Effect of Fermented Angelica gigas Nakai on Lipid Metabolism in Orotic Acid Model Rats (발효당귀가 Orotic acid 유발 흰쥐 지질 대사에 미치는 영향)

  • The aim of this study was to identify the effect of fermented Angelica gigas Nakai (A. gigas) on lipid metabolism in orotic acid-induced fatty liver model rats. Sprague-Dawley male rats were randomly divided into four dietary groups (n=6 per group): a normal (N) group fed a standard diet only, OA control, OA acid plus 5% (w/w) A. gigas (OAG), and OA plus 5% (w/w) fermented A. gigas (OFAG). OA treatment induced enlargement of the liver and accumulation of hepatic triglycerides. The consum ption of fermented A. gigas reduced triglyceride concentrations in the liver and increased the serum lipid concentrations to normal levels. Furthermore, OA treatment significantly decreased serum triglyc eride concentrations without diminishing mRNA expression of microsomal triglyceride transfer protei n (MTP) and protein disulfide isomerase (PDI). Hepatic MTP mRNA expression increased 1.08-fold in response to OA treatment, despite triglyceride accumulation in the liver relative to that of the normal group. OFAG administration was slightly lower as compared to the OA treatment. This result suggests that MTP mRNA expression is not always correlated with hepatic triglyceride accumulation in the OA-induced fatty liver model. However, PDI mRNA expression was significantly increased in the OAG and OFAG groups (1.62-fold and 1.63-fold, respectively) compared with the normal group. The hepatocytes in the OA group contained numerous large fat droplets. These were slightly reduced in the OFAG group.

  • 새로운 간장질환 치료제로 개발중인 YH439가 orotic acid, nicotinamide 및 ethionine에 의해 유발되는 지방간에 대한보호 및 치료효과를 관찰하였다. 웅성 SD계 rat에 orotic acid(1%) 또는 nicotinamide(2%)가 첨가된 사료를2주간 섭취시켜 유발된 지방간에 대한 YH439의 보호 및 치료효과와 ethionine(100mg/kg, i.p.)투여로 유도되는 지방간에 대한 YM439의 보호효과를 관찰하기 위하여 간 조직 중 triglyceride, cholesterol 및 phospholipid의 함량을 측정하여 비교하였다. 그 결과, erotic acid 또는 nicotinamide의 섭취로 인해 랫드의 간 조직 중 triglyceride, cholesterol 및 phospholipid의 함량은 1.5-3배정도 증가하였으며, 이 증가된 지질들은 YH439 100, 200mg/kg 투여에 의해 유의성 있게 억제되었다. 또한 ethionine투여에 의해서도 랫드의 간 조직 중 triglyceride, cholesterol 및 phospholipid의 함량은 약 2배정도 증가되었으며, YH439 전저치에 의해 이 현상들이 억제되는 효과를 나타내었다.

Improvement Effect of Fermented Orostachys malacophyllus against Orotic Acid-induced Fatty Liver Model Rats (발효 와송 급여 흰쥐의 orotic acid 유발 지방간 개선 효과)

  • This study aimed to evaluate the protective effect of Orostachys malacophyllus (OM) and fermented O. malacophyllus (FOM) in Sprague-Dawley rats who had been intoxicated with 1% (w/w) orotic acid (OA) for 10 days. The activities of several hepatic enzymes, aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), and cholinesterase, were increased when OA was given, but these parameters were significantly decreased by FOM treatment. In addition, OA treatment resulted in an increased lipid peroxidative index (thiobarbituric acid-reactive substances, TBARS). A worsened antioxidant status (reduced glutathione) in the liver and serum was also observed. FOM treatment improved the antioxidant status of OA-induced fatty-liver rats, which was evaluated by decreased levels of the lipid peroxidative index and improved antioxidant status in the liver and serum. The contents of liver non-heme iron were increased with OA treatment and significantly decreased with FOM treatment, which suggested that the lipid peroxidation contents were inversely correlated with liver non-heme iron content. Based on these results, FOM is considered a material with significant potential for development into a functional health food that can improve fatty-liver conditions.

  • This study was to investigate the potential effects of fermented Angelica gigas Nakai (FAG) at 5% (w/w) levels in Sprague-Dawley strain rats, which were intoxicated with 1% (w/w) orotic acid (OA) for 10 days. The activities of several hepatic enzymes, including aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), and cholinesterase were increased when OA was treated, but these parameters were significantly decreased by FAG administration. OA treatment induced a significant increase in the thiobarbituric acid reactive substances (TBARS) levels, which was attenuated by FAG administration. Liver nonheme ion was decreased in the OA treatment group and was significantly increased in FAG administration, which suggests that lipid peroxidation contents are inversely correlated with liver nonheme ion content. The glutathione concentration was significantly decreased in the OA treatment group compared with the normal group, but this concentration was significantly increased in the FAG group, and it showed the antioxidant ability of glutathione. Based on these results, fermented Angelica gigas Nakai is a material with significant potential for development into a health food that can improve fatty liver conditions.

