Optimal Culturing and Enhancement of Lipid Accumulation in a Microalga Botryococcus braunii (미세조류 Botryococcus braunii의 배양조건 최적화 및 지질축적 향상)

  • Several tests and experimental work have been done for identifying the best growth conditions and accumulated amount of lipid moiety in B. braunii, a microalga(UTEX 572) in terms of media composition. The specific growth rate was found to be the highest at 0.15 g/L-day when the phosphorus concentration was doubled with the other ingredients at the normal level. Experiments for varied media compositions revealed that the accumulation of lipid was the highest at 48% (dry cell weight based) in the nitrogen deficient medium and its corresponding specific growth rate was comparative to that in the normal BG 11 medium. In the bubble column experiments, carbon dioxide containing air produced four times more cell mass than air only. Light and glucose addition also enhanced cell mass with maximum, 1.8 g/L and accordingly 42% of lipid composition, which turned out to be a better strategy for higher lipid-producing microalgal culture.

  • Microalgae have been explored as potential host species for biofuel production. Environmental factors affect algal growth and cellular composition. The effects of several key environmental factors, such as temperature, light, and pH of the medium on the growth and biochemical composition of Scenedesmus obliquus were investigated in this study. The highest growth rate of microalgae was observed at an optimal temperature of 25℃, 150 μmol/(m2·s) light intensity, and pH 10.0. The biochemical composition analysis revealed that the carbohydrate content decreased at lower (20℃) or higher temperature (35℃), whereas the protein and lipid contents increase at these temperatures. The fluctuation of light intensity significantly affected the contents of protein, carbohydrate, and lipid. The protein levels varied greatly when the pH of the medium was below 7.0. The carbohydrate and lipid contents significantly increased at pH above 7.0.

An Optimal Design of Gas Lift in Offshore Oil Reservoirs Considering Oil and Injected Gas Composition (해저 오일 저류층 내 오일 및 주입가스 조성에 따른 가스리프트 공법의 최적 설계)

  • This study presents optimal design of gas lift considering composition of reservoir oil and injected gas which can affect gas lift performance in offshore oil reservoir. Reservoir simulation was conducted by using reservoir models which were built in accordance with API gravity of oil. The results of simulation reveal that oil production rate is considerably increased by gas lift when the reservoir productivity decrease. As a results of response curve analysis for gas lift using well models, gas injection rate to improve the production rate increases as the API gravity of oil decreases and the specific gravity of injected gas increases. The optimal design of gas lift was carried out using multiple lift valves. Consequently, gas lift can be operated at relatively low injection pressure because of decrease in injection depth in comparison to the single lift valve design. The improved oil production rates were analyzed by coupling between reservoir model and well model. As a results of the coupling, it is expected that natural gas injection in the heavy oil reservoir is the most efficient method for improving oil production by gas lift.

  • The fluctuation of inlet flow to a water treatment plant makes a serious problem that it can change the filtration rate abruptly, and ultimately occur the breakthrough of the detained particles inside filter media. Also, since it takes very short time (about 10 minutes) for the surface wave occurred from the fluctuation of inlet flow to reach the filtration process, it is impossible to control the filtration rate stably. Therefore, this study was conducted to evaluate the effect of daily flow-rate fluctuation on the performance of sand filtration process, and to suggest the dual media composition for coping with that effect. Comparative column tests have been carried out for various dual media (sand and anthracite) compositions. From the results of column tests, dual media, especially in the case of sand 45cm/anthracite 30cm, is more effective to cope with the effect of flow-rate fluctuation on the performance of filtration than single media (only sand). In addition, irrespective of dual media composition, managing ability to cope with that fluctuation tends to be weak at the end of allowable filtration duration time,

  • Optimal lyophilization process was developed for manufacturing the dried product of Lactobacillus acidophilus with high cell viability. Three major factors, freezing rate, specific surface area of samples, and stabilizer type and their synergy were shown to play a crucial role in the development of an effective lyophilization process. Finally we found an optimal combination among three process parameters mentioned above; an exceptionally high cell survival percentage of 90% was achieved using the 8.28 cm-1 specific surface area of samples, slow freezing rate, and a stabilizer composition of 4% skim milk +1% glycerol +0.1% calcium chloride.

