Identification of Colletotrichum spp. associated with pepper anthracnose in Korea (oral)

  • Pepper anthracnose is one of the major limiting factors in pepper production. Boring last over 10 years, Colletotrichum gloeosporioides has been known as the most prevalent species among five Colletotrichum spp. involved as anthracnose causing agents. Recently, however, the change of major species with pepper anthracnose has been proposed. Identification study was peformed on 12 test isolates collected from anthracnose disease symptoms on pepper during 2001-2002 and 25 reference isolates obtained from several other host plants. The identification of the isolates with morphological observation and IfS region sequence comparison resulted that 11 ones from 12 test isolates colleted from pepper anthracnose during 2001-2002 were identified as C. acutatum. PCR using species-specific primers designed from ITS region sequence suggested a rapid diagnosis method in identifying C. acutatum from C. gloeosporioides.

ABA Increases Susceptibility of Pepper Fruits to Infection of Anthracnose by Collectotrichum acutatum

  • To examine the relationship between plant hormones and the development of pepper anthracnose, we investigated the effects of several plant hormones on the progression of disease symptoms. Of the five plant hormones examined, abscisic acid (ABA) increased the lesion length and disease incidence on detached fruits of Capsicum annuum cv. Nokkwang. The simultaneous application of ABA with inoculation of Colletotrichum acutatum JC24 resulted in increased lesion length, depending the concentration of ABA applied. Additionally, application of ABA caused the development of pepper anthracnose in fruits of Capsicum baccatum cvs. PBC80 and PBC81, which were previously resistant to the disease. Furthermore, ABA administration rendered increased pathogenicity of other isolates of C. acutatum BAC02063, PECH10, and TCBNU3 obtained from the Chinese matrimony vine, peach, and tea tree, respectively. Our data suggest that exogenous ABA may result in the suppression of defense mechanisms of pepper fruits against anthracnose, which leads to a change in the susceptibility of pepper fruits and the development of pepper anthracnose.

The Cell Wall Integrity MAP Kinase Signaling Pathway Is Required for Development, Pathogenicity, and Stress Adaption of the Pepper Anthracnose Fungus Colletotrichum scovillei

  • The cell wall integrity (CWI) signaling pathway plays important roles in the dissemination and infection of several plant pathogenic fungi. However, its roles in the pepper fruit anthracnose fungus Colletotrichum scovillei remain uninvestigated. In this study, the major components of the CWI signaling pathway-CsMCK1 (MAPKKK), CsMKK1 (MAPKK), and CsMPS1 (MAPK)-were functionally characterized in C. scovillei via homology-dependent gene replacement. The ΔCsmck1, DCsmkk1, and ΔCsmps1 mutants showed impairments in fungal growth, conidiation, and tolerance to CWI and salt stresses. Moreover, ΔCsmck1, ΔCsmkk1, and ΔCsmps1 failed to develop anthracnose disease on pepper fruits due to defects in appressorium formation and invasive hyphae growth. These results suggest that CsMCK1, CsMKK1, and CsMPS1 play important roles in mycelial growth, conidiation, appressorium formation, plant infection, and stress adaption of C. scovillei. These findings will contribute to a better understanding of the roles of the CWI signaling pathway in the development of pepper fruit anthracnose disease.

Control Effects of 3-(4-Hydroxyphenyl)-propionic Acid Isolated Xenorhabdus nematophila K1 against Phytophthora Blight and Anthracnose of Red Pepper (Xenorhabdus nematophila K1 대사물질 3-(4-hydroxyphenyl)-propionic acid의 고추 역병과 탄저병에 대한 방제 효과)

