Bacillus subtilis 발효대두박의 사료 내 첨가가 흰다리새우(Litopenaeus vannamei)의 성장, 면역력 및 질병저항성에 미치는 영향 (Effect of Fermented Soybean Meal by Bacillus subtilis in Pacific White Shrimp Litopenaeus vannamei Diets on Growth Performance, Innate Immunity and Disease Resistance)

  • A feeding trial was conducted to evaluate the effect of dietary supplementation with fermented soybean meal by Bacillus subtilis (BFSBM) on growth performance, feed utilization, innate immunity, water quality, and disease resistance of Litopenaeus vannamei. A basal fish meal-based diet was regarded as a control and two other diets were prepared by replacing 30% fish meal with either soybean meal (SBM) or BFSBM. Triplicate groups of shrimp ($0.80{\pm}0.010g$) were fed one of the experimental diets for 6 weeks. No differences were found in final body weight, feed conversion ratio, or survival among all dietary treatments after the feeding trial. Shrimp fed the BFSBM diet had significantly higher phagocytic and phenoloxidase activities than those of shrimp fed the SBM diet. Total ammonia concentrations in culture water during a zero water exchange test tended to decrease in tanks where BFSBM diet was provided. Disease resistance of shrimp to Vibrio harveyi tended to increase in the BFSBM group during 14 days of challenge compared to that in the control (CON) and SBM groups. These results show that dietary supplementation with BFSBM can enhance the innate immunity of Pacific white shrimp and replace at least 30% of dietary fish meal.

주걱송편버섯(Pycnoporus cinnabarinus SCH-3)의 Laccase에 의한 Acridine 산화 (Oxidation of Acridine by Laccase of Pycnoporus cinnabarinus SCH-3)

  • Acridine은 fungal laccase의 기질이 아님에도 불구하고 acridine을 Pycnoporus cinnabarinus SCH-3 배양액에 첨가했을 때 acridone으로 산화되었다. P. cinnabarinus SCH-3균주는 배양 중에 다량의 laccase와 3-hydroxyanthranilic acid (3-HAA)와 cinnabarinic acid (CA)를 생성했다. 정제된 laccase와 acridine을 완충용액에서 직접 반응시켰을 때 acridine은 변화되지 않았다. 그러나 laccase의 기질인 2,2'-azino-bis-(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS)나 3-HAA를 laccase와 acridine 혼합액에 첨가했을 경우에는 acridine이 acridone으로 산화되었다. 특히 ABTS 첨가구는 3-HAA 첨가구보다 acridine 산화율이 2배 이상 높았다. 한편 정제된 laccase와 3-HAA를 완충액에서 반응시켰을 때 3-HAA는 CA로 전환되었다. 이와 같은 실험결과들은 P. cinnabarinus SCH-3의 laccase가 배양중에 생산된 3-HAA를 매개체로 사용하여 acridine을 acridone으로 산화하고 CA는 laccase에 의하여 3-HAA로부터 합성됨을 나타낸다.

몇 가지 PBTs (Persistent, Bioaccumulative, Toxic Chemicals)가 생태계 곤충에 미치는 영향

  • Pollutants that are persistent, bioaccurnulative, and toxic have been linked to numerous adverse effects in human and animals, PBTs include heavy metals, polychlorinated biphenyls (PCBs), dioxins, polycyclic aromatic compounds (PACs) in addition to pesticides. This study focuses on toxic effects of the PBTs except pesticides on insects. Eight PBTs were selected from subgroups: three heavy metals (Pb, Hg, and Cd), two PCB mixtures (Aroclor mixtures 1 and 2), 2,3,7,8-tetrachlorodibenzo-p-dioxin, two monophenols (4-octylphenol and 4-nonylphenol), and tetrabutyltin, Beet armyworm, Spodoptera exigua, was used as test target insect species. Three physiological markers (metamorphosis, immune reaction, and follicle patency) were assessed in each exposure to different doses of the PCBs. Heat-shock proteins as molecular markers were also analyzed in response to the PCBs. All tested PBTs were toxic to metamorphosis from larvae to pupae when they were applied with diet. Two PCB mixtures were the most toxic compounds in this assay by giving significant toxicity at 0.005 ppm, while others had from 10 to 1000 ppm. Dioxin (0.1 ppb), tetrabutyltin (0.1 ppb), Pb (10 ppb), and Hg (0,01 ppb) were potent to inhibit immune reactions analyzed by inducing phenoloxidase activity and blocked phospholipase $A_2$ enzyme, Tetrabutyltin and dioxin significantly induced follicle cell patency, but their effects were lower than that of endogenous juvenile hormone, Dioxin, Pb, Hg, and Cd could induce the expression of heat shock proteins that were detected by immunoblotting against human HSP70 monoclonal antibody. HSP78 and HSP80 were upregulated in response to the PBTs. This expression was detected from the fat body and epidermis at as fast as 4h after injection. All these results clearly suggest that PBTs give significant ecotoxicity to insects that are valuable organisms in our environment.

