Presently, we report a simple, reproducible and high frequency plant regeneration in Hibiscus syriacus L. using axillary buds. H. syriacus was regenerated from axillary buds directly or through a callus phase. Regenerated shoots were directly induced from young and fresh axillary buds cultured on Murashige and Skoog medium (MS) supplemented with 0.01 mg/L of the growth regulator thidiazuron (TDZ) after 2 weeks of culture. Directly induced shoots were transferred to hormone-free MS medium and root development was observed after 6 weeks. On the other hand, old and stale axillary buds were regenerated to shoots via callus induction on MS medium containing 0.01–2 mg/L TDZ after 4 weeks. A TDZ concentration of 0.01 mg/L was most effective in callus formation. Green callus was transferred to MS medium containing 0.01 mg/L α-naphthalene acetic acid (NAA) and 0.5 mg/L benzylaminopurine (BA). After 4 weeks, callus had developed into multiple shoots. Plantlets were formed from 10 week cultures of single shoots on hormone-free MS medium. Regenerated plantlets were cultured on MS medium for one month and then transferred to pots containing garden soil. Potted plants were acclimatized for one month and grown to maturity under greenhouse conditions. The present study has shown that various concentrations of plant growth regulator can be effective for in vitro plant regeneration of H. syriacus. The direct and indirect regeneration protocol presented here will be useful for understanding the manipulation and propagation of H. syriacus.
High frequency plant proliferation system via direct frond regeneration of endemic duckweed plants Lemna paucicostata and Spirodela polyrhiza was established. Fronds of L. paucicostata and S. polyrhiza were able to multiply half-strength MS basal medium without plant growth regulators. However, addition of BA at a range of 1 to 3 mg/L was more effective than high concentration of BA treatments for fronds proliferation. Also half-strength MS salts was suitable for the fronds proliferation. Increase of salts concentration had inhibitory effect on fronds proliferation. Also the frequency of callus formation from fronds of L. paucicostata was 3.3%, when they cultured onto 1/2 MS medium supplemented with 1 mg/L of BA. Similarly the frequency of callus formation from S. polyrhiza was very low. After subculture of white globular structures derived from fronds of L. paucicostata, numerous globular somatic embryos and calluses were developed onto the surface of fronds. However these somatic embryos did not fully develop into normal plants when transferred to 1/2 MS basal medium. Therefore direct frond regeneration system was more efficient for mass proliferation of L. paucicostata and S. polyrhiza. The plant regeneration system of L. paucicostata and S. polyrhiza established in this study, might be applied to mass proliferation and genetic transformation for molecular breeding.
벼 육종을 위한 약 배양은 기존 육종 방법을 사용하여 새로운 품종을 개발하는데 최소 6세대에 필요한 시간을 크게 줄여 동형접합자를 신속하게 생산하는 방법이다. 이 약 배양기술은 방법론적 관점에서 더 많은 녹색 식물을 얻을 기회를 제공하며, 약 배양에서 시간과 노력을 절약하는 배양의 효율성을 높이기 때문에 중요하다. 본 연구에서는 배양액과 배양 방법이 다른 1단계와 2 단계 배양의 캘러스 유도율과 녹색 식물 재분화율로 비교하였다. 1단계 배양은 하나의 배지에서 캘러스 유도 및 식물 재생을 허용하는 반면, 2 단계 배양은 두 개의 다른 배지에서 유도 및 식물 재분화가 필요하다. 이 연구에서는 1 단계 및 2 단계 배양으로 벼 꽃밥의 캘러스 유도와 식물 재생률을 비교했다. 캘러스 형성률은 1 단계 배양의 경우 13.0 %, 2 단계 배양의 경우 8.6%로 2 단계 배양보다 1 단계 배양에서 4.4% 더 높았다. 식물 재분화율은 1 단계 배양에서 1.0%, 2 단계 배양에서 3.0 %로 1 단계 배양보다 2단계 배양에서 식물체 재분화율이 3배 더 높았다. 이것은 2 단계 배양이 반수체 생산을 위한 1 단계 배양보다 더 효율적임을 시사한다.

In order to enhance the system of potato transformation and further regeneration, potato was transformed using the Agrobacterium tumefaciens harboring $\beta$-glucuronidase (GUS) gene. We found that a series fo modified medium ttained 100% shoot regeneration within 5 weeks after the preincubated explants on stage I medium were infected with Agrobacterium. Callus appeared at the cut edges of stem segments on stage II medium, mainly at the basal parts. Some explants started to form shoots after two to three weeks on stage III medium containing kanamycin (50 mg/L). When transferred to MS medium containing 200 mg/L kanamycin, 81% of the transformed shoots formed roots at the cut edge of the plantlets. In contrast, untrasformed shoots never rooted and became yellowish after few weeks under the same conditions. Southern and northern analysis indicated in vitro shoot regeneration on the callus derived from the potato explants, which were incubated with Agrobacteria. The regeneration cycle was shortened after the transformatin and finally the transformation efficiency was highly enhanced.
