)하였다. (4) 보존년수(保存年數)에 따른 PT/PDratio를 보면 홍삼(紅蔘)은 0.646~0.694%의 범위내에 또 백삼(白蔘)은 0.414~0.428%의 범위내에 있어 홍삼(紅蔘)이 백삼(白蔘)에 비(比)해 PT/PD ratio가 약 50% 정도가 높았다. 이것이 홍삼(紅蔘)의 약리작용(藥理作用)에 깊이 관계(關係)할 것으로 생각된다. (5) 홍삼(紅蔘)과 백삼(白蔘)의 추출액(抽出液)의 가시부(可視部)의 변화(變化)를 보면 백삼(白蔘)이 홍삼(紅蔘)보다 증가폭(增加幅)이 더 크므로 불안정(不安定)함을 알 수 있고, 또 자외부(紫外部)의 변화(變化)를 보면 278nm에서 홍삼(紅蔘)의 흡광도(吸光度)가 백삼(白蔘)보다 크므로 홍삼(紅蔘)의 향미(香味)가 백삼(白蔘)보다 좋음을 알 수 있다.