  • 3-(4-Hydroxyphenyl)-propionic acid (HPP) is a bacterial metabolite synthesized and released by an entomopathogenic bacterium Xenorhabdus nematophila K1. In this study, the control efficacy of HPP was tested against Phytophthora blight and anthracnose of red pepper plants. HPP suppressed mycelial growth of Phytophthora blight and anthracnose pathogens. Under natural sunlight condition, HPP maintained the antifungal activity on the diseases for at least twenty five days. The antifungal activity was not decreased even in the condition of soil-water. It was proved that HPP was able to penetrate the roots and travel upward of the red pepper plants. When HPP suspension was applied to soil rhizosphere before transplanting the red pepper seedlings or was regularly sprayed to the foliage of the plants with ten days interval, it resulted in significant reduction of the disease occurrences (Phytophthora blight and anthracnose) without any phytotoxicity. These results suggested that HPP can be developed to a systemic agrochemical against Phytophthora blight and anthracnose of red pepper plants.

Selection of Antifungal Bacteria Burkholderia lata CAB13001 for Control on Red Pepper Anthracnose and Its Control Efficacy in Field (고추 탄저병 방제제 Burkholderia lata CAB13001 선발 및 포장방제 효과)

  • To control the pepper anthracnose caused by Colletotrichum acutatum, antifungal bacterium strains which was selected among bacterium from natural soil, was tested the antimicrobial activity against various pathogens and its control efficacy on anthracnose disease in the fields. We confirmed that antagonistic activity of CAB13001 strain to pathogens such as Sclerotinia cepivorum, Sclerotinia sclerotium and Botrytis cinerea including Colletotrichum acutatum was remarkable superior with the dual culture method in the artificial medium. In vitro bioassay using the green pepper fruit, CAB13001 strain suppressed the lesion development of Anthracnose disease, and its control value compared to the untreated one was 82.4% on pepper fruit in field test. These results suggested that CAB13001 strain could be a very useful biological control agents to anthracnose disease caused by air born plant pathogens of pepper. By the way, analysis of nucleotide sequence of the gene 16S rDNA, antagonistic bacterium CAB13001 strain used in this study was identified as Burkholderia lata.

Biocontrol of Anthracnose of Chili Pepper by Bacillus sp. NAAS-1 (Bacillus sp. NAAS-1을 이용한 고추 탄저병 생물학적 방제)

  • Bacillus sp. NAAS-1 isolated from the field of Chili pepper was tested for biocontrol activity against anthracnose pathogen of Chili pepper caused by Colletotrichum acutatum. The antifungal activity of Bacillus sp. NAAS-1 culture broth was compared with synthetic fungicide containing carbendazim (40%) and kasugamycin (3.45%). Bacillus sp. NAAS-1 showed a similar fungicidal activity against the anthracnose pathogen at the concentration of 50 ${\mu}L/mL$ in comparison to the fungicide containing carbendazim (40%) and kasugamycin (3.45%) using a cup method. Bacillus sp. NAAS-1 also exhibited its potent fungicidal activity against the anthracnose in vivo test at the concentration of 50 ${\mu}L/mL$ when compared to the fungicide containing carbendazim (40%) and kasugamycin (3.45%).

Studies on the suppression of transmission of anthracnose with covering method and environment friendly agricultural materials (EFAM) in pepper field

  • We studies a model for management of pepper anthracnose based covering method and spraying system in field. 1. Among 82 organic fungicides, 42 materials showed most effective inhibition against mycelia growth of the Colletotrichum acutatum in vitro. 23 formulated biocontrol agents were chosen to control the disease from 42 biocontrol agents in greenhouse. In the end, five kinds (2 plant extracts, 2 biopesiticides, 1 Bordeaux mixture) were selected from 23 materials in the field. 2. The mulching materials of bed covering in fruit season were thin non-woven fabric sheet and black plastic. The use of a fabric sheet was reduced the spread of anthracnose as compared to the plastic covering. 3. The application with the chosen materials was reduced 34% of anthracnose for 7 times sprays to planting 70 days as compared to the untreated control. In yield, nonwoven fabric sheet with formulated biopesticides was increased 17% than black plastic. 4. This result indicated that the developed biocontrol strategy could be an effective and economic crop protection system in organic pepper cultivation field.

