The reactive oxygen species (or free radicals) generated by ultraviolet radiation cause damage on cellular components and pigment of skin. The aim of this study was to investigate the skin-whitenig effect of Okyong-san. Inhibitory effects of okyong-san extracts on melanin synthesis were studied. Namely, UV-absorbing ability, free radical scavenging activity and tyrosinase inhibitory activity of okyong-san extracts were investigated. As a result, the extracts of okyong-san were found to inhibit the activity of tyrosinase and they showed an absorbance in the UV-B region and UV-C region. We also observed that extracts of okyong-san had free radical scavenging activity.

The aim of this study was to investigate the skin-whitening effect of okyong-san and to develop new drug delivery carrier The extracts of okyong-san were found to have the whitening effect and Eudragit$\^$ⓡ/ L 100-55 (EUD) coated solid lipid nanoparticle (E-SLN) was prepared by solvent evaporation method and melt dispersion technique. As a result, E-SLN have a 144-170 nm of particle size, spherical shape, and 33-41% encapsulation efficiency, After release test in vitro, release profile of E-SLN depended on pH and temperature. Lastly, closed patch test and skin-whitening test was peformed clinically. In conclusion, test sample had non-stimulation and high % whiteness. The results suggest that okyong-san and E-SLN is useful as cosmeceuticals for whitening cosmetics.

Solid lipid nanoparticle (SLN) system has been attracted increasing attention during last few years as a potential drug delivery carrier However, the SLN have disadvantage of low encapsulation efficiency for hydrophilic drug. In this study, for increase it's encapsulation efficiency, we prepared the $Eudragit^{\circledR}$ L100-55 (eudragit) coated SLN(E-SLN) based on solvent evaporation method and melt dispersion technique, and analyzed their physicochemical properties in terms of particle size, morphology, and encapsulation efficiency. As a result, they have a ${\pm}150$ nm particle size, spherical shape, and $10^{\sim}25$ % loading efficiency. SLN consists of coconut oil as core material, ascorbic acid and okyong-san as hydrophilic drug.

In the literatual studies on the causes and treatments of the melasma, the results were as follows. 1. Melasma is the blackish patch on the face, is belong to the cartegory of the 'Myunjin(面塵)', 'Myunganzung(면간증)', 'Jakban(雀斑)' etc. in Oriental medicine 2. Melasma is deeply associated with Yangming channel in Meridian, with the spleen, stomach, heart and kidney in Viscera. 3. The pathogenic factors of Melasma is divided five parts. One is insufficient of Yangming's energy in Neijing(內經). Two is incoordination between vital energy and blood caused by wind-evil and phlegm-retention syndrome. Three is anxiety impairing the spleen. Four is kidney-asthenia and fire-hyperactivity. Five is heat-evil. 4. In the treatments of Melasma, Sthenia-syndrome was used cooling blood and activating blood circulation, or dispelling wind-evil and promoting meridian, or expelling fire-evil and removing toxic material etc. Asthenia-syndrome was used invigorating the liver and kidney, or nourishing yin and keeping fire downwards etc. 5. Melasma is concerned with sun-light, is mostly seen in female. 6. In the prescription of Melasma, it was used Jujesamultang(酒製四物湯加減), Okyong -san(玉容散), Chunghwasungitang(沖和順氣湯), Okyongseosisan(玉容西施散), Yukmiji-hwanghwan(六味地黃丸) etc.
