양식장 배출수에 첨가된 항생제 제거 위한 수중 비열 유전체장벽 방전 플라즈마 처리 효과 (Effect of plasma treatment using underwater non-thermal dielectric barrier discharge to remove antibiotics added to fish farm effluent)

  • 본 연구의 목적은 양식장 배출수 내에 포함된 5종의 항생제(tetracycline, doxycycline, oxytetracycline, clindamycin 및 erythromycin)를 제거하기 위해 사용한 수중 비열 유전체장벽 방전 플라즈마(Dielectric Barrier Discharge plasma, DBD plasma) 장치 안으로 공기와 산소를 각각 주입했을 때 항생제의 제거효율을 비교하는 것이다. DBD plasma를 발생시키기 위해서 주어진 전압은 27.8 kV이었고, 처리간격은 0, 0.5, 1, 2, 4, 8, 16 및 32분이었다. 3종의 tetracycline계 항생제는 공기를 주입했을 때는 4분만에 유의하게 감소하였고, 산소를 주입했을 때는 30초만에 유의성을 나타내었다. 32분째 공기와 산소를 각각 주입한 결과, tetracycline은 78.1%와 95.8%, doxycycline은 77.1%와 96.3% 그리고 oxytetracycline은 77.1%와 95.5% 감소하였다. Clindamycin은 공기를 주입했을 때 59.6%가 감소되었고, 산소는 83.0% 감소되었다. 또한, erythromycin은 공기주입 시 53.3%가 감소되었고 산소 주입 시 74.3%가 감소하여 두 항생제 모두 tetracycline계 항생제보다 낮은 제거 효율을 보였다. 결론적으로 수중 DBD plasma는 양식장 배출수 내에 포함된 5종의 항생제를 감소시킬 수 있고, 제거 효율은 공기보다 산소를 주입하는 것이 더 효과적이다.

  • This survey was carried out to detect the residual antibiotics and sulfonamides in emergency slaughtered cattle(n=265) from slaughter houses in Kyeonggi province by EEC 4-plates method, Charm II and HPLC. The results were summarized as follows ; 1. Antimicrobial substances were detected from 24 samples(9.1%) by EEC 4-plates method and the detection ratio were highest in summer(13.8%). 2. Twenty-two of the 24 positive samples were classified as sulfonamide(34.4%), TCs(31.3%), $\beta$-lactam(23.5%) and aminoglycoside(9.3%) by Charm II test. 13(59.1%) of the 22 samples contained single agent and 9 samples(40.9%) contained 2 or more agents. 3. Oxytetracycline(27.3%), penicillin G (27.3%) and sulfathiazole(18.2%) were detected in 20 from 22 samples by the HPLC and Charm II test. 4. The residual concentration of oxytetracycline, penicillin G and sulfonamide were 0.29~9.30 ppm, 0.05~9.58 ppm and 0.04~7.59 ppm, respectively and 19 samples(7.2%) were exceeded tolerance levels.

넙치에서 분리된 Edwardsiella tarda의 약제내성 전달성 R plasmid (Transferable R plasmid of Edwardsiella tarda isolated from diseased flounders, Paralichithys olivaceus)

  • 양식넙치에서 분리된 Edwardsiella tarda가 갖는 약제내성을 전달하는 R plasmid의 특성을 알아보기 위하여, 16종의 화학요법제에 대한 E. tarda의 감수성 정도를 비교하였으며, 내성형태 및 내성전달을 확인하고 transferable R plasmid를 분리하였다. E. tarda는 ampicillin, amoxicillin, erythromycin, flumequine, doxycycline(DOXY), nalidixic acid, novobiocin, oxolinic acid, oxytetracycline(OTC), thiamphenicol(TP) 그리고 sulfonamide의 11약제에 대하여 다양한 조합으로 복합내성을 보였으며, DOXY, OTC, TP 내성이 Escherichia coli로 전달되었다. 복합내성의 두 균주에서 transconjugant가 형성되었으며 이들로부터 분리된 transferable R plasmid는 서로 상이한 DNA 구조였다. 이는 넙치에서 분리되는 E. tarda에는 적어도 두 종류 이상의 thansferable R plasmid가 분포한다는 것을 의미한다.

Streptomyces rimosus가 생산하는 Protease의 정제와 특성 (Purification and Properties of Extracellular Protease from Streptomyces rimosus)

  • Oxytetracycline 생산균주인 Streptomyces rimosus를 maltose 2%, NH$_4$Cl 0.5%, yeast extract 0.4 %, MGSO$_4$.7$H_2O$ 0.2% 조성의 배지를 배양초기 PH 6.5로 하여 3$0^{\circ}C$, 72시간 진탕배양하여 얻은, 세포외 protease를 유안분획, Sephadex A-50 이온교환, Sephadex G-100 gel 여과를 행하여 정제하였다. 효소의 최적온도는 5$0^{\circ}C$이며, 최적 pH는 8.0이었으며, Co$^{2+}$ 이온에 의해 활성화되며 Hg$^{2+}$, Fe$^{2+}$ 및 EDTA에 의해 저해를 받으며 casein 분자량을 23,600으로 추정하여 구한 Km값은 2.7$\times$$10^{-4}$M이었다.

