Construction of Plasmids for Overproduction of L-Phenylalanine (L-페닐알라닌 대량생산을 위한 재조합 플라스미드 구성)

  • For the overproduction of L-phenylalanine using Escherichia coli, the authors constructed various recombinant plasmids including pMW 10, pMW 11 and pMW 12. The $aroF{FR}$ and $pheA^{FR}$ genes for the production of L-phenylalanine were isolated from Escherichia coli MWEC 101-5 strains. The productivity and atability of Escherichia coli regulatory mutants containing recombinant plasmids were investigated to evaluate the efficiency of the $aroF^{FR}$ and $pheA^{FR}$ genes. The MWEC 101-5/pMW 11 strain produced 24.3g/l of L-phenylalanine while its stability was 73.8 percent. The specific activity of prephenate dehydratase in the MWEC 101-5/pMW 11 strain increased by 26-fold compared with that of Escherichia coli K-12.

Promoter Methylation of CDKN2A, $RAR{\beta}$, and RASSF1A in Non-Small Cell Lung Carcinoma: Quantitative Evaluation Using Pyrosequencing

  • Background: While qualitative analysis of methylation has been reviewed, the quantitative analysis of methylation has rarely been studied. We evaluated the methylation status of CDKN2A, $RAR{\beta}$, and RASSF1A promoter regions in non-small cell lung carcinomas (NSCLCs) by using pyrosequencing. Then, we evaluated the association between methylation at the promoter regions of these tumor suppressor genes and the clinicopathological parameters of the NSCLCs. Methods: We collected tumor tissues from a total of 53 patients with NSCLCs and analyzed the methylation level of the CDKN2A, $RAR{\beta}$, and RASSF1A promoter regions by using pyrosequencing. In addition, we investigated the correlation between the hypermethylation of CDKN2A and the loss of $p16^{INK4A}$ immunoexpression. Results: Hypermethylation of CDKN2A, $RAR{\beta}$, and RASSF1A promoter regions were 16 (30.2%), 22 (41.5%), and 21 tumors (39.6%), respectively. The incidence of hypermethylation at the CDKN2A promoter in the tumors was higher in undifferentiated large cell carcinomas than in other subtypes (p=0.002). Hyperrmethylation of CDKN2A was significantly associated with $p16^{INK4A}$ immunoexpression loss (p=0.045). With regard to the clinicopathological characteristics of NSCLC, certain histopathological subtypes were found to be strongly associated with the loss of $p16^{INK4A}$ immunoexpression (p=0.016). Squamous cell carcinoma and undifferentiated large cell carcinoma showed $p16^{INK4A}$ immunoexpression loss more frequently. The Kaplan-Meier survival curves analysis showed that methylation level and patient survival were barely related to one another. Conclusion: We quantitatively analyzed the promoter methylation status by using pyrosequencing. We showed a significant correlation between CDKN2A hypermethylation and $p16^{INK4A}$ immunoexpression loss.

Effects of Electrodeposition Conditions on Properties of Ni Thin Films Electrodeposited from Baths Fabricated by Dissolving Metal Ni Powders (금속 Ni 분말을 용해한 도금용액으로부터 전기도금 된 Ni 박막 특성에 미치는 도금조건의 영향)

  • Chloride plating solution was fabricated by dissolving metal Ni powders in solution with HCl and deionized water. Effects of deposition conditions on the properties of Ni films electrodeposited from chloride baths were studied. Current efficiency of Ni films electrodeposited from the baths containing saccharin was decreased with increasing the current density. Residual stress of Ni thin films ware measured to be about 230 ~ 435 MPa in the range of current density of $10{\sim}25mA/cm^2$. Cathode current efficiency in baths without saccharin was initially increased with increasing pH, while it was decreased with increasing pH further. Cathode current efficiency in baths with saccharin (except at pH 2) exhibited less 10 ~ 20% than that in baths without saccharin. Residual stress of Ni films electrodeposited from baths without saccharin was measured to be 388 ~ 473 MPa in the range of pH 2 ~ pH 5 and then was increased to 551 MPa at pH 6. On the other hand, residual stress of Ni films electrodeposited from baths with saccharin was increased with increasing pH. Surface morphology was strongly affected by the change of current density, but slightly by solution pH and addition of saccharin.

