A Study on the Worst Stress Condition Test Evaluation of Blowers for Small Stationary Fuel Cell System (소용량 건물용 연료전지시스템 블로워의 가혹조건 평가에 관한 연구)

  • The fuel cell is one of the renewable energy sources. And it is a new source of energy that can be applied to various fuels and continuously supported by the excellent city-gas infrastructure. It is important to improve performances and reliabilities, and reduce the cost of fuel cell systems for commercialization. And, some safety performances of blower domestically produced are evaluated and some improvements are researched to save the cost of fuel cell systems. In this paper, the performance and worst stress condition of blowers are evaluated in operating environment similar to the fuel cell systems. Actually, the correlation of flow, leakage and thermal behavior are evaluated in the worst stress condition at $70^{\circ}C$ and, some major factors of blower degradation such as a motor deterioration, material and structures of the outlet are examined.

Effect of Hydropsyche kozhantschikovi Extracts on Oxidative Stress (줄날도래 추출물이 산화적 스트레스에 미치는 영향)

  • The present study aimed to investigate effects of ethanol extracts from Hydropsyche kozhantschikovi on cell and DNA damage caused by oxidative stress. In a radical scavenging assay, compared with ascorbic acid used as a control, the level of DPPH (1,1-diphenyl-2-picrylhydrazyl) and that of hydroxyl radicals in H. kozhantschikovi extracts were 60.0% and 43.7%, respectively. The ferrous iron chelating level was 37.5% compared to the chelating value of EDTA (ethylenediaminetetraacetic acid) as a positive control at the same concentration. To verify inhibitory effects of oxidative cell damage induced by reactive oxygen species (ROS), the relative level of lipid peroxidation and the expression level of the p21 protein were compared in extracts-treated and untreated groups. Lipid peroxidation was completely inhibited in the extracts-treated group compared with the radical-only treated group. The level of p21 protein expression was restored to 92.2% of p21 protein expression in the control sample. In addition, DNA cleavage inhibition in the H. kozhantschikovi extracts was 74.1% compared with that of the control group, suggesting that H. kozhantschikovi extracts repress DNA cleavage induced by ROS. Moreover, the phosphorylation ratio of the H2AX protein was 16.7% in the radical-treated group, indicating that the ethanol extracts inhibited 83.3% of DNA damage. Our findings suggest that ethanol extracts from H. kozhantschikovi are effective not only in repressing the oxidation of free radicals and highly toxic hydroxyl radicals, but also in decreasing cell and DNA damage caused by oxidative stress.

Oxidative Stress and Antioxidant Responses in Poplar Clones Irrigated with Livestock Waste Leachate (양돈폐수 처리에 의한 포플러클론의 산화스트레스와 항산화반응)

  • We studied the influence of livestock waste leachate on oxidative damage and antioxidative responses in poplar clones in August which increase the demand of antioxidants because of high temperature and high light during this period. We measured ion leakage, antioxidant enzyme activities (APX, GR), and carotenoid contents. Oxidative damage and antioxidative responses by treated livestock waste leachate in poplar clones showed various results. We divided poplar clones into three groups using the criteria based on ion leakage which represent cell damage induced oxidative stress. Eco 28, 62-10, Bonghwa1 and Dorskamp belonged to the first group in which the cell damaged level was lower than that of the control. The results suggest that this group augmented for demand of antioxidative in summer because high concentration of nitrogen induced by treatment of live stock wastes acted as environmental stress. Consequently, they failed to keep up the homeostasis of reactive oxygen species. The second group in which the cell damaged level was similar to that of the control was Suwon, 72-30 and 72-31 clones. Finally, 97-18 clone belonged to the third group in which the cell damaged level was lower than that of the control group. In this case, nitrogen treated by livestock waste leakage decreased oxidative stress. 97-18 clone was the clones with the least damage in summer oxidative stresses treated by livestock waste leakage. These results suggest that the high concentration nitrogen due to the livestock waste leakage can act differently upon the clones. We speculate that the added oxidation damage in the summer (growing season) may have an effect on the total fresh weight and also influence the purification ability for livestock waste leakage. However, further studies are needed for the confirmation.

