The study was conducted to determine the efficiency in producing spontaneous polyploids in some mandarin hybrids with different seed embryony. Seed formation by open pollination, frequency of spontaneous polyploids, and plant growth characteristics were evaluated in four mandarin hybrids with polyembryony such as 'Amakusa', 'Haruka', 'Hayaka', and 'Seminole' and two with monoembryony such as 'Benibae' and 'Harehime'. The mean number of the developed seeds per fruit was 10.0 and frequency of small seeds was 25.1%. Polyploids were selected from plants germinated in vitro by a flow cytometry and confirmed by chromosome analysis. One triploid was produced from 'Harehime', one tetraploid, 'Amakusa', and one tetrapoid, 'Benibae'. There were little differences in leaf shape, thickness, petiole length, and internode length between diploids and polyploids such as tri- or tetraploid. However, polyploids had larger stomata and lower density of stomata in abaxial epidermis than diploids. SPAS indicating chlorophyll content and photosynthetic rate were significantly affected by ploidy level. The results indicated that spontaneous polyploids might be produced by open pollination in some mandarin hybrids and monoembryony had higher frequency in polyploid occurrence than polyembryony.
This study was conducted to investigate the morphological and anatomical characteristics of needle and the genetic diversity of Pinus pumila Regel which is a unique and the southern peripheral population in South Korea. ANOVA test showed that there were significant differences among individuals within population in all 8 needle characteristics. Average characteristics of 66 individuals were 53.59 mm in needle length, 0.78 mm in needle width, 68.98 in needle index, 0.65 mm in needle thickness, 4.56 ea. in maximum stomata row, 3.80 ea. in minimum stomata row, 8.36 ea. in total stomata row and 1.71 ea. in resin canals, respectively. Resin canal per needle of this species ranged from one to three, depending on external type. Especially, arrangement types were 69.47% in two resin canals and 30.45% in a single resin canal. A total of 78 bands was generated from 9 selected I-SSR primers. The estimates of genetic variation were 61.5% in proportion of polymorphic bands (P), 1.698 in effective number of alleles ($A_e$), 0.388 in expected heterozygosity ($H_e$) and 0.567 in Shannon's information index (S.I.), respectively.
In bioreactor cultures of plants, inoculum density is an important factor affecting growth and proliferation of the plantlets. To maximize shoot growth and proliferation of grape rootstock '5BB' in bioreactors, inoculum density varied at 15, 30, 45 and 60 single nodes in a 3-liter scale balloon type bioreactor, respectively and cultured for 40 days. Results suggested that the growth and the photosynthesis of the plantlet were greatly affected by inoculum density in the bioreactor. The inoculum density of 45 nodes resulted in the greatest growth (910.4 mg/shoot FW, 764.4 mg/root FW) followed by 30 nodes. $CO_2$ assimilation rate, stomatal conductance, transpiration rate of the plantlet were also highest at the inoculum density of 45 nodes. Significant reduces in shoot and root growth (426.5 mg/shoot FW, 248.4 mg/root FW) were observed at the inoculum density of 60 nodes. When the inoculum density decreased by 15 nodes, plantlets were malformed due to hyperhydricity, resulting in the highest transpiration rate and the lowest $CO_2$ assimilation rate. The plantlets stressed by the inoculum density at 15 nodes and 60 nodes showed larger number and irregular shape of stomata compared to the plantlets inoculated with 45 nodes.
This study was carried out to know the effect of air temperature on photosynthesis and growth of Korean Valerian (valeriana fauriei var. dasycarpa HARA). The results are as follows; 1. Photosynthesis of V fauriei had highly significant relation to temperature in a quadratic regression model, from which the optimum temperature for the plant growth were estimated to be $17.7^{\circ}C$. During the daytime in the field condition, the photosynthesis rate was highest at 9 a.m, then reduced to minimum at 2 p.m., and increased again thereafter. 2. Number of stomata was about $25/mm^2$ on the upper side and $85/mm^2$ on the lower side of the leaf, and the size was $21{\sim}30/{\mu}m$ in diameter. 3. A highly significant quadratic regression was noted between temperature and leaf width or root weight of V. fauriei. It was estimated from the regression equation that the optimum temperature for root growth was $20.3^{\circ}C$.
