Clinical sign and transmission of foot-and-mouth disease in deer, Review (사슴에서의 구제역 증상과 전파 가능성)

  • Foot-and-mouth disease (FMD) commonly infects cloven-hoofed livestock animals such as cattle, pig, sheep, and goat and its clinical signs are well-known. Besides livestock, FMD can be transmitted among cloven-hoofed animals in the wild. FMD mostly affects livestock animals in farms, but, wild animals are likely to play a pivotal role in spreading the disease due to their way of free living. In the case of deer, the clinical signs of FMD vary widely from subclinical to severe infections. Thus, in some deer species, it may be hard to verify clinical signs of FMD. A deer may carry the virus up to 11 weeks after exposure, shedding the virus during the period. However, deer is not considered as a typical host for persistent infection like buffalo, cattle or sheep. In Korea, small-scale livestock farms which have less than 10 animals make up 63.6% of the entire livestock farms. Considering raising environment in deer farms, it is assumed that the risk of virus excretion and consequent transmission of FMD among deers is relatively lower than other cloven-hoofed animals. However, Sika deer and Elk which are typical deer species in Korea would manifest mild to subclinical symptoms upon FMD infection. Therefore, laboratory testing is necessary to confirm FMD in these animals because of difficulty in verifying clinical signs and the risk of virus shedding during inapparent infection.

Efficient Cryopreservation of Porcine Blastocysts produced by In Vitro Fertilization

  • Cryopreservation has been applied successfully in many mammalian species. Nevertheless, pig embryos, because of their greater susceptibility to cryoinjuries, have shown a reduced developmental competence. The aim of this study was to evaluate the survival status of vitrified-warmed porcine embryos. Forced blastocoele collapse (FBC) and non-FBC blastocysts are vitrified and concomitantly cultured in culture media which were supplemented with/without fetal bovine serum (FBS). Porcine vitrified-warmed embryos were examined in four different methods: group A, non-FBC without FBS; group B, non-FBC with FBS; group C, FBC without FBS; group D, FBC with FBS. After culture, differences in survival rates of blastocysts derived from vitrified-warmed porcine embryos were found in group A~D (39.5 (A) vs 52.5 (B) and 54.8 (C) vs 66.7% (D), respectively, p<0.05). Reactive oxygen species (ROS) level of survived blastocysts was lower in group D than that of another groups (p<0.05). Moreover, total cell number of survived blastocysts was higher in group D than that of other groups (p<0.05). Otherwise, group D showed significantly lower number of apoptotic cells than other groups ($2.0{\pm}1.5$ vs $3.2{\pm}2.1$, $2.8{\pm}1.9$, and $2.7{\pm}1.6$, respectively, p<0.05). Taken together, these results showed that FBS/FBC improves the developmental competence of vitrified porcine embryos by modulating intracellular levels of ROS and the apoptotic index during the vitrification/warming procedure. Therefore, we suggest that FBS and FBC are effective treatment techniques during the vitrification/warming procedures of porcine blastocysts.

Isolation of Enterobacteriaceae from bile, urine and intestine in slaughtered pigs and its susceptibility to antibiotics (도축돈의 담즙, 뇨 및 장관에서 장내세균의 분리 및 항생물질 감수성)

  • Seventy-nine strains of Enterobacteriaceae isolated from 117 slaughtered pigs (bile, urine, small intestine, cecum and rectum) in 1995 were examined for biotypes and susceptibility to 19 antibiotics with MicroScan WalkAway 40/96. The obtained results were as follows : 1. Among the twenty-two species isolated from the samples, Proteus mirabilis, E. coli and Enterobacter cloacae were commonly encountered. 2. The distribution frequency of isolates from cecum, small intestine, rectum, bile, and urine was 31(38.8%), 25(31.3%), 18(22.8%), 3(3.7% ), and 2(2.5% ), respectively. 3. A majority of isolates were sensitive to 16 antibiotics, singly or in combination. And these isolates were commonly susceptible to various antibiotics such as Cp, Ts, Azt, Caz, To, Gm, Cfz, Crm, Am and Cfx, in order. Whereas the Salmonella spp was susceptible to Cf, Ti and Pi, and Proteus mirabilis to Imp, Tim, Cft and Cz. Meanwhile, no effect was found to Cf, Ak and Cax. 4. Among the antibiotic resistant strains, a total of 17 reistant patterns was noted End of these Ak Tim 45(57.0%), Ak Am Cf Cfx Cfz Tim 8(10.1%) and Ak Ti Tim 6(7.6% ) were frequently encountered.

