Eudragit으로 코팅된 초다공성 하이드로젤의 제조 및 pH 의존형 팽윤거동 (Preparation of Eudragit Coated Superporous Hydrogels and Their pH Dependent Swelling Behavior)

  • 초다공성 하이드로젤은 다공성 공극구조를 이용하여 기존 수화젤의 팽윤성을 획기적으로 향상시킨 것으로, 빠른 팽윤거동과 높은 흡수율로 다양한 의약용 응용분야에 유용한 재료이다. 본 연구에서는 장용 코팅제인 Eudragit 계열 고분자들을 사용하여 poly(acrylic acid-co-acrylamide)계 초다공성 하이드로젤을 코팅함으로써 pH 의존성 팽윤거동을 보이는 초다공성 하이드로젤을 제조하고자 하였다. 서로 다른 pH 영역에서 작용하는 Eudragit L100과 S100을 이용하여 딥 코팅에 의해 표면을 코팅한 후 SEM을 이용해 공극구조를 관찰하고 pH에 따른 초다공성 하이드로젤의 팽윤거동을 관찰하였다. Eudragit 계열 고분자들로 코팅된 하이드로젤은 낮은 pH 환경하에서는 팽윤이 억제되다가, 특정 pH 이상에서 팽윤성이 향상되는 pH 의존성 팽윤거동을 보였고, 이러한 pH 의존성은 사용한 장용 코팅제용 고분자들의 pH 특성에 의존하였다.

  • Drug release experiments were performed based on pH-sensitive swelling behaviors of polymethacrylic acid. 5-Fluorouracil as a nonionic model drug revealed release patterns depending solely on pH-dependent swelling kinetics of polymethacrylic acid. In contrast, release of propranolol hydrochloride as a cationic model drug was significantly affected by ionic drug-polymer interaction as well as the swelling kinetics. Accordingly, a zero-order release pattern was obtained at pH 7, which was distinguished from the general matrix type drug release pattern.

pH-민감성 삼성분계 공중합체 젤의 합성 및 팽윤 속도론 (Synthesis and Swelling Kinetics of a Cross-Linked pH-Sensitive Ternary Copolymer Gel System)

  • A pH sensitive ternary copolymer gel was synthesized in the presence of ethylene glycol dimethacrylate (EGDMA) as a crosslinking agent through radical polymerization of vinyl acetate (VA), acrylic acid (AA) and methyl acrylate(MA) with a weight ratio of 1 : 1.3 : 1. A number of experiments were carried out to determine the swelling behavior of the gel under a variety of pH conditions of the swelling medium. As the pH of the swelling medium was changed from 1.0 to 8.0 at $37^{\circ}C$, the gel showed a shift in the pH-dependent swelling behavior from Fickian (n=0.3447) to non-Fickian (n=0.9125). The resulting swelling parameters were analyzed using graphical and statistical methods. The results showed that the swelling of the gel was controlled by the pH of the medium, i.e. $n=n_o{\exp}(S_{C}pH)$, where n is the diffusion exponent, $n_o(=28.9645{\times}10^{-2})$ is the pre-exponential factor and $S_C$(=0.1417) is pH sensitivity coefficient. The swelling behavior of the gel was also examined in aliphatic alcohols. The results showed that the rate of swelling increased with increasing number of carbon atoms in the alcoholic molecular chain.

  • Equilibrium swelling and pH-sensitivity of a polyelectrolyte copolymer hydrogel were controlled by employing copolymers with different hydrophilic-hydrophobic balances. Model pH-sensitive hydrogels, e.g., poly(methacrylic acid), poly(methacrylic acid-co-acrylamide), poly(methacrylic acid-co-2-hydroxyethylmethacrylate), poly(methacrylic acid-co-styrene) were synthesized at various monomer compositions. As hydrophobicity of the copolymer hydrogels increased, the equilibrium swelling decreased while the pH-sensitivity increased. In the case of poly(methacrylic acid-co-acrylamide), polymer-polymer interaction significantly affected the equilibrium swelling and provided a wide range control of pH-sensitivity.

