Immune-enhancing Activity of Paeonia lactiflora through TLR4-dependent Activation of p38, JNK, and ERK1/2 RAW264.7 Cells

  • Paeonia lactiflora roots (PLR) are a medicinal plant widely used for treating inflammatory diseases. However, PLR has been recently reported to increase the production of proinflammatory mediators and activates phagocytosis in macrophages. Thus, in this study, we tried to verify the macrophage activation of PLR and elucidate its mechanism of action. PLR upregulated the production of proinflammatory mediators and activated phagocytosis in RAW264.7 cells. However, these effects were reversed by inhibition of TLR2/4. In addition, the inhibition of p38, JNK, and ERK1/2 reduced the PLR-mediated production of proinflammatory mediators, and the PLR-mediated activation of p38, JNK, and ERK1/2 was blocked by the TLR4 inhibition. These findings indicate that PLR may activate macrophages through TLR4-dependent activation of p38, JNK, and ERK1/2. These indicate that PLR has immunostimulatory activity. Thus, it is believed that PLR can be used as a functional food agent that enhances the immune system.

Effects of Thermotherapy and Shoot Apical Meristem Culture, Antiviral Compounds for GLRaV-3 Elimination in Grapevines (열처리와 생장점 배양 및 항바이러스제 처리에 의한 포도 GLRaV-3의 무독화효과)

  • Grapevine leafroll-associated virus 3(GLRaV-3) is one of the most severe pathogens for viral diseases found in Korea. This study was conducted to establish the virus-free stock production system for the virus disease control. The effects of thermotherapy, merestem culture and chemotheratpy to eliminate the GLRaV-3 in gratevines were tested. Thermotherapy at 37$\pm$2$^{\circ}C$ for 6∼8 weeks combined with 0.5∼1.0mm size of meristem culture method was the most effective for virus elimination. Thermotherapy alone was not effective. In chemotheratpy, DHT and Amantadine (20, 40mg/L) treatment in medium was more effective than Ribavirin to eliminate the GLRaV-3 in grapevine. However, Ribavirin spraying to potted was not available for virus elimination. Therefore, virus-free stock production system using the thermotherapy combined with shoot apical meristem culture was the most effective in grapevine.

Metabolic Engineering of Medicinal Plants tov Tropane Alkaloid Production (Tropane alkaloid의 생합성과 분자육종)

  • The tropane alkaloids hyoscyamine (its racemic form being atropine) and scopolamine are used medicinally as anticholinergic agents that act on the parasympathetic nerve system. Because they differ in their actions on the central nervous system, currently there is a 10-fold higher commercial demand for scopolamine, in the N-butylbromide form, than there is for hyoscyamine and atropine combined. Several solanaceous species have been used as the commercial sources of these alkaloids, but the scopolamine contents in these plants often are much lower than those of hyoscyamine. For this reason there has been long-standing interest in increasing the scopolamine contents of cultivated medicinal plants. Naturally occurring and artificial interspecific hybrids of Duboisia have high scopolamine contents and are cultivated as a commercial source of scopolamine in Australia and other countries. Anther culture combined with conventional interspecific hybridization also has been used to breed high scopolamine-containing plants in the genera Datura and Hyoscyamus, but without much success. The use of recombinant DNA technology for the manipulation of metabolic processes in cells promises to provide important contributions to basic science, agriculture, and medicine. In this review, I introduce on the enzymes and genes involved in tropane alkaloid biosynthesis and current progress in metabolic engineering approaches for tropane alkaloid, especially scopolamine, production.

Effect of Triiodobenzoic Acid on Broomrape (Orobanche ramosa) Infection and Development in Tomato Plants

  • Branched broomrape (Orobanche ramosa) is a holo-parasitic flowering plant that attaches to the root of its host, green plant, by means of a specialized structure known as haustorium. Following successful contact and penetration on susceptible plant root, complex tissue of Orobanche cells is formed which is known as the tubercle. Newly formed tubercles contain high activity ofindole-3-acetic acid (IAA). Triiodobenzoic acid (TIBA), as an inhibitor of IAA polar transport, was utilized to investigate the supply and requirement of auxin to the developing O. ramosa on tomato plant. There was no significant reduction in the incidence of O. ramosa per pot of different TIBA treatments. However, infection severity in terms of the number of O. ramosa shoots that emerged per plant and number of attachments per plant root system were significantly reduced by 60 % and 45 % on TIBA treated plants, respectively. Histo-logical studies revealed conspicuous delay in the initiation of xylem vessel differentiation inside tubercles of the TIBA treated tomato plants. Also, differentiated vessels showed thinner secondary wall deposition, and improper alignment within bundles inside those tubercles. They were wider and shorter in diameter in comparison to those of untreated plants. These findings were attri-buted to the short supply of IAA required for normal development, and to the xylem vessel differentiation of O. ramosa tubercles on infected tomato. Hence, this parasitic flowering plant seems to depend upon its host in its requirements for IAA, in a source to sink relation-ship.

