A taxonomic review of Adonis pseudoamurensis W. T. Wang (Ranunculaceae) (개복수초(미나리아재비과)의 분류학적 재고)

  • Morphological characters of Adonis pseudoamurensis W. T. Wang and its related species were examined to illustrate its taxonomic entity, and their new key was made. As a result, it shows that A. ramosa Franch., an endemic species in Japan, was sparsely pubescent on the abaxial surface of the leaf and petiole, and its sepal was as long as its petal. However, A. pseudoamurensis is glabrous on the same surfaces and its sepal was shorter than its petal. In addition, A. multiflora Nishikawa et Ko. Ito was clearly distinguished in that it had acuminate leaf apices, linear shape of the final lobes of leaf and scales on the branched stem. Meanwhile, A. pseudoamurensis featured petiolate at cauline leaves and pubescent on pistils, and these characters were common with their allied groups. But diagnostic key characters suggested by the original description such as sessile at cauline leaves and glabrous on pistils were not observed in this study.

Analysis of Leaf Morphological Variation of 11 Natural Populations of Acer pictum subsp. mono (Maxim.) Ohashi (고로쇠나무 11개 자연집단의 잎의 형질 변이 분석)

  • Characteristics of leaf morphology was examined in 11 natural populations of Acer pictum subsp. mono (Maxim.) Ohashi in Korea. Nested Anova showed that there were statistically significant differences among populations as well as among individuals within populations in all 14 quantitative characters. In all the leaf characters, variance components among individuals within populations were higher than those among populations. Some morphological characters such as the ration of Blade length to Petiole length (0.90), Uower lobe width (0.85), Blade length (0.83), and Middle lobe width (0.83) showed the relative high differences in the values of differentiation (Dij) among populations. Of the ten populations, Gwangyang (1.36) and Cheongyang (1.30) were highly differentiated based on the total mean Dij of fourteen morphological characters, while Ulju (0.30) and Jeongseon (0.44) were scarcely differentiated.

Factors Affecting the Production of In Vitro Plants from the Nodal Pieces of Chinese Yam (Dioscorea Opposita Thunb)

  • This study was carried out to establish The regeneration of healthy seedlings from the nodal segment culture of Chinese yam (Dioscorea opposita cv. Danma), cultivated in Korea. Different explants such as leaves, petioles, roots and nodal pieces, excised from the in vitro grown seedlings of Chinese yam, were cultured on MS medium supplemented with various combinations of growth regulators. All the growth regulators used induced plantlet regeneration from the nodal segments at a high frequency, while there was no induction of shoot or callus from leaf, petiole or root tissues. The medium supplemented with 0.01mg/L NAA, 0.5mg/L BA, 0.5-1.0mg/L kinetin and without plant growth regulator was effective for shoot development of buds from the nodal segment culture. The concentration of BA and NAA was an important factor in the bud induction of buds from the nodal segments of Chinese yam. Nodal segments cultured on the medium containing 1.0mg/L NAA and 0.5-1.0mg/L BA gave the best response to bud formation. The addition of GA$_3$ to the culture medium suppressed shoot induction and growth, while it increased microtuber formation. The shoot growth and microtuber formation were also affected by medium strength and solidity. The MS basal medium containing 1 g/L gelrite was suitable for microtuber formation from the nodal segment of Chinese yam.

Identification and Antibiotic Susceptibility Test of Bacteria from In Vitro Cultures of Pinellia ternata and Tea Plant (반하 및 차나무의 기내배양시 발생하는 세균의 동정 및 항생제 감수성 검정)

