Effects of Resistance Exercise Training and High Protein Diet on Anabolic Factors of Skeletal Muscle in Sarcopenic Obese Rats (저항성 운동과 고단백식 혼합처치가 sarcopenic obese 쥐의 골격근 내 단백질 합성요인에 미치는 영향)

  • The purpose of this study was to investigate the effects of resistance exercise training and high-protein diet on anabolic factors and insulin resistance of skeletal muscle in sarcopenic obese rats. 50wks of male Sprague-Dawley rats were randomly assigned for 4 groups(Chow, HP, Ex, HPEx) after 6 weeks of high-rat diet induced obesity period. The 8-week of ladder climbing exercise significantly reduced body fat and insulin resistance, significantly increased mTOR activity. However hind limb muscles weight were not changed. When treat with exercise and high-protein diet, body fat and insulin resistance did not improve, but rather the effect of exercise training appeared to be inhibited. Therefore high protein diet for improving the sarcopenic obesity may be need more study about the amount and composition of protein.

The Effect of Ephedrae Herba Pharmacopuncture on Adipocyte Metabolism (마황약침(麻黃藥鍼)이 지방세포 대사에 미치는 영향)

  • Objectives : This study was carried out to investigate the effects of Ephedrae Herba pharmacopuncture (EHP) on the adipogenesis in 3T3-L1 cells, lipolysis in rat epididymal adipocytes and histological changes in porcine adipose tissue. Methods : Inhibition of preadipocyte differentiation and/or stimulation of lipolysis play important roles in reducing obesity. 3T3-L1 preadipocytes were differentiated with adipogenic reagents by incubating for 3 days in the absence or presence of EHP ranging from 0.01 to 1.0 $mg/m{\ell}$. The effect of EHP on adipogenesis was examined by measuring glycerol-3-phosphate dehydrogenase (GPDH) activity and by oil red O staining. Mature adipocytes from rat epididymal fat pad were incubated with EHP ranging from 0.01 to 1.0 $mg/m{\ell}$ for 3 days. The effect of EHP on lipolysis was examined by measuring free glycerol released. Fat tissue from porcine skin was injected with EHP ranging from 0.1 to 10.0 $mg/m{\ell}$ to examine the effect of EHP on histological changes under light microscopy. Results : The following results were obtained from present study on adipogenesis of preadipocytes, lipolysis of adipocytes and histological changes in fat tissue. Proliferation of preadipocytes was significantly inhibited by EHP at the concentration of 1.0 $mg/m{\ell}$. Lipolysis of adipocytes was increased by EHP at the concentration of 0.1, 1.0 $mg/m{\ell}$. Porcine fat tissues were widely injured by EHP at the concentration of 10.0 $mg/m{\ell}$. Conclusions : From the above results, EHP efficiently induces inhibition of preadipocytes proliferation, lipolysis of adipocytes and histologic injury in fat tissues. Therefore, EHP may be useful to treat localized obesity.

Biochemical mechanism of Jijang-Kimch on anti-obesity effect and blood lipid metabolism in obesity model rats (비만모델 랫드에서 지장김치의 혈액 지질대사, 항비만 효능 및 메카니즘)

  • This study investigated action mechanism and biological effect of Jijang-kimch, including its anti-obesity effect and blood lipid-decreasing effect in a high-fat diet-induced obese model animals. There were four treatment groups: CD (chow diet as normal control), HFD (high fat diet as obesity control), HFDCK (HFD plus commercial kimchi extracts), and HFDJK (HFD plus Jijang-kimchi extract). Kimchi extracts were orally administered for 28 days. Body weight, liver, and adipose tissue weight declined in HFDJK compared to those in HFDCK(p<0.05). Blood triglyceride, total cholesterol, low density lipoprotein-cholesterol (LDL-C), and glucose level decreased in CD, HFDJK, and HFDCK compared to those in HFD(p<0.05). Those in HFDJK were lower than those in HFDCK(p<0.05). Sizes of liver and adipose cells increased in HFD, HFDCK, and HFDJ that those in CD(p<0.05). Those in HFDJK were greatly decreased than those in HFDCK(p<0.05). These results indicate that ingestion of Jijang-kimchi in obese model animals has anti-obesity effect by lowering blood lipid and glucose levels and decreasing adipocyte size compared to that of commercial-kimchi.

