The Effect of Ammonia and Sodium Hydroxide Treatment on the Storage and Rumen Microbial Fiber Degradation in Silage of Rice Straw Contaminated Mycotoxin (암모니아 및 가성소다 처리가 Mycotoxin 오염 사료용 볏짚의 사일레지 저장 및 반추위 미생물의 섬유소 분해에 미치는 영향)

  • This study was conducted to research on the efficacy of chemical treatment as an effective method for reducing mycotoxin in rice straw silage. As a chemical treatment method, ammonia and sodium hydroxid were treated at 4% level of rice straws contaminated with mycotoxin, and the effects of silage storage on fungal toxin reduction, fermentation quality, and fiber digestion were evaluated. Aflatoxin B1, B2, G1, G2 and fumonisin B1, B2 as well as deoxynivalenol were not detected in all experimental groups, and ochratoxin A and zearalenone were detected. Ochratoxin A was detected lower in the chemical treatment than control (41.23 g / kg) (p<0.05). Zearalenone showed lower results in sodium hydroxide treatment (297.44 ㎍ / kg) than control (600.33 ㎍ / kg) and ammonia treatment (376.00 ㎍ / kg) (p<0.05). The pH of rice straw silage was the lowest in ammonia treatment and the highest in sodium hydroxide treatment (p<0.05). The lactic acid contents of control and ammonia treatments were similar, but sodium hydroxide treatment was the lowest (p<0.05). Propionic acid was higher in the control than in the chemical treatments (p<0.05), and showed similar contents in the ammonia and sodium hydroxide treatment. Both the rumen microbial degradation rate of NDF and ADF showed the highest in sodium hydroxide treatment, followed by ammonia treatment, and the control showed the lowest level (p<0.05). Therefore, the results of this study are demonstrated to have a good effect on the treatment of ammonia and sodium hydroxide to reduce the mycotoxins and increase the rumen microbial degradation rate in the rice straw silage. Sodium hydroxide treatment was more effective in reducing mycotoxins and improving fiber degradation rate than ammonia treatment, but it is thought to have an inefficient effect on silage fermentation in rice straw silage.

Effect of Varying Levels of Aflatoxin, Ochratoxin and Their Combinations on the Performance and Egg Quality Characteristics in Laying Hens

  • A 50 day feeding trial was conducted with White Leghorn (WL) laying hens, 42 weeks old, to determine if feeding of varying levels of aflatoxin (AF), ochratoxin A (OA) or their combinations has any effect on their performance and egg quality parameters. Feeding of $T_4$, $T_7$, $T_8$, $T_9$ and $T_10$ caused significant reduction in feed intake of hens. Hen day egg productions were significantly reduced at all the levels of toxins except 0.5 ppm of AF. Maximum reduction in egg production was noticed at 2 and 4 ppm of AF and OA, respectively. Average body weight and egg weight were not affected by toxin feeding. The feed efficiency in terms of net feed efficiency and feed consumed per dozen egg produced was significantly reduced at higher levels of both the toxins and their combinations. Feed consumption for production of 1 kg egg mass remained uninfluenced due to aflatoxin feeding whereas significant increase in the value of the same was noticed at 4 ppm level of OA and combination of 1 and 2 ppm of AF and 2 and 4 ppm of OA ($T_9$ and $T_10$), respectively. Various levels of OA (1-4 ppm) and all the combination of two toxins ($T_8$, $T_9$ and $T_10$) significantly altered the shape index of eggs in laying hens. The shell thickness was significantly reduced by higher level of AF (2 ppm), OA (2 and 4 ppm) and their combination. Albumen index, Haugh Unit and yolk index remained unchanged due to incorporation of toxins in the diet. It is concluded that AF, OA either singly or in combination at higher levels could depress the performance in terms of egg production and feed efficiency significantly. The egg quality parameters i.e. shape index and shell thickness were also significantly affected.