  • Poloxamer-poly (acrylic acid) (PAA) interpenetrating polymer networks (IPNs) were prepared via matrix polymerization of acrylic acid with poloxamer prepolymer. The equilibrium s welling of poloxamer/PAA IPNs was determined in various pH medium. The swelling of poloxamer/PAA IPNs was more affected by pH difference compared with the swelling of homo PAA gel due to protonation and deprotonation of the PAA network, followed by reversible formation and dissociation of the interpolymer complex due to hydrogen bonding between acidic hydrogens and ether oxygens. Nonionic/anionic/cationic drugs were incorporated into IPN matriceds as a model drug and their release behavior was studied. Nonionic, drug revealed release patterns depending solely on pH dependent swelling kinetics. In contrast, the release of ionic drugs was significantly affected by ionic drug-polymer interaction as well as the swelling kinetics.

글루코스를 함유한 음이온 하이드로젤의 pH 감응성 동적 팽윤거동 (pH-Sensitive Dynamic Swelling Behavior of Glucose-containing Anionic Hydrogels)

  • 음이온 하이드로젤은 그들이 가지고 있는 pH 감응성 팽윤거동 때문에 단백질 약물의 경구투여용 전달물질로써 많은 주목을 받고 있다. 본 연구에서는 음이온 하이드로젤의 pH 변화에 따른 용매의 침투 메커니즘을 규명하기 위하여 methacrylic acid와 2-methacryloxyehtyl glucoside를 공중합하여 P(MAA-co-MEG) 하이드로젤을 합성한 후 pH 변화에 따른 하이드로젤의 동적 팽윤거동을 관찰하였다. 용매의 침투 메커니즘이 Fickian 또는 non-Fickian 인지를 설명할 수 있는 특성지수 n을 $M_t/M_{\infty}=kt^n$ 관계식으로부터 계산하였다. 하이드로젤에 대한 용매의 침투 메커니즘은 주위 pH의 영향을 많이 받았으며, 젤의 $pK_a$ 보다 높은 pH인 7.0에서는 침투 메커니즘이 상대적으로 고분자사슬의 이완에 의한 지배를 많이 받는다는 것을 알 수 있었다. 한편, pH 7.0에서 고분자 이완에 의한 용매의 침투 메커니즘은 하이드로젤에 존재하는 카르복실산의 이온화에 기인한 것임을 ATR-FTIR 분광분석을 이용하여 확인하였다.

Drug Release from the Enzyme-Degradable and pH-Sensitive Hydrogel Composed of Glycidyl Methacrylate Dextran and Poly{acrylic acid)

  • Hydrogels composed of glycidyl methacrylate dextran (GMD) and poly(acrylic acid, PM) were prepared by UV irradiation method for colon-specific drug delivery. GMD was synthesized by coupling of glycidyl methacrylate to dextran in the presence of 4-(N,N-dimethylamino)pyridine. GMD was photo-polymerized by ammonium peroxydisulfate as initiating system in phosphate­buffered solution (0.1 M, pH 7.4). And then, acrylic acid monomer was added and subsequently heat-polymerized by 2,2'-azobisisobutyronitrile as an initiator. The hydrogels exhibited high swelling ratio (about 20) at $37^{\circ}C$, and showed a pH-dependent swelling behavior. In addition, the swelling ratio of the hydrogel was remarkably enhanced to about 45 times in the presence of dextranase at pH 7.4. The swelling-deswelling behavior proceeded reversibly for the GMD/PM hydrogels between pH 2 and pH 7.4. Release of 5-aminosalicylic acid from the GMD/PAA hydrogels was evaluated in simulated gastrointestinal pH fluids in the absence or presence of dextranase. We concluded that the hydrogels prepared could be used as a dual-sensitive drug carrier for sequential release in gastrointestinal tract.

