Effect of the Ethanol Extract from Steamed Roots of Rehmannia Glutinosa on the Antioxidant Enzyme Activities in HEI-OC1 Auditory Cells (숙지황 에탄올 추출물이 HEI-OC1 세포의 항산화 효소 활성에 미치는 영향)

  • A mechanism of hair cell damage caused by noise and ototoxic agents is mediated through generation of free radicals and reactive oxygen species (ROS). It is known that most of animals have defense systems to protect against ROS, and the cochlea of inner ear in animals also has ROS defense systems including several antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX), glutathione reductase (GR), and glutathione (GSH), which efficiently detoxifying ROS generated under normal condition. Steamed roots of Rehmannia glutinosa have been traditionally used in Oriental medicine for the treatment of auditory disease such as tinnitus, vertigo, and hearing loss as well as inflammatory diseases, hectic fever, night sweat, and headache. In the present study, we showed that the ethanol extract from steamed roots of R. giutinosa (ESRG) increased the antioxidant enzymes such as SOD, CAT, GPX, and GR activities and GSH level in HEI-OC1 auditory cells. This extract itself did not show any significant cytotoxicity up to $50{\mu}g/ml$. Our results further support the view that ESRG is promising sources of potential antioxidants. Future studies will be aimed at investigating the effects of ESRG on the regulation of cellular mechanisms and isolating and identifying the substances responsible for the regulation of antioxidant enzyme system from the plant extracts.

The Antioxidative Effects of Rhododendron brachycarpum Extracts (만병초(Rhododendron brachycarpum) 추출물의 항산화 효과)

  • The objective of this study was to investigate the antioxidative capacity of Rhododendron brachycarpum 95% ethanol extracts. Total antioxidant status was examined by total antioxidant capacity against ABTS radical reactions. Total antioxidant capacities of R. brachycarpum extract at the concentrations of 0.2 and 1 mg/mL were 0.33 and 2.26 mM Trolox equivalents, respectively. Superoxide scavenging activities of R. brachycarpum extract at the concentrations of 0.2 and 1 mg/mL were 45.0 and 77.0%, respectively. Oxygen radical absorbance capacities of R. brachycarpum extract at the concentrations of 5 and 100 ${\mu}g/mL$ were 40.88 and 131.00 ${\mu}M$ Trolox equivalents, respectively. Total phenolic contents of R. brachycarpum extract at the concentrations of 0.2 and 1 mg/mL were 0.37 and 1.25 mM gallic acid equivalents, respectively. R. brachycarpum extract at the concentration of 0.1 mg/mL inhibited 0.2 mM and 0.5 mM tert-butyl hydroperoxide induced cyototoxicity by 52.1 and 30.3%, respectively, in HepG2 cell culture system. Thus, strong antioxidant and cytotoxicity-inhibiting effects of R. brachycarpum extract seem to be due to, at least in part, the prevention from free radicals-induced oxidation as well as high levels in total phenolic contents.

  • In this study, the antioxidant effect of black sea ginseng containing a large amount of eicosapentaenoic acid, a natural physiologically active substance, was investigated, and its superiority as a functional cosmetic raw material was presented. The ability of the sea cucumber extract to remove active oxygen was evaluated for the inhibitory effect on intracellular nitric oxide (NO) production. The physiologically active substances of sea cucumber have an antioxidant effect that removes free radicals, and as a result of evaluation, it was confirmed that excellent NO production was suppressed. Studies have shown that bioactive substances are involved in antioxidant effects, immune and inflammatory response regulation by showing high antioxidant efficacy even at low concentrations. It is thought that the antioxidant removal ability of sea cucumber greatly contributes to the antioxidant efficacy of phenolic components containing EPA (eicosapentaenoic acid) of the sea cucumber extract. It was found to be similar to the report that the antioxidant component of sea cucumber works. In addition, it is excellent for wound healing with the eicosapentaenoic acid (EPA) component contained in a large amount of sea cucumber extract, which has been proven to be involved in the inhibitory effect of inflammatory activity. As a result of this study, it was confirmed that the effect of the phenol component contained in the sea cucumber extract had a close causal relationship with the antioxidant activity.

Antibacterial and Antiviral Activities of Multi-coating Polyester Textiles (다중 코팅 폴리에스터 섬유 여재의 항균 및 항바이러스 특성)

  • The effect of coated polyester (PET) textiles with metal oxide, chitosan, and copper ion on the antibacterial and antiviral activities was evaluated to investigate the applicability of multi-coated PET textiles as antiviral materials. Compared to coated PETs with a single agent, multi-coated PETs reduced the loading amount of coating materials as well as the contact time with bacteria for a bacterial cell number of < 10 CFU/mL, which was not detectable with the naked eyes. Metal oxides generate reactive oxygen species (ROS) such as free radicals by a catalytic reaction, and copper ions can promote contact killing by the generation of ROS. Chitosan not only enhanced antibacterial activities due to amine groups, but enabled it to be a template to load copper ions. We observed that multi-coated PET textiles have both antibacterial activities for E. coli and S. aureus and antiviral efficiency of more than 99.9% for influenza A (H1N1) and SARS-CoV-2. The multi-coated PET textiles could also be prepared via a roll-to-roll coating process, which showed high antiviral efficacy, demonstrating its potential use in air filtration and antiviral products such as masks and personal protective equipment.

