For the survey of viruses infected in peanut cultivated in Korea, peanut seeds and leaves showing viral symptoms were collected from their growing areas. Typical symptoms on virus infected peanut leaves including mosaic, mottle with necrosis, yellowing, stripe or vein banding and stunts were observed. Two viruses isolated from the naturally infected peanuts were identified as Bean common mosaic virus(BCMV-PSt) and Peanut mottle virus(PeMoV) by their host range, immunosorbent elcetron microscopy(ISEM), direct immuno staining assay(DISA), RT-PCR, and intracellural symptoms. Direct negative staining method by electron microscope showed filamentous particles of about 780 m in length as well as inclusion bodies. In ultrathin sections of BCMV-PSt and PeMoV infected tissues, cytoplasmic cylindrical inclusions as well as filamentous virus particles were observed in the cytoplasm of parenchyma cells. ISEM revealed filamentous particles strongly decorated with antiserums of BCMV-PSt and PeMoV Peanut seeds were stained with BCMV-PSt and PeMoV antisera indicating the possibility of seed transmission far these viruses. Seedlings germinated from peanut seeds which reacted with antiserums of BCMV-PSt by DISA showed mild mottle or stripe symptoms while mosaic and necrotic mottle symptoms were observed for PeMoV-positive seedlings. Filamentous particles were strongly decorated with each antiserum under ISEM observation. BCMV-PSt coat protein gene of about 1.2 Kbp was amplified by RT-PCR. Altogether these results indicate that BCMV-PSt is the most prevalent virus infecting peanut in Korea.
One hundred Capsicum accessions were screened for symptomatic response and resistance to Tomato spotted wilt virus-pb1 (TSWV-pb1). Symptom and its severity rating were checked by visual observation at 9, 12, 14, and 45 days after inoculation, respectively. Enzyme-linked immune-sorbent assay was performed all tested individuals on non-inoculated upper leaves after the third rating to indentify viral infection. Leaf curling was predominant in almost susceptible individuals of each accession. Stem necrosis was most frequent in wild species while yellowing in commercial hybrids and Korean land race cultivars. Ring spot, a typical symptom of TSWV, was rarely detected in some of a few accessions. Different levels of resistance to TSWV-pb1 were observed among the tested accessions. High level of resistance was detected in 4 commercial cultivars of Kpc-35, -36, -57, and -62, and 8 wild species of PBI-11, C00105, PBC076, PBC280, PBC426, PBC495, PBC537, and PI201238 through seedling test by mechanical inoculation.

Development and validation of crop models often require measurements of biomass for the crop of interest. Considerable efforts would be needed to obtain a reasonable amount of biomass data because the destructive sampling of a given crop is usually used. The Kinect sensor, which has a combination of image and depth sensors, can be used for estimating crop biomass without using destructive sampling approach. This approach could provide more data sets for model development and validation. The objective of this study was to examine the applicability of the Kinect sensor for estimation of chinese cabbage fresh weight. The fresh weight of five chinese cabbage was measured and compared with estimates using the Kinect sensor. The estimates were obtained by scanning individual chinese cabbage to create point cloud, removing noise, and building a three dimensional model with a set of free software. It was found that the 3D model created using the Kinect sensor explained about 98.7% of variation in fresh weight of chinese cabbage. Furthermore, the correlation coefficient between estimates and measurements were highly significant, which suggested that the Kinect sensor would be applicable to estimation of fresh weight for chinese cabbage. Our results demonstrated that a depth sensor allows for a non-destructive sampling approach, which enables to collect observation data for crop fresh weight over time. This would help development and validation of a crop model using a large number of reliable data sets, which merits further studies on application of various depth sensors to crop dry weight measurements.
