Profiles of Toxin Genes and Antimicrobial Resistance of Bacillus cereus Strains Isolated from Commercial Jeotgal (시판 젓갈에서 분리한 Bacillus cereus의 독소 유전자 및 항균제 내성 분석)

  • Twenty-three Bacillus cereus strain isolated from commercial jeotgal were investigated for 11 toxin genes and susceptibility to 25 different antimicrobials. The hemolytic enterotoxins hblA, hblC, and hblD were detected in 13.0%, and non-hemolytic enterotoxins nheA, nheB, and nheC were detected in 26.1%, 100%, and 100% of the isolates, respectively. The positive rates of cytK, entFM, becT, hlyII, and ces were 73.9%, 60.9%, 26.1%, 8.7%, and 0.0%, respectively. According to the disk diffusion susceptibility test, all of the strains studied were resistant to cefuroxime, followed by cefoxitin (78.3%), oxacillin (78.3%), ampicillin (69.6%), penicillin G (69.6%), and amoxicillin (65.2%). However, all the strains were susceptible to 11 other antimicrobials, including amikacin, chloramphenicol, and ciprofloxacin. The average minimum inhibitory concentrations of amoxicillin, ampicillin, and cefuroxime against B. cereus were 462.9, 235.0, and 135.0 ㎍/mL, respectively. These results highlight the need for sanitizing commercial jeotgal, and provide evidence to help reduce the risk of jeotgal contamination by antimicrobial-resistant bacteria.

  • Bacteria have been regarded as major etiolgic factors in root canal infections. Infected root canal flora from thirteen patients who had visited to conservative department of Wonkwang dental hospital were cultured on blood agar plates. Cultued microorganisms were isolated and identified with Gram stain and biochemical tests using Vitek Systems(BioMeriux, MO, USA); Antibiotic susceptibillity was performed with disk diffusion and broth microdilution using Vitek Systems. Gram positive cocci(65 %) were predominant, which were composed of 6 Streptococcus viridans group, 5 Staph. spp., and 4 Enterococcus faecium, in the isolatd 23 strains. Gram negative rods (26 %) were the next common bacteria, which were composed of 5 non - fermentative Gram negative rods, and 1 Enterobacter cloacae. Most strains of S. viridans group and E. faecium were susceptible to antibiotics including penicillin. But strains of Staphylococcus spp. and non - fermentative Gram negative rods showed marked resistance to antibiotics except tetrancyclin and cefotaxime. Most results between disk diffusion and microdilution were all agreed, but the results of non - fermentative Gram negative rods were susceptible to cefotaxime in disk diffusion method but resistant in microdilution.

Isolation, Identification and Drug Susceptibility of Bacteria from Genital Organs of Slaughter Sows (도축돈의 생식기내 세균분리 동정 및 약제함수성시험)

  • This study was conducted to determine the microorganisms inhabitating in sow genital organs and their anti-microbial drug susceptibility During the period between February, 1991 and November 1991, 128 sow genital organs were sampled at six abattoirs. Gross pathological examination and bacterial isolation and identification were performed from the genital organ. In addition, antimicrobial drug susceptibility for the major organisms isolated were examined. 1. Among the bateria isolated from normal genital organs, E. coli(30.7%) Stahylococcus spp.(29.4%), Corynebarterium pyogenes(C. pyogenes) (14.7%), Streptococcus spp.(13.3%) were most freqently isolated, whereas the genera of Klebsiella, Actinobacillus, and Serratia were detected less freqently. 2. Among the bacteria isolated from abnormal genital organs, C. pyogenes,(37.7%), Stahylococcus spp.(30.2%), Proteus spp. (26.4%) , Pasteurella spp. (18.9%) , Steptococcus spp. (9.4%) were most freqently isolated whereas the genera of Pseudomonas, Serratia and Klebsiella were detected less freqently. 3. From sow genital organs showing lesion of endometritis and purulent endometritis C. pyogenes were most freqently isolated, the isolation rate being 67.7% and followed by Stahylococcus spp., E. coli, Proteus spp., Steptococcus spp. and Pasteurella spp. in the order. 4. Antimicrobial drug susceptibility of the major organisms showed that all the isolates were susceptible to cephalothin, ampicillin, chloramphenicol and sulfamethoxazole / trimethoprim, but resistant to penicillin and streptomycin.