  • Broccoli is considered as one of the functional foods to offer a hearty supply of the nutrient-rich vegetable in the world. Broccoli contains high level of phytochemicals, and that is selected as one of the top 10 vegetables for human health promotion. Especially, glucosinolates and flavonoids are well known as anti free oxygen radicals in vegetables and fruits. In Korea, broccoli consumption has increased to well known on the health-beneficial vegetables since 2000. However, broccoli has many problems of postharvest management since the quality of harvested heads quickly declines. Major problems are the floret yellowing, wilting, off-odor, and decay. The multiple postharvest applications improve broccoli quality and cold treatment including pre-cooling extends on the shelf-life with circumstance of optimum storage which is 0℃ temperature and a range of 95-100% relative humidity. Controlled atmosphere or modified atmosphere packaging can be used as supplemental treatments to extend postharvest life. 1-2% O2 + 5-10% CO2 is currently recommended for broccoli. Postharvest management is important for broccoli because price fluctuations depend on harvest time and quality. In this study, we tried to review physiological change of broccoli after the harvest, storage method, and various techniques to optimize quality during distribution.

Identification and functional prediction of long non-coding RNAs related to oxidative stress in the jejunum of piglets

  • Objective: Oxidative stress (OS) is a pathological process arising from the excessive production of free radicals in the body. It has the potential to alter animal gene expression and cause damage to the jejunum. However, there have been few reports of changes in the expression of long noncoding RNAs (lncRNAs) in the jejunum in piglets under OS. The purpose of this research was to examine how lncRNAs in piglet jejunum change under OS. Methods: The abdominal cavities of piglets were injected with diquat (DQ) to produce OS. Raw reads were downloaded from the SRA database. RNA-seq was utilized to study the expression of lncRNAs in piglets under OS. Additionally, six randomly selected lncRNAs were verified using quantitative real-time polymerase chain reaction (qRT-PCR) to examine the mechanism of oxidative damage. Results: A total of 79 lncRNAs were differentially expressed (DE) in the treatment group compared to the negative control group. The target genes of DE lncRNAs were enriched in gene ontology (GO) terms and Kyoto encyclopedia of genes and genomes (KEGG) signaling pathways. Chemical carcinogenesis-reactive oxygen species, the Foxo signaling pathway, colorectal cancer, and the AMPK signaling pathway were all linked to OS. Conclusion: Our results demonstrated that DQ-induced OS causes differential expression of lncRNAs, laying the groundwork for future research into the processes involved in the jejunum's response to OS.

Effects of Culture Media and Oxygen Concentration on In Vitro Development of Porcine IVM/IVE Embryos (배양액 및 산소농도가 돼지 체외수정란의 발달에 미치는 영향)

  • During in vitro culture of mammalian oocytes and embryos, the cells are exposed to the risks that cause cell injury or death. Numerous studies have been reported that the cell injury may be induced by the action of free radicals generated by auto-oxidation. This study was undertaken to investigate the optimal culture condition system for in vitro culture of porcine embryos. We first evaluated the effect of culture media on the porcine embryo development. NCSU-23 and PZM-5, culture medium tested, were failed to produce significant difference on the rate of blastocyst formation. In NCSU-23, the developmental rate was slightly higher than that in PZM-5. During in vitro maturation (IVM), fertilizaton (IVF), and culture (IVC) under 5 or 20% oxygen ($O_2$), the rates of cleavage and development were insignificantly different from each other under our culture condition (20% $O_2$, in NCSU-23), the mean cell number per blastocyst was $40{\pm}10$. These results showed that medium and $O_2$ concentration had no significant effect on the development of porcine embryos.

