Prevention of UV-induced Skin Damage by Activation of Tumor Suppressor Genes p53 and p14ARF

  • UV radiation is the most dangerous stress factor among permanent environmental impacts on human skin. Consequences of UV exposure are aberrant tissue architecture, alterations in skin cells including functional changes. Nowadays new kinds of outdoor leisure-time activities and changing environmental conditions make the question of sun protection more important than ever. It is necessary to recognize that self-confident consumers do not consider to change their way of life, they demand modern solutions on the basis of new scientific developments. In the past one fundamental principle of cosmetics was the use of physical and organic filter systems against damaging UV-rays. Today new research results demonstrate that natural protecting cell mechanisms can be activated. Suitable biological actives strongly support the protection function not from the surface but from the inside of the cell. A soy seed preparation (SSP) was proven to stimulate natural skin protective functions. The major functions are an increased energy level and the prevention of DNA damage. These functions can I be defined as biological UV protection. The tumor suppressor protein p53 plays a key role in the regulation of DNA repair. p53 must be transferred into the phosphorylated form to work as transcription factor for genes which are regulating the cell cycle or organizing DNA repair. A pretreatment with SSP increases the phosphorylation rate of p53 of chronically UV-irradiated human keratinocytes significantly. According to the same test procedure SSP induces a dramatic increase in the expression of the tumor suppressor protein p14ARF that is supporting the p53 activity by blocking the antagonist of p53, the oncoprotein Mdm2. Mdm2, a ubiquitin E3-ligase, downregulates p53 and at the same time it prevents phosphorylation of p53. The positive influence of the tumor suppressor proteins explains the stimulation of DNA repair and prevention of sunburn cell formation by SSP, which was proven in cell culture experiments. In vivo the increased skin tolerance against UV irradiation by SSP could be confirmed too. We have assumed, that an increased repair potential provides full cell functionality.y.

Beginning Earth Science Teachers' Perceptions about Learning in Geologic Field Trip (야외 지질 학습에 대한 초임 지구과학 교사의 인식)

  • The purpose of this study was to investigate beginning earth science teachers' perceptions about Learning in Geologic Field Trip. The hundred and sixteen high school earth science teachers who were in the training for their promotion answered the questionnaire of Learning in Geologic Field Trip. It was divided into five parts: needs and educational values, teachers' experiences and their difficulties, and desirable teaching methods of Learning in Geologic Field Trip. Results indicated that the participants perceived Learning in Geologic Field Trip important in earth science education and responded positively to the educational values of Learning in Geologic Field Trip. Most of the beginning teachers had experience of geologic field trip, but did not learn how to teach it to their students during the geologic field trip in the pre-service training program. It was shown that only a few teachers had experience of geologic field trip and of teaching students in the outdoor of geologic fields after they became a classroom teacher. Most of the participants perceived that teaching students about the geologic field trip is difficult because of lack of their experience. In order to overcome the difficulty, teachers suggested that they need to learn how to teach the geologic field trip to students through their in-service training. leachers suggested that a desirable teaching method of the geologic field trip is to provide teachers with a teacher's manual including geological explanation and information about field trip sites before their visit so that they can scaffold it and facilitate the students' cooperative group activity during the field trip. As a follow up activity, the participants agreed that the presentation and discussion about the results should concur.

Relaxing Effect of Evening Primrose Root on Skin Irritation Caused by Particulate Matter in Subway Tunnel (지하철 미세먼지에 의하여 유발되는 피부염증에 대한 달맞이꽃 뿌리 추출물의 완화 효과)

