Development and Validation of an Analytical Method for Ametoctradin Residue Determination in Domestic Agricultural Commodities by HPLC-PDA (HPLC-PDA를 이용한 국내 유통 농산물 중 ametoctradin 잔류량 분석법 개발 및 검증)

  • This study was carried out to validate the safety of ametoctradin residues in agricultural commodities by developing an official analysis method. An analytical method was developed and validated using HPLC-PDA detectors. The samples were extracted with methanol, subsequently partitioned with dichloromethane and purified with florisil column chromatograph using acetone/hexane (30/70, v/v) as solvent. The method was validated by using grape, hulled rice, mandarin, and potato spiked with ametoctradin at 0.05 and 5.0 mg/kg, and pepper at 0.05 and 2.0 mg/kg. Average recoveries were 76-114.8% with relative standard deviation less than 10%, and the limit of detection and limit of quantification were 0.0125 and 0.05 mg/kg, respectively. The result of recoveries and overall coefficient of variation of the laboratory results from Gwangju regional Food and Drug Administration (FDA) and Daejeon regional FDA was accorded with Codex Alimentarius Commission Guideline (CAC/GL 40). Based on these results, this method was found to be appropriate for ametoctradin residue determination and can be used as the official method of analysis.

Physicochemical and Functional Properties of Commercial Whey Powders (시판 유청분말의 이화학적 및 기능적 특성)

  • In order to understand some physicochemical and functional properties of whey powders, imported and domestic products were analyzed. The pH values of imported whey powder solution were $5.85{\sim}6.33$, while those of domestic $5.70{\sim}6.43$. The titratable acidity values of imported whey powders were $0.11{\sim}0.18%$, while those of domestic products $0.10{\sim}0.24%$. The contents of moisture, crude ash, protein, lipid and lactose of the imported whey powder were $1.31{\sim}2.10%,\;7.37{\sim}7.49%,\;11.54{\sim}12.14%,\;0.82{\sim}1.40%\;and\;64.43{\sim}72.66%$, respectively, while those of domestic products $2.11{\sim}2.81%,\;5.39{\sim}8.03%,\;10.41{\sim}20.03%,\;1.88{\sim}2.54%\;and\;54.32{\sim}68.42%$, respectively. The active SH group contents of imported whey powders were $0.36{\sim}0.82{\mu}M/g$, while those of domestic products ranged $0.29{\sim}4.83{\mu}M/g$. The protein solubility of imported whey powders were $54.50{\sim}82.26%$, while that of domestic products $26.93{\sim}68.44%$. The emulsifying capacity and the emulsion stability of imported whey powders were $5.83{\sim}12.53cm^{2}/g$ and $10.24{\sim}12.45%$, respectively, while those of domestic products $6.19{\sim}11.28cm^{2}/g$ and $7.28{\sim}9.93%$, respectively. The foam overrun and stability of imported whey powders were $4.34{\sim}5.54%$ and $0.49{\sim}0.66%$, respectively, while those of domestic products $2.56{\sim}4.24%$ and $0.15{\sim}0.35%$, respectively.

A Demonstrative Study on the Intake Habits of Dairy Goats (Saanen) Fed with Roughages (유산양의 조사료 채식습성에 관한 실증적 연구)

  • The experiment was conducted from 2007 to 2008. The experimental roughages include five sources and 25 species in all; grasses and legumes: 5 species (mixed grasses, orchardgrass, tall fescue, alfalfa, white clover), native grasses and weeds: 5 species (mixed native grasses, Miscanthus sinensis Anderss, Arundinella hirta (Thunb.) Tanaka, barnyard grass, short awn, forage crops and straw: 5 species (barley 4- hairy vetch, wheat 4- hairy vetch, rye silage, barley silage, baled rice straw), browse and fallen leaves: 5 species (mixed browse, oriental white oak browse, Quercus serrta Thunb., browse oriental cherry fallen leaves, Japanese chestnut fallen leaves), and imported hay and straw: 5 species (timothy hay, tail fescue straw, annual ryegrass straw, klinegrass hay, alfalfa hay). Ten dairy goats (Saanen) were selected which had nearly the same body weight (25kg). The experiment was carried out on the dairy goats farm at Geumsan-Cun in Chungnam province. The chemical composition and dry matter digestibility of each roughage source and species were significantly different at the sampling area, plant species, growth stages and cutting period. Among all the 25 species of roughages, the favorite intake species order by dairy goats was observed like this: mixed grasses, white clover, alfalfa and the lower intake species order was baled rice straw and rye silages. The dairy goats ate more roughages which had low fibrous contents, but high dry matter digestibility. On the other hand, compared to each roughage source, the goat's favorite roughage were grasses and legumes (34.6%) among the trial species. Based on the result, it is reconfirmed that the food habit of dairy goats seems to be closer to that of graters.

