Tacrine is an acetylcholinesterase inhibitor that is approved for the treatment of Alzheimer's disease. However, tacrine treatment for Alzheimer's disease results in reversible hepatotoxicity in 30-50% of patients, which seriously limits its clinical use. Accordingly, the identification of constituents in natural products that have protective effects on tacrine-induced hepatotoxicity would be valuable. In the present study, an immortalized human hepatoma cell line, HepG2 was employed to screen for agents that protect against tacrine-induced hepatotoxicity. The bioassay-guided fractionation of water extract of Cassiae Semen furnished two anthraquinones, aurantio-obtusin (1) and obtusifolin (2). Compounds 1 and 2 showed hepatoprotective effects with the protection ratio values of 55.3 +/- 0.5% and 41.2 +/- 0.8% at $160{\mu}M$, respectively.
Our continuing interest on Garcinia and Mesua species has led us to carry out a detail study on the chemistry of the root bark of Garcinia mangostana (Guttiferae) since this part of the plant has not been investigated before, and the strm bark of Mesua corneri (Guttiferae) an uninvestigated species. This study has yielded six xanthones, ${\alpha}-mangostin$ (1), ${\beta}-mangostin$ (2), ${\gamma}-mangostin$ (3), garcinone-D (4), mangostanol (5) and gartanin (6) from Garcinia mangostana and two xanthones rubraxanthone (7) and inophyllin B (8) from Mesua corneri. Structural elucidations were achieved using $^1H,\;^{13}C$ NMR and MS data. The crude hexane and chloroform extracts of the root bark of Garcinia mangostana and the hexane extract of the stem bark of Mesua corneri were found to be active against CEM-SS cell lines with $IC_{50}$ values less than $30\;{mu}g/ml$. Moreover, ${\gamma}-mangostin$ gave a very low $LC_{50}$ value of $4.7\;{mu}g/ml$ while rubraxanthone gave an $LC_{50}$ value of $5.0\;{mu}g/ml$ indicating these two compounds to be potential lead compounds for anti-cancer activity against the CEM-SS cell line. This paper reports the isolation and identification of these compounds as well as bioassay data for the crude extracts, ${\gamma}-mangostin$ and rubraxanthone.

The fruits of Piper cubeba L. are used traditionally to treat respiratory disorders in Indonesia. In order to determine the compounds responsible for this activity, the fruits were extracted with nhexane followed by ethanol to give n-hexane and ethanol extracts. Based on tracheospasmolytic assay on these two extracts, the n-hexane extract was more active to inhibit trachea contraction than that of ethanol extract. Upon bioassay guided isolation of the n-hexane extract, a tracheospasmolytic active compound was isolated and identified as dihydrocubebin [(3,4),(3',4')-bis-methylenedioxy-9,9'dihydroxylignan] $(\underline{1})$. Compound $\underline{1}$ was tested further for its ability to inhibit histamine released from mast cells, using rat basophilic leukemia (RBL-2H3) cell line and rat peritoneal mast cells RPMCs) as models; and $DNP_{24}$-BSA, thapsigargin, ionomycin, compound 48/80 and PMA were used as inducers for histamine released from mast cell. The test result showed that $\underline{1}$ inhibited histamine release from RBL-2H3 cells induced by $DNP_{24}$-BSA, thapsigargin and ionomycin. In addition, $\underline{1}$ suppressed histamine release from RPMC induced by either thapsigargin or ionomycin. However, $\underline{1}$ did not inhibit histamine release from RPMC induced by either compound 48/80 or combination PMA-sub optimum dose of ionomycin. Therefore, it was concluded that the inhibitory effects of $\underline{1}$ on the histamine released from mast cells may involve mechanisms related to intracellular $Ca^{2+}$ signaling events or downstream processes of intracellular $Ca^{2+}$ signaling in mast cells.
The in vitro cytotoxicity of SKI 2053R was evaluated against human tumor cell lines along with those of cisplatin and carboplatin using MTT assay. The cell lines tested were two human lung cancer cell lines and five human stomach cancer celt lines. The level of cytotoxic effects of SKI 2053R against two human lung cancer cell lines was located between cisplatin and carboplatin. However, the cytotoxic activity of SKI 2053R against five human stomach cancer cell lines was similar to that of cisplatin. SKI 2053R is considered to be selectively cytotoxic toward human stomach cancer cell lines. We carried out pharmacokinetic and ex vivo phrmacodynamic studies of SKI 2053R in beagle dogs to predict the clinical antitumor effect of SKI2053R, comparing with those of cisplatin and carboplatin. In ex vivo pharmacodynamics which used MTT assay as bioassay on the 2 lung and 5 stomach cancer cell, mean antitumor indexes (ATIs) of SKI 2053R were highest among three compounds in both lung and stomach cancer cell lines, especially in stomach cancer cell. Much higher ATI profile and maximal inhibition rates of SKI 2053R appeared in the stomach cancer cells will give desirable advantages to clinical trial s against gastric carcinoma. The anti tumor activity and target organ toxicity of SKI 2053R were compared with those of cisplatin on stomach cancer cell line, KATO III xenografted into nude BALB/c(nu/nu) mice. All groups of cisplatin and SKI 2053R showed active tumor regression. The inhibition rates(IR) of SKI 2053R were higher than that of cisplatin on the basis of mean IR. Though the loss of body weight was observed in all groups from the first week, the SKI 2053R group recovered it soon from the third week after the initiation of treatment, maintaining the most active anti tumor activity among three groups.