Effects of Extracts of Monascus-fermented Angelica gigas Nakai in High-fat Diet-induced Obesity in Rats (고지방 식이 유도 비만 흰쥐에서 홍국발효 참당귀 추출물의 항비만 효과)

  • This study aimed to identify the effects of extracts of fermented Angelica gigas Nakai (A. gigas) with a Monascus purpureus strain on anti-obesity in a high-fat diet (HFD)-induced obesity rat model. Male Sprague-Dawley rats were randomly divided into seven dietary groups (n=8 per group), as follows: a normal diet group (N) and six HFD groups (C: control, HFD and no treatment; AG: HFD +10% A. gigas extracts; FAG2.5: HFD +2.5% fermented A. gigas extracts; FAG5: HFD +5% fermented A. gigas extracts; FAG10: HFD +10% fermented A. gigas extracts; GC: HFD + Garcinia cambogia extracts). FAG-fed rats exhibited effectively lowered rates of increasing body weight and visceral fat accumulation in the HFD-induced obesity model. The activities of several hepatic marker enzymes, including aspartate aminotransferase (AST), alanine aminotransferase (ALT), lactate dehydrogenase (LDH), and alkaline phosphatase (ALP), were increased with HFD-induced obesity, but levels of these marker enzymes were significantly decreased in FAG-fed rats. The consumption of FAG reduced triglyceride concentrations in serum to normal levels. FAG-fed rats showed effectively increased leptin concentrations in the HFD-induced obesity model. HFD ingestion induced a significant increase in the thiobarbituric acid reactive substances (TBARS) levels, which was decreased in FAG-fed rats. Hematoxylin and eosin staining and Oil Red O staining of the liver showed that the lipid deposits were decreased via FAG feeding. Moreover, hematoxylin and eosin staining of epididymal adipose adipose tissue showed that the adipocyte were decreased by feeding FAG.

  • Under normal conditions, women produce a single dominant follicle that participates in a single ovuation each menstrual cycle. But Polycystic ovary syndrome(PCOS) conditions, folliculogenesis does not proceed normally. This condition leads to the accumlation of large numbers of small graffian follicles in which the theca interstitial cells (TIC) produce abnormally large amounts of androgen. PCOS is probably the most common endocrine disorder, affecting women of reprodutive age with 5-10% prevalence estimate. Chronic anovulation, hyperandrogenism, hirsutism, obesity, infertility and polycystic ovaries are clinical hallmarks of women with PCOS. Its etiology remains unknown. To investigate the gene expression pattern of ovary in PCO-induced rat, we used cDNA expression analysis. Total RNA was extracted from the ovary of PCO-induced rat and reverse-transcribed in the presence of[$\alpha$$^{32}$P]-dATP Which were hybridized to Atlas$^{TM}$ Rat Toxicology 1.2 array (Clontech) representing approximately 1176 rat genes. We compared gene expression between ovary of pco-induced immature female rats and control. Differential gene expression profiles were revealed (LIFR-alpha, ADRA1A, Heat shock 90-kDa protein A, PDGFRA). Reverse transcription-polymerase chain reaction(RT-PCR) was used to validate the relative expression pattern obtained by the cDNA array. The precise relationship between the altered expression of genes and PCO is a matter of further investigation. This study was supported by Korea Science and Engineering Foundation(KOSEF)

Effect of Ginseng and Herbal Plant Mixtures on Anti-Obesity in Obese SD Rat Induced by High Fat Diet (홍삼 및 천연물 혼합소재의 비만 유도 흰쥐에 대한 항비만 효과에 관한 연구)

  • Anti-obesity effects of ginseng and herbal plant mixtures were investigated to develop natural materials for anti-obesity. After inducing obesity with high fat diet for 8 weeks in male SD rats, ginseng and herbal plant mixtures DM1 (ginseng, puer tea, opuntia) and DM2 (ginseng, puer tea) were administrated orally to rats for another 8 weeks. During administration, food efficacy ratio and body weight of rat were measured twice weekly. After administration, body weight, body fat contents, and serum lipid level were estimated for anti-obesity effect and hematological analysis blood level of ALP and ASP was checked for safety. Body weight in rats fed high fat diet was significantly increased. Body weight in obese rats induced by high fat diet was significantly decreased by DM1 and DM2 feedings. The amount of body fat (epididymal, perirenal and visceral fat, brown adipose tissue) was significantly reduced by DM1 and DM2 treatments. The amount of TG, the concentration of leptin in blood plasma, and the concentration of insulin in blood plasma were significantly diminished by DM1 and DM2. Lipid accumulation on liver was reduced in DM2. There were no side effects among all groups according to blood analysis, hematological findings, and body weight. The findings of this study suggest that DM1 and DM2 may be effective materials for anti-obesity through reducing plasma triglyceride and body fats, and also decreasing body weight without side effects.