  • Contamination of bacterial infection is one of serious problems in in vitro culture system of root crops. From the contaminated tubes over 140 of petiole cultures of Pinellia ternata, a medicinal plant, 4 genera 8species 48 strains of bacteria, including Aeromonas and Pseudomonas, were isolated and identified and another 8 strains were not fully identified. Most of them were motile Gram positive bacteria as in common in early stage of in vitro cultures. Six strains of bacteria, 5 of Gram negative, including Enterobacter, and 1 of Gram positive, were identified from the embryonic axes cultures of tea plant. From the susceptibility test to pre-screened 5 antibiotics, all of the bacteria except for 2 species of Pseudomonas were susceptible to cefotaxime 60∼100mg/L. While 60mg/L erythromycin only was effective to Pseudomonas. Combination of erythromycin 20mg/L and cefotaxime 60mg/L totally suppressed the growth of all bacterial strains tested. Susceptibility test of bacteria from tea embryonic axes cultures showed similar results. Combination of erythromycin 35mg/L and cefotaxime 60mg/L was effective to 15 bacterial strains and partially effective to 1 unidentified.

GUS gene expression and plant regeneration via co-culturing with Agrobacterium in grapevine (Vitis vinifera) (Agrobacterium 공동배양을 이용한 포도 재분화율 향상과 GUS 유전자의 발현)

  • Efficient transformation and regeneration methods are a priority for successful application of genetic engineering to vegetative propagated plants such as grape. In this study, methods for Agrobacterium tumefaciens-mediated transformation and plant regeneration of grapevine (Vitis vinifera) were evaluated. Tamnara, Heukgoosul, Heukbosek, Rizamat were co-cultivated with Agrobacterium strains, LBA4404 containing the vector pBI121 carrying with CaMV 35S promoter, GUS gene as reporter gene and resistance to kanamycin as selective agent. Seven percent of the maximum regeneration frequency was obtained from co-cultivated with explants from Rizamat with LBA4404 strain on selection medium with kanamycin. The addition of acetosyringone, 200 ${\mu}m$ in virulence induction step was a key factor for successful GUS reporter gene expression in grapevine transformation. Transgenic plants showed resistance to kanamycin and the GUS positive response in leaf ($T_0$) stem ($T_0$) and petiole ($T_0$).

Establishment of high frequency plant regeneration system from leaf explants of Pinellia koreana via bulblets formation

  • Pinellia koreana K-H Tae & J-H Kim is a recently discovered Korea endemic medicinal plant species whose natural habitat is rapidly destroyed by industrial development. Described in this paper are culture conditions for high frequency plant regeneration via bulblet formation from leaf explant cultures of P. koreana. Leaf explants formed white nodular structures and off-white calluses at a frequency of 91.2% when cultured on MS medium supplemented with 2 mg/L BA and 0.5 mg/L NAA. However, the frequency of white nodular structures and off-white calluses formation was slightly decreased with an increasing concentration of NAA up to 4 mg/L, where the frequency reached 31.7%. Most petiole explants did not form white nodular structures and off-white calluses except the combination treatment of 2 mg/L BA and 2 mg/L NAA. Upon transfer onto MS basal medium, over 90% of nodular structures gave rise to numerous bulblets and developed into plantlets. Plantlets regenerated from bulblets were transplanted to potting soil and grown to maturity at a survival rate of over 95% in a growth chamber. Therefore, the in vitro plant regeneration system of P. koreana obtained in this study will be useful for mass propagation and long-term preservation of genetic resources of P. koreana.

Survey of Overwintering Inoculum Potential of Anthracnose of Sweet Persimmon Caused by Colletotrichum gloeosporioides (감나무 탄저병균(Collectotrichum gloeosporioides)의 월동 전염원 조사)

  • In 2006 to 2007, the potential inoculum source of the anthracnose of sweet persimmon caused by Colletotrichum gloeosporioides was surveyed. The infected twigs, buds, dead twigs, petiole, leaves, dropped fruits were collected and tested for their possibility as overwintering inoculum. The detection rates of the pathogen from various parts of sweet persimmon tree were varied. When the collected samples were examined in April. Over than 93.3% of infected twig samples were harbored mycelia of C. gloeosporioides, and 46.7% of infected buds, 36.7% of dead twigs, 23.3% of petioles, and 16.7% of leaves were beared pathogenic fungus. No pathogenic fungus were detecded from healthy twigs and buds. Infected twigs and bud was important overwintering sites and formed conidia actively in next spring. The infected twigs, leaves, petioles, and fruits in growing season produced great number of conidia and caused active dissemination of the anthracnose disease in sweet persimmon. In growing season, all of the infected parts, such as twigs, leaves, petioles, and fruits produced pathogenic fungus.