Application of Volatile Antifungal Plant Essential Oils for Controlling Pepper Fruit Anthracnose by Colletotrichum gloeosporioides

  • Anthracnose caused by Colletotrichum gloeosporioides has been destructive during pepper fruit production in outdoor fields in Korea. In vitro antifungal activities of 15 different plant essential oils or its components were evaluated during conidial germination and mycelial growth of C. gloeosporioides. In vitro conidial germination was most drastically inhibited by vapour treatments with carvacrol, cinnamon oil, trans-cinnamaldehyde, citral, p-cymene and linalool. Inhibition of the mycelial growth by indirect vapour treatment with essential oils was also demonstrated compared with untreated control. Carvacrol, cinnamon oil, trans-cinnamaldehyde, citral and eugenol were among the most inhibitory plant essential oils by the indirect antifungal efficacies. Plant protection efficacies of the plant essential oils were demonstrated by reduced lesion diameter on the C. gloeosporioides-inoculated immature green pepper fruits compared to the inoculated control fruits without any plant essential oil treatment. In planta test showed that all plant essential oils tested in this study demonstrated plant protection efficacies against pepper fruit anthracnose with similar levels. Thus, application of different plant essential oils can be used for ecofriendly disease management of anthracnose during pepper fruit production.

Evaluation on Red Pepper Germplasm lines (Capsicum spp.) for Resistance to Anthracnose Caused by Colletotrichum acutaum

  • We evaluated 209 Capsicum accessions from the National Agrobiodiversity of RDA, for their reaction against of anthracnose disease caused by Colletotrichum acutatum. Two hundred nine accessions of Capsicum were almost composed with Capsicum annuum var. annuum originated from 37 countries. The percentage of infection due to C. acutatum ranged from 11.1% to 100% on immature and matured pepper fruits at 21 days after inoculation. On immature fruits of pepper, one hundred seventy three accessions of tested pepper were found infected with C. acutatum. Out of the 173 accessions, eighty five accessions were susceptible to anthracnose with dark brown to black lesions and slowly increased in size at 21 days after inoculation. Only thirty six accessions showed resistance against C. acutatum. On mature fruits of pepper, one hundred ninety eight accessions were infected with C. acutatum at 21 days after inoculation. Twenty one accessions showed a resistance reaction against C. acutatum. The results of this work indicated that all of resistance accessions to C. acutatum were have a genetic potential for the resources of resistance can be further used in pepper breeding programme against anthracnose and also demand more detailed investigation in this.

Occurrence and Ecological Characteristics of Red Pepper Anthracnose (고추 탄저병의 발병 생태 특성)

  • Occurrence of anthracnose on red pepper fruit was examined at 6 red pepper-growing regions including Yecheon, Andong, Euisong, Cheongsong, and Ponghwa in Kyungbuk province, Korea in 1999. The disease occurred firstly June 13 at Cheongsong, progressed gradually thereafteri and increased rapidly from late August. The average disease incidence with mid September was 30.4%. There was no significant difference in disease incidence among the regions examined. Correlation coefficient analysis of the disease incidence with weather factors revealed that it was significantly related to relative humidity, precipitation, rainy days, and duration of continuous rainy days. Most fungal isolates from the diseased fruit were Colletotichum gloeosporioides. In the in field experiment, unmature fruit was somewhat more susceptible to the disease than mature fruit. In the field experiment, the disease occurred severely in open fields, having the disease incidence of 12.1%; however, no disease was observed in the rain-proof fields. The anthracnose on red pepper fruit developed earlier and more severely in the successive cropping field with red pepper than the field having other crops. Removal of infected plant debris also delayed and reduced the disease occurrence, suggesting that plant debris infested with the anthracnose fungus serve as a primary inoculum source which may govern the severity of the disease in the field.