Heterologous Expression of Hybrid Type II Polyketide Synthase System in Streptomyces Species

  • Polyketides are an extensive class of secondary metabolites with diverse molecular structures and biological activities. A plasmid-based minimal polyketide synthase (PKS) expression cassette was constructed using a subset of actinorhodin (act) biosynthetic genes (actI-orfl, actI-orf2, actI-orf3, actIII, actⅦ, and actIV) from Streptomyces coelicolor, which specify the construction of an orange-fluorescent anthraquinone product aloesaponarin II, a type II polyketide compound derived from one acetyl coenzyme A and 7 malonyl coenzyme A extender units. This system was designed as an indicator pathway in S. parvulus to generate a hybrid type II polyketide compound via gene-specific replacement. The act ${\beta}-ketoacyl$ synthase unit (actI-orfl and actI-orf2) in the expression cassette was specifically replaced with oxytetracycline ${\beta}-ketoacyl$ synthase otcY-orfl and otcY-orf2). This plasmid-based hybrid PKS cassette generated a novel orange-fluorescent compound structurally different from aloesaponarin II in both S. lividans and S. parvulus. In addition, several additional distinctive blue-fluorescent compounds were detected, when this hybrid PKS cassette was expressed in S. coelicolor B78 (actI-orf2 mutant), implying that the expression of plasmid-based hybrid PKS cassette in Streptomyces species should be an efficient way of generating hybrid type II polyketide compounds.

Elfvingia applanata 엑스가 수종 항생물질의 항균력에 미치는 병용효과 (Antimicrobial Activity of Elfvingia applanata extract alone and in Combination with Some Antibiotics)

  • As part of our search for less toxic antimicrobial agents from natural resources, the carpophores of Elfvingia applanata$(P_{ers}.)K_{ARST}.$ was extracted with hot water. EA, the aqueous extract from the carpophores of E. applanata, was lyophilized and a dark brownish powder was obtained. Antimicrobial activity of EA was tested in vitro against Gram positive and Gram negative bacteria by serial broth dilution method, and the antimicrobial activity was expressed by minimal inhibitory concentration(MIC). Among fourteen species of bacteria tested, the antimicrobial activity of EA was the most potent against Proteus vulgaris showing MIC of 1.250 mg/ml. To investigate the effect of antimicrobial combinations of EA with four kinds of antibiotics(ampicillin, cefazolin, oxytetracycline and chloramphenicol), the fractional inhibitory concentration index(FICI) was determined by checkerboard assay for each strain. The antimicrobial combinations of EA with four kinds of antibiotics resulted in synergism in four instances, but no antagonism was observed. Four instances of synergism were observed when EA was combined with ampicillin against Micrococcus luteus, with cefazolin against Bacillus subtilis, with cefazolin against Micrococcus luteus and with oxytetracycline against Staphylococcus aureus.

Cloning and Characterization of the Tetracycline Resistant Gene, tetB, from Vibrio parahaemolyticus

  • A tetracycline resistant Vibrio parahaemolyticus, capable of growing on TCBS medium containing tetracycline, was isolated from cultivated fishes. A gene responsible for the tetracycline resistance was cloned from chromosomal DNA of the V. parahaemolyticus strain using Escherichia coli KAM3, which lacks major multi-drug efflux pumps (${\Delta}acrB$) as host cells. The nucleotide sequence and homology analysis revealed an open reading frame (ORF) for tetracycline resistance protein (TetB). In order to characterize the antibiotic resistance of TetB originated from the V. parahaemolyticus strain, the gene was sub cloned into plasmid pSTV28. The resulting plasmid was designated as pSTVTetB and transformated into E. coli KAM3. E. coli KAM3 cells harboring the recombinant plasmid pSTVTetB are able to grow on plates containing tetracycline and oxytetracycline but not doxycycline, indicating that the tetB gene confers the tetracycline- and oxytetracycline-resistance to the host cell.

Photocatalytic Degradation of Oxytetracycline Using Co-precipitation Method Prepared Fe2O3/TiO2 Nanocomposite

  • $Fe_2O_3/TiO_2$ nanocomposite were successfully synthesized by co-precipitation method using $Fe(NO_3)_3{\cdot}9H_2O$ and $Ti(SO_4)_2$ as raw materials. Structural and textural features of the mixed oxide samples were characterized by X-ray diffractometer, field emission scanning electron microscopy and energy-dispersive X-ray. The effects of initial concentration of oxytetracycline (OTC), different competitive ions and organics on the photocatalytic degradation rate of OTC by the $Fe_2O_3/TiO_2$ nanocomposite were analyzed under UV and visible light irradiation. The results indicate that the optimized initial concentration of OTC was 50 mg/L to achieve the best photocatalytic efficiency. $Cu^{2+}$, $NH_4{^+}$, $C_3H_8O$ and EDTA in the aqueous suspension were found to suppress the degradation rate of OTC, whereas the effect of $NO_3{^-}$ and $H_2C_2O_4$ can be ignored.

Determination of antibiotics by SPE-LC-MS/MS in wastewater and risk assessment

  • In this study, conditions of solid phase extraction (SPE) for determination of some antibiotics such as trimethoprim, oxytetracycline, erythromycin, clarithromycin, azythromycin, doxycycline, sulfamethazine, ciprofloxacin, chlortetracycline, sulfamethoxazole in wastewaters were optimized. After the optimum volume and pH of the sample were determined, the effect of the concentration of the compounds and matrix were investigated. The highest recovery rates for antibiotic compounds were determined between 82% and 105% in 200 mL sample volume and pH 2.5. Then, antibiotic compounds were investigated in influent and effluent samples taken from Konya Urban Wastewater Treatment Plant.
