Activation of Macrophages by Exopolysaccharide Produced by MK1 Bacterial Strain Isolated from Neungee Mushroom, Sarcodon aspratus

  • Background: The MK1 strain, a novel bacterial isolate from soft-rotten tissue of the Neungee mushroom, produces copious amounts of exopolysaccharide (EPS) in a dextrose minimal medium. This study examined the molecular characteristics and immunomodulatory activity of MK1 EPS. Methods: The EPS in the culture supernatant was purified by cold ethanol precipitation, and characterized by SDS- PAGE/silver staining and Bio-HPLC. The immunomodulatory activities of the EPS were examined using the mouse monocytic cell line, RAW 264.7 cells. Results: The molecular weights of the purified EPS were rather heterogeneous, ranging from 10.6 to 55 kDa. The EPS was composed of glucose, rhamnose, mannose, galactose, and glucosamine at an approximate molar ratio of 1.00 : 0.8 : 0.71 : 0.29 : 0.21. EPS activated the RAW cells to produce cytokines, such as TNF-${\alpha}$ and IL-$1{\beta}$, and nitric oxide (NO). EPS also induced the expression of co-stimulatory molecules, such as B7-1, B7-2 and ICAM-1, and increased the phagocytic activity. The macrophage-activating activity of EPS was not due to endotoxin contamination because the treatment of EPS with polymyin B did not reduce the macrophage-activating activity. Conclusion: The EPS produced from the MK1 strain exerts macrophage-activating activity.

Induction of Differentiation of the Human Histocytic Lymphoma Cell Line U-937 by Hypericin

  • Hypericin, a photosensitizing plant pigment, was found to be a potent inducer of differentiation of human myeloid leukemia U-937 cells. At a concentration of $0.2{\mu}M$, hypericin exhibited 50% growth inhibition. An effect on cell differentiation by hypericin was assessed by its ability to induce phagocytosis of latex particles, and to reduce nitroblue tetrazolium (NBT). Approximately 51% of $0.2{\mu}M$ hypericin-treated cells were stained with NBT and 63% showed phagocytic activity. In order to establish whether hypericin induces differentiation of U-937 cells to macrophage or granulocyte, esterase activities and cell sizes were measured. When U-937 cells were treated with $0.2{\mu}M$ and $0.15{\mu}M$ of hypericin, the .alpha.-naphthyl acetate esterase activity was increased by 38.4% and 48.1%, respectively, but naphthol AS-D chloroacetate esterase activity was not influenced. The size of hypericin-treated cells in terms of cell mass was larger than that observed in untreated cells as determined by flow cytometry. Protein kinase C (PKC) inhibitor, NA-382, decreased the NBT reducing activity of hypericin, whereas a cAMP-dependent protein kinase A (PKA) inhibitor, H-89, did not show any influence on the differentiations. These results indicate that hypericin triggers differentiation toward monocyte/macrophage lineage by PKC stimulation.

월견초종자유가 생쥐의 면역반응에 미치는 영향 (Effects of Evening Primrose Oil on the Immune Responses in Mice)

  • The purpose of this experiment was to investigate both the immunomodulatory effect of evening primrose(EP) oil and the effects of EP oil on immunoregulation by cyclophosphamide in mice. EP oil at doses of 0.1, 0.2 and 0.4 ml/kg were orally administered to ICR male mice once daily for 28 consecutive days. Cyclophosphamide was injected intraperitoneally to ICR mice with a single dose of 5 mg/kg at 2 days before secondary immunization. Mice were sensitized and challenged with sheep red blood cells(S-RBC). Immnune responses were evaluated by humoral and cellular immune responses and non-specific immune response. The results of this study were summarized as follows; (1) The humoral immune responses such as hemagglutination titer(HA), hemolysin titer(HY), Arthus reaction and plaque forming cell(PFC) were significantly enhanced in the low dose EP oil administered groups(0.1 and 0.2 ml/kg). However, in the high dose EP oil administered group(0.4 ml/kg) the responses were significantly lowered. (2) In the case of cellular immune responses, delayed type hypersensitivity reaction(DTH) was significantly decreased in EP oil whereas rosette forming cell(RFC) was remarkably enhanced. (3) Activities of natural killer cells and phagocyte were generally enhanced in EP oil. In addition, serum albumin and globulin were also increased.

