Improvement of Growth of Potato (Solanum tuberosum L. cv. Dejima) Plants at In Vitro and Ex Vitro and Energy Efficiency by Environmental Control with Growth Stage in Photoautotrophic Micropropagation System (광독립영양 기내 미세증식 시스템에서 생육단계별 환경조절을 통한 감자의 기내 및 기외 생육과 에너지 효율 향상)

  • This study was conducted to evaluate the effect of optimized environment conditions with growth stage in photoautotrophic micropropagation on the growth of potato (Solanum tuberosum L. cv. Dejima) plantlets and energy efficiency. Optimum environment conditions at each stage were decided in our previous study. For the evaluation of optimized environment control, potato plantlets were cultured under four different conditions: photoautotrophic optimum conditions of photosynthetic photon flux density (PPFD) and $CO_2$ levels with growth stage (POG), photoautotrophic constant condition with average PPFD and $CO_2$ levels (PCA), photoauototrophic constant condition with maximum PPFD and $CO_2$ levels (PCM), and photomixotrophic conventional condition with 3% sucrose (PMC) as control. As a result, environment control with growth stage (POG) significantly promoted all the growth characteristics such as the number of nodes and unfolded leaves, shoot height, shoot diameter, and fresh and dry weights of potato grown in vitro. In addition, based on dry weight consumed electricity and $CO_2$ were the lowest in POG suggesting the highest energy efficiency among the treatments. After transferring potato plantlets to greenhouse, the plantlets under POG showed vigorous growth, which was pretty similar with those under PMC. The accumulations of dry matter in POG were 4.7 times in vitro and 3.8 times in greenhouse as much as those in the conventional control (PCM). Thus, we concluded that in vitro environment control with growth stage induced vigorous growth of potato plantlets both in vitro and in greenhouse with less energy consumption.

The Photoautotrophic Culture System Promotes Photosynthesis and Growth of Somatic Embryo-derived Plantlets of Kalopanax septemlobus (독립영양방식 액체대량배양 시스템하에서 배양한 체세포배 유래 음나무 기내묘의 생장과 광합성)

  • A photoautotrophic micropropagation methodology in liquid culture medium has a number of advantages for large-scale propagation of plants. This paper describes an improved system for the mass propagation via somatic embryogenesis of the medicinal plant Kalopanax septemlobus Nakai. Somatic embryo-derived young plantlets of K. septemlobus were cultured either under heterotrophic conditions with sucrose on half-strength MS medium with $30gL^{-1}$ sucrose, under heterotrophic conditions without sucrose, or under photoautotrophic conditions (MS liquid medium without sucrose under forced aeration) for four weeks before transferring the plantlets for acclimatization. Plantlets grown under photoautotrophic conditions had more leaves, higher chlorophyll content, a higher net photosynthetic rate (NPR), and a higher survival rate. The results indicate that the photoautotrophic conditions with a forced ventilation system are effective in enhancing the growth of plantlets and the rate of net photosynthesis. The plantlets grown under photoautotrophic conditions had a high survival rate (92%) upon ex vitro transplantation. Our study shows that autotrophically produced plantlets acclimatize better and sooner upon ex vitro transplantation than conventionally cultured plants.

Mixotrophic Production of Marine Microalga Phaeodactylum tricornutum on Various Carbon Sources

  • We investigated the potential use of various carbon sources (fructose, glucose, mannose, lactose, and glycerol) for culturing Phaeodactylum tricornutum UTEX-640 in mixotrophic and heterotrophic batch cultures. Concentrations of carbon substrates tested ranged from 0.005 M to 0.2 M. P. tricornutum did not grow heterotrophically on any of the C-sources used, but successive additions of organic carbon in mixotrophic growth mode substantially increased the biomass concentration and productivity relative to photoautotrophic controls. The maximum biomass productivities in mixotrophic cultures for glycerol, fructose, and glucose were 21.30 mg/l h, 15.80 mg/l h, and 10.20 mg/l h, respectively. These values were respectively 10-, 8-, and 5-fold higher than those obtained in the corresponding photoautotrophic control cultures. Mannose and lactose did not significantly affect microalgal growth. The biomass lipids, eicosapentaenoic acid (EPA) and pigments contents were considerably enhanced with glycerol and fructose in relation to photoautotrophic controls. The EPA content was barely affected by the sugars, but were more than 2-fold higher in glycerol-fed cultures than in photoautotrophic controls.

Effects of Several Culture Conditions on in vivo Growth and Development in Gerbera hybrida (기내 배양환경이 거베라 유묘의 기외이식 후 생육에 미치는 영향)

  • Propagules grown at different in vitro culture conditions such as heterotrophic, mixotrophic and photoautotrophic conditions were investigated for growth, total photosynthesis ratio and flowering. Survival rate of propagules after transplanting was higher in photoautotrophic propagules than in the heterotrophic and mixotrophic ones. Total photosynthesis was higher plantlets growth in photoautotrophic (154 mg$CO_2$.mgDW$^{-1}$ h$^{-2}$ ) those grown than in mixotrphpic (148 mg$CO_2$.mgDW$^{-1}$ h$^{-2}$ ) and heterotrophic (102 mg$CO_2$.mgDW$^{-1}$ h$^{-2}$ ) 30 days after transplanting into fields. Day to flowering of the plant cultured in photoautotrophic condition was shortened by 7~10 days than those of heterotrophic and mixotrophic ones. Length of the petiole, number of leaves, leaf area and chlorophyll content were also increased.