Effect of Cordycepin-Enriched Cordyceps militaris Powder on Tissues Lipid Peroxidation and Antioxidative Activity in Orotic Acid-Induced Fatty Liver Model Rats (Cordycepin이 orotic acid 유발 지방간 흰쥐의 과산화 지질 농도 및 항산화 활성에 미치는 영향)

  • This study is aimed to evaluate the protective effect of Cordycepin-enriched Cordyceps militaris (CM${\alpha}$) strain on orotic acid (OA)-induced fatty liver in rats. OA treatment induced the retardation of body weight gain and enlargement of the liver. The activities of liver marker enzymes, alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH), Alk. phosphatase and Cholinesterase were increased when treated with OA, but these parameters were significantly decreased in the CM${\alpha}$ group. The current study observed significant elevations of thiobarbituric acid-reactive substance (TBARS) levels. However, among the OA groups, the CM${\alpha}$ group showed significantly low TBARS levels of hepatic homogenate. The OA group resulted in a significant decrease in the levels of plasma and hepatic glutathione, but these reductions were significantly increased in the CM${\alpha}$ group. These effects were more pronounced in the CM${\alpha}$ group than in the PJ or CM groups in Orotic acid treated rats. Accordingly, Cordycepin-enriched Cordyceps militaris (CM${\alpha}$) may be an ideal candidate for hepatoprotective effects in animal models.

Improvement Effect of Capsaicin Against Orotic Acid-Induced Fatty Liver in Rats (Orotic Acid-유발 지방간에 미치는 Capsaicin의 개선효과)

  • The effect of capsaicin (0.02%) on the concentrations of liver and serum lipids, GOT and GPT activities, and hepatocyte morphology in male Sprague Dawley rats fed diets with or without erotic acid (1.0%) for 28 days was studied. Liver triglyceride concentration was increased by approximately 4.7-fold after erotic acid-feeding, wherease simultaneous feeding of capsaicin and erotic acid reduced its concentration by 38%. Orotic acidfeeding elevated liver cholesterol concentration, but an addition of capsaicin to the erotic acid diet resulted in the significant decrease in liver cholesterol. Capsaicin alone did not affect the liver triglyceride and cholesterol concentrations. Reduction of the serum concentrations of triglyceride and cholesterol by capsaicin was also observed in rats fed diets with erotic acid. The serum GOT and GPT activities were not significantly different among the experimental groups. Liver tissue morphology showed that the hepatocytes of erotic acid-feeding rats were a typical fatty liver with numerous fat droplets, whereas simultaneous feeding of capsaicin with erotic acid reduced the size and numbers of fat droplets. The present study demonstrated that the diet containing 0.02% capsaicin can prevent the erotic acid-induced fatty liver.

Content of Orotic Acid in Raw milk and Reduction of OA by Lactic Acid Bacteria during Fermentation (Orotic acid의 원유내 함량과 발효중 유산균에 의한 감소)

  • Changes in erotic acid (OA) contents of raw milk and during cultivation and storage at refrigerated temperature were determined. OA contents of raw and 10% reconstituted milk held at $121^{\circ}C$ for up to 15 min decreased by 17.6 and 22.4%, and those fermented at 40 and $37^{\circ}C$ for 72 hr with Lactobacillus helveticus 166 and Lactobacillus casei 955 decreased 37.8-43.2 and 41.8-76%, whereas when fermented at $40^{\circ}C$ for 72 hr with Streptococcus thermophilus ST-37, did not decrease significantly compared with fermentation by other lactobacilli. OA content did not change during storage at refrigeration temperature. OA reduction by Lactobacillus sp. was dramatically increased at the initiation of stationary phase during fermentation. OA reduction varied among different lactobacilli. These results show decrease in cultivation time, rapid cooling of yogurt, and proper selection of Lactobacillus sp. prevent OA content reduction.