The Effects of Foreign Workers' Health Beliefs on Health Behavior (외국인 근로자의 건강신념이 건강행위에 미치는 영향)

  • Objectives: This study was conducted to investigate the effects of personal characteristics and health beliefs on health behaviors of foreign workers living in Korea. Methods: The subjects of this study were foreign workers who understood the purpose of this study and allowed participation in the workplaces in northern Gyeonggi Province. The final subjects were 206, and collected all data were analyzed by SPSS 23.0. Results: The personal characteristics of the subjects were 51.0% for male and 49.0% for female, and the average age was 31.76(±6.96) years old. The mean for each health belief factors was perceived benefits 4.03(±.56), perceived severity 3.75(±.73), perceived susceptibility 3.65(±.64), self-efficacy 3.56(±.83), and perceived barriers 3.34(±.73). The mean of each health behavior factors was non-smoking 4.02(±.83), drinking 3.92(±.80), health responsibility 3.42(±.83), exercise 3.37(±1.01), dietary habits 1.91(±.11), and stress management 1.72(±.27). The correlation between sub-factors of health belief was all at the level of p <0.01. The sub-factors of health belief were health behavior and perceived susceptibility(r=.773, p<0.01), followed by self-efficacy(r=.760, p<0.01), and perceived severity(r=.574, p<0.01). The factors affecting health behavior were self-efficacy(β=.540, p<.001), perceived susceptibility(β=.461, p<.001), perceived benefits(β=.152, p<.05), marriage status(β=-. 100, p <.05), and income(β=-. 120, p <.05) in order. Conclusion: In-depth consideration of factors that can affect the physical, psychological and social health of foreign workers, and various program development and intervention strategies based on these factors should be sought.

Separation of Ni(II), Co(II), Mn(II), and Si(IV) from Synthetic Sulfate and Chloride Solutions by Ion Exchange (황산과 염산 합성용액에서 이온교환에 의한 니켈(II), 코발트(II), 망간(II) 및 실리케이트(IV)의 분리)

  • Reduction smelting of spent lithium-ion batteries at high temperature produces metallic alloys. Following solvent extraction of the leaching solutions of these metallic alloys with either sulfuric or hydrochloric acid, the raffinate is found to contain Ni(II), Co(II), Mn(II), and Si(IV). In this study, two cationic exchange resins (Diphonix and P204) were employed to investigate the loading behavior of these ions from synthetic sulfate and chloride solutions. Experimental results showed that Ni(II), Co(II), and Mn(II) could be selectively loaded onto the Diphonix resin from a sulfate solution of pH 3.0. With a chloride solution of pH 6.0, Mn(II) was selectively loaded onto the P204 resin, leaving Ni(II) and Si(IV) in the effluent. Elution experiments with H2SO4 and/or HCl resulted in the complete recovery of metal ions from the loaded resin.

  • According to a survey on the infringement of SMEs, the level of technology protection capability is improving every year, but technology leaks and damage continue to occur. This shows that there is a need for a security management and supervision system that can strengthen the security awareness of SME executives and employees and maintain the security level continuously. The Personal Information & Information Security Management System(ISMS-P) authentication systems is the latest related standard, which has the problem of applying the same certification criteria without considering the types of certification target organizations such as ISPs, IDC, hospitals and schools, and SMEs.. In this paper, 73 evaluation items that can be specialized and applied to SMEs were derived by referring to ISMS-P certification and Personal Information Protection Management System (PIMS) certification. The results of the study show that the number of evaluation items decreased by 28.4% compared to the existing ISMS-P certification.

  • We introduce some new type of admissible mappings and prove a coupled coincidence point theorem by using newly defined concepts for generalized compatible pair of mappings satisfying ${\alpha}-{\psi}$ contraction on partially ordered metric spaces. We also prove the uniqueness of a coupled fixed point for such mappings in this setup. Furthermore, we give an example and an application to integral equations to demonstrate the applicability of the obtained results. Our results generalize some recent results in the literature.

  • Vertical shortening in tall buildings would be of little concern if all vertical elements shortened evenly. However, vertical elements such as walls and columns may shorten different amounts due to different service axial stress levels. With height, the differential shortening may become significant and impact the strength design and serviceability of the building. Sometimes column transfers or other vertical structural irregularities may cause differential shortening. If differential shortening is not addressed properly, it can impact the serviceability of the building. This paper takes the perspective of a structural engineer in planning the design, predicting the shortening and its effects, and communicating the information to the contractor.

  • In this work we have investigated temperature dependence of spin-rotational relaxation rate, $(1/T_1)_{SR}$, of methyl carbon-13's in 2-bromo-p-xylene, 2,5-dimethylaniline, and 2,5-dimethylanisole and have found that temperature behaviors of two methyl carbon-13's in ortho- and meta-position, respectively, are substantially different. It has been confirmed that the modified Burke-Chan model proposed by Park et al. can nicely explain different temperature dependence of $(1/T_1)_{SR}$ for these two methyl carbon-13's while the original Burke-Chan model fails to do so.
