Nitrogen Source Investigation for Economical Production of Cellulolytic Enzymes

  • Trichoderma inhamatum KSJ1, a filamentous fungus, isolated from rotten wood showed high ability to hydrolysis of cellulosic materials. Enzyme productivity by strain KSJ1 was high in the cultivation using carbon sources such as cellulosic materials and lignocellulosic wastes as rice straw and paper waste. In previous study peptone was one of optimum organic nitrogen sources in producing cellulases for saccharification of food wastes. However, it was too expensive using peptone as organic nitrogen source, so, in this study, soybean and yeast were applicated to substitute peptone. Yeast showed producing high enzyme activity, so it was estimated that yeast is available in producing cellulase using Trichoderma inhamatum KSJ1 at industrial Production.

Application of Response Surface Methodology for Optimization of Lactic Acid Production Using Date Juice

  • Media components, including date juice, sodium acetate, peptone, and $K_{2}HPO_4$, which were screened by Plackett-Burman fractional factorial design, were optimized for lactic acid production from date juice using the response surface method (RSM). Sodium acetate, peptone (p<0.0001), and $K_{2}HPO_4$ (p=0.0029) were highly significant in influencing the lactic acid production. Close correlationship between predicted and experimental values was observed. When the optimum values of the parameters obtained through RSM (25.0 g/l date sugar, 15.0 g/l sodium acetate, 19.1 g/l peptone, and 4.7 g/l $K_{2}HPO_4$) were applied, lactic acid production (22.7 g/l) increased by 50.33%, compared with unoptimized media (15.1 g/l). The subsequent validation experiments confirmed the validity of the statistical model.

  • This study was performed to evaluate initial pH and substrate concentration on hydrogen fermentation of protein. The optimum initial pH and substrate concentration of hydrogen fermentation using protein was 8.0 and 1.0 g peptone/L, respectively. The maximum hydrogen yield at initial pH 8.0 and 1.0 g peptone/L was $19.2{\pm}0.8mL\;H_2/g$ peptone. As results of VFAs analysis, percentages of valerate was similar to hydrogen yield. Also, C. stickalandii, which was hydrogen and valerate producing bacteria, was dominated.

  • The mycelial growth of Agaricus bitorquis and Pleurotus ostreatus in synthetic media were carried out by ordinary methods. The optimum pH and temperature for mycelial growth were from pH 6.0 to 6.5 and 25 to $30^{\circ}C$, and from pH 5.0 to 6.5 and $25^{\circ}C$ in A. bitorquis and P. ostreatus, respectively. Among the carbon and nitrogen sources, glucose, starch, and peptone showed the good result for the mycelial growth of A. bitorquis, and glucose, fructose, starch and peptone were good for the mycelial growth of P. ostreatus. The yield of mycelium decreased under lower or higher C/N ratio. Also, at the same C/N ratio, the higher the concentration of glucose and peptone, the more the yield was increased. Among various vitamins thiamine, Ca-pantothenate and folic acid were suitable for the mycelial growth of A. bitorquis, and thiamine, folic acid and ino­sitol for the mycelial growth of P. ostreatus. Although pH, total nitrogen and glucose contents of media decreased gradually during culture period the yield of mycelium increased.

Effect of Medium Compositions on the Mycelial Growth of Inonotus obliquus (차가버섯의 균사체 성장에 대한 배지성분의 영향)

  • Effect of temperature($22-32^{\circ}C$), pH(5-7) and medium composition on the mycelial growth for the submerged culture of Inonotus obliquus. The concentrations of glucose, starch, peptone, yeast extract, $K_2HPO_4$, $MgSO_4{\cdot}7H_2O$ and $CaCl_2$ were examined in the ranges of 30-120 g/L, 0-10 g/L, 0-20 g/L, 0-15 g/L, 0-2 g/L, 0-1.5 g/L and 0-0.5g/L, respectively. The maximum mycelial growth of Inonotus obliquus was obtained for $26-27^{\circ}C$ and pH 6. The concentrations of glucose, yeast extract and $CaCl_2$, which gave the maximum mycelial growth of Inonotus obliquus, were 70 g/L, 5 g/L and 0.1 g/L, respectively. In the cases of starch, peptone and $K_2HPO_4$, the mycelial growth of Inonotus obliquus increased with increasing the concentrations. However, as the concentration of $MgSO_4{\cdot}7H_2O$ increased, the mycelial growth of Inonotus obliquus decreased. The medium for maximum mycelial growth of Inonotus obliquus consisted of (per 1 L): glucose, 70 g; peptone, 5-20 g; starch, 10 g; yeast extract, 5 g; $K_2HPO_4$, 2 g and $CaCl_2$, 0.1 g.

