Purification of Anti-allergic Compound from Mori Cortex Radicis Extract (상백피 추출물 중의 항알러지 활성 성분의 분리)

  • Mori Cortex Radicis, the root bark of mulberry tree, has been used in the treatment of bronchial asthma and other lung diseases in traditional medicine. There was a recent repor t that the water soluble part with molecular weight of above 10,000 has anti-allergic activity. Therefore, we intended to isolate and purify the anti-allergic compound from hot water extract of the Mori Cortex Radicis. Crude extract of Mori Cortex Radicis was prepared by hot-water extraction, and anti-allergic compound was further purified by alcohol precipitation, successive ultrafiltration, anion exchange chromatography and gel filtration chromatography. This compound had homogeneity which was shown by the sharp single peak in HPLC chromatogram (TSK-GEL G400OPW column, RI detector). The molecular weight of the compound was estimated as 23Kda on the basis of calibration curve plotted against protein standards. This compound was identified as complex of sugar, protein and lignin (19.2: 5.9: 72.7), and proteolysis could not decrease the anti-allergic activity but mild delignification decreased the activity remarkably. Therefore, we concluded that the anti-allergic compound of Mori Cortex Radicis was a lignin-carbohydrate complex.

  • A simple, reproducible, and rapid gas chromatographic method for putrefactive metabolite determination in feces was developed. The method involves the direct injection of fecal supernatants into the gas chromatograph, without pretreatment. The mass spectra of these metabolites were obtained using an HP 5971 mass selective detector operated in electron impact (EI) ionization mode. This method produced sharp peaks and allowed the simultaneous determination of fecal putrefactive metabolites.

Analysis of On-Line Partial Discharge Patterns in High Voltage Motor Stator Windings (고압전동기 고정자 권선의 운전중 부분방전 패턴 분석)

  • During normal machine operation, partial discharge(PD) measurements were performed with turbine generator analyzer(TGA) in two high voltage motors. Two 6.6kV motors were installed with 80pF capacitive couplers at the terminal box. The PD patterns were displayed two dimensional and three dimensional. TGA summarizes each plot with two quantifies such as the normalized quantity number(NQN) and the peak PD magnitude(Qm). Off-line PD measurements were conducted on one 4.16kV motor. The motor was energized to 2.4kV, 3.0kV, 3.5kV and 4.16kV, respectively. The PD levels in pC were measured with a conventional digital PD detector. The comparison of positive to negative PD indicates whether the defect elements of PD are within the insulation or on the insulation surface. Discharge at conductor surface was discovered in No. 1 motor. Internal discharges were generated in phase A, B and C of No. 2 motor, Slot discharges occurred in three phases of No. 3 motor.

  • Partial discharge(PD) tests are used to evaluate the insulation condition of stator windings in two 4.16kV and three 6.6kV motors. These tests were conducted using a conventional partial discharge detector(PDD) and turbine generator analyzer(TGA). Off-line PD measurements were performed on five high voltage motors. PD magnitudes ranged from 1000 pC to 5400 pC at the normal line-to-ground voltage. Five high voltage motors have been equipped with 80pF epoxy-mica coupler on the motor terminal box. The PD pulse from sensors were measured with the TGA instrument. TGA summarizes each plot with two Quantities such as the peak PD magnitude(Qm) and the total PD activity(NQN). The defect mechanisms of high voltage motor can be associated with PD patterns such as internal, slot and conductor surface discharges. The PDD and TGA test results of No. 4 motor showed that internal discharge was detected in voids of the groundwall insulation.

Investigation of the relationship between earthquakes and indoor radon concentrations at a building in Gyeongju, Korea

  • This article measured and analyzed the indoor radon concentrations at one university building in Gyeongju, Republic of Korea, to investigate if there is any relationship between earthquakes and indoor radon concentration. Since 12 September 2016, when two 5.1 and 5.8 magnitude earthquakes occurred, hundreds of aftershocks affected Gyeongju until January 2017. The measurements were made at the ground floor of the Energy Engineering Hall of Dongguk University in Gyeongju over a period between February 2016 and January 2017. The measurements were made with an RAD7 detector on the basis of the US Environmental Protection Agency measurement protocol. Each measurement was continuously made every 30 minutes over the measurement period every month. Among earthquakes with 2.0 or greater magnitude, the earthquakes whose occurrence timings fell into the measurement periods were screened for further analysis. We observed similar spike-like patterns between the indoor radon concentration distributions and earthquakes: a sudden increase in the peak indoor radon concentration 1-4 days before an earthquake, gradual decrease before the earthquake, and sudden drop on the day of the earthquake if the interval between successive earthquakes was moderately longer, for example, 3 days in this article.

An integrated DNA barcode assay microdevice for rapid, highly sensitive and multiplex pathogen detection at the single-cell level

  • Here we report an integrated microdevice consisting of an efficient passive mixer, a magnetic separation chamber, and a capillary electrophoretic microchannel in which DNA barcode assay, target pathogen separation, and barcode DNA capillary electrophoretic analysis were performed sequentially within 30 min for multiplex pathogen detection at the single-cell level. The intestine-shaped serpentine 3D micromixer provides a high mixing rate to generate magnetic particle-pathogenic bacteria-DNA barcode labelled AuNP complexes quantitatively. After magnetic separation and purification of those complexes, the barcode DNA strands were released and analyzed by the microfluidic capillary electrophoresis within 5 min. The size of the barcode DNA strand was controlled depending on the target bacteria (Staphylococcus aureus, Escherichia coli O157:H7, and Salmonella typhimurium), and the different elution time of the barcode DNA peak in the electropherogram allows us to recognize the target pathogen with ease in the monoplex as well as in the multiplex analysis. In addition, the quantity of the DNA barcode strand (~104) per AuNP is enough to be observed in the laser-induced confocal fluorescence detector, thereby making single-cell analysis possible. This novel integrated microdevice enables us to perform rapid, sensitive, and multiplex pathogen detection with sample-in-answer-out capability to be applied for biosafety testing, environmental screening, and clinical trials.

