Synthesis of the 7,8-Dihydro-7-deazapurine Derivatives and Their Antibiotic Activity

  • The cis- and trans-diastereomers of the 7,8-dihydro-7-deazapurine derivatives were synthesized from the corresponding diastereomers of 4-tran5-cyano-2-methyl-3-phenyl-5-oxopyrrolidine (5), which were reduced from the 2-cis- and 2-tran5-diastereomers of 4-trans-cyano-2-hydroxymethyl-3-phenyl-5-oxopyrrolidine (2) via tosylation, iodination and following elimination respectively. The prepared cis- and ttans-diastereomers of 6-amino-2-mercapto-8-methyl-7-phenyl-7,8-dihydro-7(9H)-deazapurine (8) were transferred to the corresponding 2-methylthio-diastereomers 9 and following desulfurization with Raney-nickel leaded to the cis and trans-diastereomers of 6-amino-8-methyl-7-phenyl-7,8-dihydro-7(9H)-deazapurine (10), respectively. The synthesized 7-deazapurine derivatives were tested for their antibiotic activity by the serial two-fold dilution method.

Ring Transformation Reaction of Thiazolo[3,2-$\alpha$][1,3]diazepinium-Betaine (티아졸로[3,2-$\alpha$[1,3]디아제핀-베타인의 고리변환 반응)

  • 3-Phenyl-5, 6, 7, 8-tetrahydrothiazolo[3, 2-$\alpha$][1, 3]diazepine, which has pharmaceutical activities, was reacted with phenyl isothiocyanate to give 3-phenyl-9-phenyl(thiocarbamoyl)-5, 6, 7, 8-tetrahydrothiazolo[3, 2-$\alpha$][1, 3]diazepi nium-betaine. New biheterocyclic compounds were prepared by the ring transformation reaction of the above reactive betaine with $\alpha$-haloesters and $\alpha$-haloketones such as ethyl bromoacetate, methyl bromoacetate, chloroacetone, and 4'-methoxyphenacyl bromide, respectively. In each ring transformation reaction, two major products supposed to be geometrical isomers were obtained.

  • Among the efforts to increase the efficiency of organic light-emitting device (OLED), there is a way: doping phosphorescent materials. As a phosphorescent material, complexes of heavy transition metal, platinum, were synthesized. $Cl^-$ ion and phenyl group were used as ancillary ligands with 2-(2-pyridyl)benzimidazole (pbi) as a chromophore. The complexes were analysed by FAB-mass spectrometer and absorption and emission spectra were obtained. A phenyl group was able to shift the emission band of the complex even if it's not a chromorphore.

Recognition of the Importance of Imidazolidinone Motif for Cytotoxicity of 4-Phenyl-1-arylsulfonylimidazolidinones Using Thiadiazolidine-1, 1-Dioxide Analogs

  • For probing the importance of planarity of imidazolidinone motif of 4-phenyl-1-(N-acylindoline-5-sulfonyl)imidazolidinones 1 for their cytotoxicity, 4-phenyl-1-(N-acylindoline-5-sulfonyl)[1,2,5]thiadiazolidine-1,1-dioxides 2 were prepared and their cytotoxicity were measured against human lung carcinoma (A549), human colon carcinoma (COLO205), human ovarian cancer (SK-OV-3), human leukemic cancer (K562), and murine colon adenocarcinoma (Colon26) cell lines in vitro. Although only carbonyl moiety of imidazolidinone ring was replaced with sulfonyl group, compounds 2 do not show any activity against all five cancer cell lines unlike 1. Therefore the planarity of imidazolidinone ring of 1 should be an important factor for their cytotoxic activity.

  • In this study, 6-[( 4-arylidene-2-phenyl-5-oxoimidazolin-1-yl)phenyl]-4,5-dihydro-3(2H)-pyridazinone and 4-[(4-arylidene-2-phenyl-5-oxoimidazolin-1-yl)phenyl]-1(2H)-phthalazinone derivatives were synthesized by reacting 6-(4-aminophenyl)-4,5-dihydro-3(2H)-pyridazinone or 4-(4-aminophenyl)-1(2H)-phthalazinone compound with different 4-arylidene-2-phenyl-5(4H)-oxazolone derivatives. The vasodilator activities of the compounds were examined both in vitro and in vivo. Some pyridazinone derivatives showed appreciable activity.

  • The main object of this experiment is to provide a systematic approach to the reaction mechanism in the photolysis of 5-phenyl-1,2,3,4-tetrazole during which the formation of C-phenyl-nitrile-imine of 1.3-dipole was expected. So the occurrence of 1,3-dipole-addition was examined but not observed despite the formation of nitrile-imine. 3,6-diphenyl-1,2,4,5-tetrazine (IV); 3,6-diphenyl-1,4-dihydro-1,2,4,5-tetrazine (III); 3,5-diphenyl-1,2,4-triazole; 4-amino-3,5-diphenyl-1,2,4-triazole; benzonitrile; ammonia and nitrogen were isolated as final products of this reaction.

Development of Recombinant Saccharomyces cerevisiae Expressing Epoxide Hydrolase for the Preparation of Chiral Phenyl Oxirane (광학활성 Phenyl Oxirane 제조용 유전자 재조합 생촉매 개발)

  • Epoxide hydrolase (EH) gene from Aspergillus niger #33 was cloned from cDNA library generated by reverse transcriptase-polymerase chain reaction (RT-PCR). The nucleotide sequence analysis revealed that the deduced amino acid sequence was almost similiar to that of A. niger LCP521 previously reported. The cloned EH gene was transformed into Saccharomyces cerevisiae and expressed by addition of galactose. The recombinant S. cerevisiae showed hydrolytic activity toward racemic phenyl oxirane substrate based on chiral GC analysis, and can be used as a potential biocatalyst for the preparation of chiral phenyl oxirane.

  • In view of the growing medicinal importance of pyrazole and its derivatives, the single crystal X-ray diffraction study was carried out for the potential active 2-Benzyl-3-[3-(4-bromo-phenyl)-1-phenyl-1H-pyrazol-4yl]-4,6-dioxo-5-phenyl-octahydro-pyrrolo[3,4-C]pyrrole-1-carboxylic acid ethyl ester ($C_{37}H_{31}BrN_4O_4$, H2O). In the title compound are two molecules exist in the asymmetric unit. It crystallizes in the monoclinic space group $P{\hat{i}}$ with unit cell dimension $a=13.361(18){\AA}$, $b=13.424(17){\AA}$ and $c=21.649(2){\AA}$ [${\alpha}=80.745(9)^{\circ}$, ${\beta}=79.770(10)^{\circ}$ and ${\gamma}=60.788(6)^{\circ}$]. The pyrazole ring adopts planar conformation. The sum of the bond angles at nitrogen atom of the pyrazole ring indicates the $Sp^2$ hybridized state. The crystal structure is stabilized by intramolecular C-H...O hydrogen bond interaction.

  • A series of N-{2,4-dichloro-5-(3-pyrazolyl)Phenyl}imides 7 and 8 was prepared and evaluated their herbicidal activities. Those compounds in which either carboxylate or carboxamide moiety is on pyrazole moiety 7c-71 showed no herbicidal activity whereas, the compounds in which trifluoromethyl group is substituted in pyrazolyl moiety showed moderate herbicidal activities against barnyardgrass, monochria and flat-sedge under paddy conditions with good rice tolerance at rate 63-250 g/ha.