감귤박 및 감태추출물의 사료첨가제 급여에 따른 둥근전복 (Haliotis discus discus)의 성장 및 생리적 변화 (Effects of dietary supplementation with citrus pomace and Ecklonia cava residue on the physiological changes and growth of disk abalone, Haliotis discus discus)

  • Here, we report the physiological changes and growth in disk abalone, Haliotis discus discus, in relation to dietary supplementation with citrus pomace (CP) 6%, Ecklonia cava residue (ECR) 6%, and CP + ECR (3% + 3%). The composition and nutrient content, survival rate and growth rate were measured 0, 4, 8 and 12 weeks after feeding the supplemented diets of CP and/or ECR. Moreover, the experiment of low salinity stress (25psu) for environmental resistance was examined for a period of 48 hours after feeding the supplemented diets for 12 weeks. The activities of superoxide dismutase (SOD), catalase (CAT), lysozymes, respiratory burst, and phenoloxidase were measured. The moisture content and crude protein condition of the body were increased with the addition of ECR only (P<0.05). We observed higher levels of survival in the experimental group compared with the control group. Moreover, the growth disk abalone that were fed a diet containing ECR was higher compared with the control group. However, the growth of abalone fed a diet containing CP was similar to the control group. With a rearing condition of low salinity stress, survival rate and lysozyme activity were increased in the ECR group compared with the control group. Dietary ECR reduced the level of CAT activity to approximately 30% of the control, however the level of CAT activity in the ECR group was similar to the start level of the previous stress. These results suggest that dietary ECR gives rise to an enhanced immunity in disk abalone, as a result of the decrease in CAT and lysozyme activity in particular. Accordingly, the growth and survival rate were increased by feeding an ECR-supplemented diet in the rearing of disk abalone, Haliotis discus discus.

  • This study was conducted to determine the supplemental effects of two insect meals, mealworm (MW) and black soldier fly (BSF), with high or low lipid levels in diets, on Pacific white shrimp Litopenaeus vannamei. Sardine and tuna by-product meals were used as the fish meal source in a control (Con) diet. The fish meals were replaced with MW, defatted MW (deMW), BSF or defatted BSF (deBSF), respectively. The shrimp (body weight: 0.47 g) were stocked into 20 acryl tanks (215 L) and fed the diets six times a day. After 45 days of the feeding trial, the shrimp that were fed insect meals had significantly higher phenoloxidase and superoxide dismutase activities than the shrimp fed Con diet. The gene expressions of prophenoloxidase, crustin and penaeidine-3c in shrimp hepatopancrease were also higher in shrimp that were fed the insect diets, regardless of defatting than those in shirmp that were fed Con diet. The survival against Vibrio parahaemolyticus was higher in shrimp that were fed the diets containing defatted insect meals than in shrimp that were fed Con diet. These results indicate that MW and BSF, regardless of lipid levels, could be good protein sources for the enhancement of innate immunity and anti-oxidant capacity of the shrimp.

흰다리새우(Litopenaeus vannamei) 사료 내 동애등에 유충과 갈색거저리 유충의 어분대체효과 (Replacing Fish Meal with Black Soldier Fly Larvae and Mealworm Larvae in Diets for Pacific White Shrimp Litopenaeus vannamei)

  • This study was conducted to partially replace fish meal with black soldier fly (BSF) larvae or mealworm (MW) larvae in the diet for Pacific white shrimp Litopenaeus vannamei. A tuna by-product meal (27%) was used as the protein source in a control (Con) diet, which was replaced with 20%, 40%, or 60% BSF (designated as BSF20, BSF40, and BSF60, respectively), or MW (MW20, MW40, and MW60, respectively). The shrimp (average body weight, 0.09 g) were randomly stocked in 28 acrylic tanks and quadruplicate groups were fed one of the experimental diets six times daily. After 57 days of the feeding trial, growth and feed efficiency were significantly higher in shrimp that were fed the BSF40 and BSF60 diets than those in shrimp fed the Con diet. Phenoloxidase, superoxide dismutase, glutathione peroxidase and gene expression of crustin were significantly higher in shrimp fed BSF or MW diets than those in shrimp fed the Con diet. The results indicate that BSF or MW could be used as a fish meal replacement or as a functional protein source in diets and can help improve the growth, feed utilization, innate immunity and antioxidant capacity of Pacific white shrimp.

Toxic effects of ammonia on the survival, growth, and oxidative and immune responses in the Pacific abalone, Haliotis discus hannai

