The identification of optimum condition for direct regeneration in black raspberry

  • Adventitious buds appeared within 2 weeks on the base of the petiole explants and increased for two months. A maximum of regeneration (15.6%) was obtained on the medium containing $1.5\;{\mu}M$ TDZ in combination with $1\;{\mu}M$ IBA. To know which explants are the best for the induction of regeneration, three explants such as leaf, petiole and leaf-petiole were used. Among the explant types, the leaf-petiole explant was significantly more effective than leaf and petiole for promoting adventitious shoots, with leaf-petiole inducing at the highest regeneration frequency (33.7%). The regeneration frequency of adventitious shoots in the leaf-petiole explants was significantly affected by leaf size and the position of explants. The leaf-petiole smaller than 5 mm leaf in width was induced at the highest regeneration frequency (68.9%). The smaller leaf sizes, the greater regeneration frequency. Also when the leaves are nearer to the shoot tip, the regeneration frequency is higher. When the rooted micro-shoots were transferred to the soil after growing for 6 weeks in the media, the survival rate was 90%.

  • Petiole explants from 20 cultivars of Catharanthus roseus were cultured on various shoot-inducing media to assess their competence for adventitious shoot formation. After eight weeks of culture on Murashige and Skoog' s medium supplemented with 4.4 $\mu\textrm{m}$6-benzyladenine and 0.5 $\mu\textrm{m}$ $\alpha$-naphthaleneacetic acid, petiole explants from 'Cooler Icy Pink' exhibited the greatest frequency of adventitious shoot formation at 40%, which was followed by 'Little Bright Eye'. By comparing with a previous study on assessment of competence for adventitious shoot formation in hypocotyl explant cultures of various cultures, it is indicated that the relative degree of their competence among cultivars varies to the organ used for the source of explant. Excised adventitious shoots were readily rooted on half-strength MS basal medium. Regenerated plantlets were successfully transplanted to potting soil and grown to maturity in a greenhouse.

  • An efficient plant regeneration of C. asiatica was achieved from organogenesis using petiole explants of in vitro plantlet on MS basal medium controled with different plant growth regulators (NAA,2,4-D, IAA kinetin, and BA). Best results that 50%, efficiency of regeneration per explant for regeneration were obtained with IAA $17.13\;{\mu}M$ and BA $8.9\;{\mu}M$. Formation of adventitious shoots via organogenesis from the petiole explant was verified by histological sectioning of plantlets. Regenerated plants were transplanted into soil.

  • A rapid and efficient in vitro regeneration system was established for Hibiscus syriacus L. The successful regeneration protocol employs induction of shoot organogenesis on leaf, petiole, and root explants. Among the various plant growth regulators evaluated, thidiazuron (TDZ) was the most effective for inducing rapid shoot formation. Most efficient shoot regeneration frequency was obtained from Murashige and Skoog (MS) media containing 0.01 mg/L TDZ. Regeneration efficiency was highest in the roots, and lowest in the leaves. A combination of 0.01 mg/L TDZ with benzyladenine (BAP) markedly improved the frequency of shoot differentiation from the root (up to 98%) and petiole (up to 88%) explants. Furthermore, leaf and petiole explants showed the highest frequency of shoot induction in half-strength MS media containing 0.01 mg/L TDZ and 1.0 mg/L BAP, while root explants formed the greatest number of shoots when 0.01 mg/L TDZ and 0.1 mg/L BAP were added to half-strength MS media. Although the frequency of shoot differentiation from leaf explants was only 50%, the leaf is considered the most efficient plant organ for use in tissue culture because leaves are easier to obtain than roots and petioles. Our findings show that various organs of H. syriacus can be used for plant regeneration, and the protocol developed in this study may be applicable in the horticulture industry.

Effects of different types and ages of explants and cytokinins on shoot regeneration in Brassica juncea L. (절편부위와 사이토키닌이 갓의 재분화에 미치는 영향)

  • To establish an efficient protocol for plant regeneration of Brassica juncea L. Czern, the effects of explant types, explant ages and cytokinins on shoot regeneration were examined in this study. Shoot regeneration was markedly affected by the explant types used in the following order: cotyledon with petiole> hypocotyl> leaf with petiole> cotyledon> leaf. Five-day-old seedlings of cotyledon with petiole explants showed the maximum shoot regeneration frequency. Of the six cytokinins-6-${\gamma}$-${\gamma}$-Dimethylallylamino-purine (2-ip), 6-${\gamma}$-${\gamma}$-Dimethylallylamino-purine riboside (2-ip riboside), 6-Benzyl amino-purine (BAP), Thidiazuron (TDZ), Zeatin, Zeatin riboside-TDZ ($8{\mu}M$) was found to be the best cytokinin for shoot regeneration with the highest shoot induction frequency (80%) from cotyledon with petiole after 4 weeks. All the regenerated plantlets were developed well and they produced morphologically normal flowers.