Properties in Formation of Protein-Binding Polysaccharide in Cordyceps militaris (동충하초(冬蟲夏草)의 단백다당류 생성 특성)

  • The effects of liquid culture conditions and nutrient sources on the formation of protein-binding polysaccharide (PS) in Cordyceps militaris were examined. The formation amount of PS was increased in proportion to the growth rate of mycelium, in case of higher aeration or lower acidity. The optimum growth temperature of the mycelia was 25$^{\circ}C$ for the formation of PS. The optimum carbon source and nitrogen source were glucose and peptone, respectively. The ratio of C/N was optimal with 3% glucose to 0.5 % peptone. The sugar composition in the PS was greatly changed according to the carbon sources. The mycelium of Cordyceps militaris by liquid culture showed a higher electron donating ability than that by solid culture.

Characteristics of Culture and Isolating Lactic Acid Bacteria and Yeast from Sourdough (Sourdough로부터 젖산균과 효모의 분리 및 배양 특성)

  • This study was conducted to investigate the optimum conditions for the medium composition after isolating and identifying yeast and lactic acid bacteria from sourdough. It was found that the best quality lactic acid bacterium with acid product and flavor was identified as Leuconostoc species among isolated 115 lactic acid bacteria, the best Quality yeast with good fermentation and flavor was identified as Saccharomyces species among isolated 8 yeast. While the microbial growth with glucose or sucrose as sugar source was good, it was, selected that sucrose which is using commercially is better than glucose. The growth of lactic acid bacterium ; was good with 1% added sucrose whereas yeast needed more growth. Additionally, the medium for the optimum 1 growth of the yeast was composed of 0.5% wheat flour, 0.5% peptone and 3% sucrose, whereas lactic acid bacterium was composed of 0.5% wheat flour and 1% sucrose without peptone.

Production of Bioflocculant by Agrobacterium sp. KF-67 (Agrobacterium sp. KF-67에 의한 미생물 응집제 생산)

  • Among 120 microorganisms isolated from soil, KF-67 was the best producer of flocculant and was examined for flocculating ability in the kaolin clay and CaCl2 suspension. KF-67 was identified to be a species belong to the genus Agrobacterium sp. The influence of components of the culture medium for flocculant production by Agrobacterium sp. KF-67 was studied. The favorable carbon and inorganic nitrogen source for production of the flocculant were glucose and NH4NO3 and their addition concentrations were 2% and 0.1%, respectively. Addition of the organic nitrogen such as yeast extract, peptone and inorganic salt such as CaCO3 significantly increased the production of flocculant. These result indicated that the production of flocculant by Agrobacterium sp. was significantly affected by both organic nitrogen and inorganic salt. The components of the optimum culture medium were 2% glucose, 0.1% NH4NO3, 0.01% yeast extract, 0.01% peptone, 0.04% CaCO3, 0.03% NaCl in initial pH 7.5 when cultured with rotary shaker controlled at 3$0^{\circ}C$ and 120 rpm. Under the optimum culture medium, flocculant production was highly improved about 76% than that isolation medium.

  • This study was excuted to decide the physiological characteristics of Lentinus lepides. L. lepides was tested to select pertinent substract, temperature and pH range for the growth. Mycelial growth of Lentinus lepides was mostly supported on MYPA among other tested synthetic or semi-synthetic media. The temperature range for pertinent mycelial growth was about 22~32$^{\circ}C$ and mostly stimulated at $25^{\circ}C$. And the pertinent pH range of MYPA was 3.0~4.0. The required carbon and nitrogen source for mycelial growth of Lentinus lepides was tested. The mycelial growth was mostly stimulated by soluble starch at content. The carbon sources for pertinent mycelial growth was maltose. And the nitrogen source for pertinent mycelial growth was peptone.