60종(種)의 약용식물과 식용작물을 대상으로 항종양활성등의 생리활성을 조사함으로서 약용식물과 식용작물의 유용적인 측면의 확인뿐만 아니라 나아가서 새로운 생리활성물질 탐색의 가능성을 검토하기 위해서 실험하였으며 결과는 다음과 같다. 약용식물 및 식용작물에 대한 80% EtOH 추출물을 이용하여 항종양효과를 보면 PKC법에서는 명아주(73.4%) 및 antibleb형성억제력검정에서는 검정콩이, PLC법에서는 검정콩(91.9%), MTT법에서 50%의 억제력을 나타내는 농도$(IC_{50})$가 검정콩과 쑥의 경우 각각 $4.7{\mu}g/ml$을 보였다.
Activated microglia by neuronal injury or inflammatory stimulation overproduce nitric oxide (NO) by inducible nitric oxide synthase (iNOS) and reactive oxygen species (ROS) such as superoxide anion, resulting in neurodegenerative diseases. The toxic peroxynitrite (ONOO$^-$), the reaction product of NO and superoxide anion further contributes to oxidative neurotoxicity. We tried to evaluate the effects of two kinds of varieties of Perilla frutescens var japnica Hara on the NO production in lipopolysaccharide (LPS)-activated microglia. The perilla cultivars of Namcheondeulkkae (NC) and Boradeulkkae (BR) were developed by pure line from the local variety and by a cross between 'deulkkae' and 'chajogi', respectively. Spirit, hexane, chloroform and butanol fractions of the leaves of NC and BR inhibited the production of NO in LPS-activated microglia. The fractions of BR showed stronger activity than NC and the spirit extracts was the most potent in both cultivars. The solvent fractions of BR suppressed the expression of protein and mRNA of iNOS in LPS-activated microglial cells. Moreover, the extracts of NC and BR showed the activity of peroxynitrite scavenging in cell free bioassay system. These results imply that Namcheondeulkkae and Boradeulkkae might have neuroprotective activity through the inhibition of NO production by activated microglial cells and peroxynitrite scavenging activity.

Leaf extracts of Cassia alata L (akapulko), traditionally used for treatment of a variety of diseases, were evaluated for their potential antitumor properties in vitro. MTT assays were used to examine the cytotoxic effects of crude extracts on five human cancer cell lines, namely MCF-7, derived from a breast carcinoma, SK-BR-3, another breast carcinoma, T24 a bladder carcinoma, Col 2, a colorectal carcinoma, and A549, a nonsmall cell lung adenocarcinoma. Hexane extracts showed remarkable cytotoxicity against MCF-7, T24, and Col 2 in a dose-dependent manner. This observation was confirmed by morphological investigation using light microscopy. Further bioassay-directed fractionation of the cytotoxic extract led to the isolation of a TLC-pure isolate labeled as f6l. Isolate f6l was further evaluated using MTT assay and morphological and biochemical investigations, which likewise showed selectivity to MCF-7, T24, and Col 2 cells with $IC_{50}$ values of 16, 17, and 17 ${\mu}g/ml$, respectively. Isolate f6l, however, showed no cytotoxicity towards the non-cancer Chinese hamster ovarian cell line (CHO-AA8). Cytochemical investigation using DAPI staining and biochemical investigation using terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL)-a method used to detect DNA fragmentation-together with caspase assay, demonstrated apoptotic cell death. Spectral characterization of isolate f6l revealed that it contained polyunsaturated fatty acid esters. Considering the cytotoxicity profile and its mode of action, f6l might represent a new promising compound with potential for development as an anticancer drug with low or no toxicity to non-cancer cells used in this study.
To ascertain the effect of over-expressed maize calreticulin in tomato plant on tobamovirus movement in addition to validating potentiality of the gene (ZmCRT) as a means for the virus-resistance resource, four ZmCRT-expressing homozygous lines were generated from the T0 plants as using an Agrobacterium-mediated transformation, nucleic acid analyses, and a conventional breeding method. Of them, a line was subjected to the bioassay for tolerances to tobacco mosaic virus-U1 (TMV-U1) and tomato mosaic virus (ToMV) followed by RT-PCR and a chlorophyll fluorescence quenching analyses. Both transgenic plants transcribing ZmCRT and wild-type plants showed no symptom by 20 days after viruses inoculation, however the photosystem II quantum yield parameter measured from the upper leaves of ToMV-inoculated plants revealed that ZmCRT transgenic plants have higher photosynthetic ability than wild-type ones at that time, which indirectly implies that over-expressed ZmCRT product acts as a barrier to the cell-to-cell and/or systemic movement of ToMV. Moreover, ZmCRT transgenic plants showed remarkably longer shoot length than wild-type ones in 40 days after TMV-U1 or ToMV inoculation each, which might be resulted from higher photosynthetic ability during the phase not yet showing any external symptoms. Collectively, over-expressed ZmCRT protein in tomato plants is able to interrupt the systemic movement of infected TMV-U1 and ToMV even though not perfect.