Physicochemical Characteristics and Volatile Compounds of Glutinous Rice Wines Depending on the Milling Degrees (도정도에 따른 찹쌀발효주의 이화학적 특성 및 휘발성 향기성분)

  • In order to investigate the effects of different milling degrees on the quality of glutinous rice wines, the physicochemical properties and volatile compounds of various wines were evaluated. Sample wines prepared from glutinous rice with 90, 80, and 70% milling yields were analyzed for ethanol, pH, total acids, amino acids, soluble solids, coloring degree, UV absorbance, reducing sugars, organic acids, free sugars and volatile compounds. After fermentation for 17 days, ethanol contents in the wines ranged from 15.2 to 15.85%, while total acid levels ranged from 0.31 to 0.35%. The amino acid contents in four samples ranged from 0.63 to 0.73%, while soluble solid contents ranged from 11.4 to $13.1^{\circ}Bx$. The wine prepared from glutinous rice with a 30% degree of milling showed the highest coloring degree, UV absorbance and reducing sugar content among four samples. Furthermore, this wine had the highest levels of malic acid and acetic acid, while the glutinous rice wine prepared from rice with a 0% degree of milling had the highest levels of succinic acid and lactic acid. In all the glutinous rice wines tested, the most abundant free sugar was glucose followed by maltose. With increasing degree of milling, the alcohol, amino acid and organic acid contents of the glutinous rice wines decreased, whereas soluble solids, coloring degree, UV absorbance, reducing sugar and free sugar contents increased. Volatile compounds were identified using GC-MSD, and thirty-nine esters, seven alcohols, six acids, one aldehyde, four alkanes, one alkene and two miscellaneous compounds were identified in the glutinous rice wines. Using relative peak area, it was determined that other than ethyl alcohol, hexadecanoic acid ethyl ester was the major component and was primarily found in the range of 11.566-18.437%. Succinic acid diethyl ester and isoamyl laurate decreased with an increasing degree of milling, whereas hexanoic acid ethyl ester and 2-octenoic acid ethyl ester increased. Overall, it was shown that different milling degrees greatly affected the physicochemical and volatile characteristics of the glutinous rice wines.

MICROTENSILE BONDING OF RESIN FIBER REINFORCED POST TO RADICULAR DENTIN USING RESIN CEMENT (레진 시멘트를 이용한 레진 파이버 강화 레진포스트의 치근 상아질에 대한 미세인장결합강도)

  • Object The purpose of this study were to evaluate the microtensile bond strength of resin fiber reinforced post to radicular dentin using resin cement according to various dentin surface treatment and to observe the inter face between post and root dentin under SEM Material and Method A total 16 extracted human single rooted teeth were used. A lingual access was made using a #245 carbide bur in a high-speed handpiece with copious air water spray. The post space was mechanically enlarged using H-file(up to #60) and Gates Glidden bures(#3). This was followed by refining of the canal space using the calbrating drill set provided in ER Dentinpost(GEBR, BRASSELER GmbH&Co. KG). The 16 teeth were randomly distributed into 4 group of 4 teeth. Group 1 teeth had their post space prepared using 10% phosphoric acid as root canal surface treatment agent during 20s. The canal was then rinsed with saline and dried with paper point. Group 2 teeth had their post space prepared using 3% NaOCl as root canal surface treatment agent during 30min. The canal was then rinsed with saline and dried with paper point. Group 3 teeth had their post space prepared using 17% EDTA as root canal surface treatment agent during 1min. The canal was then rinsed with saline and dried with paper point. Group 4 teeth had their post space prepared using 17% EDTA as root canal surface treatment agent during 1min. After rinsing with saline, the canal was rinced 10m1 of 3% NaOCl for 30min. After drying with paper point, the post(ER Dentinpost, GEBR, BRASSELER GmbH&Co. KG) was placed in the treated canals using resin cement. Once the canal was filled with resin cement(Super bond C&B sunmedical co. Ltd.), a lentulo was inserted to the depth of the canal to ensure proper coating of the root canal wall. After 24 hours, acrylic resin blocks($10{\cdot}10{\cdot}50mm$) were made. The resin block was serially sectioned vertically into stick of $1{\cdot}1mm$. Twenty sticks were prepared from each group. After that, tensile bond strengths for each stick was measured with Microtensile Tester. Failure pattern of the specimen at the interface between post and dentin were observed under SEM. Results 1. Tensile bond strengths(meen{\pm}SD$) ) were expressed with ascending order as follows group 4, $12.52{\pm}6.60$ ; group 1, $7.63{\pm}5.83$ ; group 2, $4.13{\pm}2.31$ ; group 3, $3.31{\pm}1.44$. 2. Tensile bond strengths of Group 4 treated with 17% EDTA +3%NaOCl were significant higher than those of group 1, 2 and 3 (p<0.05). 3. Tensile bond strengths of Group 1 treated with 10% phosphoric acid were significant higher than those of group 2 (p<0.05). Tensile bond strengths of Group 4 treated with 17% EDTA +3% NaOCl was significant higher than those of other groups.