At the survey of June, 2002, total 294 dead frees were found in 20 ha of Black pine (Pinus thunbergii) forest located in the western coastal region of Korean peninsula. The dead trees were scattered over the 27 place as groups of about ten trees each. As a result of the field survey on the relationship between the conditions of dead trees and the occurrence of fruiting bodies of Rhizina undulata around the dead and/or dying trees, from June 2002 to August 2004 in the four plots, the occurrence of infected trees was observed as a shape of an irregular concentric circle from the first infected tree and R. undulata was found mainly around the dead tree. Because there was no observation of any other insects and pathogens which would kill trees, the cause of tree death in groups was considered owing to R. undulata. From the analysis of the physical and chemical proper ties of the soil collected from the damaged areas, the pH of soil was between 4.6 and 5.8 and the contents of soil nutrients were very low. Any "fire" trace was not found at all the 27 damaged places in the area, Taean, Chungcheongnam-Do, which are generally known as an important factor to initiate development of the disease. Therefore, further examination is needed to verify precisely about other environmental factors related with the group dying of the Black pines in this area beside 'fire'.
The tribe Neillieae, a small group of about 18 species in the Rosaceae, comprises three taxonomically difficult genera, Neillia, Physocarpus, and Stephanandra. The tribe, characterized by lobed leaves with persistent or deciduous stipules and ovoid, shiny seeds with copious endosperm, is strongly supported as a monophyletic group by a variety of lines of molecular evidence. Due to the high amount of morphological variation across the three genera and the species in tribe Neillieae, conflicting classification schemes and numerous species have been proposed over the past three centuries. However, no comprehensive systematic study of the group, including all species across their geographic ranges, has ever been undertaken. As part of a taxonomic revision of tribe Neillieae, a revision of Physocarpus based on the morphological examination of herbarium specimens, including types, and field observation is presented. Artificial keys, comprehensive nomenclatural treatments, descriptions, distribution maps, and lists of specimens examined are provided. Six species in Physocarpus are recognized. A lectotype is here designated for the following species: Opulaster pubescens, Opulaster ramaleyi, Spiraea opulifolia var. parvifolia, Spiraea opulifolia var. tomentella, Physocarpus michiganensis, and Physocarpus missouriensis.
The in vivo effect of tyrosinase inhibitors in the melanogenesis of gold fish (jet black color) was evaluated by measuring surface color and observing melanin pigment. The fish was firstly cultivated in 0.9% NaCl solution for 1 week to induce melanogenesis, and then, it was transferred to each treatment group containing tyrosinase inhibitor. The fish was grouped into control. food additive group (addition of 5 mM glutathione, 5 mM cysteine, and 1 mM benzoic acid), microbial inhibitor group (addition of culture broth of Aspergillus oryzae in shiitake and glucose medium), and plant extract group (addition of the mixed extracts of green tea, beet, red chicory, and nameko). After 6 days, the fish was anesthetized by electric shock, and color of pectoral region, lateral region, and dorsal fin was measured. Hunter's L and b values of treated group were generally higher than those of control group, indicating that the tyrosinase inhibitors could inhibit the melanogenesis of the fish. Effect of plant extract was apparent, though relatively weak, not because it did not work in vivo, but because a sufficient amount of extract could not be added to fish globes. If a large amount of extract was added, fish gradually died due to a microbial contamination. Microscopic observation of melanin in lateral scale and dorsal fin showed that in the treated groups with tyrosinase inhibitors, the number of melanophore per unit area and the size of one melanophore decreased.
Studies on the anatomical observation of Bamboos distributed in East-South Asia for systematic botany were scarecely reported except for the morphological studies on Bamboos, which was investigated by Dr. Hiroshi Usui, present author and others. Recently it has been certain that it could be hardly classify some species of bamboo family without this work. For this reason the author reported the anatomical studies on Korean bamboos in 1959, and furthermore the author intended to identify the two species of Pleioblastum Simoni Nakai in Korean bamboos by studying the characteristics of internal structure and qualitative assay of free amino acid containg in Pleioblastus Simoni Nakai(1), (2). In view of fact that there is some differences between two species, it is likely to be identifiable completely different species one another. Furthermore, a lot of characteristics surveyed by the author were as follow: It was reassumed that two species cultivated under same conditions for three years were different alternately in the species of Pleioblastus Simoni Nakai. At the same time, these studies could clarify some evolutional processes of bamboo family. On the other hand, it was found that two species had not only the obvious difference in aerenchyma, the size of thickwalled parenchyma and bulliform cell, but also the internode of Pleioblastus Simoni Nakai(1) is longer than the other, the speed of growing is more rapid, the leaf of former is narrow and that of latter is wide. The free amino acids containg in each species were found as 18 kinds, especially Pleioblastus Simoni Nakai (2) had not Histidine in spite of containg in Pleioblastus Simoni Nakia(1). From the characteristics and the experiments described above, it seemed to the author that Pleioblasus Simoni Nakai (1) which was growing at the region of Ulsan in Korea.