Drug Resistance of Bacterial Flora Isolated from Kunsan Bay (군산 내만에서 분리된 세균총에 대한 약제 내성 조사)

  • One hundred and twenty three strains of bacterial flora collected from Kunsan bay and examined for drug resistance to 9 antibiotics. The isolated and examined bacteria were Vibrio spp.(44 strains), Pseudomonas spp.(42 strains), Aeromonas spp.(26 strains), Moraxella spp.(9 strains), Enterobacteria spp.(6 strains), Bordetella spp.(3 strains), Alkaligenesis spp.(3 strains), Staphylococcus spp.(3 strains), and Flavobacterium spp.(2 strains). The drugs used were Ampicillin(AM), Penicillin-G(PM), Rifampicin(RF), Streptomycin(SM), Oxolinic acid (OA), Nalidixic acid(NA), Oxytetracycline(OT), Amikacin(AK), and Enorfloxacin(EF). Forty two strains were found to be sensitive to all drugs. The remaining strains showed resistance to various combinations of drugs. Among the resistant strains were mostly restricted to AM(54 strains/43.9%), PM(47 strains/38.2%), RF(35 strains/28.4%), SM(9 strains), OA(5 strains/ 4.06%), and NA(1 strains/0.8%), in combination at high degree showing 15 different drug resistant patterns. The most frequently showed resistant patterns were AM-PM-RF(16 strains/13.4%), AM-PM(8 strains/6.5%), and PM-RF(7 strains/5.6%). These results suggested that Kunsan bay were contaminated with various strains of highly resistant strains to drugs(AM, PM and RF). These results suggest that high levels of various antibiotics have already been introduced to Kunsan bay. Furthermore it seems that chemotherapy of fish disease has become extremely difficult because of the acquirement of multi-drug resistance to wide range of antibiotics.

A monitoring survey on antimicrobial resistance of bacterial isolates from companion dogs in Incheon (인천지역 반려견 유래 세균의 항생제 내성 모니터링 조사)

  • This study was performed to investigate antimicrobial resistance in bacterial isolates obtained from companion dogs in veterinary hospitals and an animal shelter in Incheon. Drug resistance was examined respectively with the isolates of Escherichia coli, Enterococcus faecalis, and Staphylococcus pseudintermedius. The prevalence of drug resistance was calculated for each bacterial species towards 163 E. coli isolates, 156 E. faecalis isolates, and 86 S. pseudintermedius isolates by using selected antimicrobials. E. coli isolates were highly resistant to ampicillin, ciprofloxacin and tetracycline (47.9%, 28.2% and 28.2%, respectively). E. faecalis isolates were highly resistant to quinupristin-dalfopristin, tetracycline, kanamycin, rifampicin (69.8%, 66.0%, 53.8% and 51.9%, respectively). Higher levels of resistance were detected for ampicillin, penicillin, tetracycline, erythromycin, trimethoprim/sulfamethoxazole, telithromycin in S. pseudintermedius isolates (83.7%~52.6%, respectively). Occurrence of methicillin-resistant S. pseudintermedius (MRSP) was confirmed by oxacillin disc diffusion method, resulted in 23.3% occurrence among the S. pseudintermedius isolates (20/86 strains). The occurrence ratio of multidrug-resistance in the isolates of E. coli, E. faecalis, and S. pseudintermedius was 34.5%, 56.9%, and 67.9%, respectively. In this study, higher levels of antimicrobial drug resistance were observed in bacterial isolates obtained from dogs in Incheon. A regular monitoring and surveillance program should be implemented to prevent the emergence and spread of the drug-resistant bacteria carried in companion dogs.

Characteristics of Staphylococcus aureus Isolated from Patients with Diarrhea. (설사 환자에서 분리된 Staphylococcus aureus의 특성)

  • The major causative bacteria of food poisoning were Salmonella spp. $(35.6\%)$, Staphylococcus aureus $(11.3\%)$ and Vibrio parahaemolyticus $(3.2\%)$ in our country. In this study we attempted isolation of S. aureus from stools of patients with diarrhea. Sixty-four strains $(9.1\%)$ were isolated from 704 the stools of patients with diarrhea. The enterotoxin was detected from 29 isolates $(45.3\%)$: 24 isolates $(37.5\%)$, 3 isolates $(4.7\%)$ and 2 isolates $(3.1\%)$ were A, B and C type, respectively. In the antibiotic susceptibility, 63 isolates $(98.4\%)$ were resistant to penicillin, 60 isolates $(93.8\%)$ to ampicillin, 35 isolates $(54.7\%)$ to erythromycin, 32 isolates $(50.0\%)$ to gentamycin, 22 isolates $(34.4\%)$ to tetracycline and 20 isolates $(31.3\%)$ to oxacillin. All of S. aureus isolates were susceptible to chloramphenicol and vancomycin, 20 isolates $(31.3\%)$ were methicillin-resistance S. aureus (MRSA). MRSA isolation rate was higher in male $(35.7\%)$ than female $(26.3\%)$. With the exception of two isolates which were resistant only to penicillin, sixty-one isolates were multiple antibiotic resistance.