Evaluation of clinical status of removable partial dentures (가철성 국소의치의 임상적 상태에 대한 평가)

  • Statement of Problem: Although many efforts have been continually made to estimate long term prognosis of removable partial dentures, the complication of removable partial dentures was still found because of inaccurate fabrication procedure and improper maintenance care. Purpose: The purpose of this study was to evaluate the clinical status of removable partial dentures. Material and methods: A total of 112 individuals with 153 removable partial dentures (35 - 87 years, 64 women and 48 men) were examined by intra-oral examination, diagnostic cast and radiographic examination. Results and conclusion: The results of this study were as follows: 1. Length of service of removable partial dentures was $5.3{\pm}4.3$ years (mean), 4.0 years (median). 2. A total of 45 removable partial dentures were considered failures. The loss of 18 abutments of 369 was founded. 3. Type of arch, Kennedy classification and type of opposite dentition were found to have no influence on longevity and success rate of removable partial dentures (P > .05). 4. Most common major connector was the palatal plate in maxilla and the number of lingual bar and linguoplate designed in mandible were similar. 5. The circumferential type retainer was the most commonly used retainer. 6. Sixty-three percent of the class I and II removable partial dentures incorporated indirect retention into the design. 7. Approximately 81% of the removable partial dentures had at least one defect. Excessive wear of posterior teeth (27.9%), lack of integrity (23.2%), lack of stability (22.6%) were frequent defects of removable partial dentures.

Studies on the Feed Value of Yukmijihwangtang Meal (육미지황탕박의 사료가치에 관한 연구)

  • Two experiments were conducted to investigate feed value of Yukmijihwangtang (herbal medicine) meal. In experiment 1, the chemical composition of Yukmijihwangtang ingredients was determined before and after extraction, and investigated the effects of partial substitution of dried Yukmijihwangtang meal on TMR for fattening cattle. In experiment 2, nine Korean native male goats were employed to investigate the palatability and ruminal parameters of Yukmijihwangtang meal silages. In experiment 1, the crude protein content after water extraction was higher in Alisma orientale Juzepczuk (Teaksa), and lower in Pachyma hoelen Rumph (Bokryeong). the crude fat content after water extraction was higher in Cornus officinalis Sieb. et Zucc (Sansuyu), and lower in other ingredients. ADF content after water extraction of Cornus officinalis Sieb. et Zucc (Sansuyu) was higher compared to those of Pachyma hoelen Rumph (Bokryeong) and Paeonia suffruticosa Andrews (Mokdanpi). Dioscorea japonica Thunberg (Sanyak) was the lowest in ADF content. Dry matter disappearance was increased by adding Yukmijihwangtang meal to TMR. In experiment 2, feed intakes of Yukmijihwangtang silage containing molasses (0.5 and 1%) for 30 min and 6 hr were significantly higher than that of control (0%) in Korean native goats. In various ruminl parameters, molasses addition to silage materials trended to increase total VFA, propionate and valerate contents, but trended to decrease iso-butylate, butylate and iso-valerate contents in Korean native goats. In conclusion, Yukmijihwangtang meal could be used as partial substitution of low level in TMR for fattening cattle. Molasses supplementation to silage materials contained with herbal medicine meal could improve its nutritional quality and palatability for Korean native goats. One percent level of molasses addition could give a useful effect in ensiling.

