Effects of Fish Oil Supplementation to Korean Lactating Women -III. The Effects of Fatty Acid Composition, Plasma Lipid Concentration and Fatty Acid Composition of Plasma Phopspholipids and Erythrocyte of Infants- (한국인 수유부에 어유의 보충 급여 효과에 관한 연구 -III. 영아의 지방산 섭취, 혈장 지질 농도 및 혈장과 적혈구 인지질의 지방산 조성에 미친 영향-)

  • This experiment was conducted to examine the effects of fish oil supplementation with low does on the lipid concentration and fatty acid composition of plasma and the fatty acid composition of plasma phospholipid and erythrocyte of infants. Among 18 breast-fed infants, 6 were in control group and 12 were in fish oil groups. The subjects in fish oil groups were nursed by their mothers who supplemented with fish oil 1.96g/d or 3.92g/d, respectively for 2 weeks from 10 to 12 weeks postpartum. The nursing mothers consumed their usual diets at home. Blood samples were collected at the final day of experiment. There were no significant changes in daily intakes of total lipid, triglyceride, free fatty acid, phospholipid and cholesterol of infants by fish oil supplementation. However, the content of EPA (eicosapentaenoic acid)increased and that of ARA (arachidonic acid) decreaed significantly in plasma PC(phophatidylchline). And also, there were tendencies to increase triglyceride concentration and to decrease cholesterol and phopholipid concentrations of plasma. As the above results, atherogenic index (AI) showed a tendency to decrease, but not significant. DHA (docosahexaenoic acid) and EPA contents in plasma PC and PE (phosphatidylethanolamin) as well as those of erythrocyte tended to increase. In these results, we concluded that fish oil supplementation with low dose to lactating women does not obviously affect of the plasma lipid concentrations and fatty acid composition of plasma PC and PE as well as erythrocyte. However the increase of EPA content of plasma PC and the tendency to increase DHA and EPA contents of plasma as well as erythrocyte membrane indicate that there may be some beneficial effect on infant lipid metabolism of fish oil intake of nutsing mother were increased.

  • Perilla (Perilla frutescens Britton var. Crispa Decasne) cultivated in three geographical areas of Korea, Gwangju, Taegu and Jeju, was analyzed for carbohydrate, lipid, protein, inorganic components and fatty acid composition. Carbohydrate, lipid and protein content of perilla seed ranged from 40 (Taegu) to 44% (Jelu), from 28 (Jeju) to 34% (Taegu) and from 15 (Gwangju) to 16% (Jeju), respectively. The overage contents of potassium, silicate, calcium, phosphate, iron, zinc, magnesium and mangane in the perilla seed varied between $426{\sim}446$, $197{\sim}229$. $124{\sim}136$, $46{\sim}56$, $30{\sim}49$, $42{\sim}45$, $40{\sim}45$, and $30{\sim}36mg%$, respectively and those variations different cultivation areas were not significant except iron. Saponification number, iodine value and acid value of the perilla oil were between $194{\sim}198$, $196{\sim}200$ and $4{\sim}5$, respectively. Those variations among the cultivation areas were not significant. The composition of the perilla oil was observed to be composed of $92{\sim}95%$ of triglyceride, $1.2{\sim}1.3%$ of phospholipid, $1.7{\sim}1.9%$ of unsaponifiables, $0.7{\sim}0.9%$ of free fatty acid, and $1.1{\sim}1.4$ linolenic, linoleic and oleic acid as $55{\sim}56$, $16{\sim}18$ and $16{\sim}20%$ of total fatty acids, but in phospholipid, the content of saturated fatty acids, $12{\sim}24$ of total fatty acid, was higher than that in triglyceride, $8{\sim}19%$ of total fatty acids. The content of saturated fatty acids in sterylester $(14{\sim}19%)$ was higher than that in sterylglycoside ($6{\sim}7%$ of total fatty acids). The variation in fatty acid composition was not significant in the composition of total fatty acid but a significant difference was observed in the composition of phospholipid, in which the content of palmitate ranged from 11.8%(Taegu) to 24%(Gwangju) of total fatty acids. No significant variation was found in the fatty acid composition among the cultivation areas, while a significant difference was observed in phospholipid.