Prevalence of mycotoxin contamination in pig feedstuffs (양돈장 사료의 곰팡이독소 오염률 조사)

  • To analyze prevalence of mycotoxins, a total of 74 feedstuff samples were collected from silos (n=37) and hoppers (n=37) in nine pig farms. Six mycotoxins were tested with commercialized ELISA kits. All samples were contaminated with four or more mycotoxins. Zearalenone was detected in all of the tested samples. Ochratoxin, deoxynivalenol and H-2/HT-2 toxin were detected in more than 90% of the samples. And also, fumonisin was positive in 89.2% of the samples from the silos, 75.2% from the hoppers, respectively. On the other hand, aflatoxin was detected in about 40% of the samples. When the behavior of lactating sows was observed, possible mycotoxicosis was suspected. It was confirmed that their feedstuffs were contaminated with high levels of mycotoxins such as ochratoxin and T-2/HT-2 toxin. After cleaning the feedline, the clinical symptoms in sows suspected with mycotoxicosis were disappeared. Although mycotoxin concentration in most of the feedstuffs was below the acceptance level, these data indicate that what are required is more monitoring and continuous management for mycotoxins in pig feedstuffs.

Isolation of Bacillus amyloliquefaciens Strains with Antifungal Activities from Meju

  • Bacilli with fibrinolytic activities were isolated from traditionally-prepared Meju and some of these strains showed strong antifungal activities. One isolate, MJ1-4, showed the strongest antifungal activity. MJ1-4 and other isolates were identified as B. amyloliquefaciens strains by recA gene sequencing and RAPD-PCR results. B. amyloliqufaciens MJ1-4 efficiently inhibited an Aspergillus spp.-producing aflatoxin B1 ($AFB_1$) and a Penicillium spp.-producing ochratoxin (OTA) in addition to other fungi. Antifungal activity of B. amyloliquefaciens MJ1-4 culture reached its maximum (40 AU/mg protein) in LB or TSB medium around 48 hr at $37^{\circ}C$. Antifungal activity of the concentrated culture supernatant was not decreased significantly by protease treatments, implying that the antifungal substance might not be a simple peptide or protein. Considering its antifungal and fibrinolytic activities together, B. amyloliquefaciens MJ1-4 can serve as a starter for fermented soyfoods such as Cheonggukjang and Doenjang.

Investigation of Unintentionally Hazardous Substance in Teas (식수대용차 재료의 비의도적 유해물질 오염도 조사연구)

  • This study was conducted to survey levels of heavy metals, benzopyrene and mycotoxins in teas purchased from online markets, supermarkets, and traditional markets in Gwangju, Korea. We examined the arsenic, cadmium and lead content in heavy metals. We also tested for benzo(a)pyrene, which is classified as a carcinogen. In addition, we tested for the mycotoxins aflatoxin (AFB1, AFB2, AFG1, AFG2), ochratoxin A (OTA), fumonisin (FUB1, FUB2), and zearalenone (ZON). According to our results, heavy metals were detected within the allowable range (below 5.0 mg/kg for lead) and all samples met with the standards for tea as presented in the Korean Food Code. The Benzo(a)pyrene showed a detection rate of 12.8% in 16 of 125 samples and mycotoxins showed 23.2% in 29 of 125 samples. However, safe levels regarding benzo(a)pyrene and mycotoxin in tea were not ascertainable since the standards are not set in the Korean Food Code.

Occurrence of Fungal Contamination in Ginseng Sprout and Mycotoxigenic Potential (새싹삼의 곰팡이 발생과 독소생성능)

  • In order to investigate frequency of fungal contamination in ginseng sprout, we collected 18 types of retail ginseng sprouts and analyzed them. Overall frequency of fungal contamination ranged from 113.3 to 174.1% with the highest occurrence of Penicillium spp. Fungal detection rate was significantly higher in moss than in stem, leaf and root of ginseng sprout. Penicillium spp. occurred in leaf and stem with the highest incidence and Fusarium spp., in root. Among Penicillium spp. and Fusarium spp., P. olsonii and F. oxysporum were dominant, respectively. Nine Fusarium species, Aspergillus westerdijkiae, Aspergillus flavus, and 11 Penicillium species were identified by phylogenetic analysis. PCR screening of mycotoxigenic potential revealed that 19 out of 25 isolates tested were positive for respective mycotoxin biosynthetic gene. Two 2 A. flavus and 11 A. westerdijkiae isolates produced varying amount of aflatoxin or ochratoxin A in czapek yeast extract brothsome of which showed high levels of mycotoxin production. These results suggests a need for continuous monitoring and management program to control fungal contamination in the ginseng sprout production chain.

