  • Orchardgrass의 종자배양에서 캘러스형성과 식물체 재분화에 적합한 조건을구명하기 위하여 품종간의 차이와 배지조성 및 생장조절물질의 조성 등에 대한 일련의 실험을 수행한 바, 공시품종중 "Amba"의 식물체 재분화율이 가장 높았고, $N_6$ 배지가 MS, $B_5$ 배지보다 종자에서 형성된 캘러스 생체중이 무거웠으며, 식물체 재분화율도 높게 나타났다. 또한 3 mg/L의 dicamba가 첨가된 배지에서 형성된 캘러스를 재분화배지에 이식하였을 때 식물체 재분화율이 30%로 가장 높게 나타났다.

  • 딸기 '수홍'의 기내배양된 잎과 탁엽 절편체로부터 기관형성을 통한 식물체 재생 방법을 확립하고자 실험을 수행하였다. 엽절편체 배양에서는 생장조절제 처리에 따라 NAA 첨가시에는 뿌리 재생, BA 첨가시에는 신초 재생이 이루어졌으며, BA 1.0㎎/L + NAA 0.2 ㎎/L 처리구에서 재생률 31.1%, 절편체당 신초수 1.7개로 가장 양호하였다. 탁엽절편체 배양에서는 생장조절제와 광조건의 처리를 실시하였는데, 명조건에서는 생장조절제 처리에 따른 뚜렷한 경향을 발견할 수 없었으나, 암조건에서는 고농도 BA와 저농도 NAA의 혼용처리가 신초 재생에 필요함을 알 수 있었다. 탁엽절편체는 BA 2.0 ㎎/L + NAA 0.1㎎/L 처리구에서 재생률 44.4%, 절편체당 신초수 4.0개로 가장 양호한 반응을 나타내었다. 재생된 신초는 NAA 0.1㎎/L 가 첨가된 MS배지에서 발근되었으며, 이어서 인공토양(vermiculite : perlite = 1 : 1)에서 성공적으로 활착을 유도할 수 있었다.

In vitro Regeneration of Phragmites australis through Embryogenic Cultures

  • Phragmites australis (reed) has received much attention as being one of the principle emergent aquatic plants for treating industrial and civil wastewater. Plant regeneration via plant tissue culture in p. australis was investigated. Three types of callus were identified from seeds on N6 medium plus 4.5 UM 2,4-dichlorophenoxyacetic acid (2,4-D). Yellow compact type showed the best redifferentiation, whereas white compact type and yellow friable were not competent to differentiate into plane. Solid medium culture was better than liquid suspension culture for enhancing callus growth when N6 medium supplemented with 4.5 ${\mu}M$ 2,4-D was used. Phytagel, as a gelling agent, was superior to agar in plant regeneration on N6 medium, supplemented with 9.4 ${\mu}M$ kinetin and 0.54 ${\mu}M$ $\alpha$-naphthaleneacetic acid (NAA). Transfer of the plantlets regenerated from kinetin and NAA-supplemented N6 medium to growth regulator-free MS medium enhanced the further development of the plantlets. Plantlets on subsequently grown to maturity when tansferred to potting soil. The regenerated plants exhibited morphologically normal. The system for plant regeneration of P. australis enables to propagate elite lines on a large scale for water purification in the ecosystem

금잔디 포복경으로부터 식물체 재분화에 있어서 식물생장조절물질과 배지첨가물질의 영향 (Effect of Plant Growth Regulators and Medium Supplements on Plant Regeneration from Stolon of Zoysia matrella Merr)

  • 금잔디(Zoysia matrella (L.) Merr.)의 포복경 조직으로부터 식물체 재분화를 위한 최적 조직배양조건을 확립하기 위하여 온실에서 재배한 식물체의 포복경 절편체로부터 식물체 재분화에 미치는 몇 가지 요인을 조사하였다. 포복경 조직으로부터 식물체 재분화에 있어서 기본배지로는 MS배지가 N6배지나 SH배지에 비해 높은 효율을 보였으며, 배지에 첨가되는 식물생장조절물질로는 0.5 mg/L 2,4-D와 1 mg/L of kinetin을 첨가했을 때 33.3%로 가장 높은 재분화율을 보였다. 또한 배지에 첨가되는 탄소원 종류로는 sucrose를 3% (v/v) 농도로 첨가해 주었을 때 재분화 효율이 향상되었으며, 재분화배지에 첨가되는 항산화물질로는 5 mg/L $AgNO_3$와 10 mg/L cysteine을 동시에 첨가해 주었을 때 40%의 가장 높은 식물체 재분화 효율을 나타내어 재분화효율이 향상되었다. 본 연구를 통하여 확립한 금잔디의 포복경 재분화 시스템은 캘러스 배양기간 없이 단기간 내에 재분화 식물체를 높은 효율로 얻을 수 있음으로써 유전자 형질전환을 통한 잔디의 분자육종에 있어서 유용하게 사용되어질 수 있을 것이다.

Plant Regeneration via Organogenesis from Seed Explants in Red Pepper (Capsicum annuum L.)

