Development of Antimicrobial Edible Film from Defatted Soybean Meal Fermented by Bacillus subtilis

  • In order to extend shelf-life of the packaged or coated foods, an antibacterial edible film was developed. Antimicrobial activities of 9 bacteriocin-like substance (BLS)­producing strains were evaluated after growing them on defatted soybean meal medium (DSMM). Bacillus subtilis was selected among those, because it showed the biggest inhibition zone against 6 problem bacteria in food. The antimicrobial edible film, containing $0.32\%$ of BLS, was produced from the fermented soybean meal with B. subtilis at the optimum condition of pH 7.0-7.5 and $33^{\circ}C$ for 33 h. The antimicrobial activity of the film was over $50\%$ of the maximum activity after film production with heat treatment at $90^{\circ}C$ and pH adjustment to 9. When the soy protein film with BLS was applied on the agar media containing E. coli, the growth inhibition was much higher than the ordinary soy protein film. These results indicate that the soy protein film with BLS from B. subtilis can be used as a new packaging material to extend the shelf-life of foods.

Electrochemical treatment of cefalexin with Sb-doped SnO2 anode: Anode characterization and parameter effects

  • In this study, it was aimed to evaluate direct oxidation of aqueous solution containing cefalexin antibiotic with new generation Sn/Sb/Ni: 500/8/1 anode. The fact that there is no such a study on treatment of cefalexin with these new anode made this study unique. According to the operating parameters evaluation COD graphs showed clearer results compared to TOC and CLX and thus, it was it was chosen as major parameter. Furthermore, pseudo-first degree kd values were calculated from CLX results to show more accurate and specific results. Experimental results showed that after 60 min of electrochemical oxidation, complete removal of COD and TOC was accomplished with 750 mg L-1 KCl, at pH 7, 50 mA cm-2 current density and 1 cm anode-cathode distance. Also, the stability of the Sn/Sb/Ni anode was evaluated by taking SEM and AFM images and XRD analysis before and after of electrochemical oxidation processes. According to the results, it was not occurred too much change on the anode surface even after 300 h of electrolysis. Thus, it was thought that the anode material was not corroded to a large extent. Furthermore, the removal efficiencies were very high for almost all the time and conditions. According to the results of the study, electrochemical oxidation with new generation Sn/Sb/Ni anodes for the removal of cefalexin antibiotic was found very successful and applicable due to require less reaction time complete mineralization and doesn't require pH adjustment step compared to other studies in literature. In future studies, different antibiotic types should be studied with this anode and maybe with real wastewaters to test applicability of the process in treatment of pharmaceutical wastewaters containing antibiotics, in a better way.

  • This paper aims to investigate the potential use of cempedak durian peel (CDP) from Negara Brunei Darussalam, which is low-cost, locally available, eco-friendly and highly efficient to remove methyl violet (MV) dye from aqueous solutions. The time required for equilibrium to be reached is 2.0 h with no adjustment of pH necessary. FTIR analysis was indicative of the involvement of -COOH and C=O functional groups in adsorption process. The Langmuir model provided the best fit with maximum adsorption capacity of $0.606mmol\;g^{-1}$. Thermodynamics data indicate that the adsorption is spontaneous, feasible and endothermic in nature. Best regeneration of CDP's adsorption ability is achieved by base solution, showing about 95% removal efficiency of MV even after 5 cycles, indicating that CDP can be regenerated and reused. This, together with its high adsorption capacity, makes CDP a potential adsorbent for the removal of MV in wastewater.

  • Chemical-mechanical polishing (CMP), one of the dominant technology for ULSI planarization, is used to flatten the micro electro-mechanical systems (MEMS) structures. The objective of this paper is to achieve good planarization of the deposited film and to improve deposition efficiency of subsequent layer structures by using surface-micromachining process in MEMS technology. Planarization characteristic of poly-Si film deposited on thin oxide layer with MEMS structures is evaluated with different slurries. Patterns used for this research have shapes of square, density, line, hole, pillar, and micro engine part. Advantages of CMP process for MEMS structures are observed respectively by using the test patterns with structures larger than 1 urn line width. Preliminary tests for material selectivity of poly-Si and oxide are conducted with two types of silica slurries: $ILD1300^{TM}\;and\;Nalco2371^{TM}$. And then, the experiments were conducted based on the pretest. A selectivity and pH adjustment of slurry affected largely step heights of MEMS structures. These results would be anticipated as an important bridge stone to manufacture MEMS CMP slurry.

  • This study investigated the effects of combined treatment of immersion liquids (adjusted pH 4.5 with Citrus junos concentrate (CL), pH 5.5 with Prunus mume concentrate (PL), or no pH adjustment, pH 6.5, control) and high hydrostatic pressure (HHP, 450 MPa) on shelf-life and quality of mackerel. In this study, we measured changes in viable cell counts, pH level, volatile basic nitrogen, thiobarbituric acid reactive substances, color analysis, volatile organic compounds, and sensory evaluation of mackerel at $4^{\circ}C$ for 20 days. CL/HHP and PL/HHP treatments reduced viable cell counts by 3 log cycles during storage compared to the control. Mackerel treated with CL/HHP and PL/HHP showed significantly lower TBARS and VBN levels as compared to the control. After combined treatment, lightness and whiteness increased but redness decreased. VOCs tests showed that contents of alcohol, acid, and ketones in mackerel fillet treated with CL/HHP and PL/HHP were relatively reduced. Especially, CL/HHP and PL/HHP suppressed production of sulfur. In the sensory evaluation, aroma, taste, salinity, hardness, and springiness of mackerel treated with combined HHP showed higher scores than the control. These results suggest that immersion liquids and HHP treatments may increase shelf-life of mackerel and maintain quality during storage at $4^{\circ}C$.

