Effects of dietary lipid sources on apoptotic and immune gene expression in head kidney of olive flounder (Paralichthys olivaceus)

  • It can be hypothesized that dietary fatty acids can modulate immune responses in fish by inducing apoptosis of immune cells since dietary polyunsaturated fatty acid (PUFA) increase apoptosis by oxygen radicals generated by peroxidation. Thus we examined the effects of deferent dietary lipid sources such as squid liver oil (FO), linseed oil (LO) and soybean oil (SO) on oxidation (Cytochrome C oxidase; COS), apoptosis (TNF-${\alpha}$ Scinderin like) and immune (IL-$1{\beta}$ and NKEF) gene expression in the main immune organ (head kidney) in olive flounder (Paralichthys olivaceus) by Q-PCR analysis after feeding diets containing each oil (5%) for 15 weeks. Linseed oil and soybean oil were chosen to compare n-3 or n-6 enriched vegetable oils, respectively. Consequently, COS, TNF-${\alpha}$ and Scinderin like gene expression was increased in SO group, indicating the induction of oxidation and apoptosis. Meanwhile, no significant difference was found in immune gene expression. In conclusion vegetable oils containing n-3 PUFA like linseed oil seems to be more suitable lipid source than soybean oil for replacement of fish oil in flounder since n-6 PUFA in SO leads to activation of apoptosis pathways within the cellular damage in head kidney.

올리브유 대체 비율이 저지방 돈육 패티의 품질 특성에 미치는 영향 (Effect of Olive Oil Substitution Ratio on Quality Characteristics of Low-fat Pork Patty)

  • 본 연구는 돼지 지방 대체제로 올리브유 겔 첨가 비율을 달리하여 제조한 저지방 돈육 패티의 이화학적 품질 특성을 검토하였다. 돈육 패티는 돼지 지방 20%(대조군, T0), 돼지 지방 15%와 올리브유 겔 5%(T1), 돼지 지방 10%와 올리브유 겔 10%(T2), 돼지 지방 5%와 올리브유 겔 15%(T3), 그리고 올리브유 겔 20%(T4)를 첨가하여 제조하였다. 올리브유 겔 첨가 비율에 따라 수분 함량은 증가하고, 지방 함량은 감소하였다. 가열 전후 돈육 패티의 명도(L*)와 적색도(a*)는 T0가 가장 높았으며, 가열 후 돈육 패티의 황색도는 T4가 가장 높았다. 올리브유 겔 대체 비율이 높을수록 보수력, 수분 보유율 및 지방 보유율은 증가하였고, 조리 감량, 직경 감소율 및 수축율은 유의하게 감소하였다. 경도, 탄력성 및 씹힘성은 T4가 가장 높았고, 응집성 및 검성은 T0가 가장 높았다. 콜레스테롤 함량은 T4가 가장 낮았으며, 대조군(T0)에 대한 감소율은 T1, T2, T3 및 T4가 각각 13.8%, 12.6%, 34.5% 및 49.0%였다. 포화지방산은 palmitic acid가 가장 높았고, 불포화지방산은 oleic acid가 가장 높았다. 돈육 패티의 불포화지방산은 올리브유 겔의 첨가 비율에 따라 증가하였다. 그러므로 돼지 지방을 올리브유 겔로 대체하면 이화학적 특성들이 향상되고, 콜레스테롤 함량이 감소하며, 지방산 조성이 개선된 육제품의 제조가 가능하다.

  • 폐식용유에서 지방분해효소를 생산하는 세균을 분리하였고, PIBWIN 세균동정 방법으로 생리 생화학적 특성을 조사하여 확인한 결과 Pseudomonas sp. OME로 동정하였다. 여러 기질로 지방분해효소 생산을 조사한 결과 올리브유에서 6.1 U/ml의 생산력을 나타내었다. 물리적 인자인 배양시간, 온도. pH 및 올리브유와 효모 추출액의 영양인자에 의한 지방분해효소 생산 조건을 조사 하였다. 효소의 분비는 배양시간. 올리브유 와 효모 추출액의 농도에 강한 영향을 받았으며, RSM을 이용한 최적화는 이들 인자를 가지고 조사하였다. RSM을 이용한 지방분해효소 생산은 배양시간. 올리브유와 효모 추출액의 농도가 48 hr, 0.3 g, 및 0.9 ml에서 최적 생산조건을 나타냈다.