  • Effects of rf power, pressure, sputtering gas composition, and substrate temperature on the deposition rate of the $WC_x$ coatings were investigated. The effects of rf power and sputtering gas composition on the hardness and corrosion resistance of the $WC_x$ coatings deposited by reactive sputtering were also investigated. X-ray diffraction (XRD) and Auger electron spectroscopy (AES) analyses were performed to determine the structures and compositions of the films, respectively. The hardnesses of the films were investigated using a nanoindenter, scanning electron microscopy, ana a salt-spray test, respectively. The deposition rate of the films was proportional to rf power and inversely proportional to the $CH_4$ content of $Ar/CH_4$ sputtering gas. The deposition rate linearly increased with increasing chamber pressure. The hardness of the $WC_x$ coatings Increased as rf power increased. The highest hardness was obtained at a $Ar/CH_4$ concentration of $10 vol.\%$ in the sputtering gas. The hardness of the $WC_x$ film deposited under optimal conditions was found to be much higher than that of the electroplated chromium film, although the corrosion resistance of the former was slightly lower than that of the latter.

  • Bioheavy oil, which is expanding its range of use as an alternative fuel to reduce environmental pollutant emissions, has a lot of difficulty in combustion due to its low emission of pollutants such as nitrogen oxide (NOx) and sulfur oxide (SOx), while its low dissipation and high oxygen content in fuel. many studies have been conducted on change in characteristics by mixing rate combustion characteristics and combustion reactions, but there have been no specific and effective studies on the composition of control system, optimization of control, development of logic for mixing and burning, minimizing environmental pollutantants discharge. In this study, we intend to consider systemmatic and empirical considerations on the composition, logic development, solve the problem of manual switching of bioler master due to excessive oxygen content and tuning of the control system for optimal combustion of bioheavy oil.

Selection of the Optimal Transplanting Method and Time for Restoration of Zostera marina Habitats (잘피(Zostera marina)서식지 복원을 위한 최적 이식방법 및 시기 선정에 관한 연구)

  • Seagrass bed is an important component in coastal and estuarine ecosystems, providing food and shelter to a wide variety of fauna. Recently, seagrass coverage has declined significantly due to anthropogenic influences such as reclamation, dredging, and eutrophication and consequently, necessity of seagrass habitat restoration is rising. Transplantation experiments with Zostera marina using TERFS, staple method, and shell method have been conducted at Dadae Bay, Kosung Bay and Jindong Bay on the south coast of Korea to select an optimal transplanting method for restoration of Z. marina habitat. Three experimental sites located at the vicinity of natural Z. marina beds with an average water depth of about 4m. Z. marina plants, which were collected from donor bed in Koje Bay were also transplanted at 7 different time from October 2003 to July 2004 to find appropriate transplanting time. Density of Z. marina was monitored monthly at both transplanted areas and natural beds. Transplantation using the staple method showed the highest survival rate of transplant. Shell method was also an effective transplanting method at muddy areas in Kosung Bay and Jindong Bay, but not suitable at sandy areas in Dadae Bay. These results suggest that sediment composition of transplanting areas should be considered for the selection of the optimal transplanting method. Z. marina transplanted during fall usually showed the highest survival rate, while most Z. marina plants transplanted in summer died due to high lethal temperature during this period.

Establishment of Optimal {100} Si Etching Condition for $N_2H_4-H_2O$ Solutions and Application to Electrochemica Etching ($N_2H_4-H_2O$용액의 {100} Si에 대한 최적식각조건의 설정과 전기화학적 식각에의 응용)

  • Using the anisotropic etching characteristics of N2H4-H2O solutions, Si diaphragm was fabricated for the integrated sensors. The optimal composition and temperature of the etching solution in (100) Si etching process was established to be 50mol% N2H4 in H2O at 105\ulcorner\ulcorner for both higher etch rate (=2.6\ulcorner/min) and better surface quality of etched (100) planes. Based on the above optimal etching condition, the electrochemical etch-stop technique was employed to form n-type Si diaphragm having a thickness of 20\ulcorner and the thickness of diapragm could exactly be controlled to 20\ulcorner\ulcorner.
