Four Fatal Cases of Amiodarone-Induced Pulmonary Toxicity (치명적인 Amiodarone 폐독성 4예)

  • The lungs are frequently the site of adverse drug reactions because of their higher oxygen concentration, the distinctive properties of the pulmonary circulation, and the close proximity of the alveolar epithelium to the blood. Amiodarone, an iodinated benzofuran derivative, is an effective antiarrhythmic drug commonly used for refractory tachyarrhythmia. However, it has a wide range of adverse effects, the most serious of which is lung disease. Most patients present with the insidious onset of dyspnea and a nonproductive cough, and generally recover after withdrawing the drug. We recently experienced four fatal cases of amiodarone pulmonary toxicity. Therefore, we discuss these unusual drug-induced pulmonary toxicity cases with a review of the relevant literature.

Molecular Mechanism of Tetrabromobisphenol A (TBBPA)-induced Target Organ Toxicity in Sprague-Dawley Male Rats

  • Brominated flame retardants (BFRs) are present in many consumer products ranging from fabrics to plastics and electronics. Wide use of flame retardants can pose an environmental hazard, which makes it important to determine the mechanism of their toxicity. In the present study, dose-dependent toxicity of tetrabromobisphenol A (TBBPA), a flame retardant, was examined in male prepubertal rats (postnatal day 18) treated orally with TBBPA at 0, 125, 250 or 500 mg/kg for 30 days. There were no differences in body weight gain between the control and TBBPA-treated groups. However, absolute and relative liver weights were significantly increased in high dose of TBBPA-treated groups. TBBPA treatment led to significant induction of CYP2B1 and constitutive androstane receptor (CAR) expression in the liver. In addition, serum thyroxin (T4) concentration was significantly reduced in the TBBPA treated group. These results indicate that repeated exposure to TBBPA induces drug-metabolising enzymes in rats through the CAR signaling pathway. In particular, TBBPA efficiently produced reactive oxygen species (ROS) through CYP2B1 induction in rats. We measured 8-hydroxy-2'-deoxyguanosine (8-OHdG), a biomarker of DNA oxidative damage, in the kidney, liver and testes of rats following TBBPA treatment. As expected, TBBPA strongly induced the production of 8-OHdG in the testis and kidney. These observations suggest that TBBPA-induced target organ toxicity may be due to ROS produced by metabolism of TBBPA in Sprague-Dawley rats.

Acute Toxicity of Oncheon Stream Water to the Sea Urchin, Hemicentrotus pulcherrimus (말똥성게에 대한 온천천수의 급성독성)

  • This research was conducted to evaluate the effect of polluted Oncheon Stream on the marine organisms in the Suyeong Bay. Water quality and 96 hr acute toxicity to the sea urchin, Hemicentrotns pulcherrimus by recirculation bioassay were examined from Feb. 20 to Apr. 15, 1984. The 96 hr $50\%$ effective concentration($EC_{50}$) on the attachment of the podia of the sea urchin was observed to occur at test concentrations between 40.0 and $51.0\%$ (v/v), and safe concentrations may be assumed to be within 4.0 and $5.1\%$. These values indicate as follows : 1. Oncheon Stream was extremely polluted by oxygen-demanding wastes and synthetic organic compounds from sewage and industrial waste water. 2. Linear alkylbenzene sulfonate(LAS) which has not been yet included in water quality standard was discharged above the TLm. 3. Unknown toxicity may be synergy among complex substances. In consideration of the relationship between COD values of Oncheon Stream and dilution water, the effect of toxicity of Oncheon Stream water reached to the area of the Suyeong Bay where the COD value was found to be 12.2 ppm.

Toxicity assessment of food additive(E171) in aquatic environments (식품첨가물 E171이 수생물에 미치는 독성 평가)

  • E171, a mixture of titanium dioxide, has been widely used as a food additive due to its whitening effect and low toxicity. However, it has been proven that E171 is no longer safe for public health. So far, there are insufficient studies on the toxic effects of E171 on organisms especially using standardized test methods. In this study, toxicity assessments of E171 to two aquatic species, water flea (Daphnia magna) and zebrafish (Danio rerio), were performed using modified standardized test methods based on the physicochemical properties of E171. The hydrodynamic diameter, polydispersity index, and turbiscan stability index (TSI) were measured to ensure the dispersion stability of E171 in exposure media during the test period. The EC50 for immobilization of water flea was 141.7 mg L-1 while zebrafish was not affected until 100 mg L-1 of E171. Measurements of reactive oxygen species (ROS) and antioxidant enzyme activities confirmed that E171 induced oxidative stress, leading to the activation of superoxide dismutase and catalase in both water flea and zebrafish, although the expression of antioxidant enzyme genes differed between species. These results suggested the potential risk of E171 to aquatic organisms and provided toxicological insights into the impacts of E171 on the environment.