  • If human skin is exposed to high concentrations of particulate matter (PM2.5 and PM10) for a long time in the outdoor environment such as subway tunnel, it will be adversely affected. In particular, fine particles can damage the skin, causing inflammation and allergic reactions. This study investigated the ability of evening primrose root (EEPR) extract to suppress the skin damages caused by the fine particles. PM was collected from a subway tunnel, where high concentrations have been reported per day over the course of a study. The EEPR had higher antioxidant activity than that of control group (62.6%). The mixture of EEPR and PM inhibited the production of nitric oxide (NO), thereby alleviating skin inflammation caused by fine particle dust. EEPR had weaker cytotoxic activity than the positive control. When cells were exposed to particulate-type dust (PM10), the levels of free radicals were decreased with the increased concentrations of the extract (5, 10, 20 ㎍/mL). While at the same time more effective than positive controls. Therefore, this study proved that the Moonlight flower root extract can be used as a cosmetic material for skin by providing an effect to alleviate skin damage caused by fine particle-type dust.

The Effects of Sunscreen Use Patterns and Awareness by Type on Product Satisfaction (자외선 차단제 유형에 따른 사용 인식 요인과 재구매 및 만족도 조사에 관한 연구)

  • In this study, 340 Korean women were surveyed to analyze the impact of the type of sunscreen and use status on the satisfaction of products and resale. Studies have shown that indoor activity is higher than outdoor activity, and both simple and complex sunscreen is often used to compensate for skin tone. Out of single sunscreen, sun cream was used and air cushion was used a lot. Although product usability and UV protection indices were important factors in purchasing, 30% experienced side effects while product satisfaction and willingness to repurchase were high. The recognition of safety, Effectiveness, convenience, usability, satisfaction, and repurchase were all higher than that of composite UV-blocking products, and among subcomponents of product usage recognition, the level of convenience, efficacy, usability, and safety affected satisfaction and repurchase in order. Consequently, the perception of simple UV protection products was higher when using products for UV protection, and its convenience had a significant impact on satisfaction and resale.

Radiological and Geochemical Assessment of Different Rock Types from Ogun State in Southwestern Nigeria

  • Background: This paper deals with the study of natural radioactivity in rocks from Ogun State in Southwestern Nigeria. The aim is to determine radiation emissions from rocks in order to estimate radiation hazard indices. Objectives: The following objectives were targeted: 1. To determine radiation emissions from each type of rocks; 2. To estimate radiation hazard indices based on the rocks; 3. To correlate the activity concentrations of radionuclides with major oxides. Methods: The samples were analyzed using a NaI (Tl) gamma ray spectrometric detector and PerkinElmer AAnalyst 400 AAS spectrometer. Results: The activity of 40K, 226Ra, and 232Th were found in order of decreasing magnitude from pegmatite>granite>migmatite. In contrast, lower concentrations were found in shale, phosphate, clay stone, sandstone and limestone. The mean absorbed doses were 125±23 nGyh-1 (migmatite), 74±13 nGy/h (granite), 72±13 nGyh-1 (pegmatite), 64±09 nGyh-1 (quartzite), 45±16 nGyh-1 (shale), 41±09 nGyh-1 (limestone), 41±11 nGyh-1 (clay stone), 24±03 nGyh-1 (phosphate), and 21±10 nGyh-1 (sandstone). The outdoor effective dose rates in all rock samples were slightly higher than the world average dose value of 0.34 mSvy-1. The percentage composition of SiO2 in the rock samples was above 50 wt% except for in the limestone, shale and phosphate. Al2O3 ranged from 4.10~21.24 wt%, Fe2O3 from 0.39~7.5 wt%, and CaO from 0.09-46.6 wt%. In addition, Na2O and K2O were present in at least 5 wt%. Other major oxides, including TiO2, P2O5, K2O, MnO, MgO and Na2O were depleted. Conclusions: The findings suggest that Ogun State may be described as a region with elevated background radiation. It is recommended that houses should be constructed with good cross ventilation and residences should use home radiation monitoring instruments to monitor radon emanating from walls.