An efficient plant regeneration protocol is developed for a standard-type chrysanthemum. When petal segments derived from flower buds (4 or 8cm in diameter) were used as the culture material, the highest shoot regeneration frequency (96%) was obtained on a Murashige and Skoog (MS) medium supplemented with 0.5 mg/L IAA, 2 mg/L BA, 3% sucrose, and a 0.8% agar. Pre-culturing the explants under dark conditions for 14 days produced better results for the shoot regeneration frequency than the explants cultured under a continuous 16 h photoperiod ($40{\mu}molm^{-2}s^{-1}$). The shoot regeneration frequency ranged from 19.0% for the Shinmato cultivar to 89.1% for the Baeksun cultivar. Activated charcoal (0.2%) enhanced the root formation of the regenerated shoots in a hormone-free MS medium. The rooted plantlets were acclimatized and successfully established in a greenhouse.
Explants of button daisy were screened for their regeneration potential and transient GUS gene expression. Medium containing MS salts minerals and $B_5$ vitamins supplemented with $0.1\;\cal{mg/L}$ BA and $0.1\;\cal{mg/L}$ TDZ showed the best regeneration. Disc florets and receptacles were the most responsive explants in regeneration and transient gene expression respectively. Regenerated plants were successfully rooted and established in the green-house conditions. Infection and co-cultivation of explants with Agrobacterium tumefaciens containing pCAMBIA 1301 resulted in transient GUS foci. Among the different explants, receptacles showed the highest percentage of transient GUS gene expression. Enzymatic and molecular analyses of transformed calli confirmed the integration of GUS gene.
A method for plant regeneration via organogenesis from Pelagonium inquinans leaf disc has been developed. Mature leaf explants were collected from field-grown plants and used for the induction of adventitious shoot regeneration on Murashige and Skoog (MS) medium supplemented with 3% (w/v) sucrose plus plant growth regulators. Maximum shoot organogenesis, with $11.8{\pm}1.5$ shoots (98.6%) per leaf disc, was obtained with $2\;mg/l$$N^6-benzyladenine$ (BA) and $0.5\;mg/l$${\alpha}-naphthyleneacetic$ acid (NAA) in 30 days. For rooting, the in vitro proliferated and elongated shoots were excised into 1.5-2 cm in length microcutting, which were plated individually on an half-strength MS (1/2MS) medium supplemented with 2% (w/v) sucrose plus various concentrations of indole-3-butyric acid (IBA). Shoots rooted with a frequency of 100% following culture on 1/2MS medium containing $0.5\;mg/l$ IBA.
Our study was carried out for plant regeneration via somatic embryogenesis from immature seeds of Bletilla striata. The highest frequency of embryogenic callus formation was obtained from the immature seeds (at 150 days after pollination) cultured on Hyponex and VW medium supplemented with 3 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D) and 1 mg/l kinetin under the dark condition. Multiple somatic embryos were induced when embryogenic callus was transferred to VW medium without growth regulators under continued illumination. Somatic embryos were observed histologically with scanning electron microscopy. Regeneration of Bletilla striata was obtained from somatic embryos with a well-defined scutellum and coleoptile as well as with one or more shoot primordia and root primordia. We think that these methods for orchid multiplication must be useful to access clonal propagation of orchids.
Root cluster section culture, showing high efficient Protocorm-like body (PLB) formation capacity, were established in Doritaenopsis hybrids. Three types of root were obtained from excised shoots in 1/2MS medium containing different concentrations of NAA; \circled1normal roots, \circled2multiple roots and \circled3abnormal root clusters. Those were placed on 1/2MS medium supplemented with 0.5 mg/L thidiazuron for PLB regeneration. PLB regeneration rate was greater in root cluster section cultures (77.8%) compare to normal root tip cultures(30%). Number of PLBs regenerated from root cluster sections were counted over 11 per explant (5.3 per normal root tip).High frequency of PLB regeneration was achieved in root cluster section culture. This result can be used as an efficient method for clonal proliferation of Doritaenopsis hybrids.
The effect of carbenicillin on the dedifferentiation and the regeneration efficiency of plant tissues of horseradish(Armoracia rusticana) was evaluated, Inhibition effect for callus initiation was observed when leaf blade, root and petiole segments were grown on MS medium containing 500 mg/L to 2000 mg/L carbenicillin and 0.5 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D). The regeneration of horseradish shoots from leaf blade, root and petiole explants were decreased as the addition of carbenicillin increased from 1000 mg/L to 2000 mg/L in MS medium containing 0.5 mg/L of 6-benzylaminopurine (BAP) or kinetin. Especially, 500 mg/L carbenicillin treatment significantly inhibited shoot induction when leaf blade explants were grown on hormone-free MS medium. It was suggested that the toxic effects of combinations of carbenicillin and 2,4-D may be due to high auxin activity levels.