  • The accumulation of ammonia, a consequence of intensive aquaculture activity, can damage cultured animals. We analyzed the survival rates, growth, antioxidant responses, and immune responses of abalones, Haliotis discus hannai (mean shell length 70.2 ± 4.9 mm; mean body weight 36.9 ± 3.6 g), biweekly for four weeks, to determine the accumulated concentrations of ammonia (0, 0.1, 0.2, 0.4, 0.8, and 1.6 mg/L). In our biweekly measurements, a survival rate of ≥ 95% was observed for all concentrations of ammonia. The specific growth rate (SGR) decreased at ≥ 0.8 mg/L (p < 0.05), but the daily increment in shell length (DISL) showed no significant change (p < 0.05). Regarding antioxidant responses, the superoxide dismutase (SOD) activity of the hepatopancreas was increased at ammonia concentrations over 0.8 and 0.2 mg/L (p < 0.05) at two and four weeks, respectively, and the SOD activity of the gills increased at concentrations over 0.4 and 1.6 mg/L (p < 0.05) at four weeks. The catalase (CAT) activity of the hepatopancreas and gills increased at ammonia concentrations > 0.8 mg/L (p < 0.05). Phenoloxidase (PO) activity increased at ammonia concentrations over 0.8 mg/L (p < 0.05), and Lysozyme (LZM) increased at concentrations over 0.8 and 0.4 mg/L (p < 0.05). Overall, our findings indicated that ammonia concentrations over 0.8 mg/L in seawater might damage the SGR, antioxidant responses, and immune responses in H. discus hannai. Our findings suggest the necessity of improvement or reinstallation for abalone aquaculture systems and can be used to assess the toxic effects of ammonia on H. hannai.

사료 내 Silymarin의 첨가가 흰다리새우(Litopenaeus vannamei)의 성장, 사료효율, 비특이적 면역력 및 항산화력에 미치는 영향 (Effects of Dietary Silymarin on Growth Performance, Feed Utilization, Innate Immunity and Antioxidant Capacity of Pacific White Shrimp Litopenaeus vannamei)

  • This study was conducted to evaluate the effects of dietary silymarin supplementation on the growth performance, feed utilization, innate immunity and antioxidant capacity of Pacific white shrimp Litopenaeus vannamei. Four experimental diets were formulated to contain 0, 0.025, 0.05 and 0.1% silymarin (designated as Con, S0025, S005 and S01, respectively). Triplicate groups of shrimp (initial body weight: 0.70 g) were fed each of the diets for 6 weeks. After the feeding trial, weight gain and specific growth rate were significantly higher in silymarin-supplemented groups compared to Con group. Dietary silymarin significantly enhanced protein efficiency ratio of S01 group and reduced feed conversion ratio of S005 and S01 groups. Phenoloxidase and anti-protease activities were significantly higher in S01 group compared to Con group. Glutathione peroxidase and catalase activities were significantly higher in silymarin-supplemented groups compared to those of Con group. The results of this study indicate that dietary silymarin could improve the growth performance, feed utilization, innate immunity and antioxidant capacity of Pacific white shrimp. The optimum level of silymarin in diet for Pacific white shrimp is suggested to be ≥0.025%.

Effects of bioflocs on immune responses of Fleshy shrimp, Fenneropenaeus chinensis postlarvae and adults as related to the different feeding abilities

  • The present study compared the structure of mixilliped of Fenneropenaeus chinensis between the larval and adult stage and investigated the effect of the structural difference on the immunity of F. chinensis. A fourteen day and a one-month long culture trial were conducted each with postlarvae and adults of F. chinensis in the biofloc, mixed water (50% biofloc:50% clear seawater) and seawater control. Immune-related genes mRNA expressions of postlarvae was analysed by quantitative reverse transcription polymerase chain reaction (qRT-PCR). And the analysis of adult stage immunity was carried out using phenoloxidase (PO) enzyme activation in haemocyte. In the postlarvae, the final body weights were 51.43 and 58.47 mg for the biofloc water and the control seawater, respectively. On the other hand, the final body weights of the adults were significantly different between biofloc water and seawater. The survival rate showed the opposite trend to the growth rate. Immune related genes mRNA expression levels in the postlarvae in the biofloc water were significantly lower than those in the seawater. While, the adult stage showed significantly higher PO enzyme activations in the biofloc water than in the seawater with the PO enzyme activation increasing proportionally to the biofloc concentration. This result is considerably explained by the observations of setal morphological structures of the third maxilliped: postlarvae have short serrulated setae that compose the small 'net' structure while adults had long and dense plumose setae. It is understood that the morphological difference of the maxilliped structure resulted in the different feeding abilities in the postlarvae and the adult F. chinensis to use bioflocs as food source.

  • This study investigated the effects of dietary supplementation with Sargassum horneri celluclast hydrolysate (SHCH) on the growth performance, feed utilization, non-specific immunity and antioxidant capacity of the Pacific white shrimp Penaeus vannamei. Six diets were prepared by SHCH supplemention levels of 0, 5, 10, 20, 40 and 80 g/kg (designated as Con, S5, S10, S20, S40 and S80, respectively). A total of 540 shrimp were randomly distributed in triplicate across 18 tanks and fed one of the diets for five weeks. Weight gain did not differ significantly among the groups except for S80. The feed conversion ratio was significantly lower in S10 than in S80. Phenoloxidase and nitro-blue tetrazolium activities were significantly higher in S5 than in Con. The prophenoloxidase gene was significantly upregulated in S10 compared to that in Con. The gene expression of crustin, glutathione peroxidase, lipopolysaccharide and β-1,3-glucan-binding protein were significantly upregulated in S5 compared to Con. The superoxide dismutase gene was significantly upregulated in S20 and S80 compared to Con. Supplementing shrimp diets with up to 8% of SHCH did not impair non-specific immunity or antioxidant capacity. However, based on growth results, supplementation up to 4% was considered appropriate.