Effect of Whole Egg Spray-drying Conditions on Physical and Sensory Properties of Sponge Cake (계란분말 제조 조건에 따른 스펀지케이크의 특성 변화)

  • The objective of this study was to determine the optimum manufacturing conditions of whole egg powder with high foaming property sufficient for making sponge cakes. Whole eggs were either desugarized and/or adjusted pH before spray-dried at a temperature of $70^{\circ}C$. The physical properties of the cake were measured, and sensory evaluation was conducted on a 9-point scale. The pH and specific gravities of the foam and batter made from desugarized egg powder did not differ from those of the control, whereas batter made from commercial powder experienced significant loss of cake characteristics. The pH adjustment did not improve the foaming properties of the egg powders. The volume and textural properties of the cakes made from spray-dried egg did not differ from those of fresh egg. The taste and sensory characteristic scores for fresh egg, desugarized whole egg powder, and commercial powder were 5.00, 4.78, and 1.89, respectively. These results indicate that egg powders desugarized and spray-dried at $70^{\circ}C$ are sufficient for making sponge cakes with acceptable physical and sensory attributes.

Spectrofluorimetric Determination of Fe(Ⅲ) with 4,5-Dihydroxy-1,3-Benzenedisulfonic Acid (4,5-Dihydroxy-1,3-Benzenedisulfonic Acid를 이용한 Fe(Ⅲ)의 분광형광법 정량)

  • A spectrofluorimetric method for the determination of Fe(III) in aqueous solution with 4,5-dihydroxy-1,3-benzenedisulfonic acid(Tiron) as a fluorimetric reporter was developed. Tiron, which is very soluble in water,is a good fluorimetric reagent. However, when Tiron was complexed with Fe(III), the fluorescent intensity was decreased proportionally with the concentration of Fe(III) by a quenching effect. The excitation and fluorescene wavelength of Tiron showing the quenching effect by Fe(III) at pH 4.5 were 312 nm and 341 nm, respectively. The highest sensitivities were shown at Tiron concentration of $1.0{\times}10^{-2}M$. To enhance the quenching effect, the Fe(III)-Tiron complex solution was heated to 80$^{\circ}C$ for 90 minutes. As for Fe(III), the most interfering ion was Cu(II). The interference effects could be mostly eliminated by pH adjustment or by adding EDTA. The concentration ranges showing the linear response to Fe(III) was from $5.0{\times}10^{-7}M\;to\;6.0{\times}10^{-5}M$ With this proposed method, the detection limits of Fe(III) was $2.8{\times}10^{-6}M$. Recovery of Fe(lII) in a synthetic sample was almost quantitative. Based on experimental results, it is proposed that the above technique can be applied to the practical determination of Fe(III).

Rapid Cell Death Phenotype of Streptococcus mutans under Prolonged Growth Conditions (장시간 생장 조건에서 Streptococcus mutans의 급격한 세포사 표현형 분석)

  • The oral pathogen Streptococcus mutans is considered a major causative agent of dental caries in humans. The use of dental hygiene products, including toothpaste and mouthwash, is used for caries control. However, food intake can lead to the recurrence of oral microorganisms. This study aimed to explore why this bacterium dies so quickly during prolonged incubation and to assess whether this growth characteristic is closely associated with the secretion of metabolic products. Notably, the number of live S. mutans cells rapidly declined after 24 hr during the entire period tested, whereas the number of Escherichia coli cells, an indicator strain, remained steady over the same period. To test whether the S. mutans supernatants contained possible signals that accelerated the death of neighbor cells, we obtained the individual supernatants at the above time points. Following pH neutralization, the cells in which the supernatant was supplemented with glucose grew well. However, pH adjustment alone could not fully recover cell growth in conditions in which the supernatant was supplemented, with or without glucose. These phenotypes of S. mutans may be associated with signaling, not only resulting from nutrient depletion. The findings on the survival phenotype of S. mutans provide new insights into cell-cell communication in the biology of this bacterium.

Influence of Probiotics Supplementation on Egg Quality and Excretal Noxious Gas in Laying Hens (산란계에 있어 생균제의 첨가가 계란품질 및 배설물내 유해가스 함량에 미치는 영향)

  • This study was conducted to investigate the effects of feeding probiotics on the egg quality and excretal noxious gas in laying hens. One hundred forty four, 36 weeks old ISA brown commercial layer, were employed in a 28 d feeding trial with a 7 d adjustment period. Dietary treatments are 1) control(basal diet), 2) PB0.3(basal diet+0.3% probiotics), 2) PB0.6(basal diet+0.6% probiotics). For overall period, hen-day egg production, egg weight and egg shell breaking strength tended to increase (P>0.05) by dietary probiotic supplementation. Egg shell thickness was improved by supplementation of probiotics(linear effect, P$<$0.01). Diets PB0.3 and PB0.6 improved the yolk color compared to control diet(linear effect, P$<$0.02). As supplementation level of probiotics increased in the diets, egg yolk index tended to increase(linear effect, P$<$0.04). $NH_3$-N concentration in excreta fed PB0.6 diet was significantly (P$<$0.04) lower than either control or PB0.3 diet. In conclusion, supplementing probiotics to a Corn-SBM diet for laying hens increased egg shell thickness, yolk color and decreased egg yolk index, and decreased excretal $NH_3$-N concentration.
