Studies on the Antioxidant and Whitening Effects of Cheongyang Pepper based on a Bibliometric Approach

  • Red pepper (Capsicum annuum L.) is one of the most consumed vegetable worldwide. In this study, we tried to suggest the possibility of Cheongyang pepper as a functional cosmetic material by identifying the physiological activity, especially antioxidant and whitening effects of Cheongyang pepper through bibliometric analysis and experimental studies. A bibliometric analysis was performed through co-word analysis of 8,892 papers retrieved from SCOPUS. 4 research fields were obtained by cluster mapping from VOSviewer software, among which we noted the antioxidant activity of extracts from Capsicum annum L.. Phenol as a useful ingredient of Cheongyang pepper was analyzed using HPLC (high-performance liquid chromatography). Antioxidant and whitening effects were evaluated by measuring DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity, hydrogen peroxide scavenging activity, and tyrosinase inhibitory activity. Cheongyang pepper extract showed contents of 0.106 ± 0.01 and 0.105 ± 0.02 mg/g, respectively, in the order of gallic acid and protocatechuic acid. The extract exhibited 56.95% DPPH scavenging activity and 43.97% hydrogen peroxide scavenging activity at a concentration of 1,000 ㎍/ml. In addition, 1,000 ㎍/ml of the extract inhibited tyrosinase activity by 52.44% and 42.61%, respectively in a whitening efficacy test using L-tyrosine and L-DOPA (L-3,4- dihydroxyphenylalanine) as substrates. These results suggest that Cheongyang pepper extract and its active ingredients have antioxidant and whitening effects, and the possibility of future development as a whitening functional cosmetic material.

Effect of Red Pepper [Capsicum annuum L.] on Growth Rate and Resting Metabolic Rate in Chickens (고추 [Capsicum annuum L.]가 병아리의 성장률(成長率)과 휴식시대사(休息時代謝)에 미치는 영향(影響))

  • The effect of administration of red pepper Capsicum annuuml on the growth rate and resting metabolic rate in male chickens was investigated while varying the dose of red pepper. Growth of the chickens fed the rations containing small amount of red pepper(0.1% or 0.5%) was significantly enhanced. In the chickens given the ration containing 1% of red pepper, which showed the result similar to that of control chickens fed the ration without red pepper, retarded growth was seen as compared with the growth of the chickens fed the rations containing small amount of red pepper. Chickens administered the rations containing large amount of red pepper (5%, 10%, 60%) showed significantly retarded growth as compared with the growth of control chickens. There was no effect on the resting metabolic rate of chickens when administered varying amount of red pepper. The oxygen of resting chickens was varied $9.0-10.5ml/kg^{0.75}/min$ and rectal temperature was varied within the normal range ($41-42^{\circ}C$).

High Frequency Embryogenesis and Plant Regeneration in Anther Cultures of Pepper (고추 약배양을 통한 고빈도 배발생 및 식물체 재분화)

  • Culture conditions for high frequency embryogenesis and plant regeneration in anther cultures of various F$_1$ hybrid and homozygous lines of pepper (Capsicum annuum L.) are described. Anthers pigmented less than halfway from the distal end were dissected from the flower bud in which petals elongated 2 mm higher than the receptacle. They were placed on Dumas medium supplemented with 0.1 mg/L 2,4-D and 0.1 mg/L kinetin. After four weeks of culture, embryos began to appear on anthers. After eight weeks of culture, frequencies of embryo formation reached up to 58.3%. Upon transfer to MS basal medium, greater than 95% of embryos developed into plantlets.

Isolation and Characterization of Pathogen-Inducible Putative Zinc Finger DNA Binding Protein from Hot Pepper Capsicum annuum L.

  • To better understand plant defense responses against pathogen attack, we identified the transcription factor-encoding genes in the hot pepper Capsicum annuum that show altered expression patterns during the hypersensitive response raised by challenge with bacterial pathogens. One of these genes, Ca1244, was characterized further. This gene encodes a plant-specific Type IIIA - zinc finger protein that contains two Cys$_2$His$_2$zinc fingers. Ca1244 expression is rapidly and specifically induced when pepper plants are challenged with bacterial pathogens to which they are resistant. In contrast, challenge with a pathogen to which the plants are susceptible only generates weak Ca1244 expression. Ca1244 expression is also strongly induced in pepper leaves by the exogenous application of ethephon, an ethylene releasing compound. Whereas, salicylic acid and methyl jasmonate had moderate effects. Pepper protoplasts expressing a Ca1244-smGFP fusion protein showed Ca1244 localizes in the nucleus. Transgenic tobacco plants overexpressing Ca1244 driven by the CaMV 355 promoter show increased resistance to challenge with a tobacco-specific bacterial pathogen. These plants also showed constitutive upregulation of the expression of multiple defense-related genes. These observations provide the first evidence that an Type IIIA - zinc finger protein, Ca1244, plays a crucial role in the activation of the pathogen defense response in plants.