Mechanical Properties and Microstructure of Mg-Zn-(Mn)-Ca Alloys (Mg-Zn-(Mn)-Ca 합금의 미세조직 및 기계적성질)

  • The microstructure and tensile properties of Mg-Zn-Ca and Mg-Zn-Mn-Ca alloys have been investigated. The alloys were obtained by melting in a low carbon crucible coated with boron nitride under an Ar gas atmosphere to prevent oxidation and combustion. The Mg alloy melt was cast into the metallic mold at room temperature, and cooling part was located at the bottom of mold. The phase formed during solidification of the Mg-Zn-(Mn) alloys containing 0.5%Ca is $Ca_2Mg_6Zn_3$. The yield strength and ultimate tensile strength of the alloys increased with increasing Zn content, but the ductility did not change with increasing Zn content. The addition of Mn improves the yield strength and ultimate tensile strength of the alloys, but the ductility did not change. Tensile fracture of the alloys revealed brittle failure, with cracking along the $Ca_2Mg_6Zn_3$ phase. The variation of stress with strain obeyed the relationship of the ${\sigma}=K{\varepsilon}^n$.

Molecular Cloning and Expression of Sequence Variants of Manganese Superoxide Dismutase Genes from Wheat

  • Reactive oxygen species (ROS) are very harmful to living organisms due to the potential oxidation of membrane lipids, DNA, proteins, and carbohydrates. transformed E.coli strain QC 871, superoxide dismutase (SOD) double-mutant, with three sequence variant MnSOD1, MnSOD2, and MnSOD3 manganese superoxide dismutase (MnSOD) gene isolated from wheat. Although all QC 871 transformants grown at $37^{\circ}C$ expressed mRNA of MnSOD variants, only MnSOD2 transformant had functional SOD activity. MnSOD3 expressed active protein when grown at $22^{\circ}C$, however, MnSOD1 did not express functional protein at any growing and induction conditions. The sequence comparison of the wheat MnSOD variants revealed that the only amino acid difference between the sequence MnSOD2 and sequences MnSOD1 and 3 is phenylalanine/serine at position 58 amino acid. We made MnSOD2S58F gene, which was made by altering the phenylalaine to serine at position 58 in MnSOD2. The expressed MnSOD2S58F protein had functional SOD activity, even at higher levels than the original MnSOD2 at all observed temperatures. These data suggest that amino acid variation can result in highly active forms of MnSOD and the MnSOD2S58F gene can be an ideal target used for transforming crops to increase tolerance to environmental stresses.

Antimicrobial and Antioxidant Properties of Secondary Metabolites from White Rose Flower

  • Low-molecular-weight secondary metabolites from plants play an important role in reproductive processes and in the defense against environmental stresses or pathogens. In the present study, we isolated various volatiles and phenolic compounds from white Rosa rugosa flowers, and evaluated the pharmaceutical activities of these natural products in addition to their ability to increase survival in response to environmental stress and pathogen invasion. The DPPH and hydroxyl radical-mediated oxidation assay revealed that the white rose flower extract (WRFE) strongly scavenged free radicals in a dose dependent manner. Moreover, WRFE inhibited the growth of E. coli and fatally attacked those cells at higher concentration (>0.5 mg/mL). FITC-conjugated Annexin V stain provided further evidence that WRFE had strong antimicrobial activity, which may have resulted from a cooperative synergism between volatiles (e.g. 1-butanol, dodecyl acrylate and cyclododecane) and phenolic compounds (e.g. gallic acid) retained in WRFE. In conclusion, secondary metabolites from white rose flower hold promise as a potential natural source for antimicrobial and non-chemical based antioxidant agents.