The Role of Cyclooxygenase Metabolites in the Pathogenetic Mechanism of Endotoxin-Induced Acute Lung Injury in Domestic Pigs (내독소에 의한 돼지의 급성 폐손상에서 Cyclooxygenase 대사물의 역할에 관한 연구)

  • Background:It has been suggested that the cyclooxygenase metabolites play an important role in changes of early hemodynamic parameters in the endotoxin-induced acute lung injury. But there have been many debates about their role in the late increase of alveolar-capillary permeability, and it is not known whether they act directly or indirectly through oxygen free radicals which have been known to be produced during the metabolic process of cyclooxygenase pathway. So we performed this study to identify the pathogenetic role of cyclooxygenase metabolites in the endotoxin-induced acute lung injury in domestic pigs. Method: We infused endotoxin into 8 domestic pigs; endotoxin only (n=3), and pretreatment with indomethacin (n=5). We observed the sequential changes in hemodynamic parameters, the concentration of plasma oxidized glutathione (GSSG) in pulmonary arterial and venous blood, and albumin content in bronchoalveolar lavage fluid (BALF). Results: 1) While cardiac output decreased, mean pulmonary arterial pressure, pulmonary vascular resistance, and alveolar-arterial oxygen difference increased over phase 1 (0-2hr) and phase 2 (2-4.5hr) by endotoxin, indomethacin attenuated the decrease in cardiac output during phase 1 and increase in mean pulmonary arterial pressure, pulmonary vascular resistance, and alveolar-arterial oxygen difference during both phases. 2) The increase in plasma GSSG content during phase 2 was not attenuated by indomethacin. 3) The content of BALF albumin was significantly lower in indomethacin groups than that of endotoxin group. Conclusion: These results suggest that it is likely that cyclooxygenase metabolites have an effect on endotoxin-induced acute lung injury during both phases probably through direct action.

Comparison of Anti-oxidative Activity in a Single Serving Size of the Commercial Coffees and Teas

  • The aim of this work was to study the comparison of anti-oxidative activity in a single serving size of commercial coffees and teas. Commercial regular coffees and teas, including, brand regular coffees ($BC_A$, $BC_B$, $BC_C$, $BC_D$, and $BC_E$), green tea ($GT_A$, $GT_B$, $GT_C$, and $GT_D$), black tea ($BT_A$, $BT_B$, and $BT_C$), pu-erh tea ($PT_A$, $PT_B$, and $PT_C$), chamomile tea ($CT_A$, $CT_B$, and $CT_C$), peppermint tea ($P_A$, $P_B$, and $P_C$), polygonatum odoratum tea ($POT_A$, $POT_B$, and $POT_C$), and jujube tea ($JT_A$, $JT_B$, and $JT_C$) were assayed for the levels of ascorbic acid, caffeine, total content of polyphenols and flavonoids, and ability to scavenge free radicals, using two in vitro antioxidant assays. The scavenging abilities of $BC_A$ and $BC_C$ were $664.91{\pm}48.87mg$ ascorbic acid equivalent/serving size and $624.36{\pm}16.18mg$ ascorbic acid equivalent/serving size, respectively. The four beverage samples ($BC_A$, $BC_C$, $GT_D$, and $BT_A$) significantly reduced the production of reactive oxygen species (ROS) and intracellular oxidative stress induced by $H_2O_2$. These results suggest that the beverages possess significant radical scavenging ability, which may be due to the presence of antioxidants. Furthermore, the significant reducing level of ROS evidences the potential antioxidant effects of these beverages in human cells.

Advanced Treatment of Sewage and Wastewater Using an Integrated Membrane Separation by Porous Electrode-typed Electrolysis (분리막/다공 전극형 전기분해 조합공정을 이용한 하.폐수의 고도처리)

  • To treat nitrate and non-biodegradable organics effectively in sewage, industrial wastewater and livestock wastewater, the activated sludge process integrated by a membrane separation and a porous electrode- electrolysis was proposed and its efficiency was investigated. The proposed system was consisted of 3 processes; activated sludge, membrane filtration and electrolysis. In the study, the membrane filtration played a role in reducing the load of the electrolysis to operate the proposed process stably. The electrolysis consisted of a porous electrode to increase the efficiency due to the extension of the specific surface area. Additionally, redox reaction in the electrolysis was induced by decomposing influent water as current was applied. As a result, hydrogen free radicals and oxygen radicals as intermediates were produced and they acted as oxidants to play a role in decomposing non-degradable organics. It was environmentally-friendly process because intermediates produced by porous electrode were used to treat waste matters without supplying external reagent. Experimental data showed that the proposed process was more excellent than activated sludge process. SS removal efficiencies of the proposed process, membrane filtration and activated sludge process were about 100%, about 100% and about 90%, respectively. COD removal efficiencies of the proposed system, membrane filtration and activated sludge process were about 92%, about 84% and about 78%, respectively. T-N removal efficiencies of the proposed system, membrane filtration and activated sludge process were about 88%, about 67%, and about 58%, respectively. The SS data showed that SS was efficiently removed in the single of the membrane filtration. The COD/T-N data showed that COD/T-N of membrane hybrid process was treated by removing a little soluble organics and SS, and that COD/T-N of electrolysis hybrid process was treated by oxidize organics with high removal rate.
