Study on Antioxidant Potency of Green Tea by DPPH Method (DPPH 방법을 통한 녹차의 항산화 활성에 대한 연구)

  • The present study was conducted to compare antioxidant activity of green teas, fermented teas and other related common teas by examining radical scavenging activity using DPPH (2,2 diphenyl l-picryl hydrazyl). Scavenging activity ($SC_{50}$/) of epigallocatechin gallate (EGCG) for 0.1 mM DPPH radical was 5.5 $\mu$M or 4.2 mg/L by weight, then catechin, 14 $\mu$M or 2.5 mg/L and vitamin C, 22 $\mu$M or 3.9 mg/L, respectively. Kyokuro tea (okro) powder of 24.2 mg/L or green tea powder of 25.2 mg/L was used to reach $SC_{50}$/ for 0.1 mM DPPH. One serving of 2 g green tea provides antioxidant activity equivalent to 109∼147 mg EGCG, 145∼185 mg catechin or 131∼168 mg vitamin C. Teas from the first harvest had the highest radical scavenging activity when compared with later harvest green teas grown in the same region, but there is virtually no difference by the harvest time. A Chinese green tea, Dragon well had the highest antioxidant activity among other green teas tested providing antioxidant capacity equivalent to 168 mg EGCG or 188 mg vitamin C per 2 g serving, but partially fermented Chinese teas had much lower antioxidant activity than any green tea tested. Black tea which is fully fermented showed as strong antioxidant activity as green teas (76.3 mg vs 86.7∼67.6 mg per tea bag). One tea bag of green teas from market provided antioxidant capacity equivalent to 52∼86 mg EGCG, 70∼105 mg catechin or 63-96 mg vitamin C. Teas made of persimmon leaf, pine needle, mulberry leaf had comparatively low anti-oxidant activity equivalent to 2.5∼4.8 mg EGCG or 15∼21 mg vitamin C per teabag. The third brewed green tea still had enough antioxidant activity, while tea from tea bag brewed for 3 min or 5 min did not have any difference in their antioxidant activity. More systemic studies are needed to clarify the relationship between tea catechins and antioxidant capacity focusing on how growing, harvest time, fermentation and other processes can influence on this.

Isolation of Polyphenol Compounds from the Leaves of Korean Persimmon (Diospyrus kaki L. Folium) (한국산 감잎로부터 Polyphenol계 생리활성물질 분리)

  • We purified polyphenols from persimmon leaf and tested their biological activity. The 60% acetone extract was lyophilized and applied to test enzyme inhibition of glucosyltransferase and tyrosinase. GTase was 82.4% inhibited at $1.8{\times}10^{-1}$ mg/ml and tyrosinase 21.7% inhibited at 0.8 mg/ml. The acetone extract was fractionated into F-1, 2, 3, 4, 5 by Sephadex Q-50 gel filtration and the fraction-1 and 2 showed higher enzyme inhibition activity than the other fractions. To the Proteinase K treatment and autoclaving of the two fractions had no effect on the enzyme activity, but these results suggested that active fraction was not protein but phenol ring completed compounds. By Sephadex LH-20, MCI-gel and Bondapak $C_{18}$ column chromatographies, compouds 1, 2, 3 and 4 from F-1 fraction, compounds 5 and 6 from F-2 fraction and compounds 7 , 8 from F-3 fraction were purified and re-crystallized. The purified compounds was assumed to be condensed tannins of frame flavan-3-ol frame on the basis of color reagent reaction and to be a mixture of monomer, dimer and trimer according to TLC analysis.

Effect of Korean Traditional Tea Materials on Minerals Content and Histological Changes in Pb-Administered Rats (전통음료 소재가 납투여한 흰쥐의 체내 무기질 함량 및 간과 신장조직의 헝태학적 변화에 미치는 영향)

  • This study was designed to investigate the effect of Korean traditional tea materials on lead (Pb) accumulation and histological changes in Pb-administered rats. Male rats were divided into six groups: a normal, Pb, and Pb-water extract of Korean traditional tea materials (green tea: GT, persimmon leaf: PL, safflower seed: SS, Eucommia ulmoides: EU) groups. Pb (25 mg/kg BW) was orally administered once a day for 4 weeks. The extract of GT, PL, SS, and EU were administered based on 1.26 g of raw traditional tea materials/kg Bw/day. Pb contents of serum and liver were significantly higher in the Pb group than in the normal group. However the water extracts of GT, PL, SS, and EU administration lowered Pb contents of serum and liver in the Pb-administered rats, respectively. Serum Zn content was significantly higher in the Pb group than in the normal group, whereas, Cu and Fe contents were lower in the Pb group. Hepatic Ca content was significantly lower in the Pb group compared to the normal group. In the present study, water extract of Korean traditional tea materials administration effectively improved Cu and Fe contents of serum and liver. Infiltration of the local inflammation was found in Pb group, whereas water extracts of Korean traditional tea materials administration attenuated histological changes of tissues.

