Three parathion-degrading bacteria and eight pairs of bacteria showing syntrophic metabolism of parathion were isolated from rice field soils, and their genetic and phenotypic characteristics were investigated. The three isolates and eight syntrophic pairs were able to utilize parathion as a sole source of carbon and energy, producing p-nitrophenol as the intermediate metabolite during the complete degradation of parathion. Analysis of the 16S rRNA gene sequence indicated that the isolates were related to members of the genera Burkholderia, Arthrobacter, Pseudomonas, Variovorax, and Ensifer. The chromosomal DNA patterns of the isolates obtained by polymerasechain-reaction (PCR) amplification of repetitive extragenic palindromic (REP) sequences were distinct from one another. Ten of the isolates had plasmids. All of the isolates and syntrophic pairs were able to degrade parathion-related compounds such as EPN, p-nitrophenol, fenitrothion, and methyl parathion. When analyzed with PCR amplification and dot-blotting hybridization using various primers targeted for the organophosphorus pesticide hydrolase genes of previously reported isolates, most of the isolates did not show positive signals, suggesting that their parathion hydrolase genes had no significant sequence homology with those of the previously reported organosphophate pesticide-degrading isolates.
Among the environmental pollutants, parathion, chloroform and 2, 6-dichlorophenol may impair human health; they may inhibit or reduce the metabolic function of human body and may furthermore cause diseases directly or indirectly. This study was undertaken to investigate the effects on the immune response by intoxication of parathion, chloroform or 2, 6-dichlorophenol. Parathion(1.3mg/kg, olive oil 10ml), chloroform (100mg/kg, olive oil 10ml) were administered via intraperitoneal injection to rats. And 2, 6-dichlorophenol (13mg/kg, olive oil 10ml) was administered via oral injection. After 3 weeks, the rats were intoxicated with the above chemicals and immunized with sheep RBC. After 4 weeks the immune response of rat spleen cells was measured by the Jerne's technique. The results were obtained as follows. 1. There was no change of leukocyte counts by the intoxication of parathion, chloroform and 2, 6-dichlorophenol. 2. Parathion, chloroform and 2, 6-dichlorophenol reduced hemoglobin contents for most intoxicated and immunized groups. 3. Hematocrits were decreased by the intoxication of parathion, chloroform or 2, 6-dichlorophenol significantly. 4. It was determined that total protein, A/G (albumin/globulin), .alpha.-, .betha.-and .gamma.-globulins in rat serum were not changed. 5. Intoxication by parathion, chloroform or 2, 6-dichorophenol reduced the number of hemolytic plaque to the sheep RBC in rat spleen cells. Therefore, the capacity of erythrocyte production and the immune response of rat spleen cells were decreased by the intoxication of parathion, chloroform, or 2, 6-dichlorophenol.

The toxic impact of parathion on bagrid catfish (Pseudobagrus fulvidraco) juvenile was examined 96 hr $LC_{50}$ value of parathion estimated as 1 90 mg/L. Chronically, bagrid catfish were exposed for 30 days to concentrations of parathion, i.e. 63, 95, 190 and $380\;{\mu}g/L$, representing 3.3, 5.0, 10.0 and $20.0{\%}$ of the 96 hr $LC_{50}$. Survival rate was significantly affected by the concentration over $63\;{\mu}g/L$ after 10 days. Growth rate, specific growth rate (SGR) and feed efficiency were reduced in time as concentration dependent manner. Growth rates of the exposed groups were significantly reduced as $28.06-60.63{\%}$ to the control fish, SGR was decresed as $1.50-4.72{\%}$ and feed efficiency was declined as $6.10-11.90{\%}$. It can be concluded that if p. lulvidraco is chronically exposed to parathion over $63\;{\mu}g/L$ concentration, the physiological damages might affect growth factors of the fish.

A bacterium, Burkholderia sp. JBA3, which can mineralize the pesticide parathion, was isolated from an agricultural soil. The strain JBA3 hydrolyzed parathion to p-nitrophenol, which was further utilized as the carbon and energy sources. The parathion hydrolase was encoded by a gene on a plasmid that strain JBA3 harbored, and it was cloned into pUC19 as a 3.7-kbp Sau3AI fragment. The ORF2 (ophB) in the cloned fragment encoded the parathion hydrolase composed of 526 amino acids, which was expressed in E. coli DH10B. The ophB gene showed no significant sequence similarity to most of other reported parathion hydrolase genes.