Analysis and Improvement of 'Life cycle of Butterfly' in the Science Textbook for 3rd Grade Elementary Students (초등학교 3학년 과학교과서에 제시된 배추흰나비의 한살이 내용분석 및 개선 방안)

  • The life cycle of cabbage-white butterfly is presented in textbooks as an activity to understand the life cycle of animals for 3rd graders in elementary schools. This study was intended to analyze the contents of the current science textbook in the 2015 revised curriculum and suggested improvement of contents. The life cycle of cabbage-white butterfly presented in the textbook consists of eggs, caterpillars, pupae and adult insects in linear steps, and there is a shortage of activity to show the full life cycle. To improve the contents, we checked life cycle of cabbage-white butterfly through a series of experiments. To collect eggs in the out field, kale and cabbage seedlings were placed next to outdoor flowers, next to artificial butterfly feeder, and in grasses without flowers during June. The largest number of butterfly eggs were found from kale beside flowers. The sponges soaked in sugar water or honey water were provided for adult butterflies in ground-type, hook-type and ceiling-type form. Most absorbent activities were observed on the sponge soaked in honey water installed on the ceiling. From the third day, butterfly started to lay eggs. In order to incorporate these facts in the textbook, we suggest to reorganize the relevant contents of the textbook as follows. The kale seedlings should be placed next to the flowers on the school flower bed from June to collect the cabbage white butterfly eggs. A sponge soaked with honey should be placed on top of the breeding net for butterflies. Breeding net should be big enough to keep butterfly flying freely and several kale flowerpots. After laying eggs, butterflies should be released in the field. This process will give students an better understanding of the life cycle of animal.

Anti-oxidative and protective effects of Arthrospira platensis ethanol extracts on zebrafish ROS Induced by UVB Induction (UVB 로부터 ROS를 유도한 제브라피쉬에 스피룰리나 에탄올 추출물의 항산화 및 보호효과에 관한 연구)

  • Arthrospira platensis is one of the oldest algae in the world and has been reported to have anti-aging properties, including phycocyanin, tocopherol and beta-carotene. In this study, we tried to search protective activities against UVB-induced reactive oxygen species(ROS) of Arthrospira platensis under indoor cultivation ethanol extracts(ICAE) and outdoor cultivation ethanol extracts(OCAE). The anti-oxidative capacities were evaluated by DPPH radical scavenging activity and SOD-like activities at various concentrations(0.1, 0.5, 1mg/m) of ICAE and OCAE. Zebrafish embryos and HaCaT cells were exposed to UVB radiation and treated with various concentrations(0, 0.01, 0.05, 0.1, 0.5, 1mg/m) of ICAE and OCAE. ROS levels of zebrafish and HaCaT cells were generated by UVB radiation. ROS levels were detected using a fluorescent microscope after DCFH-DA staining. The DPPH radical scavenging activity of ascorbic acid was 73% and SOD-like activity was 86% in the positive control group. ICAE and OCAE at 1mg/m concentration showed 43, 57% DPPH radical scavenging activity and 20, 19% SOD-like activity. Anti-oxidative of ICAE and OCAE had lower effects than the positive control ascorbic acid but significant results. ROS of UVB-induced zebrafish embryos and HaCaT cells were higher than negative control. ICAE and OCAE treated group decreased ROS concentration dependently than UVB-induced positive control group. These results suggest that Arthrospira platensis ethanol extract may have usability value as a cosmetic material for skin protection.

Airborne Bacteria Concentration and Species Identification in Residential Living Spaces (주택내 주거공간에 따른 부유세균 농도 분포 및 종 동정 연구)