Anti-Obesity Effect of Fermented Detoxified Rhus verniciflua Vinegar Supplementation in Diet-Induced Obese Rats (무독화한 옻발효초가 고지방식이를 급여한 흰쥐에 미치는 항비만 효과)

  • The study aimed to investigate the anti-obesity effects of 1% Rhus verniciflua vinegar (RV) supplementation in high-fat-diet (60% fat)-induced obese rats. A total of 50 4-wk-old male Sprague-Dawley rats were fed normal chow diet or maintained on high-fat diet (HFD) for 12 weeks to induce obesity and were then randomized into five groups as follows: normal diet+ultra-pure water (CON), HFD+ultra-pure water (OB-DW), HFD+1% acetic acid (OBAA), HFD+1% RV (OB-RV), and HFD+0.1% caffeine (OB-CF). AA was used as a control for RV, and caffeine was used as a positive control due to its weight reducing effect. After 2 months, body weight, organ and adipose tissue weights, serum lipids, hepatic lipids, adipocyte size, and cell number per spot level were analyzed. As a result, food efficiency ratio, abdominal adipose tissue weight, serum levels of total cholesterol, triacylglycerol, free fatty acids, coronary artery index, and fecal lipid were significantly reduced in the RV treatment group. In this study, we found that dietary RV improved obesity by increasing lipid excretion and reducing lipogenesis. These results suggest that RV has potential as a functional anti-obesity food.

Effect of Allium Victorialis Var. platyphyllum Leaves on Triton WR-1339-Induced and Poloxamer-407-Induced Hyperlipidemic Rats and on Diet-Induced Obesity Rats (식이성 고지혈 및 비만에 대한 산마늘 추출물의 효과)

  • The herb of Allium victorialis var. platyphyllum (Liliaceae) has been used as an edible wild herb and to treat heart failure and gastritis. We have already reported antihyperlipidemic anti-tumor effects of this plant. To enlarge the commercial availability of this food, it was investigated whether the extracts of A. victorialis var. platyphyllum reduce hyperlipidemia and obesity or not. The plants tested in this experiment were collected from two eco-types of Is. Ullung and Mt. Odae cultivated at Pyongchang. Extracts were prepared by extracting the fresh leaves and those dried at $36^{\circ}C$ and $90^{\circ}C$, respectively. Pretreatment with the ethanolic extracts for two weeks(p.o.) reduced serum triglyceride-, total cholesterol- and LDL-cholesterol contents in rats induced by Triton WR-1339, respectively. Furthermore, oral administration of the extracts also inhibited the hyperlipidemia induced with oral diet of 30% corn oil. In the other attempt to find to alleviate the obesity, the model rats with obesity were induced by the high fat-diet for six weeks. Post-treatment with the extracts for two weeks significantly reduced the hyperlipidemia. Retroperitoneal-, epidymal- and total abdominal fat pad weights were considerably reduced at 100 mg/kg oral administration of the extracts. Increased feces lipid contents were also found in the rat treated with the extracts. The extract may lead to the higher activity in treatment of hyperlipidemia and obesity than of the dried one.

The Effect of Germanium Complex on the Body Fat Weight, Body Weight and Serum Biochemical Value in Rats Fed High Fat Diets (게르마늄 복합물이 비만유도 흰쥐의 체지방 및 체중과 생화학적 변화에 미치는 영향)

  • Germanium is found in a range of minerals and ores and is present in foods including beans, tomato juice, oysters, tuna and garlic. Germanium is a non-metallic element, which can exist in valence states of 2 and 4. Clinical trials and use in private practices for more than a decade have demonstrated organic germanium's efficacy in treating serious disease including cancer, arthritis and senile osteoporosis. But it was rarely reported that inorganic germanium has biological properties. STB-BM contains mineral complex, rare earth elements and a little amount of Inorganic germanium. The experiment was carried out the anti-obesity effect. To investigate anti-obesity effect of STB-BM, we measured the effect of body weight, fat weight (subcutaneous fat, epididymal fat, visceral fat, kidney fat and total fat) and serum biochemical level in rats fed high fat diets. STB-BM 35 mg/kg suppressed the increasing ratio of body weight, epididymal fat weight, visceral fat weight, total fat weight, triglyceride and LDL-cholesterol (p<0.05).

Weight Loss Effect of Wax Gourd (동아의 체중조절 효과)

  • This study attempted to evaluate the effectiveness of function of Wax Ground for obesity and the related disease of adult people can be developed by the combination of the 17 food materials screened in this study. The experiments using Sprague-Dawley male rats show that seventeen kinds of food material inhibited the increase of body weight gain, energy intake, fat pads weight, liver triglycerides content, atherogenic index, and blood glucose concentration induced by cafeteria diet. Oral administration of wax gourd decreased most body weight gain, energy intake, fat pads weight, and blood glucose concentration. Liver triglycerides content was the lowest in the group administered mulberry stem extract and the group administered pine leaves showed the lowest atherogenic index. The combined administration of wax gourd, mulberry stem extract and pine leaves improved more the obesity and related symptoms than the individual administrations.
