Changes of Properties in Cinnamon Extracts Prepared by Enzyme Hydrolysis and Addition of Salts, Sugars and Antioxidant Synergists (효소분해와 염과 당 및 항산화 작용 상승제의 첨가에 의한 계피 추출액의 특성 변화)

  • The dried cinnamon was extracted with enzymes, salts, sugars and additives in order to find the most effective extraction material. Enzymatic hydrolysis of cinnamon suspension with cellulase, hemicellulase, pectinase, ${\beta}-1.4-glucosidase$, tannase and lipase showed a little increase of their cinnamic aldehyde contents. Solid yield, antioxidant activity and degree of browning were increased in hemicellulase treatment. Acid and alkali extraction of cinnamon showed a some increase in solid yields and antioxidant activity was increased by addition of glucose and Na-ascorbate. Cinnamic aldehyde contents and degree of browning were increased in extraction with Na-citrate addition.

  • Soft-rot bacterial pathogen, Erwinia sp., was isolated from chinese cabbage tissue showing soft-rot symptom. This bacterial strain caused soft-rot to chinese cabbage and potato, and identified as Erwinia chrysanthemi PY35(Ech PY35). Ech PY35 have extracellular CMCase, pectinase, pectate lyase, and protease activity, but not hemicellulase activity. The results of the microscopy showed that chinese cabbage tissue and potato tissue were macerated by infection of Ech PY35. In analysis of the CMCases activity in the total protein of Ech PY35, three CMCases were detected as intracellular protein while two CMCases were as extracellular protein by CMC-SDS-PAGE direct stain method.

Quality Characteristics and Antioxidant Effect of Sugar Preserved Wild Peach (Prunus persica L.) Juice by Enzymatic Treatment (효소 처리에 따른 야생복숭아(Prunus persica L.) 당절임액의 품질 특성 및 항산화 효과)

  • This study was to investigate the effects of enzyme on quality characteristics and antioxidant activities of wild peach (Prunus persica L.) juice. pH levels and S.S (soluble solid) values in all samples ranged from 3.86 to 4.13 and from 48.0 to $55.0^{\circ}Brix$, respectively. The TA (total acidity) values of control (not treatment enzyme) were higher than those of the others. The highest 'L', 'a' and 'b' values were observed on PWP (preserved wild peach (Prunus persica L.) juice of cellulase/pectinase (1:1, w/w) sample. Glucose (26.65 g/100 g) and fructose (17.42 g/100 g) in PWP product were determined, however sucrose and maltose were not detected. The predominating organic acid components analyzed in PWP sample were tartaric (32.36 g/100 g) and lactc acids (209.34 g/100 g), whereas citric acid, acetic acid and malic acid were not detected. Higher scores for taste, flavor, color and overall acceptance were found for PWP products compared to other samples. The total phenolic content (13.31 mg GAE/mg dry weight) analyzed using Folin-Ciocalteu's reagent, of PWP sample was higher than those of the others and the total flavonoid concentrations were also 10.95 mg CE/mg dry weight. The DPPH radical scavenging and ABTS radical scavenging activities in all samples ranged from 55.16 to 74.29% and from 39.59 to 82.79%, respectively. The antioxidant activities were affected by addition of enzyme. These results indicate that the use of the mixture of cellulase and pectinase could increase the quality, and antioxidant potentials of sugar preserved wild peach (Prunus persica L.) juice by enzymatic treatment.

  • The yield, viability and continuous culture of isolated Italian ryegrass protoplasts were investigated. The effects of cold treatment (4^{\circ}C.$) for 7 days and basic LS medium supplemented with 5mg/l $AgNO_3$ showed effectively on embryogenic callus induction and regeneration responses of immature and mature embryos or young inflorescences subcultured every 4 weeks on basic medium. The optimum combinations of growth regulator on the regeneration responses was 0.2mg/l BAP and 2mg/l 2, 4-D. Calli induced inflorescences were suspended in its liquid medium for 5 days before enzyme treatment. Maximum protoplast yield and viability were obtained after digestion in enzyme solution contained 4% cellulase R10. 2% macerozyme and 2% pectinase in 0.6M mannitol. Cell division and microcalli development were observed in isolated protoplasts cultured in agarose culture of KM8P medium.

A Study on the Extraction of Thermostable Pectinesterase from Valencia Orange (Valencia 오렌지로부터 내열성 Pectinesterase의 추출)

  • Low yield of a thermostable pectinesterase(TSPE) from citrus fruits has made its detailed study extremely tedious and difficult; therefore, maximizing TSPE extraction is desirable. It is assumed that TSPE is bound to the cell components via ionic linkage and covalent bonds. Therefore, in this study, variations in extraction time, pH, NaCl concentration and commercial enzyme preparations were used to increase the yield of TSPE from Valencia orange. The largest recovery of TSPE, obtained by heating extracted pectinesterase(PE) at $70^{\circ}C\;for\;5{\sim}10$ minute, was achieved using actate buffer(pH 4.14) with 1 M NaCl and 0.2% $Cytolase^{TM}$ 104(a mixture of cellulase, hemicellulase and pectinase; Genecor, Inc). The two aquous phase partitioning with 5.0% Triton X-114 could be used as a tool for separation of thermolabile pectinesterase(TLPE) and TSPE from crude PE. Also, water extraction and $0{\sim}0.3$ ammonium sulfate fractionation could be used for removing non-pectinesterase protein.