  • Green peppers (Capsicum annuum L. cv, Soonjung) were grown under different combinations of $CO_2$ concentration and temperature levels and examined on the effect of elevated $CO_2$ and temeprature on plant growth, carbon and nitrogen concentrations. Plant height was stimulated by elevated $CO_2$ levels at 20.3 and 22.6$^{\circ}C$. Leaf area and fresh weight were remarkedly increased by high $CO_2$ concentration at 22.6$^{\circ}C$. Dry weights of leaf, stem, root, and whole plant were increased as temperature increased at 611 ppm $CO_2$, but those values decreased at 22.6$^{\circ}C$ in 397 ppm $CO_2$ concentration. Elevated $CO_2$ increased plant growth by 1.5 times at 20.5$^{\circ}C$ and 22.6$^{\circ}C$. C/N ratio increased with increasing temperature under elevated $CO_2$ levels.

Quality Characteristics of Doenjang Added with Red Pepper (Capsicum annuum L.) Seed (고추씨 첨가 된장의 품질 특성)

  • This study investigated the physico-chemical and sensory quality of Doenjang added with red pepper (Capsicum annuum L.) seed. The initial moisture content and salt content of Doenjang were 52.03-53.79% and 13.28-14.05%, respectively. The moisture and salt contents of Doenjang slightly decreased and increased, respectively, as fermentation periods increased. According to increasing fermentation periods, pH of Doenjang showed a little decreasing value although there were no difference between samples with various red pepper seed contents. On the other hand, titratable acidity of Doenjang increased as fermentation periods increased. Also, Doenjang added with red pepper seed had higher titratable acidity value than control Doenjang without red pepper seed. In the color of Doenjang, ‘L’ of lightness, ‘b’ of yellowness, and ‘a’ of redness were decreased as fermentation periods increased. Also, generally, increase of red pepper seed concentration resulted in higher values of redness (a), yellowness (b) and lightness (L) of Doenjang. Also, amino-type nitrogen content, which was 497-623 mg% in initial fermentation period samples, increased to 1,000 mg% for control and to 756~896 for red pepper added at the fermentation 120 days. In the total microbes, total cell count in the Doenjang samples was in the range of $10^7{\sim}10^8$ CFU/g regardless of fermentation periods. In contrast, yeast and mold number of Doenjang samples showed the range of $10^5$ CFU/g at the initial fermentation periods and decreased thereafter. In the sensory evaluation of Doenjang, unique Doenjang flavor increased regardless added red pepper seed as fermentation periods increased.

Inhibitory effects of Capsicum annuum L. water extracts on lipoprotein lipase activity in 3T3-L1 cells

  • Obesity, an intractable metabolic disease, currently has no medical treatment without side effects, so studies have been actively carried out to find natural compounds that have anti-obesity activity with minimum side effects. In this study, the anti-obesity effects of water extracts of seven Capsicum annuum L. varieties being Putgochu (Pca), Oyee gochu (Oca), Kwari putgochu (Kca), Green pepper (Gca), Yellow paprika (Yca), Red paprika (Rca) and Cheongyang gochu (Cca), were examined through the evaluation of lipoprotein lipase (LPL) mRNA expression level in 3T3-L1 cells (mouse pre-adipocytes). After capsaicin elimination by chloroform defatting, freeze-dried powder of Cca was treated to 3T3-L1 cells and anti-obesity effects were examined by determining the LPL mRNA level using the RT-PCR method. Of the primary fractions, only proven fractions underwent secondary and tertiary refractionating to determine anti-obesity effects. From seven different Capsicum annuum L., there was a significant decrease of the LPL mRNA expression level of 50.9% in Cca treatment compared to the control group. A significant decrease of the LPL mRNA expression level was shown in primary fractions (Fr) 5 (36.2% decrease) and 6 (30.5% decrease) of the Cca water extracts. Due to the impurities checked by UPLC chromatography, Fr 5 and 6 were refractionated to determine the LPL mRNA expression level. Treatment of Fr 6-6 (35.8% decrease) and Fr 5-6 (35.3% decrease) showed a significant decrease in the LPL mRNA expression level. When analyzed using UPLC, major compounds of Fr 6-6 and Fr 5-6 were very similar. Subsequently, we refractionated Fr 6-6 and Fr 5-6 to isolate the major peak for structure elucidation. Treatment of Fr 5-6-1 (26.6% decrease) and Fr 6-6-1 (29.7% decrease) showed a significant decrease in the LPL mRNA expression level. Consequently, the fractions may have a possibility to ameliorate obesity through the decrease of the LPL mRNA expression level.