  • Lipid and fatty acid compositions of the free lipids in Panax ginseng (Korea, Japan and China), Panax quinquefolium (America, Canada) and Panax notoginseng (China) were studied by means of silicic acid column chromatography, thin-layer chromatography and gas-liquid chromatography. Free lipid contents were 1.13 to 1.24% in panax ginseng and 0.87 to 1.18% in Panax quinquefolium and 0.39% in panax notoginseng. Neutral lipid fractions were 81.2 to 84.4%, while glycolipid fractions 8.01% to 14.47% and phospholipid fractions 3.49 to 5.74% in free lipid contents. The major components were triglycerides, free sterols, diglyceride, free fatty acids and sterol esters in neutral lipid fractions, sterol glucoside, monogalactosyl diglyceride, digalactosyl diglyceride, esterified steryl glucoside in glycolipid fractions and phosphatidyl glycerol, phosphatide, ethanolamine, phosphatidyl choline in phospholipid fractions. The contents and compositions of neutral lipids and glyclipids were some different among various ginsengs, whereas phospholipids showed relatively similar compositions in the contents. Seventeen fatty acids were analyzed in the four free lipid fractions from the various ginsengs and the main fatty acids were linoleic acid, palmitic acid, oleic acid and linolenic acid. It was found that the amounts of some fatty acids were different among the various ginsengs, but the fatty acid patterns of these ginsengs were on the whole similar.

Gastric Mucosal Fatty Acid Composition of Phospholipids in Gastric Cancer (위암 환자에서 위 점막 인지질 분획의 지방산 조성)

  • Although the age-standardized incidence of gastric cancer has decreased in Korea, it remains the second most common type of cancer. The purpose of this study was to analyze the phospholipid fatty acid compositions of gastric mucosa in gastric cancer. Cancerous mucosa and noncancerous mucosa adjacent to cancerous tissues were obtained from 29 patients who had undergone gastrectomy for gastric adenocarcinoma. Phosphatidylcholine (PC), phosphatidylethanolamine (PE), phosphatidylinositol (PI), and phosphatidylserine (PS) were separated from phospholipids by thin-layer chromatography, and fatty acids were analyzed by gas chromatography. In cancerous mucosa, saturated fatty acids of total phospholipids and stearic acid of PE and PC contents as well as total phospholipids were significantly more abundant than in noncancerous tissues. The ratios of ω6 fatty acid products to linoleic acid of PC, PE, PI, and PS contents as well as total phospholipids were significantly higher in cancerous mucosa than in noncancerous mucosa. Arachidonic acid levels of PE and PI were significantly higher, but the PC level was lower in cancerous mucosa. These results suggest that the characteristic differences in fatty acid compositions of phospholipids and their subfractions shown in gastric cancerous mucosa may be affected by changes in lipid metabolism in gastric carcinogenesis. Further studies on structural and functional changes in phospholipids related to gastric carcinogenesis will be needed.

Effect of Antibiotics on the Phospholipid and Fatty Acid Metabolism of Chloroplast Envelope in Chlorella ellipsoidea (Chlorella ellipsoidea 엽록체막의 인지질 및 지방산 대사에 미치는 항생제의 효과)

  • The effects of amphotericin B (150 $\mu$g/ml) and cycloheximide (10 $\mu$g/ml) on the biosynthesis of phospholipid and their fatty acid composition in chloroplast isolated from Chlorella were analyzed to compare with control. The levels of total lipid, phosphatidylethanolamine (PE), and phosphatidylcholine (PC) in the group treated with antibiotics were decreased. However, the biosynthesis of phosphatidylinositol (PI) was not affected by antibiotics. The major fatty acid in chloroplast envelope was linolenic acid (27.71%) in control and stearic acid (21.59%) in the group treated with amphotericin B. It was showed that the group treated with cycloheximide contained more unsaturated fatty acid than the control.

  • This experiment was carried out to investigate comparison of total cholesterol, lipid and fatty acid composition of breast and thigh from female and male broiler meats. Total lipid and neutral lipid content of female and male broiler breast meats were lower, but phospholipid and glycolipid contents were highter than thigh meats. Unsaturated fatty acid composition of broiler thigh meats were higher than breast meats on neutral and phospholipid, but breast meat was higher than thigh meat on glycolipid. Glycolipid content in total lipid was lower in female than male broiler meat. Contents of palmitic acid in neutral lipid, palmitic stearic linolenic arachidic and arachidonic acid in phospholipid, palmitic and stearic acid in glycolipid were higher than male broiler meat. The highest content of total cholesterol in defatted tissue was thigh tissue of male and undefatted thigh tissue of female.