Effort of Capsaicin on the Lipid Peroxidation in Tissues of Rats Fed Orotic Acid. (Orotic acid 투여 흰쥐의 조직 과산화지질 농도에 미치는 capsaicin의 영향)

  • The antioxidative activities of capsaicin (Cap, 0.02 and 0.04%) on the lipid peroxidation of tissues in male Sprague Dawley rats fed diets with or without orotic acid (1.0%, w/w) were studied in vivo system by measuring thiobarbituric acid reactive substances (TBARS) concentrations. Body weight gain, food intakes, food efficiency ratio and the relative tissues weights of brain, kidney, spleen, heart, and testis were not significantly different among dietary groups. Relative weights of liver were higher in the OA group than that in the other groups. TBARS concentrations in liver were significantly higher in the OA or 0.04% Cap groups than in the normal group, while this raise was not observed 0.02% Cap group. A significant increase in TBARS concentrations was found in the liver of the OA +0.04% Cap group compared with the OA or the 0.04% Cap groups. Nonheme iron concentrations were significantly higher in the liver of the OA, 0.04% Cap, OA+0.02% Cap, OA+0.04% Cap groups than that in the normal group. TBARS concentrations in kidney were lower in the 0.02% or 0.04% Cap groups than that in the normal group, but this concentrations were higher in either the OA, OA+0.02% Cap or OA+0.04% Cap groups than that in the normal group. Meanwhile, TBARS concentrations of brain, spleen, heart and testis were not significantly different among groups. The present study suggested that the lipid peroxidation was increased in the rats liver fed diet with erotic acid, and the simultaneous supplementation of capsaicin further enhanced.

Hepatoprotective Effect of Bacillus subtilis-fermented Silkworm (Bombyx mori L.) Extract on Non-alcoholic Fatty Liver in Rats (고초균 발효누에 추출물이 비알코올성 지방간 유발 흰쥐에 미치는 간 기능 개선 효과)

  • The aim of this study was to investigate the potential effects of extracts from silkworm Bombyx mori L. fermented with Bacillus subtilis KACC 91157 at levels of 5%(v/w) and 10%(v/w) in Sprague-Dawley rats intoxicated with 1%(w/w) orotic acid (OA) for 10 days. The rats were divided into a normal group (N), a control group (C: OA), and treatment groups (SP10: OA + 10% extracts from B. mori L.; BSP5: OA + 5% extracts from B. mori L. fermented with B. subtilis KACC 91157; BSP10: OA + 10% extracts from B. mori L. fermented with B. subtilis KACC 91157). Serum activities of aspartate aminotransferase (AST), alanine transferase (ALT), alkaline phosphatase (ALP), and lactate dehydrogenase (LDH) increased following OA feeding, but the rise was slightly reduced by administration of BSP10. The total lipid, free fatty acid, phospholipid, total cholesterol, and triglyceride contents in serum were significantly lower in the OA treatment groups than in the N group. However, the contents slightly increased following the administration of BSP10. Glutathione concentrations in liver and serum were reduced in the OA-induced fatty liver, but they increased following the administration of BSP10. Hepatocytes in the OA-induced fatty liver contained numerous large droplets. However, SP10, BSP5, and BSP10 feeding prevented OA-induced lipid droplet accumulation in hepatocytes. Accordingly, extracts from silkworm powder fermented with B. subtilis could be an ideal material as a dietary supplement in healthy functional foods to improve the effects of fatty liver.

Effects of Orotic Acid and Di-(2-Ethylhexyl)Phthalate on Microsomal Triglyceride Transfer Protein(MTP) Activity and mRNA Levels in Liver and Intestine of Rats (Orotic acid와 Di-(2-ethylhexyl)phthalate 투여 흰쥐의 간장 및 소장 Microsomal Triglyceride Transfer Protein(MTP) 활성과 mRNA 수준에 미치는 영향)

  • Microsomal triglyceride transfer protein(MTP) activity and mRNA level were investigated in the liver and small intestine of rats fed on di-(2-ethylhexyl)phthalate(DEHP) and orotic acid(OA) as serum triglyceride-reducing agents. The concentration of liver triglyceride was significantly increased in the OA group, but that was not increased in the DEHP group compared with the control group. The concentration of serum triglyceride was significantly decreased in the OA and DEHP groups compared with the control group, but this reduction was more pronounced in the OA group. MTP activity and mRNA level in liver were decreased in the OA group compared with the control group, while MTP activity in the small intestine was increased in the OA group compared with the control group. MTP activities and MTP mRNA levels in both liver and small intestine had no influence by the DEHP dietary feeding, despite the triglyceride-lowering action, compared with the control dietary feeding. The activity of liver microsomal phosphatidate phosphohydrolase(PAP), the rate-limiting enzyme in triglyceride synthesis, was increased in the OA group compared with the control group, but that of cytosolic PAP was decreased in the DEHP group compared with the control group. The result suggest that MTP activity and MTP mRNA level are involved in the triglyceride-lowering action of OA, but those are not involved in that of DEHP.