Parathion is widely used in agriculture, but it is highly toxic and now clear that parathion behaves like a cholinergic drug by inhibiting the enzyme cholinesterase. In order to know the effect of toxicity and cholinesterase activity in rats injected repeatedly with parathion, cholinesterase activity in plasma, whole brain and spinal cord, and the subacute toxicity after repetitive intraperitoneal injection of parathion 20 times every 3 days were investigated. The results obtained were summerized as follows ; $LD_{50}$ value of parathion given intraperitoneally to rats was 10.5mg/kg(95% confidence limits, 6.6-16.8mg/kg). In subacute toxicity test of parathion injected intraperitoneally, mortality of parathion-pretreated rats(B : 57%, C : 83%) were increased in comparison with the control(50%). Cholinesterase activities in plasma of parathion-pretreated rats(B : 0.47 U/ml, C : 0.36 U/ml, AA : 0.31 U/ml, B : 0.26 U/ml, CC : 0.17 U/ml) were significantly decreased in comparison with the control(0.58 U/ml). Cholinesterase activities in spinal cord of parathion-pretreated rats(B : 1.87 U/g, C : 1.29 U/g, AA : 1.27 U/g, BB : 0.71 U/g, CC : 0.25 U/g) were decreased in comparison with the control(2.48 U/g). Cholinesterase activities in whole brain of parathion-pretreated rats(B : 2.52 U/g, C : 1.32 U/g, AA : 2.48 U/g, BB : 1.08 U/g, CC : 0.51 U/g) were significantly inhibited in comparison with the control(4.67 U/g). However, there were no differences in the urea nitrogen and creatinine concentrations between parathion-pretreated rats and control.
The parathion hydrolase (OPH) produced by Pseudomonas rhodesiae H5 was purified by ammonium sulfate precipitation, DEAE-Toyopearl 650M ion exchange chromatography and Sephadex gel filtration chromatography. Parathion hydrolase from crude extracts of P. rhodesiae H5 has two components designated as OPH $I_1$ and OPH $I_2$, Optimum pH and temperature of OPH $I_1$and OPH $I_2$ were pH 7.2 and $30^{\circ}C$, and pH 7.6 and $37^{\circ}C$, respectively. The activation energy of OPH $I_1$ for the hydrolysis of parathion was 3.01 ㎉/I, II, III in the temperature range of $4^{\circ}C$ to $30^{\circ}C$, and Michaelis constant ($K_m$) for parathion was 69.2 ${\mu}M$. The activation energy of OPH $I_2$ for the hydrolysis of parathion was 4.07㎉/㏖ in the temperature range of $4^{\circ}C$ to $37^{\circ}C$, and Michaelis constant for parathion was 150.9${\mu}M$. Furthermore OPH $I_1$ was completely inhibited by 1 mM $Ca^2+$, $Cu^2+$, $Mg^2+$, $Ni^2+$, but OPH $I_2$ was less inhibited than OPH $I_1$ by the metals used in this study.

Abstract The protective effects of physostigmine against the toxicity of parathion (diethyl-4-nitrophenyl phosphorothionate) were examined in male Sprague-Dawley rats. Physostigmine (100 or 1,000 ㎎/㎏, ip) injected 30 min before decreased the inhibition of acetylcholinesterase (AChE) activities in brain, lung and blood induced by parathion (2 ㎎/㎏, ip).(omitted)
The investigation of the Acetylcholinesterase (AChE) activity in tissues (brain, eye, muscle and serum) of crucian carp (Carassius auratus) exposed to the waterborne parathion was carried out for application as biomarker of organophosphate pesticides. The AChE activities were significantly inhibited in the experimental organs of C. auratus treated ${\geq}63{\mu}g/L$ of concentrations of parathion. The AChE activity of C. auratus was significantly reduced in response to brain (79.1~92.4%), eye (76.0~91.5%), muscle (89.7~97.6%) and serum (68.9~78.0%) after 30 days exposure. No significant mortality occurred during the experiment duration but behavioral changes occurred in the carp after exposure to the parathion were erratic swimming and convulsions. The anomaly in the carp exposed to parathion were observed in the form of scoliosis. The use of AChE activity and other adverse responses of the carp might be use as a reliable monitoring tool to detect parathion in aquatic ecosystem which might produce significant population changes.
The effects of altering the hepatic mixed-function oxidase(MFO) activities on the acute toxicity of parathion were examined in female rats. Phenobarbital sodium pretreatment (50mg/kg/day, i.p.) for 4 consecutive days has resulted in significant decreases in the toxicity of parathion (2 or 4 mg/kg, i.p.) as determined by lethality and cholinesterase activities wheras the toxicity arising from a single dose of CCl4(2 mmol/kg, i.p.) 24 hr prior to parathion challenge was potentiated.