Growth Characteristic Models of Collected Artemisia Resources (수집 쑥속 자원의 생육특성 모형)

  • The purposes of this dissertation were to examine relativity between growth characteristics factors and develop the model of growth characteristics. In the result of statistical summaries of the factors in growth characteristic, the highest level of correlation was in plant height, plant width, petiole and chlorophyll in the case of Artemisia iwayomogi; leaf length, in the case of A. stolonifera;, leaf width, of A. princeps var. orientalis;, diameter, of A. feddei;, leaf thickness, of A. iwayomogi, A. princeps var. orientalis, A. montana, A. stolonifera, A. sylnatica; leaf length and leaf width of the other species. In the case of correlation analysis, leaf width and diameter were most correlative in A. capillaris;, plant height and plant width, in A. montana; plant height and leaf width, in A. stolonifera; leaf length and leaf width, in other species. The results of model analysis of growth characteristics were as follows. A. capillaris proved to be about $79.4\%$ effective ; A. princeps var. orientalis, about $91.7\%$ ; A. montana, about $70.3\%$ ; A. iwayomogi, about $49.0\%$ ; A. stolonifera, about $72.5\%$; A. sylnatica, about $75.0\%$;, A. feddei about $60.4\%$ ; A. rubripes about $54.0\%$.

Ultrasturctural Study on Nectar Secretion from Extrafloral Nectary of Prunus yedoensis Matsumura (왕벚나무 화외밀선의 당액 분비에 관한 미세구조적 연구)

  • Nectar secretion from extrafloral nectary cells of Prunus yedoensis was examined by light and electron microscopy. Nectaries were composed of two or three layers of secretory cells and one layer of subsectretory cells. Vascular bundles in the petioles were connected to those of the subsectretory cell layer. Secretory cells had a number of mitochondria with poorly developed cristae. Plastids had little thylakoids and small vesicles, about 0.2 to 0.3 mm in diameter; however, no plastids had starch grains. Calcium oxalate crystals and plasmodesmata were frequently observed in the subsectretory and secretory cells, respectively. And nectar substances were observed in phloem of petiole, subsectretory, and secretory cells of the secretory gland. These results suggested that the nectar moved by symplastic transport through the plasmodesmata. On the other hand, the nectar droplets were observed in the secretory cell walls. in the cuticular layer just beyond of the former, and on the outer surface of the cuticular layer: such observations indicated that a apoplastic movement was involved in the final step of the nectar secretion. Cellular components related to the nectar transport, such as plasma membrane, cell wall and cuticle were not destroyed but intact: it was interpreted as a eccrine secretion.retion.

Evaluation of Fruit Characteristics According to Mulberry Breeding Lines for Fruit Production (오디 생산을 위한 뽕나무 계통별 과실 특성 평가)

  • We evaluated fruity characteristics to select the desirable mulberry varieties for the production of fruit. Tested items were total yield, weight of single fruit, soluble solids and fruit size including length, width, and petiole length. 'Susungppong', 'Cheongnosang', 'Suwonnosang' and 'Jeolgokchosaeng (Chungbuk)' were showed high yield for three years. In weight of single fruit, 'Jeolgokchosaeng (Chungbuk)' was 4.5 g. 4 mulberry varieties were showed move than 17 Brix $\%$ in soluble solids. From that results, eight suitable varieties such as 'Jeolgokchosaeng (Chungbuk)', 'Dangsang 7', 'Jangsosang', 'Susungppong', 'Suwonnosang', 'Palcheongsipyung', 'Kangsun', and 'Jukcheonchosaeng' were selected for the production of fruit.