Estimation of energy self-sufficiency in municipal wastewater treatment plant using photovoltaic power simulated by azimuth and hydrophilic coating (방위각과 초친수코팅에 따른 태양광발전량 시뮬레이션과 하수처리장 에너지자립율 산정)

  • This paper presents energy self-sufficiency simulated in municipal wastewater treatment plants (WWTPs) by adopting solar energy production systems that were simulated by varying azimuth and super-hydrophilic coating on the surface of photovoltaic (PV). Relative to the national average energy consumption in WWTPs, the employment of 100 kW PV system was simulated to achieve 2.75% of energy self-sufficiency. The simulated results suggested that the installation of PVs toward South or Southwest would produce the highest energy self-sufficiency in WWTPs. When super-hydrophilic coating was employed in the conventional PV, 5% of additional solar energy production was achievable as compared to uncoated conventional PV. When 100 kW of PV system was installed in a future test-bed site, Kihyeung Respia WWTP located in Yongin, South Korea, the energy self-sufficiency by solar power was simulated to be 1.77%. The simulated solar power production by azimuth and super-hydrophilic coating will be useful reference for practitioners in designing the solar PV systems in the WWTPs.

Plant Growth and Morphogenesis Control in Transplant Production System using Light-emitting Diodes(LEDs) as Artificial Light Source - Spectral Characteristics and Light Intensity of LEDs - (인공광원으로 발광다이오우드를 이용한 묘생산 시스템에서 식물생장 및 형태형성 제어 - 발광다이오우드의 분광 특성 및 광강도 -)

  • Because of their small mass, volume, solid state construction and long life, light-emitting diodes(LEDs) hold promises as a lighting source for intensive plant production system. Spectral characteristics and light intensity of LEDs were tested to investigate their feasibility as artificial lighting sources for growth and morphogenesis control in transplant production system. Blue, green, and red LEDs had a peak-emission wavelength at 442nm, 522nm, and 673nm, respectively. Their half width defined as the difference between upper and lower wavelength in the intensity equivalent to 50% of the maximum intensity showed 26nm, 41nm, and 74nm, respectively. Photosynthetic photon flux(PPE) at the distance of 9cm under the LEDs array was measured as $235{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ for red, $109{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ for green, and $75{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ for blue LEDs. At the same distance, green LEDs had the illuminance of 13,0001x, nine to ten times higher than those of red and blue LEDs. Red, green, and blue LEDs at a distance of 9cm had the irradiance of $46W{\cdot}m^{-2},\;19W{\cdot}m^{-2},\;8W{\cdot}m^{-2}$, respectively. Light intensity of blue, green, and red LEDs increased linearly in proportion to the magnitude of the current applied to the operating circuit. Thus the light intensity of LEDs was controlled by the applied current in operating circuit.

  • Most of metal cutting and processing companies are going through the difficulties such as frequent downtime and high failure rates by missing times to replace parts of CNC equipment. The reason is that the correct operation times of expensive CNC equipment can't be calculated. In this paper, to solve this problem, we try to construct the intelligent production information system for a case company and analyze its expected effects. The effects are as follows. First, it saves money about 3 million won each year during the useful life of three years by reducing downtime and failure rates, and increasing production rates etc. Second, it makes easily find and fix the cause of faulty through traceability of the production process, and enables to perform a systematic facility management through the calculation of reliable equipment's overall efficiency. This case can be easily applied in similar industries, and will contribute greatly to enhancing the competitiveness of related industries.

  • Using results from an Earth System model, we investigated change in primary production in the East China Sea, under a global warming scenario. As global warming progresses, the vertical stratification of water becomes stronger, and nutrient supply from the lower part to the upper part is reduced. Consequently, so is the primary production. In addition to the warming trend, there is strong decadal to interdecadal scale variability, and it takes a few decades before the warming trend surpasses natural variability. Thus, it would be very hard to investigate the global warming trend using data of several years' length.

Multi-functionality of honey bees for eco-friendly food production (환경친화적 식량생산을 위한 꿀벌의 다원적 가치)

  • Current food system has developed with the agricultural innovation to feed the increasing population of the world, but with high costs such as environmental contamination and inequality with low sustainability. Human has developed long history of mutualistic interaction with honey bee. This manuscript describes the multi-functionality of honey bee for food production. Firstly honey bee produces honey, bee pollen, royal jelly and propolis which are rich in functionality. Second honey bee serves as the main pollinator for crop production which is worth for 28% of total crop production values in Korea. Lastly honey bee can be an alternative meat produciton system with lower energy, carbon costs but higher nutritional security. This manuscript described those parts and discussed the multi-functionality of honey bees for eco-friendly food security pursuing lowered environmental cost and carbon-zero strategies in the climate change era.