Establishment of Marine Ecotoxicological Standard Method for Larval Fish Survival Test (어류 자어의 사망률을 이용한 해양생태독성시험 방법에 관한 연구)

  • Marine ecotoxicological standard method using fish larvae was established with the standard test species of Oryzias latipes(Japanese Medaka) and Paralichthys olivaceus(flounder) and with the 7 day $LC_{50}$ as endpoint. Test method referred to the USEPA(1994) with the replacement of test species found in the Korean water. Standard test species were selected in terms of the species supply and ecological importance in Korean waters. Japanese medaka can be reared with small tanks in the lab and has wide tolerance on salinity, and flounder eggs can be easily obtained from commercial fish hatcheries. General conditions for larval fish toxicity test are as follows. The possible salinity ranges for toxicity test were $0{\sim}35\;psu$ for medaka and >20 psu for flounder. Test type was designated as static non-renewal test if the dissolved oxygen in the test chamber does not fall below 4.0 mg/L. Ages of test species were selected as 7 days after hatched for medaka(about 5 mm TL) and 25 days for flounder(about 10 mm TL) because of the low natural mortality after these periods. Test can be accepted when the survival rates are over 80% in control. Also, species sensitivity on standard reference materials(copper, cadmium or zinc) must be provided with the toxicity test results.

Cytoprotective Constituents of Alpinia katsumadai Seeds Against Glutamate-Induced Oxidative Injury in HT22 Cells

  • Glutamate-induced oxidative injury contributes to neuronal degeneration in many central nervous system (CNS) diseases, such as epilepsy and ischemia. Bioassay-guided fractionation of the MeOH extract of the seeds of Alpinia katsumadai Hayata (Zingiberaceae) furnished three phenolic compounds, alpinetin (1), pinocembrin (2), and (+)-catechin (3). Compounds 2 and 3 showed the potent neuroprotective effects on glutamate-induced neurotoxicity and reactive oxygen species (ROS) generation in the mouse hippocampal HT22 cells. In addition, Compounds 2 and 3 showed significant DPPH free radical scavenging effect. These results suggest that compounds 2 and 3 could be the effective candidates for the treatment of ROS-related neurological diseases.

Excitation Energy Induced S-shaped PL behavior in Graphene Quantum Dots

  • Graphene quantum dots (GQDs) have attracted much attention because of various advantages such as cost-effectiveness of synthesis, low toxicity, and photostability. The origins of photoluminescence (PL) in GQDs were suggested as the intrinsic states for localized sp2 carbon domains and the extrinsic states formed by oxygen-functional groups.[1,2] Nevertheless, it is still unclear to understand the information of electric band structure in GQD. Here, we observed excitation energy induced S-shaped PL behavior. The PL peak energy position shows an S-shaped shift (redshift-blueshift-redshift) as function of the excitation wavelengths. From various samples, we only observed S-shaped PL shift in the GQDs with both luminescent origins of intrinsic and extrinsic states. Therefore, this S-shaped PL shift is related to different weight of intrinsic and extrinsic states in PL spectrum depending on the excitation wavelengths. This would be the key result to understand the electric band structure of the GQDs and its derivatives.

Biocide sodium hypochlorite decreases pigment production and induces oxidative damage in the harmful dinoflagellate Cochlodinium polykrikoides

  • The biocide sodium hypochlorite (NaOCl) is widely used for controlling algal growth, and this application can be extended to marine environments as well. This study evaluates the biocidal efficiency and cellular toxicity of NaOCl on the harmful dinoflagellate Cochlodinium polykrikoides, with emphasis on pigment production and antioxidant enzyme activity. The test organism showed dose-dependent decrease in growth rate on exposure to NaOCl, and the 72 h $EC_{50}$ was measured to be $0.584mg\;L^{-1}$. NaOCl significantly decreased pigment levels and chlorophyll autofluorescence intensity, indicating possible detrimental effects on the photosystem of C. polykrikoides. Moreover, it significantly increased the activities of antioxidant enzymes, suggesting the production of reactive oxygen species in the cells. These data indicate that NaOCl exerted deleterious effects on the photosynthetic machinery and induced oxidative damage in the dinoflagellate and this biocide could be effectively used for the control of algal blooms.

A Study on the Antimicrobial Activity of Copper Alloy Metal Fiber on Water Soluble Metal Working Fluids (수용성 절삭유의 부패 특성과 Copper Alloy Metal Fiber의 부패 방지 장치에 관한 연구)

  • Copper alloy metal fiber was incorporated into the conventional water-soluble metal working fluids to increase the antimicrobial activity. Fluid treated by copper alloy metal fiber is shown that bacteria is disappeared whereas that untreated metal fiber is increased bacteria as increasing the life time. When the electrochemical potential of Cu/Zn ion is -268mV, radicals with molecular oxygen are easily made. Especially, hydroperoxide radical shows strong toxicity to the strains, leading to the conformational change of plasma membrane. As a result antimicrobial activity of copper alloy metal fiber in metal working fluid is superior to that of copper fiber.

  • Acute respiratory failure has become an increasingly frequent cause of death following shock or trauma. Interstitial or diffuse alveolar edema, as chief pathophysiologic change of acute respiratory insufficiency, can be the result of sepsis, fat embolism, cardiac failure, lung congestion, and oxygen toxicity. These pulmonary problems are extremely difficult to treat without early recognition of their development and aggressive management. If the treatment is delayed, the progressive respiratory failure is almost uniformly fatal. Authors have experienced two cases of acute respiratory insufficiency following the blunt chest trauma, which were healed uneventfully. Literatures were briefly reviewed.