  • The lipid and fatty acid compositions of neutral lipid, glycolipid and phospholipid were identified and quantified by thin-layer chromatography and gas chromatography. Main constituents of the neutral lipid were triglyceride, free sterol and esterified sterol in the free lipid, and free fatty acid and monoglyceride in the bound lipid. In the outer part, there existed 25.7% triglyceride in the free lipid and it was not nearly found in the bound lipid. Main constitutents of the glycolipid were digalactosyl diglyceride and esterified steryl glycoside in the free lipid, and digalactosyl diglyceride in the bound lipid. Free lipid didn't contain trigalactosyl diglyceride but bound lipid contained 2.0% of it. Main constituents of the phospholipid were lysophosphatidyl choline, phosphatidyl inositol, phosphatidyl choline, phosphatidyl glycerol and phosphatidyl ethanolamine in the free lipid, and phosphatidyl choline and phosophatidyl inositol in the bound lipid. The predominant fatty acids of three fractions, neutral lipid, glycolipid and phospholipid showed almost the same pattern as that of the total free and bound lipids. The content of palmitic acid was relatively higher in the polar lipids(glyco and phospholipid). Therefore, saturated fatty acid ratio of polar lipid was higher than that of neutral lipid. found lipid contained more saturated fatty acids as compared with the free lipid.

High Fat Diet or Exercise Training Alters Hepatic Total and Phospholipid fatty Acid Compositions in Rats (고지방식이 및 운동훈련이 흰쥐 간조직의 총지방산 및 인지질 지방산패턴에 미치는 영향)

  • Effects of high fat diet and/or endurance exercise training on hepatic total and phospholipid(PL) fatty acid compositions were evaluated in rats fed one of the following diets for 31 days. control diet(CD, 5 wt% corn oil) or high fat diet(HFD, 35 wt% corn oil). Half of the rats in each group were exercise-trained regularly on a treadmill for 90 minutes/day during the entire feeding period. Total and PL fatty acid compositions of hepatic lipid extracts were determined by a gas-liquid chromatograph),. Endurance exercise training did not change the daily food intake, but significantly reduced body weight gain and feed efficiency ratio of rats, which were most prominent in animals fed HFD. Exercise training did not significantly change the percentages of ∑saturated fatty acids (SFA) and ∑polyunsaturated fatty acids(PUEA), but decreased the percentage of ∑monounsaturated fatty acids(MUFA) in hepatic total fatty acids, which might be associated with the decrease in (equation omitted) 9-desaturation index of hepatic total fatty acid metabolism. Exercise training significantly lowered the percentages of 16 : 0 and 22 : 5$\omega$3, and increased the percentages of 20 : 1 and 20 : 3$\omega$3 in both total and PL fatty acid compositions in rat liver. Both total fatty acid and PL fatty acid compositions of rat liver responded more sensitively to changes in dietary fat content than to endurance exercise training in this study. Feeding HFD, whoch contains high level of linoleic acid(LA, 18 : 2$\omega$6), significantly decreased the percentages of ∑SFA and $\Sigma$MUFA, and increased the percentages of ∑PUFA and ∑$\omega$6 fatty acids of hepatic total fatty acids. Hepatic total fatty acid composition was affected by dietary fat content and dietary fatty acid composition more sensitively than those found in hepatic PL fatty acid composition. HFD significantly decreased most of desaturation indices, while exercise training significantly decreased elongation index(20 : 5$\omega$3⇒22 : 5$\omega$3) of hepatic total and PL fatty acid metabolism in rats.   • Total lipid content, lipid class and fatty acid composition of the Antarctic krill Euphausia superba collected from the water of King George Island, Antarctica during austral summer of 1997-1998, were investigated. The overall lipid content of 5. superba was 72 mg/g dry mass similar to the reported values for most temperate species. The neutral lipid of E. superba was 29% of the total lipid and that of phospholipid was 71%. The majority of neutral lipid was triacylglycerols(31.6% of neutral lipids) while phosphatidyl choline (44% of phospholipid) was the most abundunt in phospholipids. The quantitative composition of the fatty acid in E. superba show consisting mostly of the saturated 16:0 (16.6% -22.1%), along with the polyunsaturated 20:5(n-3) (20.6% -22.1%) and 22:6(n-3) (14.9% -16.9%) acids.

  • The concentration of free fatty acids and fatty acid composition as well as cholesterol supersaturation in bile may be an important factor in the gallstone formation. Therefore, we simultaneously determinded 23 fatty acids in bile by selected ion monitoring (SIM) method of gas chromatography-mass spectrometry (GC-MS). Biliary fatty acids were extracted by aminopropyl column and the extracts with (phospholipid fraction) or without (free fatty acid fraction) alkaline hydrolysis of phospholipid were derivatized with MSTFA/TMCS (N-methyl-N-trimethylsilyl-trifluoroacetamide/trimethylsilylchloride) mixture in order to be detected on the GC-MS. The recovery range of this method was 61.1-99.0% and the RSD value of within-a-day and day-to-day test were 3.1-25.6% and 3.8-27.0%, respectively. Using this method, biliary profile was investigated in the bile of normal controls and patients with gallstones. The amounts and their distribution of free and phospholipid fatty acids showed different pattern between normal subjects and patients.