  • Efficient plant regeneration has been achieved via organogenesis in the red pepper plant (Capsicum annum L.). Shoots were induced from seed explants of cultivar 'Friendship' on Murashige and Skoog's (MS) basal medium supplemetned with; NAA or IAA, and BAP or zeatin. Seed explants on the medium supplemented with 0.1-0.3 mg/L IAA and 2-5 mg/L zeatin for 2 weeks vigorously formed normal shoots in more than 90% of the explants. When these were transferred to MS medium containing 0.5-1.0 mg/L GA, 90-100% of the shoots have elongated within 1-2 weeks. The elongated shoots rooted in media supplemented with 0.3 mg/L NAA. It was revealed that this method is very rapid and efficient regeneration system for red pepper and regenerated plants can be obtained after only 5-6 weeks of culture.

Plant Regeneration Depending on Explant Type in Chrysanthemum coronarium L.

  • Plant regeneration depending on explant type was inves-tigated with cotyledon, hypocotyl, and leaf explants of garland chrysanthemum (Chrysanthemum coronarium L.) cultured on MS basal medium supplemented with various concentrations of SAP and NAA combination. Among the three different types of explants, hypocotyl explants grown on MS medium containing $1.0{\mu}M\;NAA,\;1.0{\mu}M\;BA\;and\;6{\mu}M\;AgNO_3$ produced the highest adventitious shoots (4.67 per explant). Hypocotyl explants not only produced more vigorous shoots, which regenerated aster than the cotyledon and leaf explants. An efficient root formation was observed in MS medium containing $3\%$sucrose. The concentration of NAA did not show significant effects on root formation. Results from this experiment suggested that hypocotyl explants were efficient for the regeneration of garland chrysanthemum.

  • An efficient plant regeneration system has been developed for figleaf gourd (Cucurbita ficifolia Bouch$\'{e}$), which is exclusively used as a rootstock for cucumber. The protocol is based on results obtained from a series of culture experiments involving different parts of the cotyledons and various media. The culture of cotyledon explants was critical for the enhancement of shoot regeneration frequency. The lower parts of the cotyledon excised at the plumule base were found to display a markedly enhanced production of adventitious shoots compared to other cotyledon regions. Culture in silver nitrate-supplemented Murashige and Skoog (MS) medium was not beneficial for shoot regeneration and suppressed root regeneration. Efficient shoot regeneration was obtained on MS medium containing 1.0 $mg\;l^{-1}$ zeatin and 0.1 $mg\;l^{-1}$ indole-3-acetic acid. Regenerated shoots successfully elongated and rooted in medium containing 0.1 $mg\;l^{-1}$ 1-naphthalene-acetic acid after 10-15 days of subculturing. The plantlets were satisfactorily acclimatized in a greenhouse and grew into normal plants without any morphological alterations.

톨 페스큐의 성숙종자로부터 효율적인 캘러스 배양 및 식물체 재분화 (Efficient Callus Culture and Plant Regeneration from Mature Seed of Tall Fescue (Festuca arundinacea Schreb.))

  • 톨 페스큐의 최적 조직배양 조건을 확립하기 위하여 성숙종자로부터 최적 배발생 캘러스유도조건 및 효율적인 식물체 재분화 체계를 확립하였다. 배발생 캘러스는 6 mg/L 2,4-D와 0.1 mg/L BA가 첨가된 MS배지에서 가장 높은 빈도로 유도되었으며, 식물체 재분화율도 증가시키는 것으로 나타났다. 식물체 재분화는 배발생 캘러스를 1 mg/L 2,4-D와 3 mg/L BA가 첨가된 N6배지에서 배양했을 때 50% 이상의 재분화율을 나타내었다. 기본배지의 종류에 따른 배양효율의 차이는 캘러스유도에는 MS배지가, 식물체 재분화에는 N6배지가 효과적이었다. 품종간 배발생 캘러스의 형성율과 식물체 재분화율은 Kentucky-31 품종이 각각 58.3%와 52%로서 가장 높은 효율을 나타내어 품종 간에 큰 차이를 나타내었다. 탄소원으로는 sucrose를 첨가해주었을 때 식물체 재분화율이 55%로 증가되었다. 본 연구를 통하여 확립된 단기간 고효율 재분화 시스템은 분자육종을 통한 신품종 톨 페스큐의 개발에 유용하게 응용되어질 수 있을 것이다.

Regeneration from Storage Root Disk Culture of Purple Sweet Potato

  • Sweet potato has low regeneration capacity, which is a serious obstacle for the fruitful production of transgenic plants. Simple and rapid regeneration method from storage root explants of purple sweet potato (Ipomoea batatas L.) was investigated. The embryogenic callus was observed from 4 cultivars and its highest rate was induced at 1 μM 2,4-D after 5 weeks of culture. Result revealed that a low concentration of 2,4-D and low light intensity was important factors for embryogenic callus formation. After subculture on medium with 5 μM ABA for 4 days, subsequently, occurred the regeneration of shoots within 4 weeks when these embryogenic callus was transferred onto the MS hormone free medium. Regenerated shoots were developed into platelets, and grown normal plants in the greenhouse. We developed a simple and quickly protocol to regenerate plantlets in storage root explants of purple sweet potato. This regeneration system will facilitate tissue culture and gene transfer research of purple sweet potato.