Effects of Fucoidan on NO Production and Phagocytosis of Macrophages and the Proliferation of Neuron Cells

  • Fucoidans, high-molecular-weight sulfated polysaccharides extracted from brown seaweeds, have various biological activities. Here we examined whether fucoidan could ncrease the immunomodulation and proliferation capacity of cells in vitro. When peritoneal macrophages were treated with various concentrations of fucoidan $(1\~100{\mu}g/mL)$ for 24 hours, NO production was significantly increased. In addition, exposure of macrophages to $10{\mu}g/mL$ of fucoidan induced a phagocytic activity. Treatment of neuroblastoma cells (SK-N-SH) with fucoidan enhanced cell proliferation and NO production in a concentration-dependent manner. These results indicate that fucoidan has both immunomodulatory and cell proliferative properties, and may thus be a candidate for development as an immunomodulating agent.

Modulatory Activity of Bifidobacterium sp. BGN4 Cell Fractions on Immune Cells

  • Bifidobacteria has been suggested to exert health promoting effects on the host by maintaining microbial flora and modulating immune functions in the human intestine. We assessed modulatory effects of the different cell fractions of Bifidobacterium sp. BGN4 on macrophage cells and other immune cells from the spleen and Peyer's patches (PP) of mouse. Cell free extracts (CFE) of the BGN4 fractions induced well-developed morphological changes in the macrophages and increased the phagocytic activity more effectively than other fractions in the mouse peritoneal cells. Nitric oxide (NO) production was significantly reduced by both the cell walls (CW) and CFE in the cultured cells from the spleen and PP. The production of interleukin-6 (IL-6) and interleukin-10 (IL-10) was eminent in the spleen cells treated with experimental BGN4 cell fractions. However, in the PP cells, IL-6 was slightly decreased by the treatment with the whole cell (WC) and CW, whereas IL-10 was significantly increased by the treatment with the CW and CFE. These results suggest that different types of bifidobacterial cell fractions may have differential immunomodulatory activities depending on their location within the host immune system.

Costunolide Induces Differentiation of Human Leukemia HL-60 Cells

  • Costunolide has been reported to be a cytotoxic and chemopreventive agent. This work investigated the mechanism of the anti proliferative effect of costunolide and determined that it induced differentiation of the human leukemia cell line HL-60. Costunolide exhibited a potent antiproliferative activity against HL-60 cells. It was also found to be a potent inducer of differentiation in human leukemia derived HL-60 cells through the examination of differentiation markers, as assessed by the reduction of nitroblue tetrazolium, the increase in esterase activities and phagocytic activity, morphology change and the expression of CD14 and CD66b surface antigens. These results, accompanied by a decline in the expression of c-myc protein, suggest that costunolide induces differentiation of human leukemia cells to granulocytes and monocytes/macrophages lineage.

Immunostimulatory Activity of Syneilesis palmata Leaves through Macrophage Activation and Macrophage Autophagy in Mouse Macrophages, RAW264.7 Cells

  • Syneilesis palmata (SP) is a traditional medicinal plant. SP has been reported to have anti-inflammatory, anticancer, and anti-human immunodeficiency virus (HIV) activities. However, there is currently no research available on the immunostimulatory activity of SP. Therefore, in this study, we report that S. palmata leaves (SPL) activate macrophages. Increased secretion of both immunostimulatory mediators and phagocytic activity was observed in SPL-treated RAW264.7 cells. However, this effect was reversed by the inhibition of TLR2/4. In addition, inhibition of p38 decreased the secretion of immunostimulatory mediators induced by SPL, and inhibition of TLR2/4 decreased the phosphorylation of p38 induced by SPL. SPL augmented p62/SQSTM1 and LC3-II expression. The increase in protein levels of p62/SQSTM1 and LC3-II induced by SPL was decreased by the inhibition of TLR2/4. The results obtained from this study suggest that SPL activates macrophages via TLR2/4-dependent p38 activation and induces autophagy in macrophages via TLR2/4 stimulation.