Selective Removal of Protease from Soybean Cell Wall Degrading Enzyme Complex Isolated from Aspergillus niger CF-34 (Aspergillus niger CF-34로부터 분리한 대두세포벽분해효소 복합체 중의 Protease의 선택적인 제거)

  • By exposing the complex enzyme solution to alkaline condition, it was possible to remove the protease activity selectively without inactivation of soybean cell wall degrading activity of the crude enzyme complex produced by Aspergillus niger CF-34. Optimum reaction conditions were as follow. pH was $9.0{\pm}0.1$, temperature was $20^{\circ}C$ and reaction time was 30 min with gentle stirring. Over 90% of protease activity could be eliminated while the activities of pectinase, polygalacturonase, xylanase, carboxymethyl cellulase and soybean cell wall degrading enzyme were maintained to $80{\sim}100%$. Through alkali treatment, it was discovered that the quality and organoleptic properties of soy protein produced by this enzymes were improved because the hydrolysis of protein and formation of bitter peptide were decreased.

Characterization of Tofu-Residue Hydrolyzing Carbohydrase Isolated from Aspergillus niger CF-34 (Aspergillus niger CF-34로부터 분리한 두부 또는 두유비지 가용화 복합효소의 특성)

  • Enzymatic solubilization of tofu-residue was attempted using carbohydrase isolated from Aspergillus niger CF-34. Tofu-residue, by-product of tofu manufacture or soymilk processing was used as the model for plant cell wall. It was found that tofu-residue was rich in nurients: 46.7% carbohydrate, 32.8% protein, the rest being lipid and ashes. Carbohydrate component of tofu-residue consisted of 36.8% cellulose and 62.6% hemicellulose. The carbohydrase was found to consist of pectinase, xylanase, PGase, CMCase, and SFase when tofu-residue and pectin were used as the carbon source. Enzyme induction was maximum at 7days of culture. Optimum reaction pH was 4.0, temperature $50^{\circ}C$. The enzyme was stable to $50^{\circ}C$, above which the stability decreased rapidly.

Processing of Powdered Jujube Juice by Spray Drying (분무건조에 의한 분말대추음료의 가공)

  • Spray dried powdered juice was processed from concentrated extract of jujube(Zizyphus jujuba MILLER). Spray drying of the extract solution could not be operated to have powder product by itself over whole concentration range and required addition of some carrier or support material. The concentrated extract of 26$^{\circ}$Bx was combined with carrier material solution to have a final concentration of 30$^{\circ}$Bx, and then spray dried. Proper addition level of carrier solid for physical and flavor quality of the powder product was determined to be 1 : 1 ratio to jujube solid. Combined use of maltose and gum arabic produced the best quality product among the studied carrier materials, which were maltose, dextrin, condensed milk and gum arabic. Enzymatic treatment in extraction process could increase the yield by 13~39%, but hurt the sensory quality of powdered juice. Treatment by 0.5% pectinase(0.05 unit/ml) may be used with lesser quality change for improved yield.

  • This study was to investigate the physiological activities of bioconversion soybean embryo extract and the effects of sleeping pack containing extract on skin. The aglycone isoflavones of the soybean embryo extract bioconversed with pectinase were identified by HPLC. in vitro tests showed that they were non-toxic, anti-inflammatory, anti-melanin and anti-aging activity. After 4 weeks of human body application with the sleeping pack using extract, the skin surface roughness and eye wrinkles were so improved by low stimulation that the sleeping pack containing bioconversion soybean embryo extract has a positive effect on the skin. We confirmed that the bioconversion soybean embryo extract was effective in enhancing the physiological activity and was useful as a cosmetic material of various formulations.

Oral administration of hydrolyzed red ginseng extract improves learning and memory capability of scopolamine-treated C57BL/6J mice via upregulation of Nrf2-mediated antioxidant mechanism

  • Background: Korean ginseng (Panax ginseng Meyer) contains a variety of ginsenosides that can be metabolized to a biologically active substance, compound K. Previous research showed that compound K could be enriched in the red ginseng extract (RGE) after hydrolysis by pectinase. The current study investigated whether the enzymatically hydrolyzed red ginseng extract (HRGE) containing a notable level of compound K has cognitive improving and neuroprotective effects. Methods: A scopolamine-induced hypomnesic mouse model was subjected to behavioral tasks, such as the Y-maze, passive avoidance, and the Morris water maze tests. After sacrificing the mice, the brains were collected, histologically examined (hematoxylin and eosin staining), and the expressions of antioxidant proteins analyzed by western blot. Results: Behavioral assessment indicated that the oral administration of HRGE at a dosage of 300 mg/kg body weight reversed scopolamine-induced learning and memory deficits. Histological examination demonstrated that the hippocampal damage observed in scopolamine-treated mouse brains was reduced by HRGE administration. In addition, HRGE administration increased the expression of nuclear-factor-E2-related factor 2 and its downstream antioxidant enzymes NAD(P)H:quinone oxidoreductase and heme oxygenase-1 in hippocampal tissue homogenates. An in vitro assay using HT22 mouse hippocampal neuronal cells demonstrated that HRGE treatment attenuated glutamate-induced cytotoxicity by decreasing the intracellular levels of reactive oxygen species. Conclusion: These findings suggest that HRGE administration can effectively alleviate hippocampus-mediated cognitive impairment, possibly through cytoprotective mechanisms, preventing oxidative-stress-induced neuronal cell death via the upregulation of phase 2 antioxidant molecules.