  • A diode laser sensor system based on absorption spectroscopy techniques has been developed to measure $CO_2$ concentration and temperature non-intrusively in high temperature combustion environments using a 2.0 ${\mu}m$ DFB(Distributed Feedback) laser. Two optics was fabricated in pig-tail fashion and all optical components were implemented in a single box. The evolution of measurement sensitivity was done using test cell by changing sweep frequency and $CO_2$ concentration. Gas temperature was determined from the ratio of integrated line strengths. Species concentration was determined from the integrated line intensity and the measured temperature. The result show that the system has 2% error in wide operation frequency range and accuracy of $CO_2$ concentration was about 3%. The system was applied to measure temperature and concentration in the combustion region of a premixed $CH_4$ +air triangular flame. The measurement results of gas temperature agreed well with thermocouple results. Many considerations were taken into account to reduce optical noise, etalon effect, beam steering and base line matching problem. The evaluations results and actual combustion measurement demonstrate the practical and applicability for in-situ and real time combustion monitoring in a practical system.

No excessive mutations in transcription activator-like effector nuclease-mediated α-1,3-galactosyltransferase knockout Yucatan miniature pigs

  • Objective: Specific genomic sites can be recognized and permanently modified by genome editing. The discovery of endonucleases has advanced genome editing in pigs, attenuating xenograft rejection and cross-species disease transmission. However, off-target mutagenesis caused by these nucleases is a major barrier to putative clinical applications. Furthermore, off-target mutagenesis by genome editing has not yet been addressed in pigs. Methods: Here, we generated genetically inheritable α-1,3-galactosyltransferase (GGTA1) knockout Yucatan miniature pigs by combining transcription activator-like effector nuclease (TALEN) and nuclear transfer. For precise estimation of genomic mutations induced by TALEN in GGTA1 knockout pigs, we obtained the whole-genome sequence of the donor cells for use as an internal control genome. Results: In-depth whole-genome sequencing analysis demonstrated that TALEN-mediated GGTA1 knockout pigs had a comparable mutation rate to homologous recombination-treated pigs and wild-type strain controls. RNA sequencing analysis associated with genomic mutations revealed that TALEN-induced off-target mutations had no discernable effect on RNA transcript abundance. Conclusion: Therefore, TALEN appears to be a precise and safe tool for generating genomeedited pigs, and the TALEN-mediated GGTA1 knockout Yucatan miniature pigs produced in this study can serve as a safe and effective organ and tissue resource for clinical applications.

Effects of Carcass Weight and Back-fat Thickness on Carcass Properties of Korean Native Pigs

  • Our study analyzed the carcass properties of 170 Korean native pigs in relation to carcass weight and back-fat thickness to provide general data for the production and distribution of high quality pig meat. The 70-74 kg group showed highest yield (73.41%). The ${\geq}80kg$ group showed the highest thickest back-fat (24.13 mm) (p<0.05). The ${\geq}80kg$ group showed the best quality grade (1.00). Back-fat thickness showed significant differences in the weight among groups (p<0.05). The ${\geq}25mm$ group showed the highest carcass weight (75.93 kg). The thickest back-fat group (${\geq}25mm$) showed the highest yield (73.03%). There were significant differences in back-fat thickness among groups (p<0.05), and the ${\geq}25mm$ group showed the highest thickness back-fat (27.60 mm). We found a strong positive correlation between carcass weight and back-fat thickness (r=0.346) as well as meat quality grade (r=0.739). Backfat thickness had a relatively strong positive correlation with meat quality grade (r=0.444). Therefore, there are required to manage the breeding through selection of excellent native species for increasing their carcass weight and enhance meat quality.