  • Worldwide there is concern about the continuing release of a broad range of environmental endocrine disrupting chemicals, including polychlorinated biphenyls, dioxins, phthalates, polybrominated diphenyl ethers (PBDEs), and other halogenated organochlorines persistent organic pollutants (POPs) into the environment. They are condemned for health adverse effects such as cancer, reproductive defects, neurobehavioral abnormalities, endocrine and immunological toxicity. These effects can be elicited via a number of mechanisms among others include disruption of endocrine system, oxidation stress and epigenetic. However, most of the mechanisms are not clear, thus several number of studies are ongoing trying to elucidate them in order to protect the public by reducing these adverse effects. In this review, we briefly limited review the process, the impacts, and the potential mechanisms of dioxin/dioxin like compound, particularly, their possible roles in adverse developmental and reproductive processes, diseases, and gene expression and associated molecular pathways in cells.

Effects of a Rubus coreanus Miquel supplement on plasma antioxidant capacity in healthy Korean men

  • Korean raspberry, Rubus coreanus Miquel (RCM), contains high concentrations of phenolic compounds, which prevent oxidative stress. To determine the effect of RCM on antioxidant capacity in humans, we assessed in vivo lipid oxidation and antioxidant enzyme activities from plasma in 15 healthy men. The subjects ingested 30 g of freeze-dried RCM daily for 4 weeks. Blood was taken at baseline and at the end of the study to determine blood lipid profiles, fasting plasma glucose, liver function, lipid peroxidation, and antioxidant enzyme activities. RCM supplementation had no effect on blood lipid or fasting plasma glucose concentrations but decreased alkaline phosphatase activity. RCM supplementation increased glutathione peroxidase activities (P<0.05) but had no effect on lipid peroxidation. These results suggest that short-term RCM supplementation may offer health benefits by enhancing antioxidant capacity in a healthy population.

  • Hsp33 is a molecular chaperone achieving a holdase activity upon response to a dual stress by heat and oxidation. Despite several crystal structures available, the activation process is not clearly understood, because the structure inactive Hsp33 as its reduced, zinc-bound, monomeric form has not been solved yet. Thus, we initiated structural investigation of the reduced Hsp33 monomer by NMR. In this study, to overcome the high molecular weight (33 kDa), the protein was triply isotope-[$^{13}C$, $^{15}N$, $^2H$]-labeled and its inactive, monomeric state was ensured. 2D-[$^1H$, $^{15}N$]-TROSY and a series of triple resonance spectra could be successfully obtained on a high-field (900 MHz) NMR machine with a cryoprobe. However, under all of the different conditions tested, the number of resonances observed was significantly less than that expected from the amino acid sequence. Thus, a possible contribution of dynamic conformational exchange leading to a line broadening is suggested that might be important for activation process of Hsp33.

The Study on the Effect of Electroacupuncture at Shinmaek($BL_{62}$) on Anti-Oxidation (흰쥐의 신맥(申脈)($BL_{62}$)에 시술한 저주파 전침자극의 항산화 효과에 대한 실험적 연구)

  • Objectives : The purpose of this study is to investigate the anti-oxidative effect of electroacupuncture at Shinmaek($BL_{62}$) in rats. Methods : In order to cause oxidative stress, AAPH was administered to the abdominal cavity of rats, after we stimulated $BL_{62}$ of the rat by electroacupuncture. Blood test and anti-oxidative test(LDL-cholesterol, GOT, GPT SOD, GSH, catalase, NO, MDA) were performed at the end of treatment. Results: 1. SOD, glutathione, catalase activity were significantly increased in the $BL_{62}-EA$ group compared with the holder group. 2. NO density was significantly decreased in the $BL_{62}-EA$ group compared with the holder group. 3. Glutathione was significantly increased in the $BL_{62}-EA$ group compared with the sham-EA group. Conclusion : These results suggest that electroacupuncture at $BL_{62}$ has an anti-oxidant effect in human.