Lack of an Apparent Association between Mycotoxin Concentrations in Red Chili Peppers and Incidence of Gallbladder Cancer in India : an Ecological Study

  • Our recent studies conducted in South America have shown that mycotoxin contamination of red chili peppers (RCPs) may be associated with an increased risk of gallbladder cancer (GBC). Whether this relationship exists in India, a country with a high incidence of GBC and high consumption of RCPs, is unclear. We therefore measured concentrations of aflatoxins (AFs) and ochratoxin A (OTA) in RCPs from areas of low, medium, and high incidence of GBC in India, and compared these concentrations with GBC incidence in each area. Twenty-one RCP samples were collected from nine cities (eight from a low-incidence area, five from a medium-incidence area, and eight from a high-incidence area). Concentrations of AFs and OTA were measured using high-performance liquid chromatography. No significant differences in mean concentrations of AFs and OTA were found in the three areas. AFB1 levels in the low-incidence area ($10.81{\mu}g/kg$) and high-incidence area ($12.00{\mu}g/kg$) were more than 2.2 and 2.4 times higher compared with the maximum permitted level of AFB1 in spices ($5.0{\mu}g/kg$) set by the Commission of the European Communities, or that ($4.4{\mu}g/kg$) obtained in our previous study in Chile. Our results show that the mean concentrations of mycotoxins in RCPs are similar among the three areas in India with different incidences of GBC. Further studies with human subjects are needed to evaluate any association between AFB1 and GBC.

Safety Investigation on Foodborne Pathogens and Mycotoxins in Honeybee Drone Pupas (수벌번데기로부터 식중독 세균 및 곰팡이독소 안전성 평가)

  • In this study, safety investigations on harmful microorganisms and mycotoxins were conducted on honeybee drone pupae as a new food material, which is rich in nutrients and capable of being mass produced in apiaries. The honeybee drone pupae produced in apiaries were collected from three different regions in Korea and frozen immediately. Subsequently, the samples were subjected to freeze-drying. According to the Korean Food Code test method, coliforms, Salmonella species, Staphylococcus aureus, and enterohemorrhagic Escherichia coli were not detected in 280 honeybee drone pupas. In addition, mycotoxins, aflatoxin $B_1$, ochratoxin A, deoxynivalenol, and zearalenone were not detected. Therefore, it is proposed that the honeybee drone pupae collected from the beehives and immediately frozen as safe from harmful microorganisms and mycotoxins and can be used as a food material.

Optimization of Analytical Methods for Ochratoxin A and Zearalenone by UHPLC in Rice Straw Silage and Winter Forage Crops (UHPLC를 이용한 볏짚 사일리지와 동계사료작물의 오크라톡신과 제랄레논 분석법 최적화)

  • The objective of this study was to optimize analytical methods for ochratoxin A (OTA) and zearalenone (ZEA) in rice straw silage and winter forage crops using ultra-high performance liquid chromatography (UHPLC). Samples free of mycotoxins were spiked with $50{\mu}g/kg$, $250{\mu}g/kg$, or $500{\mu}g/kg$ of OTA and $300{\mu}g/kg$, $1500{\mu}g/kg$, or $3000{\mu}g/kg$ of ZEA. OTA and ZEA were extracted by acetonitrile and cleaned-up using an immunoaffinity column. They were then subjected to analysis with UHPLC equipped with a fluorescence detector. The correlation coefficients of calibration curves showed high linearity ($R^2{\geq_-}0.9999$ for OTA and $R^2{\geq_-}0.9995$ for ZEA). The limit of detection and quantification were $0.1{\mu}g/kg$ and $0.3{\mu}g/kg$, respectively, for OTA and $5{\mu}g/kg$ and $16.7{\mu}g/kg$, respectively, for ZEA. The recovery and relative standard deviation (RSD) of OTA were as follows: rice straw = 84.23~95.33%, 2.59~4.77%; Italian ryegrass = 79.02~95%, 0.86~5.83%; barley = 74.93~97%, 0.85~9.19%; rye = 77.99~96.67%, 0.33~6.26%. The recovery and RSD of ZEA were: rice straw = 109.6~114.22%, 0.67~7.15%; Italian ryegrass = 98.01~109.44%, 1.65~4.81%; barley = 98~113.53%, 0.25~5.85%; rye = 90.44~108.56%, 2.5~4.66%. They both satisfied the standards of European Commission criteria (EC 401-2006) for quantitative analysis. These results showed that the optimized methods could be used for mycotoxin analysis of forages.