Flavoral Essential Oil Components in the Stems of Agastache rugosa for Aromatherapy (아로마테라피용 배초향(Agastache rugosa) 줄기의 방향성 정유 성분)

  • This study was carried out to investigate the flavoral essential oil components in the stems of Agastache rugosa. These components were analyzed using gas chromatography-mass selective detector (GC-MSD). The stems of Agastache rugosa were contained alcohols, aldehydes, ketones, fatty acid esters, and terpenoids. The peak area (%) of estragole was highest among its oil components and the next were pulegone and menthone. The terpenoid alcohols found were 1-octen-3-ol, chavicol, spatulenol, 3-hexen-1-ol, 2-cyclohexen-1-ol, methyl eugenol, and octaethyllene glycol. The stems also contained ketones such as pulegone, menthone, cis-isopulegone, 2-cyclohexene-1-one, 3-octanone, 1-cyclohexanone, isoindole-1-one, t-ionone, inden-2-one, as well as the aldehydes of 4-methoxycinnam and benzaldehyde. The following esters were also detected 1-isopulegone-3-yl acetate, caryophyllene oxide, acetate and benzendicarboxylic acid ester. The terpenoids in the stems were identified as caryophyllene, limonene, cyclohexasiloxane-D, germacrene-D, anethole, cadinene, muurolene, and bourbonene. Overall Agastache rugosa contained several functional oil components including phenylpropanoids and terpenoids as flavoral essential oil components for natural aromatherapy.

CMI Tolerant Readout IC for Two-Electrode ECG Recording (공통-모드 간섭 (CMI)에 강인한 2-전극 기반 심전도 계측 회로)

  • This study introduces an efficient readout circuit designed for two-electrode electrocardiogram (ECG) recording, characterized by its low-noise and low-power consumption attributes. Unlike its three-electrode counterpart, the two-electrode ECG is susceptible to common-mode interference (CMI), causing signal distortion. To counter this, the proposed circuit integrates a common-mode charge pump (CMCP) with a window comparator, allowing for a CMI tolerance of up to 20 VPP. The CMCP design prevents the activation of electrostatic discharge (ESD) diodes and becomes operational only when CMI surpasses the predetermined range set by the window comparator. This ensures power efficiency and minimizes intermodulation distortion (IMD) arising from switching noise. To maintain ECG signal accuracy, the circuit employs a chopper-stabilized instrumentation amplifier (IA) for low-noise attributes, and to achieve high input impedance, it incorporates a floating high-pass filter (HPF) and a current-feedback instrumentation amplifier (CFIA). This comprehensive design integrates various components, including a QRS peak detector and serial peripheral interface (SPI), into a single 0.18-㎛ CMOS chip occupying 0.54 mm2. Experimental evaluations showed a 0.59 µVRMS noise level within a 1-100 Hz bandwidth and a power draw of 23.83 µW at 1.8 V.

Optimization of Enzymatic Synthesis Condition of Structured Lipids by Response Surface Methodology (반응표면분석에 의한 기능성 유지의 효소적 합성 조건 최적화)

  • Synthesis conditions were optimized using response surface methodology for producing structured lipids (SL) by interesterification of DHA-enriched algae oil derived from microalgae, Schizochytrium sp. and corn oil. Reaction was performed fer 24 hr at $55^{\circ}C$ catalyzed by immobilized lipase from Rhizomucor miehei (RM IM) in shaking water bath. Major fatty acids of SL were palmitic (21.70 mol%), oleic (20.20 mol%), and linoleic (27.34 mol%) acids, and DHA (15.06 mol%). To separate newly synthesized SL-triglycerides (TG) species, HPLC with evaporative light scatting detector (ELSD) was used. Production conditions were optimized using central composite design with reaction temperature $(35-75^{\circ}C,\;X_1)$, reaction time $(2-42\;hr,\;X_2)$, and enzyme concentration $(2-14%,\;X_3)$ as variables. When variables were $70.28^{\circ}C\;(X_1),\;28.74\;hr\;(X_2),\;and\;11.30%\;(X_3)$, maximum content of selected three peaks of synthesized SL-TG species was predicted as 6.97 area%.

Quantitative Evaluation of the Semi-Actuated Signal Control Systems (반감응 신호제어의 정량적 효과 평가에 관한 연구)

  • This study evaluated the quantitative effects of the deployment of semi-actuated signal systems using field data. For this, a semi-actuted signal system was deployed in the regional roadway network extensively. This paper investigated an operating strategy of semi-actuated signal systems for field application, and implemented the functional strategy into the standard signal controller. The performance was evaluated using three measures of effectiveness such as traffic volume, travel time, and the number of delayed vehicle. From the analysis results, traffic volume increased about 9.4% and 11.3% for morning and evening peak periods, respectively. The average travel time was reduced about 6.3% and 7.8% during morning and evening peak periods, respectively because of the expansion of bandwidths for major streets. In addition, the number of delayed vehicles was reduced about 36.4% and 23.9% for morning and evening peak periods, respectively. It is expected that the effectiveness of signal control system can be improved by incorporating a properly designed semi-actuated signal system in regional roadways with directional demand variation.