Physicochemical properties of edible cricket (Gryllus bimaculatus) in different districts (산지별 식용 귀뚜라미의 이화학적 특성)

  • Insects have gained increasing attention as an alternative protein and nutrient rich food source for humans. This study was conducted to investigate the physicochemical characteristics and harmful components of edible crickets (Gryllus bimaculatus) in the 6 districts of Yeonggwang (YG), Jeongseon (JS), Wonju (WJ), Hwaseong (HS), Geochang (GC), and Chungju (CJ). The average crude protein and crude lipid contents on a dry basis were 64.34% and 16.60%, respectively. The crude protein content of CJ was the highest (67.40%), whereas YG (59.42%) had the lowest content. On the other hand, the crude fat content of YG was the highest (20.61%), whereas CJ (14.04%) had the lowest content. The unsaturated fatty acid contents were 57.97-63.93 g/100 g of the total fatty acid content in the crickets of the 6 districts. The major fatty acids of the crickets in the 6 districts were palmitic acid, oleic acid, and linoleic acid. Among the essential amino acids, valine, leucine, and lysine were the most abundant. GC had the highest total amino acids (57.93 g/100 g), whereas YG (48.65 g/100 g) had the lowest. Major mineral contents included potassium (K, 0.92~1.01 mg/100 mg) and phosphorus (P, 0.74~0.88 mg/100 mg). The mineral composition was fairly similar among the crickets. Crickets in the 6 districts were verified to have safe levels of residual heavy metals according to the Korea Food & Drug Administration (KFDA) advisory levels.

Influence of Green Tea Powder on the Physical Properties of the Bread Flour and Dough Rheology of White Pan Bread (녹차분말 첨가가 소맥분의 물리적 특성과 제빵적성에 미치는 영향)

  • The purpose of this study was to examine physical properties of the addition of green tea powder on bread flour and dough rheology of white pan bread. Three levels(0.1, 0.5 and 1.0% ) of each green tea powder with bread flour were tested for their effects in dough mixing using rapid disco analyzer, alveogram and farinogram. Addition of green tea powder tended to reduce initial pasting temperature and increase peak viscosity, break down and set back. L(extensibility) and G(swelling index) value in alveogram showed decrement with increasing green tea powder. These meant that the volume of white pan bread would show same tendency. The use of green tea powder increased consistency and water absorption of the bread flour but decreased development time, salability and degree of softening on farinogram. White pan bread with green tea powder had higher value of hardness and springness than without it. Sensory evaluation determined that the white pan bread with 0.5% green tea powder had the highest score.

Effects of Cellulose and Pectin on Diet-induced Thermogenesis in Young Women (한국인 젊은 여성에서 셀룰로오스 및 펙틴이 식이성 발열효과에 미치는 영향)

  • The purpose of this study was to observe the effects of fiber free control diet (CD), cellulose diet (CED) and pectin diet (PTD) on diet induced thermogenesis (DIT) in healthy Korean woman for 3 hours. The three test diets were as follows: CD (carbohydrate intake: 63.4% of energy, protein intake: 14.0% of energy and fat intake: 25.8% of energy), CED (carbohydrate intake: 62.5% of energy, protein intake: 14.0% of energy and fat intake: 26.1% of energy) and PTD (carbohydrate intake: 62.7% of energy, protein intake: 14.0% of energy and fat intake: 26.2 of energy). Groups were served 10 g of cellulose for CED and 10 g of pectin for PTD, respectively. DIT was measured at fasting state and at 30, 60, 90, 120, 150 and 180 min after consuming each diet. The mean age of all subjects was $22.3{\pm}1.9$ years. Body weight was $52.5{\pm}8.6$ kg and body mass index was $20.6{\pm}2.7kg/m^2$. Preprandial resting energy expenditure was $0.79{\pm}0.02kcal/min$ and postprandial DIT were $14.05{\pm}0.62%$ for CD, $9.33{\pm}0.62%$ for CED, $11.07{\pm}1.35%$ for PTD as a percentage of the energy load. DIT of CD was significantly higher than those of CED and PTD. There was no significant difference in postprandial change in body temperature after consuming each test diets and the sympathetic nervous system activity measured by heart rate was significantly higher in CD than CED and PTD (p<0.05). With this study, it can be concluded that CED and PTD have significantly lowered in DIT (p<0.05). We didn't show the correlation of the factors that relate in DIT; thus, further experiments on that matter should be followed.