  • Objectives: Exposure to airborne bacteria is associated with adverse health effects such as respiratory and infectious diseases. This study evaluated airborne bacterial concentrations in the living rooms, kitchens, and toilets of 30 homes. Methods: Bacteria were sampled with an MAS100 impactor in three spaces in the subject homes between April 2014 and February 2015. Bacteria were grown on TSA plates for 48 hours at 35C. The bacterial strains were isolated and amplified by polymerase chain reaction. Results: The most culturable bacteria were found in toilets (624.0CFU/m3, GM: 417.3CFU/m3), followed by in the kitchen (503.8CFU/m3, GM: 324.9CFU/m3). The dominant genera identified were: Staphylococcus sp.(19%), Micrococcus sp.(16%), and Bacillus sp.(11%) in the indoor air and Bacillus sp. (30%) in the outdoor air. Gram-positive bacteria comprised more than half of all colonies. Conclusion: In this study, culturable bacteria concentrations were higher than those reported in other spaces. Therefore, it is important to control relative humidity and remove moisture to prevent bacteria from multiplying. Additionally, the dominant species in indoor air were Staphylococcus sp. and Micrococcus sp. These are found on the human skin, mucous membranes, and hair, so human activity can affect bacterial distribution. Therefore, cleaning and controlling moisture are important for reducing indoor bacterial concentrations.

Landscape Design of Gamcheon Wholesale Fish Market (감천항 수산물 도매시장 조경설계)

  • The landscape disign of Gamcheon wholesale fish Market was designed around a turnkey base to promote the economy of Busan Metropolitan City, to establish a fishery marketing center and to modernize existing facilities. The objectives of the project were to promote the mood of an old market, while preserving its functions and efficiency as a market, to design outdoor spaces with natural resources and amenities in balance, and to create attractive tourist spots in connection with the wide area development plan. The project was oriented, fast, to enhance the functions of the market. For this purpose, a multi-dimensional space layout was designed in consideration of functions as a wholesale market. The safety of pedestrians was secured by separating lathes for vehicles and for pedestrians. Tree planting with various functions such as sheltering, wind breaking and guiding was planned. Secondly, nature-friendly and human-friendly landscaping design was attempted. For this, the beautiful natural resources of Amnam Park were utilized, and green spaces such as green bridges linking buildings in the wholesale market, and rooftop gardens were to be arranged. In addition, environment-friendly facilities such as roads paved with natural materials(i.e. gravel, shells) and program parking lots were to be planned. Thirdly, landscape design was considered to create attractive tourist spots. For example, a fish farm was created as a theme street for pedestrians and various water-friendly spaces such as pedestrian ramps, observatories and seaside streets were to be secured. The main contents are as follows. First, a green bridge to Ahnnam Park was introduced for a tour source and flower garden, an event plan and viewing deck open to the sea were planned on the bridge's axis. Secondly, for the effective land use plan concerning open space and convenience to visitors, a promenade was planned, which is connected with the theme plaza and small plazas by environmental sculptures in front of the market hall and at the gate. As well, an observatory and a roof garden help create three dimensional multi leveled space, with a good view of the natural landscape of the sea, sky and park Thirdly, landscape materials, such as trees and those for facilities, strengthened for protection against the seawind and salt damage were selected. The commercial market area was intended to be transformed a traditional functional area of efficiency and economy into an attractive marine leisure area where both tourists and neighbors can make use of it.

Analysis of Hygrothermal Performance for Standard Wood-frame Structures in Korea (국내 농어촌 표준 목조주택의 hygrothermal 성능 분석)

  • As recent buildings become more air tight, the natural ventilation rate is significantly reduced and it leads to difficulty in removing accumulated moisture in buildings. Hot and humid weather in summer and the large amount of moisture caused by indoor activity are the major factors of moisture problem in Korea. The hygrothermal behavior of building environment has to be considered carefully to reduce condensation risk and mold growth potential, and comfortable indoor environment. In this study, we evaluated hygrothermal behavior of Standard Wood-frame Structure published in the Korea Rural Community Corporation Using WUFI simulation program. The results indicated that the total water contents of wood wall measured in 2014 was lower than wood wall in 2010. As a result of evaluation by separating the farming and fishing areas, Moisture problems in fishing area became larger. The walls had a significant impact on the relative humidity than the temperature each areas. Furthermore, excessive water content problem of the wood-based material was reduced in the wall that could be applied in the fishing villages by changing the outdoor finishing materials. And Mold growth risk on the interior materials could be removed through the different setting of the indoor temperature during summertime.