Comparison of Antioxidant Activities in Mugwort Teas and Commercial Teas (수집 선발한 쑥으로 만든 쑥차와 시판차의 항산화력 비교)

  • Artemisia spp. collections, AC60, AC67, and AC77, that showed the high levels of antioxidant activities and had good growth characters and productivity, were cultivated for mass production. Using selected excellent ones, AC60, AC67, and AC77, after mass production, mugwort teas were made through rubbing and drying processes. Total phenolic compound content, nitrite scavenging ability, and SOD-like activity in mugwort teas and commercial teas were compared. Total phenolic compound content was the highest in AC67 with 109.6 mg/100 ml. Nitrite scavenging ability of AC67 tea was relatively high with 92.5% at pH 1.2 and Jasmine (98.1%), Persimmon Leaf Tea (96.4%), Green tea (98.5%), and Jakseolcha (98.7%) were very high as well. In SOD-like activity, mugwort teas, AC67 (91.7%) and AC77 (93.7%), were high as well as those of Seolrokcha (92.1%) and Rooibos Tea (93.8%). This suggests that AC60, AC67, and AC77 could be used fur making high quality teas as well as Seolrokcha, Jasmine, Green tea, Jakseolcha, Rooibos Tea, Peppermint Herb Tea.

Identification of Biologically Effect and Chemical Structure of Polyphenol Compounds from the Leaves of Korea Persimmon (Diospyrus kaki L. Folium) (한국산 감잎의 Polyphenol 화합물의 생리활성물질의 화학구조 및 효소저해효과)

  • The lyophilization of the solution extracted from 60 percent of acetone applied to persimmon leaves, the compounding process in accordance with the solution's concentration, and the gel filteration through Sephadex G-50 of biologically activated substances obstructing enzyme activity, such as tyrosinase, xanthine oxidase, and angiotesin converting enzyme (ACE) led to the assumption that polyphenol was the compound serving as biologically activated substances obstructing enzyme activity. Xanthine oxidase involved in pruine metabolism oxidizes hypoxanthine to xanthine and xanthine to uric acid. In the continuous study for natural compound, nine flavan-3-ols have been isolated from the persimmon leaves. The structures of (+)-catechin, (+)-gallocatechin, procyanidin B-1, pyrocyanidin C-1, prodelphinidin B-3, gallocatechin-$(4{\alpha}{\rightarrow}8)$-catechin, procyanidin B-7-3-O-gallate, procyanidin C-1-3'-3'-3'-O-trigallate and (-)-epigallocatechin-$(4{\alpha}{\rightarrow}8)$-epigallocatechin-$(4{\alpha}{\rightarrow}8)$-catechin were established by NMR and their inhibitory effect on xanthine oxidase activity was investigated. Procyanidin B-7-3-O-gallate, (-)-epigallocatechin-$(4{\alpha}{\rightarrow}8)$-epigallocatechin-$(4{\alpha}{\rightarrow}8)$-catechin and procyanidin C-1-3'-3'-3'-O-trigallate showed 94%, 90.69%, 80.90% inhibition at $100\;({\mu})M$ and inhibited on the angiotensin converting enzyme respectively. Procyanidin B-7-3-O-gallate and procyanidin C-1-3'-3'-3'-O-trigallate showed 66%, 63% inhibition at $100\;({\mu})M$ and inhibited on the xanthine oxidase competitively. Procyanidin C-1-3'-3'-3'-O-trigallate showed 70% inhibition at $100\;({\mu})M$ and inhibited on the tyrosinase competitively.

Effect of Korean Traditional Tea Materials Water Extract on Hepatic Oxygen Free Radical Generating and Scavenging Enzyme Activities in Lead Administered Rats (한국전통차 재료의 열수추출물이 납투여 흰주의 간조직 중 유해 활성산소 생성과 제거효소 활성에 미치는 영향)

  • This study was designed to test the effect of Korean traditional tea materials on oxygen-free radical metabolism in lead (Pb) -administered rats. Male rats were divided into normal, Pb-control (Pb-Con) and Pb-water extract of green tea (Camellia sinensis; GT) , persimmon leaf (Diospyros kaki; PL) , safflower seed (Carhamus tinctorius: SS) , Du-Zhong (Eucommia ulmoides; EU) groups, respectively. Pb intoxication was induced by administration of lead acetate (25 mg/kg. B.W., oral) weekly. The extract was administered based on 1.26 g of raw material/kg B.W./day for 4 weeks. When the GT, PL, SS and EU were supplemented to the Pb-administered rats, hepatic lipid peroxide levels were significantly lower compared to the Pb-Con group. Hepatic cytochrom P-450 content and aminopyrine N-demethylase activity was lower in the Pb-Con group than in the normal group, whereas xanthine oxidase activity was significantly elevated in Pb-administered rats. The water extract of GT, PL, SS and EU supplementation attenuated changes in enzyme activities generating reactive oxygen species in the liver. Hepatic superoxide dismutase, catalase and glucose 6-phosphate dehydrogenase activities were significantly higher in the Pb-Con group than in the normal group, while monoamine oxidase activity also tended to increase in the Pb-administered rats. However, glutathione peroxidase and glutathione S-transferase activities, and glutathione content significantly decreased through Pb intoxication. The supplementation of GT, PL, SS and EU induced alleviation changes of hepatic antioxidant enzyme activity.