Plant mitochondria, irradiated with blue-colored $sunlight(350{\sim}500nm)$ under aerobic and anaerobic conditions, were assayed as to the electron transfer activity of respiratory enzyme system, and compared with those irradiated with orange-colored light(white sunlight minus blue-colored light). The respiratory activity of mitochondria was most seriousely inhibited by illumination with blue-colored light under aerobic condition. Deaeration of mitochondrial suspension resulted in substantial decrease of the photoinhibition by blue-colored light. Meanwhile, orange-colored light demonstrated much less effectiveness-almost ineffectiveness-in causing the inhibition of mitochondrial respiration system. The results of enzymatic assay revealed a strong possibility that FMN in NDH and heme group at least in cytochrome c oxidase, but not FAD in SDH, are the photodynamic sensitizers in mitochondrial inner membrane. Also worthwhile to note is the significant difference from the others of SDH in its photoinhibitory response to the light quality of visible light; that the inhibition of SDH by irradiation was not affected by atmospheric condition and that orange-colored light gave rise to considerable extents of inhibition to the enzyme. This observation was tentatively interpreted in terms of photosensitized reaction not involving molecular oxygen possibly catalyzed by Fe-S centers in the enzyme. The superoxide production and the membrane peroxidation of mitochondria under various treatments also indicated that there was blue-light photodynamic reaction in mitochondria involving active oxygens.
Molecular and cultivation techniques were used to characterize the bacterial communities of biobead reactor biofilms in a sewage treatment plant to which an Aerated Up-Flow Biobead process was applied. With this biobead process, the monthly average values of various chemical parameters in the effluent were generally kept under the regulation limits of the effluent quality of the sewage treatment plant during the operation period. Most probable number (MPN) analysis revealed that the population of denitrifying bacteria was abundant in the biobead #1 reactor, denitrifying and nitrifying bacteria coexisted in the biobead #2 reactor, and nitrifying bacteria prevailed over denitrifying bacteria in the biobead #3 reactor. The results of the MPN test suggested that the biobead #2 reactor was a transition zone leading to acclimated nitrifying biofilms in the biobead #3 reactor. Phylogenetic analysis of 16S rDNA sequences cloned from biofilms showed that the biobead #1 reactor, which received a high organic loading rate, had much diverse microorganisms, whereas the biobead #2 and #3 reactors were dominated by the members of Proteobacteria. DGGE analysis with the ammonia monooxygenase (amoA) gene supported the observation from the MPN test that the biofilms of September were fully developed and specialized for nitrification in the biobead reactor #3. All of the DNA sequences of the amoA DGGE bands were very similar to the sequence of the amoA gene of Nitrosomonas species, the presence of which is typical in the biological aerated filters. The results of this study showed that organic and inorganic nutrients were efficiently removed by both denitrifying microbial populations in the anaerobic tank and heterotrophic and nitrifying bacterial biofilms well-formed in the three functional biobead reactors in the Aerated Up-Flow Biobead process.
This experiment was carried out to examine the effects of culture medium on cell growth of the viola (Viola patrinii DC.) suspension culture. The results are as follows: The greatest cell growth rates were found with MS medium suggesting that this medium could be recommendable for the viola suspension cell culture. When the nitrogen sources (NH$_4$NO$_3$ and KNO$_3$) of MS salts were diluted at half concentrations, the cell growth rates were slightly increased, but when the combined concentration rations of NH$_4$+ and NO$_3$ions were 25 to 75 the greatest cell growth rates were obtained. This result imply that the nitrogen ion sources had slight influence on the rates. Another feature was obtained. This result implys that the nitrogen ion sources had slight influence on the rates. Another feature was that as the concentration of NH$_4$+ ion lowered, the callus color changed to pale yellow with some red spots. The addition of casein hydrolysate (5 g/L) was more effective for the cell growth. On the basis of microscopic observation, the highest cell growth rates were detected during 2-4 weeks culture and after 6 weeks of the culture, some elongated and vacuolated cells were determined.