Epizootiologic Surveys on Canine Parvovirus Infections of Dogs Kyungbuk West Area (경북서부지역에서의 개 파보바이러스 감염상황 조사)

  • The disease syndrome characterized by the acute vomiting and diarrhea with high mortality had been greatly epidemic in Kyungbuk West Area since March 1990 and it was followed serologically for the classification of the agent. The agent present in feces of dogs associated with this syndrome had characteristic feature in agglutinating pig red blood cells that was specifically inhibited by anti-CPV reference dog serum. This also showed the serological identity with the reference CPV antigen in Hemagglutinating inhibition test. The result obtained were summarized as follows : 1. During 5 years(March. 1990∼September. 1994), 1,470 dogs were investigated on the actual condition of CPV infections. The Infection rate of CPV from dogs was 62.5% and mortality rate was 59.8%. 2. Among 24 fecal samples collected from the dogs with enteric disease, all showed the hemagglutinating activity to porcine erythrocyte ranging from 40 to 5,120 of HA titers. 3. Among 12 sera samples collected from the dogs with enteric disease, all showed the serological identity with the reference CPV antigen from 5 to 5,120 of HI titers. 4. Bacteriologic examination of fecal specimens resulted in the isolation of pathogeric bacteria such as Staphylococcus sp, Streptococcus sp, Escherichia coli and Bacillus. Cultures for salmonella sp and Clostridium remaind negative. 5. The prevalence and identification of internal parasites were determined by fecal examination using the floatation methods. From 20 fecal samples 12(60.0%) were isolated and their species were Toxacara canis, Toxascaris leonina, and coccidium.

Functional Role of a Conserved Sequence Motif in the Oxygen-dependent Degradation Domain of Hypoxia-inducible Factor 1α in the Recognition of p53

  • Hypoxia-inducible factor $1{\alpha}\;(HIF1{\alpha})$ is a transcription factor that plays a key role in the adaptation of cells to low oxygen stress and oxygen homeostasis. The oxygen-dependent degradation (ODD) domain of $HIF1{\alpha}$ is responsible for the negative regulation of $HIF1{\alpha}$ in normoxia. The interactions of the $HIF1{\alpha}$ ODD domain with partner proteins such as von Hippel-Lindau tumor suppressor (pVHL) and p53 are mediated by two sequence motifs, the N- and C-terminal ODD(NODD and CODD). Multiple sequence alignment with $HIF1{\alpha}$ homologs from human, monkey, pig, rat, mouse, chicken, frog, and zebrafish has demonstrated that the NODD and CODD motifs have noticeably high conservation of the primary sequence across different species and isoforms. In this study, we carried out molecular dynamics simulation of the structure of the $HIF1{\alpha}$ CODD motif in complex with the p53 DNA-binding domain (DBD). The structure reveals specific functional roles of highly conserved residues in the CODD sequence motif of $HIF1{\alpha}$ for the recognition of p53.

Effects of Transcription Factor AP2γ on Gene Expression of Desmosome Components in Mouse Embryos

  • Transcription factor called activating enhancer binding protein 2C (AP2-gamma) is found in a variety of species and expressed from oocyte stage onwards, particularly restricted to the trophectoderm. Recent studies demonstrated that ablation of Tfap2c led to failure of tight junction biogenesis, particularly the knock-down embryos of Tfap2c did not form cavity from morula to blastocyst in mouse and pig. We speculated that the Tfa2pc may also be involved in desmosome biogenesis because blastocoel formation is coincident with the establishment of desmosome. To determine this, we depleted Tfap2c injecting siRNA into one-cell zygote and analysed the expression levels of genes that are required for desmosome complex such as PkP2, Pkp3, Dsc2, and Dsg2. We found only Pkp3 was up-regulated in the knockdowned morula embryos. Interestingly, upstream region of Pkp3 had putative Tfap2c binding sites. In conclusion, our results suggest that Tfap2c is not a crucial factor but somehow it might be involved in desmosome biogenesis directly or indirectly via Pkp3.

Survey of foot-and-mouth disease virus structural protein antibody titer in Yeongcheon (영천지역 구제역 바이러스 구조단백질 항체가 조사)

  • Three serotypes (O, A and Asia1) of the foot-and-mouth disease (FMD) vaccine were injected into domestic cloven-hoofed animals in korea after the nationwide spread at the end of 2010. The purpose of this study was survey of FMD virus stuructural protein (SP) antibody titer in Yeongcheon by enzyme-linked immunosorbent assay (ELISA). Total 1,324 samples collected from 89 farms were tested. The overall seroprevalence of FMD virus SP antibodies was 58.8% (778/1,324) The seroprevalence of FMD virus SP antibody varied with species. Results in cattle (over 12 month old) and pig (90 to 130 day old) were 58.8% and 44.9% respectively.