국산벤토나이트 현탁액의 유동학적 성질 (Rheological Studies on the Aqueous Suspension of Korean Bentonite)

  • A study has been made of the Korean bentonite aqueous suspension contrast with American bentonite by means of XRD IR swelling, gel formation and rheogram at various conditions such as concentration, temperature and pH. The Korean bentonite was identified as montmorillonite clay containig a small proprotion of crystoballite and mordenite, and its swelling power were acceptable for requirements of Korean pharmacopeia regulations though its values were not satisfied. Korean bentonite swelled to 10 times and American one did to 15 times compared to its bulkiness of powder. The rheogram of Korean bentonite suspension reveals bulged pseudoplatic flow with yield value at higher concentration and pseudoplastic flow without yield value at lower concentration. The higher the concentration, the greater were the apparent viscosity and hysteresis loop. Korean bentonite suspension showed insignificant temperature dependence on both apparent viscosity and hysteresis loop and it was more temperature dependent on viscosity but less on hysteresis loop than those of American sample. The pH dependence was so high on viscosity that apparent minimum value was near pH 7 and maximum value at pH 3 or 7. The hysteresis loop appeared minimum over the pH range 5-7 and maximum near pH 3 or 11. The Korean bentonite was inferior to the American bentonite in swelling volume, gel formation, thioxotropy, however, it would be possible to improve the quality of Korean bentonite by developing the method of purification for bentonite clay.

  • Polymers have been developed and applied in many biomedical areas as well as engineering and industrial fields. The first essential to achieve successful development and applications is that properties of such polymer materials would be investigated. In many cases, such investigations are accomplished by observing polymeric behavior arising from the environmental changes such as pH, temperature, and ionic concentration. It has long been known that NMR is extremely sensitive to many biochemical and physical changes occurring in the polymer samples. In the present study we focus our study on NMR Microimaging, which is one of the important NMR applications, to characterize the swelling effect by observing the time dependent spatial variations of polymer specimens. For the samples three kinds of polyvinyl alcohol (PVA) specimens are prepared with different degrees of cross linking density. $^1H$ NMR microimages are acquired as a function of time to visualize the swelling behavior as well as volumetric changes occurring in the specimens. From the acquired time dependent images, the swelling process is exploited.

  • Some mucoadhesive polymers such as hydroxypropylcelluose (HPC) and carbopol-934 (CP) have been employed for the preparation of mucoadhesive polymeric systems, and their physical properties including mucoadhesion, swelling, and drug release were evaluated. A new simple experimental technique that can quantitatively measure the bioadhesive properties of various polymeric systems has been developed by the methods of detachment force test. As the polymeric systems, the discs of freeze-dried HPC/CP solid dispersions were prepared. The mucosa used in these tests were upper, middle, and lower parts of small intestine of male rats weighing $300{\sim}350\;g$. Detachment forces were increased as the mole fraction of CP increased in discs of HPC/CP solid dispersions. In the points of intestinal site dependence of mucoadhesion, the solid dispersions revealed non-specific mucoadhesion to the intestine. Swelling and drug release characteristics of mucoadhesive polymeric systems were studied extensively to find out the feasibility for the oral controlled delivery systems. Swelling ratio, expressed as the final height/initial height, has been determined in various pH buffer solutions. Hydrochlorothiazide (HCT) was employed as a model drug for release study. Apparent swelling and drug release rate constants, $K_s$ and $K_r$ respectively, were obtained from the square-root time plot of either swelling ratio or released amount of drug, particularly for the time periods before reaching the equilibrium. As a result, the swelling ratio of HPC/CP solid dispersions was increased as the weight percentage of CP increased. Similarly, the release of HCT from the solid dispersions was dependent on pH changes and CP contents, resulted in the slower release of HCT with the increases of pH and CP contents.