Effect of conjugated linoleic acid in diacylglycerol-rich oil on the lipid metabolism of C57BL/6J mice fed a high-fat high-cholesterol diet

  • The effect of conjugated linoleic acid (CLA) isomers esterified in diacylglycerol (DAG)-rich oil on lipid metabolism was investigated. Since dietary DAG has been known to induce the regression of atherosclerosis, CLA-DAG and olive-DAG oils containing similar levels of DAG (51.4~54.2%) were synthesized from olive oil. Hyperlipidemic C57BL/6J mice were then fed high-fat high-cholesterol diets supplemented with these oils (5% each) for 7 wk. The CLA-DAG diet containing 2.1% CLA isomers (0.78% c9,t11-CLA; 1.18% t10,c12-CLA) remarkably increased the levels of total plasma cholesterol and glutamic oxaloacetic transaminase (GOT) along with hepatic cholesterol and triacylglycerol (TAG) contents. Furthermore, the CLA-DAG diet inhibited fat uptake into adipose tissue whereas fat deposition (especially in the liver) was increased, resulting in the development of fatty livers. Hepatic fatty acid composition in the CLA-DAG mice was different from that of the olive-DAG mice, showing higher ratios of C16:1/C16:0 and C18:1/C18:0 in the liver. The activity of hepatic acyl-CoA:cholesterol acyltransferase (ACAT) was higher in CLA-DAG mice while plasma lecithin:cholesterol acyltransferase (LCAT) activity and the ferric reducing ability of plasma (FRAP) were lower in CLA-DAG mice compared to the olive-DAG animals. Results of the present study suggest that CLA incorporation into DAG oil could induce atherosclerosis in mice.

넙치(Paralichthys olivaceus)용 고효율 곤충배합사료 양식현장적용 평가 (Effects of Worm-based Extruded Pellets on Growth Performance of Olive Flounder Paralichthys olivaceus in Commercial Aquafarms)

  • We evaluated a worm-based extruded pellet diet with black soldier fly Hermetia illucens larvae (BSF) meal and BSF oil for olive flounder Paralichthys olivaceus through field feeding experiments at a commercial aquafarm. We prepared two experimental diets by replacing fish meal and fish oil with BSF meal and BSF oil (BEP-1, BSF meal 7%, BSF oil 1%) and (BEP-2, BSF oil 2%), respectively. We prepared raw-fish based moist pellets (MP) for comparison between the two experimental diets. We distributed the olive flounder (220±6.29 g) in square (10 m×10 m×1 m) concrete, 100 ton tanks at a density of 1,600 fish per tank (n=3) in triplicate for each dietary treatment. We fed the diets to the fish to apparent satiation for 7 months. At the end of the feeding trial, we found no substantial differences between the groups in terms of growth performance, survival, or feed utilization. None of the diet groups showed any changes in either hematological or non-specific immune responses. The histological observation of the intestine showed that the goblet cell number and cholecystokinin-producing cell activity increased in the fish fed the BEP diet compared with the those of the fish fed the MP diet. These results indicated that dietary BSF meal and oil can be used for olive flounder without compromising growth or, hematological and histological parameters.

  • Our objectives in this experiment were to study the effects of oils that are high in contents of unsaturated fatty acids (olive, flaxseed, and perilla oils) and pyroligneous liquor on plasma lipid concentrations in mice. Male ICR-mouse (n=10 per group) were fed powdered form diets containing oil and pyroligneous liquor for 60 days. The cholesterol level of the PP group ($96.71{\pm}25.75$ $mg/d{\ell}$) was lower than that of the COW group ($133.56{\pm}21.53$ $mg/d{\ell}$). Levels of triglyceride of COP, CFP, and CPP groups were $121.10{\pm}50.79$ $mg/d{\ell}$, $77.80{\pm}38.58$ $mg/d{\ell}$, and $92.40{\pm}33.04$ $mg/d{\ell}$, respectively (p<0.05). The dietary addition of olive oil increased the plasma lsevels of cholesterol and LDL-cholesterol. On the other hand, the dietary addition of flaxseed oil and pyroligneous liquor increased the plasma level of HDL-cholesterol and decreased the plasma levels of LDL-cholesterol and triglyceride compared with those of the control group. Further, the dietary addition of perilla oil decreased plasma triglyceride but increased HDL-cholesterol. The decrease in the cholesterol level was much higher in the CFP group. In conclusion, the dietary addition of long-term pyroligneous liquor effectively decreased the plasma levels of cholesterol and triglyceride and increased the plasma level of HDL-cholesterol. The additional dietary administration of flaxseed and perilla oils increased the effect of pyroligneous liquor.