연구배경: 인터페론감마-(interferon-$\gamma$)는 항바이러스 효과, 암세포의 형증식 효과, 대식세포 및 B 림프구의 활성화, 주면역복합체(MHC) 항원 발현의 증가 등의 생물학적 효과를 나타낸다. 특히, 인터페론감마의 항암 효과는 이미 생체 내외에서 입증되어 실제 폐암 환자에 대한 임상 연구가 시도되고 있다. 그러나, 인터페론감마의 항암효과 기전은 여러가지 가설이 제시되기는 하고 있지만 아직 확립된 것이 없다. 세포의 괴사(necrosis)는 심한 외부의 스트레스에 의해서 발생하는 세포 사망의 형태로 잘 알려져 있다. 생명현상 중 괴사와는 전혀 다른 세포 사망의 과정으로 아포프토시스(apoptosis)가 있다. 아포프토시스는 조직의 항상성(homeostasis of tissue volume), 개체의 발생과정, 장기의 퇴행(regression), tolerance 등의 여러 생명 활동 과정에서 발생하는 세포 사망의 과정으로서, 세포질 및 핵이 분절화(fragmentation)되어 죽어가는 능동적 사망과정으로 알려져 있다. 아포프토시스에서는 수동적으로 죽어가는 괴사에서 볼 수 없는 DNA 분절화(DNA ladder pattern)가 특징적으로 관찰된다. 인터페론감마의 암세포에 대한 세포독성 기전을 연구하기 위해서 인터페론감마를 폐암세포주인 A549세 처치한 후 현미경(inverted microscope)로 A549의 변화를 관찰하였는데 A549세포가 분절화되면서 죽어가는 것을 관찰할 수 있었다. 저자들은 인터페론감마의 항암기전으로서 아포프토시스의 가능성을 평가하고자 본 연구를 시행하였다. 방법: 폐암세포주인 A549세포를 대상으로 하였다. A549세포에 여러 농도의 인터페론감마를 투여하고 24시간, 72시간, 120시간 후에 MTT(dimethylthiazolyl diphenyltetrazolium bromide) bioassay법으로 세포독성을 정량화하였다. 그리고, 100 unit/ml의 인터페론감마를 A549 세포에 120시간 처치 후, 광학 현미경으로 세포 사망의 양상을 관찰하였다. 또한, 100 unit/ml의 인터페론감마를 투여하고 120시간이 경과한 후 사망 세포의 DNA를 추출하여 1.5% agarose gel에서 전기 영동을 시행하고 ethidium bromide로 염색 후 DNA ladder pattern 유무를 관찰하였다. 결과: 1) 인터페론감마에 의한 A549 폐암세포주의 세포독성 효과는 24시간에는 거의 없다가 72시간부터 120시간 사이에 나타나기 시작하여 120시간에는 더 증가하였다. 2) 인터페론감마에 의한 A549 세포의 사망 양상은 광학현미경상 A549 세포들이 작은 분절로 나뉘면서 사망하였다. 3) 인터페론감마를 A549 폐암세포주에 처치 후 죽어기는 세포의 DNA를 추출하여 전기영동시킨 결과 아포프토시스(apoptosis)에서 특징적으로 보이는 DNA ladder pattern을 관찰할 수 있었다. 결론: 인터페론감마(interferon-$\gamma$)의 A549 폐암세포주에 대한 세포독성의 기전은 아포프토시스 과정을 통해서 일어난다.
4종류의 다공성 합성흡착제(Sepabeads 207, 700, 850, Diaion HP 20)를 사용하여 Rubus coreanus 과실의 물 추출물로부터 과당 및 비polyphenol성분을 효과적으로 제거하여 DPPH라디칼 소거능 및 화학발광법을 이용한 superoxide 라디칼 소거능이 증가된 농축물을 효율적으로 얻을 수 있었다. 또한, 실시간 HPLC $ABTS^{{\cdot}+}$ 활성분석 시스템을 활용하여 6개의 항산화 능력을 갖는 활성피크를 확인할 수 있었으며 이들 대상으로 LC-MS/MS분석을 실시하여 5개의 anthocyanin인 cyanidin-3-sambubioside, cyanidin-3-glucoside, cyanidin-3-xylosylrutinoside, cyanidin-3-rutinoside, pelargonidin-3-rutinoside를 mass pattern 및 분자량 확인 과정을 통해 잠정적 화학구조 동정을 하였다. 수율과 항산화 효능을 고려해 볼 때, 4종의 합성흡착제 중에서 Diaion HP 20이 가장 우수한 결과를 나타내었다. 이처럼 적절한 합성흡착제의 선택으로 표적물질에 대한 polyphenol성 물질의 농축뿐만 아니라 생리학적 활성증대효과를 가져올 수 있어 적절한 합성흡착제의 선택은 식품 및 의약품의 정제 및 분리에 유용하게 활용될 수 있음이 확인되었다.