Cordycepin Suppresses MHC-restricted Antigen Presentation and Leads to Down-regulation of Inflammatory Responses in Antigen Presenting Cells

  • Cordyceps militaris, a traditional medicinal mushroom, produces a component compound, cordycepin (3'-deoxyadenosine). Cordycepin has many pharmacological activities including immunological stimulating, anti-cancer, and anti-infection activities. However, the therapeutic mechanism has not yet been elucidated. In this study, we examined the effects of cordycepin on the antigen-presenting function of antigen-presenting cells (APCs). Dendritic cells (DCs) were cultured in the presence of cordycepin and then allowed to phagocytose microspheres containing ovalbumin (OVA). After washing and fixing, the efficacy of OVA peptide presentation by DCs was evaluated using CD8 and CD4 T cells. Also, we confirmed the protein levels of proinflammatory cytokines through RT-PCR and Western blot analysis. Cordycepin decreased both MHC class I and class II-restricted presentation of OVA and suppressed the expression of both MHC molecules and the phagocytic activity toward exogenous OVA. The class II-restricted OVA presentation-regulating activity of cordycepin was also confirmed using mice that had been injected with cordycepin followed by soluble OVA. Furthermore, cordycepin suppressed the mRNA and protein levels of iNOS, COX-2, pro-inflammatory cytokines in a concentration-dependent manner. These results provide an understanding of the mechanism of the T cell response-regulating activity of cordycepin through the inhibition of MHC-restricted antigen presentation in relation to its actions on APCs.

Zoogloea sp.의 다당체가 Meth A 세포에 의한 종양형성 억제 효과 (Antitumor Activities of Polysaccharudes fractuibuzed from Zoogloea sp. Against Meth A Cells)

  • The antitumor activities of the cell bound polysaccharide(CBP), water soluble polysaccharide(WSP) and sulfated polysaccharide(SP) of Zoogloea sp. were observe. The results obtained were as follows : 1) The CBP, WSP, and SP showed cytotoxic effect on the Meth A cells in vitro, however, the effect of CBP and WSP was more ten-fold greater than that pf SP. 2) When CBP, WSP, and SP was inoculated into the peritoneal cavity of the Meth A cells transplanted mice, the average survival days tended to prolonged slightly as compared with the control. 3) When Meth A cells were transplanted subcutaneously into the back side of mice, and then CBP, WSP, and Sp was inoculated into the peritoneal cavity of mice, the tumor growth inhibition ratio was 46.9% for WSP, 40.4% for CBP, and 16.2% for SP. 4) The phagocytic activity of peritoneal macrophages elicited with CBP, WSP, and SP was significantly increased than that of control. 5) The production of nitric oxide in the peritoneal macrophages stimulated with CBP, WSP, SP, and LPS aloneo was not increased than that of control. The production of nitric oxide in the peritoneal macrophages stimulated with IFN-r and CBP, IFN-r and WSP and IFN-r and SP was significantly increased than that of control, but in the case of stimulated with IFN-r and WSP was increased 50% for CBP and SP. These results suggest that the CBP, WSP and SP of Zoogloea sp. showed direct cytotoxic effect and tumor growth inhibition on Meth A cells in vitro and in vivo, and induced nitric oxide production of activated macrophages.

잔나비버섯 자실체로부터 분리한 물질의 특성 및 생리활성 (Biological Activities of Substance Extracted from the Fruit Body of Formitopsis rosea)

  • 잔나비버섯 자실체로부터 열수추출하여 얻어진 고분자 및 저분자 분획의 이화학적 특성 및 생리활성 대하여 검토하였다. 고분자와 저분자 분획은 당과 단백질 및 소량의 hexosamine을 함유하고 있었다. 구성 당은 주로 glucose, galactose, mannose와 소량의 xylose, fructose, ribose 및 arabinose 등으로 구성되었으며, 아미노산의 경우는 당과 단백질의 결합력에 관여하고 있는 Ser과 Thr를 다량 함유하고 있는 것으로 조사되었다. 고분자 분획의 분자량은 10 kD 부근에서 주를 이루고 있는 것으로 나타났다. 항보체활성은 고분자 분획에서 높았으며, 항체생성능은 대조군에 비해 약간 증가하는 양상을 나타내었다. 탐식능, NO 및 $TNF-{\alpha}$ 생성능은 서로 상관성이 있는 것으로 조사되었다. 효소적 및 비효소적 지질과산화 유발에 대한 영향은 고분자 및 저분자 분획에서 모두 억제능을 나타내었다. 이와 같이 잔나비버섯 자실체로부터 추출한 고분자 및 저분자 분획은 면역활성 및 지질 과산화 억제효과 있는 것으로 조사되었다.