오존화 올리브유의 항균성 및 변이원성 평가 (Evaluation of Antimicrobial Activity and Mutagenicity of Ozonized Olive Oil)

  • 오존화 올리브유의 여드름 치료용 소재로서의 사용 가능성을 알아보기 위하여 여드름 형성에 관여하는 미생물인 Propionibacterium acnes, Staphylococcus epidermidis, S. aureus와 Pseudomonas aeruginosa에 대한 생육 억제효과를 검토하였고, Ames test로 그의 변이원성 유무를 평가하였다. 항균활성을 paper disk 법으로 평가했을 때 P. acnes, S. epidermidis, S. aureus, P. aeruginosa에 대하여 항균활성을 나타내기 시작한 농도는 각각 4, 1, 0.5, 0,5 mg/mL이었고, 20 mg/mL 농도에서의 생육 저지환의 크기는 각각 7.7, 5.3, 5.6, 5.5 mm이었다. 50% 생육저해활성을 표시하는 $MIC_{50}$은 P. acnes, S. aureus, P. aeruginosa에 대해 2 mg/mL, S. epidermidis에 대해 4 mg/mL로 나타났으며 미생물의 생육이 관찰되지 않는 MIC 농도는 P. acnes, S. epidermidis, P. aeruginosa에 대해 8 mg/mL, S. aureus에 대해 10 mg/mL로 나타났다. 최소저해농도(MICs) 범위는 2${\sim}$10 mg/mL이었다. Salmonella typhimurium TA98, TA100, TA1535 균주에 대하여 오존화 올리브유와 오존화 메칠 올레산은 S9 mix의 첨가 유무에 상관없이 세포독성을 나타내지 않는 농도에서 $his^{+}$ 복귀돌연변이주의 수가 증가하지 않아 변이원성을 나타내지 않았다. 오존화 올리브유는 돌연변이원성을 나타내지 않으면서 여드름 형성 원인균에 대한 생육 억제효과를 나타내어 여드름 치료용 소재로서 사용할 수 있을 것으로 생각된다.

강황분말 첨가 계육 소시지의 제조조건 최적화 (Optimized Processing of Chicken Sausage Prepared with Turmeric (Curcuma longa L.))

  • The purpose of this study was to determine the optimal mixing conditions for two different amounts of turmeric (Curcuma longa L.) powder and olive oil for the processing of chicken sausage. The experiment was designed according to the central composite design of response surface methodology, with ten experimental points, including two replicates for turmeric powder and olive oil. The physicochemical and mechanical analysis of each sample, including water holding capacity, moisture content, lightness, redness, yellowness, hardness, chewiness, gumminess, and cohesiveness, showed significant differences. The results from sensory evaluations also showed very significant differences in color, flavor, tenderness, chewiness, and overall quality. The optimal formulation, calculated by numerical and graphical methods, was 1.89 g of turmeric powder and 9.77 g of olive oil. Under these conditions, the model predicted pH-6.01, salinity-0.20, WHC-94.88, $L^*$ value-61.13, $b^*$ value-37.45, hardness-$36.66{\times}10^2$ (N), springiness-8.70 (mm), chewiness-$26.88{\times}10^3$ ($N{\times}mm$).

마늘과 향신료 첨가가 난황과 감람유 유화의 관능적 특성 (Effective of Extract Garlic, Spices of Emulsified with Egg-Yolk & Olive Oil)

  • 본 연구는 Liquid egg yolk sabayon을 만들 때 버터 대신에 올리브오일과 마늘 첨가량을 달리하여 관능적 평가를 실시하여 조리표준화의 예시를 제시하고자 한다. 1, 맛에서는 마늘 10g 첨가에서 5.61로 대조군의 5.13보다 좋았다. 2. 색에서는 마늘 20g 첨가가 가장 높은 점수를 얻었고, 마늘 30g가 나쁘게 나타났다. 3. 향에서는 마늘 10g 첨가가 대조군 보다 좋았고 마늘 첨가군 모두가 대조군 보다 좋았다. 4. 종합적인 면에서도 마늘 10g첨가가 우수한 것으로 나타났다. 따라서 Liquid egg yolk sabayon을 만들 때 적당량의 마늘을 첨가하는 것이 좋을 것으로 사료되어 진다.

Facile Synthesis of Bio-Composite Films Obtained from Sugarcane Bagasse and Cardboard Waste

  • In this study, we focus on the recycling of cardboard waste and sugarcane bagasse (SCB) for the preparation of carboxymethyl cellulose (CMC) and its conversion into a biodegradable film. Sodium alginate (SA) was added to form a biodegradable composite film. SA was used to increase film permeability. Glycerol, which is a plasticizer, was used to increase the tensile strength (TS) and film expansion. To characterize the CMC, X-ray diffraction, Fourier transform infrared spectroscopy, and scanning electron microscopy were used. The addition of olive oil to the CMC-SA matrix highlighted its antimicrobial property against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). A slight decrease in tensile strength was observed with the addition of olive oil (OO), which improved the functional properties of the control films as well as lowered moisture content and water solubility. But considering all other factors, the composite films obtained from sugarcane bagasse and cardboard waste incorporated with olive oil are suitable for applications in the field of food packaging.