Effect of Plant Growth Regulators, Media and Celling Agents on In Vitro Microtuber Production of Pinellia ternata Breit (1식물생장조절제, 배지와 고형지지물이 반하의 기내 소괴경 생산에 미치는 영향)

  • The study was carried out to establish in vitro microtuber production from leaf and petiole explant cultures of Pinellia ternata. Culture conditions were optimized by investigating the effect of plant growth regulators, different media, and gelling agents on the efficiency of microtuber indurtion. Among the different combinations of plant growth regulators tested, the combination of 0.1 mg/l 2,4-Dichlorophenoxyacetic (2,4-D) and 0.5 mg/l 6-benzylaminopurine (BAP) showed the highest yield fer microtuber production from leaf (3.9) and petiole (4.7) explant culture on MS medium far 6 weeks. SH(Schenk and Hildebrandt) medium was the most effective medium for microtuber induction and the half strength SH medium was better than SH medium for microtuber production from both leaf and petiole culture. Gelrite was better than agar in the formation of microtubers and 4% Gelrite showed the highest number of microtubers per explant frome leaf (5.9) and petiole (7.8) culture. Germination rate of microtubers after cold storaged for one months long was 86% from in vitro culture and 43% from autoclaved soil.

  • Leaf and petiole explants of kiwifruit were cultured on MT basal medium supplimented with 2,4-D, kinetin, NAA, and BA. Higher organogenic callus formation was observed on the media with NAA + BA than on the media added with 2,4-D + kinetin. Adventitious buds were formed only on media with NAA and BA. Leaf was better explant than petiole. When callus and adventitious buds were subcultured, shoot formation responsed best on medium with 0.1 mg/L NAA + 2.0 mg/L zeatin. When shoots were cultured on medium with 0.5 mg/L IAA + 0.1 mg/L BA after soaking for 1 hr at IBA solution, rooting was more effective than non-IBA treatment. Rooted shoots developed into normal plants.

High Frequency Plant Regeneration from Leaf Explant Cultures of Domestic Cultivated Strawberry (Fragaria x ananassa Duch) (국내 딸기 재배품종의 잎절편 배양으로부터 고빈도 식물체 재생)

  • To develop a high efficiency plant regeneration system from in vitro cultures of strawberry, cv. Yeobong, petiole and leaf explants were cultured on MS basal medium containing a combination of 0.5 mg/L IBA and 3.2 mg/L kinetin or zeatin or benzyl amino purine (BAP) for 6 weeks, and leaf explants with dark pretreatment for a week ($T_1$), 2 weeks ($T_2$), and 4 weeks ($T_3$) were cultured on medium supplemented with 0.5 mg/L IBA and 3.2 mg/L zeatin under 16 hr photoperiod for 6 weeks. Shoot organogenesis was observed from the greenish calli containing minimal anthocyanin formed at proximal cutting edges of the leaf explant (57%) when cultured adaxial side on the medium, whereas was directly formed from a cutting edges of petiole explant (6.3%). Frequency of callus formation and shoot organogenesis at large size of leaf explant ($1.0{\sim}1.5\;cm^2$) was higher than small size ($0.5{\sim}1.0\;cm^2$), and dark pretreatment significantly improved the frequency of leaf explants that produced calli and shoots. The maximum frequency (87%) for shoot organogenesis was obtained from the leaf explants that transferred to a 16 hr photoperiod condition after the initial 4 weeks dark period. The improved frequency (87%) in comparision with control without dark pretreatment (27%). When the shoots were transferred to 1/2 MS basal medium, formed roots with 20 d of culture. The rooted plants were subsequently transferred to the pots and to the field.

Comparative Effect of Plant Growth Regulators on Callus Induction and Plant Regeneration in Glehnia littoralis Schmidt et Miquiel (갯방풍(Glehnia littoralis Schmidt et Miquiel)에서 캘러스 유도 및 식물체 재분화에 미치는 생장조절제의 효과)

  • Glehnia littoralis is known as an edible and medicinal plant using green loaves and mature roots of plant. In the present paper, the influence of plant growth regulators on callus induction and plant regeneration was investigated. Callus induction and regeneration occurred from leaf and petiole explants in Glehnia littoralis. Optimal condition of plant growth regulators for callus induction from leaf and petiole explants was MS basal medium supplemented with 2mg/L 2,4-D and 2mg/L BA. The frequency of callus induction was higher in petiole explant than leaf. When the callus was cultured on MS basal medium supplemented with 0∼1 mg/L IAA, 0∼1mg/L NAA and 0∼2mg/L BA for about 65 days, the most effective plant growth regulators on plant regeneration from callus were 1mg/L NAA and 2mg/L BA. The plantlets acclimatized successfully and grown in vermiculite matrix.