The Effect of Fertigation Setting Point on the Growth and Fruit Quality of Sweet Pepper (Capsicum annuum L.) (관비재배에서 토양수분이 착색단고추의 생육과 품질에 미치는 영향)

  • Objective of this research was to investigate the effect of fertigation setting point such as -5, -10, -20, and -30 ㎪ on the growth and fruit quality of sweet pepper (Capsicum annuum L.) in greenhouse culture. The net $CO_2$ assimilation and transpiration rate were the lowest in the treatment of -30㎪. The pH and EC in soil solution were not severly affected by irrigation setting point and no statistical differences were observed among treatments of irrigation setting point tested. The N content of above ground plant tissue was the lowest in the treatment of -30 ㎪ and those of K, Ca, and Mg were the highest in the treatment of -10 ㎪. But that of P did not show statistical differences among treatments tested. As the fertigation setting point was getting low, the growth decreased at 60th day after planting, while there were no differences among treatments at 210th day after planting. The fruit quality except sugar contents did not show differences among treatments, but sugar contents was the highest in the treatment of -30 ㎪ with $8.0^{\circ}$Brix. Above results indicated that fertigation setting point should be in the range from -10 ㎪ to -20 ㎪ to ensure good crop growth and fruit quality in sweet pepper production.

Variation of Iron Content and Ferritin Distribution during Development Stage under Conditions of Iron Nutritional Status from Hydroponic Culture in Red Pepper ( Capsicum annuum L. )

  • Total iron content and ferritin distribution have been determined in red pepper(Capsicum annuum L.) during development stage under conditions of iron nutritional status from hydroponic culture. Color of the leaves become chlorotic on iron deficient and high concentration. The plant height on each iron concentration had retarding effect at concentration lower than $25\muM$ and greater than 125$25\muM$. In normal green leaves. Total iron content was almost constant with a mean value of $2.5\mumole$ of iron/mg of dry matter, except at 63day, for which it increases slightly to $4\mumole$. Howere, iron content of chlorotic plants grew on iron free medium was not almost detectable. Also in post chlorotic leaves(++Fe), iron content was evidently increase unitl 7days after transfer on liquid medium, but decreased from after 14days. Also, ferritin protein analysed total protein extracts prepared from leaves of different ages using antibodies raised against ferritin protein. Ferritin protein deereased progressively during the first week of germination and was not detectable in vegetative tissues. Ferritin protein in post chlorotic leaves wasevidently strongly cnhanced until 11days after transfer on liquid medium but decreased until the leves became chlorotic.

Quality Attributes of Korean Red Pepper According to Cultivars and Growing Areas (한국산 고추의 품종 및 재배지역에 따른 품질 특성)

  • Seven different cultivars of Korean red pepper, Capsicum annuum L., were collected from different growing areas and analyzed for pungent principles, redness, size and weight. Length of the pod varied in the range of $7.0{\sim}9.4\;cm$ total weight, $1.2{\sim}3.1\;g$, capsaicin content, $11.5{\sim}45.0\;mg%$, dihydrocapsaicin content, $8.5{\sim}35.1mg%$, and redness (absorbance at 460 nm), $6,270{\sim}7,810$ conventional color unit. Contents of capsaicins and redness were significantly different according to the cultivars but the two components were not correlated each other. According to the growing areas, contents of capsaicins and redness were significantly different. Capsaicins content was not significantly different but redness was significantly different between sun-drying and hot air-drying of the pods.