A Closed Transplant Production System, A Hybrid of Scaled-up Micropropagation System and Plant Factory

  • Photoautotrophic micropropagation systems do not include sugar in the culture media. This characteristic provides advantages to scale up the micropropagation systems comparing photomixotrophic micropropagation systems. A closed, large-scale photoautotrophic micro-propagation for transplant production system has been developed at Chiba University, Japan. New concepts and technologies were adapted to produce high quality transplants at minimum usage of resources, and as scheduled. Newly developed software for production management was used to enhance the efficiency of the transplant production system. Currently, virus-free transplants of sweetpotato (Ipomoea batatas (L.) Lam.) are vegetatively propagated and produced under sterilized conditions in this system. This system can also be used for production of transplants of any other species including horticultural and woody plants with a minimum of modification.

Screening Method for Photosynthetic Electron Transport Inhibitors Using Photoautotrophic Cultured Cells (광학적 자가영양 배양세포를 이용한 광합성 전자전달억제자의 간이검정방법)

  • To investigate a simple and rapid screening method for photosynthetic inhibitory herbicides, responses of tobacco(Nicotiana tabacum L.) and liverwort(Marchantia polymorpha L.) PA(photoautotrophic) cells to various commercial herbicides with different modes of action and leaf extracts of four weed species were compared. PET (photosynthetic electron transport) inhibitory type of herbicides has greater inhibitory effect in liverwort photoautotrophic cells than the photomixotrophic and heterotrophic cultured cells. Similary, PET inhibitory type of herbicides inhibited the oxygen evolution more in liverwort PA cells than the other type of herbicides. Based on oxygen evolution, 60% inhibition was observed by the 10% aqueous extracts of Polygonum hydropiper, while there was 100% inhibition by the 10% methanol extracts of Polygonum hydropiper. This assay gave well correlated results to the Hill reaction data using isolated thylakoids. Thus liverwort photoautotrophic cells might be suitable materials for rapid screening method for photosynthetic inhibitory herbicides.

  • Recently, microalgae which can produce high-value products have attracted increasing attention for biological conversion of $CO_2$. However, low photosynthetic efficiency and productivity have limited the practical use of microalgae. Thus, we developed microdroplet photobioreactor for the analysis of photoautotrophic growth of model alga, Chlamydomonas reinhardtii. $CO_2$ transfer rate was increased by integrating micropillar arrays and adjusting height of microchamber. These results were identified by change of cell growth rate and fluorescence intensity. Lastly, the photoautotrophic growth kinetics of C. reinhardtii in microdroplet photobioreactor were investigated under different $CO_2$ concentrations and light intensities for 96 hours. As a result, microdroplet photobioreactor was efficient platform for isolation and rapid evaluation of microalgal strains which have enhanced productivity of high-value products and growth performance.

Structural Characteristics of Leaves and Carbohydrate Content of Propagules Grown at Different Culture Conditions in Gerbera hybrida 'Beauty' (기내 환경에 따른 거베라 'Beauty' 배양묘 잎의 구조적 특성과 탄수화물 함량의 차이)

  • Microstructure of abaxial leaf surface and carbohydrate content of propagules grown in different culture conditions such as heterotrophic, mixotrophic and autotrophic carbon source were investigated. In the leaves of propagules which were grown in the green house, autotrophic and mixotrophic conditions, wax layer was observed, but in the leaves of the heterotrophic propagules, it was not observed. Size and number of stomata of the leaves in the heterotrophic condition was larger and more numerous than that of autotrophic propagules. Especially, stomata of the leaves in the autotrophic condition was similar to the leaves of plant grown in green house. Carbohydrate content was higher in photoautotrophic condition than that in mixotrophic and heterotrophic culture. Also, Free sugar content showed higher in photoautotrophic propagules than that in mixotrophic and heterotrophic culture. In all the culture conditions, content of glucose were higher than that of other free sugars.

Plastid Transformation of Soybean Suspension Cultures

  • Plastid transformation was attempted with soybean [Glycine max (L.) Merr.] leaves and photoautotrophic and embryogenic cultures by particle bombardment using the transforming vector pZVII that carries the coding sequences for both subunits of Chlamydomonas reinhardtii Rubisco and a spectinomycin resistance gene (aadA). Spectinomycin resistant calli were selected from the bombarded leaves but the transgene was not present, indicating that the resistance was due to mutations. The Chlamydomonas rbcL and rbcS genes were shown to be site-specifically integrated into the plastid genome of the embryogenic cells with a very low transformation efficiency. None of the transformed embryogenic lines survived the plant regeneration process so no whole plants were recovered. This result does indicate that it should be possible to insert genes into the plastid genome of the important crop soybean if the overall methods are improved.

Influence of Organic Carbon Sources on Growth and Lipid Content of Marine Green Alga Dunaliella tertiolecta

  • This study investigated the potential use of various organic carbon sources (glucose, glycerol and acetate) and different concentrations of $CO_2$ for culturing marine microalga Dunaliella tertiolecta. Cell growth and lipid production were monitored under heterotrophic, mixotrophic and photoautotrophic modes of cultivation. D. tertiolecta showed the ability to grow under mixotrophic (acetate and glucose), heterotrophic (glucose) and photoautotrophic condition under high $CO_2$ concentration (15%). With all the organic carbon sources (glucose, glycerol and acetate) tested in this study, 1~5% acetate enhanced cell growth rate and lipid content, while higher concentrations of acetate (10% and 15%) were inhibitory and resulted in cell death.