Antibiotic Resistance Patterns of Enterococcus spp. Isolated From Commercial Frozen Foods (시판 냉동식품에서 분리한 장구균의 항생제 내성 양상)

  • From April to December in 2009, microbial investigation is accomplished for 100 frozen foods asked to microbial control team that corresponds with total aerobic viable bacteria, coliform group, Escherichia coli, Enterococcus spp. and antibiotic resistance patterns of Enterococcus spp. isolates are investigated. Average of total erobic viable bacteria numbers is $4.3{\times}10^4CFU/g$. Average of coliform group numbers is $4.3{\times}10^3CFU/g$. Average f Enterococcus spp. numbers is $1.8{\times}10^3CFU/g$. Escherichia coli from 100 frozen foods is not detected and detection ate is 0.0%. 22 Enterococcus spp. are isolated from 100 frozen foods. 12 of 22 Enterococcus spp. strains are identified as E. faecium. 7 of 22 Enterococcus spp. strains are identified as E. faecalis. 2 of 22 Enterococcus spp. trains are identified as E. gallinarum. 1 of 22 Enterococcus spp. strains is identified as E. hirae. Enterococcus spp. solates show a high resistance to erythromycin, rifampin, tetracycline, ciprofloxacin, chlorampenicol, penicillin and susceptibility to vancomycin, ampicillin, gentamicin, strepomycin, linezolid. 15 of 22 Enterococcus spp. strains are multi-resistant and the most frequent multi-resistant pattern is erythromycin-rifampin for 6 Enterococcus spp. strains.

Prevalence and Toxin Genes of Food-Borne Pathogens Isolated from Toothbrush in Child Care Center (보육시설 유아 사용 칫솔의 식중독 미생물 분포 및 독소 유전자)

  • This study was performed to investigate the microbiological contamination on toothbrushes, toothbrush caps, and tooth cleaning cups in the child care centers and to evaluate the toxin genes, toxin production ability and antibiotic resistance of food-borne pathogens. The average number of total aerobic bacteria and fungi were 5.3 log CFU and 3.2 log CFU. Coliform bacteria were detected in 41 (54.7%) of 75 toothbrushes, 13 (44.8%) of 29 toothbrush caps, and 29 (44.6%) of 65 tooth cleaning cups. Salmonella spp. was not detected in all of samples but Bacillus cereus was isolated from 1 (1.3%) of 75 toothbrushes and 2 (3.1%) of 65 tooth cleaning cups. Staphylococcus aureus was detected in 1 (1.5%) of 65 tooth cleaning cups. The nheA, nheB, nheC, hblC, hblD, hblA and entFM toxin genes were possessed in B. cereus isolated from toothbrush which also produce NHE and HBL enterotoxins. S. aureus was resistant to ampicillin and penicillin, while B. cereus was resistant to ${\beta}-lactam$ antibiotics. These results indicated that the sanitary conditions of toothbrushes and tooth cleaning cups in the child care centers should be improved promptly. The UV sterilization after drying and then storage in dried condition is required to improve the sanitary condition of toothbrushes and tooth cleaning cups in the child care center.

Antimicrobial Susceptibility of Gram-Negative Bacteria from Dogs and Cats (개와 고양이에서 분리된 그람음성균의 항생제 감수성 양상)

  • The purpose of this study was to determine the distribution of gram-negative bacteria isolated from companion animals with sepsis, and to investigate the antimicrobial susceptibility patterns of the isolates. Bacterial pathogens were isolated from specimens of dogs and cats submitted to National Veterinary Research and Quarantine Service between 2008 and 2009. A total of 44 gram-negative pathogens were isolated from necropsied organs. The most common isolates were E. coli (n = 33), K. pneumoniae (n = 4) and B. bronchiseptica (n = 4). Most of gram-negative isolates were susceptible to ceftiofur (68.2%), colistin (84.1%), florfenicol (84.1%) and spectinomycin (61.4%). Most of those were resistant to ampicillin (77.3%), erythromycin (86.4%), flumequine (65.9%), lincomycin (97.7%), oxytetracycline (61.4%), penicillin (100%), streptomycin (63.6%), spiramycin (97.7%), sulfamethoxazole (90.9%), tylosin (97.7%) and tiamulin (100%). In conclusion, colistin and florfenicol could be useful against sepsis due to gram-negative bacteria.

Induction of $\beta$-Lactamase on $\beta$-Lactam Antibiotics Resistant Bacterium ($\beta$-Lactam계 항생물질 내성균주의 $\beta$-Lactamase 생산의 유도)

  • Bacillus subtilis J105, high resistant bacteria against $\beta$-lactam antibiotics, become higher resistant through induction of $\beta$--lactamase in the presence of $\beta$--lactam antibiotics. When there is no antibiotics in medium, the production of resistance-inductive $\beta$--lactamase reached its plateau 15 hours later. But when there is ampicillin (500$\mu\textrm{g}$/$m\ell$) in medium, the production of enzyme reached its plateau 25 hours later since cultivating bacteria, whereas it is found that enzyme 2,900 units/$m\ell$ about 20 times as much as compared with not-presence of antibiotics was actived. In addition, as the result of MIC comparing applying ampicillin-treated and non-treated strain MIC of ampicillin-treated strain is about 2~27 times higher. It is considered that this strain induce $\beta$--lactamase production by ampicil-lin-treatment, then increasing it resistance. It is found that this resistant strain induce $\beta$--lactamase production against cephalosporin antibiotics as well as peni-cillin. As the result of examining the time of adding antibiotics for each phase of growth, it is concluded that logarith-mic phase is the most effective. As the aboves, it is suggested that this strain is a peculia strain that its resistance is induced high by various $\beta$--lactam antibiotics.