This study was intended to identify 17 taxa. (5 varieties and 12 species) of the genus Acer in Korea on the basis of the shapes of stomata, the type of trichome on the different part of leaves, the shapes, arrangement and number of stele in cross section of petiole. The results obtained were summarized as follows; 11 The shape of guard cells of stomata in the genus Acer was anomcytical, and the size of the cells ranged from 10.25 to $21.00{\mu}$ in length and from 7.57 to $11.83{\mu}$ in width. 2) Eleven types of trichome on the leaf in the genus Acer were found; pilose, sericeous, velutinous, woolly, glabrate, puberulent, bladder hair, hispid, hirsute and uncinate. Anatomical characters of the Bupleurum latissimum Nakai, an endemic species of Korea, were investigated to confirm its phylogenetic relationships. Compare to other species with anatomical characters, B. latissimum is very similar with B, euphorbioides and B, longeradiatum in point of lacking of pith in the stem, shape of involucres, number of vascular bundles in radical leaf and cauline leaf, and lacking stomata in adaxial leaf surface. The other hand, protruded pollen aperture character appears in B. latissimum and B. euphorbioides. On the based of anatomical characters, therefore, B. latissimum has closest relationships with B. euphorbioides and B. longeradiatum. It also needs molecular study including Asian species in order to confirm phylogenetic position and speciation process apparently.
Pepper plants were flooded at 0, 5, 10 and 15cm at seedling stage under the condition of greenhouse. Treatment of flooding times are 6, 12, 24, 48 and 120 hours. The results obtained are summarized as follows. Plant height, number of leaves, shoot and root fresh weight were not recovered the flooding damages regardless of flooding time and depth. Pepper plant died in flooding depth of 5cm or more for over 48 hours. Plants in fallen leaves were found at more than 5cm depth and 6 hours of floodings. Photosynthesis and respiration rate decreased in the 5cm flooding depth or more for 24 hours. Chlorophyll content and root activity decreased for 12 hours or more at all the flooding depth. Also, diffusion resistance of stomata cell increased as increased flooding time and depth. Diseases occurred remarkably in proportion to the depth and hours of flooding treatment. It was not possible to control the desease by fungicide, also then was no effects of foliar spray of urea. Weight of fruit per plant not decrease by the 12 hours of 0cm and the 6 hours of 5cm flooding but decreased at deeper and longer flooding. Average weight of a fruit increased. The yield could not expected in the depth of 5cm or more for over 48 hours, There was significant positive correlation between all the investigated characteristics of growth and yield. There was, however, negative correlation between number of leaf and diffusion resistance of stomata.

The aim of this study was to provide the basic information necessary to identify Korean fir (Abies holophylla), momi fir (A. firma), and Nikko fir (A. homolepis), and other fir trees planted in South Korea that are protected by law. Analysis of the morphological characteristics of the needles from each sample was investigated. The shape of the needle-leaf tip from A. holophylla was acute, whereas that from A. firma and A. homolepis was emarginate and that from the protected fir trees was obtuse. The number of stomata on the needles was not significantly different between A. holophylla and A. firma, and the number of stomata on the needles from A. homolepis and the protected fir trees were highly similar. In addition, the genetic differences among the Abies species were analyzed using the sequences of five chloroplast DNA regions-matK, atpF-atpH, rpoC2-rps2, rpoC1, and psbA-trnH.The atpF-atpH and psbA-trnH regions were useful for discriminating A. firma from the other species, but there were no differences among A. holophylla, A. homolepis, and the protected fir trees. The same chloroplast sequences were found in both A. holophylla and A. homolepis, which suggests that additional genetic studies might be necessary to identify the Abies species planted in both South Korea and Japan.
The albino plants of tobacco (Nicotiana tobacum L. cv. BY-4) were isolated from seed populations that were induced by heavy-ion ($^{14}N$) beam irradiation to proembryo and the in vitro growth and differentiation have been characterized. The in vitro cultured albino plants showed significant reduction of chlorophyll content and possessed larger number of stomata on both upper and lower epidermis than that of wild-type plants. Stem growth of the mutants remained dwarfed, however, the internode recovered its normal length after GA$_3$ treatment (10.0mg/L) on the MS medium containing sucrose under continuous light. When explants of leaf blades of albino plants were cultured, multiple shoots formed directly on MS medium containing 1.0mg/L of BAP or kinetin and a large number of calli were induced on the MS medium containing 1.0mg/L NAA or 1.0 mg/L 2,4-D. The albino calli regenerated multiple albino plantlets in the MS medium containing 0.1mg/L NAA + 1.0 mg/L BAP. No significant differences between the wild-type and albino plants were detected in the multiple shoot induction, callus formation from the explants and the plantlets regeneration from calli. In addition, albino plants have a similar organogenesis Pattern to that of the wild-type in the media with different combinations of NAA (0 to 5.0mg/L) and BAP (0 to 5.0mg/L) treatment. These results indicate that the albino mutant has the same normal regeneration ability as that of wild-type, although the mutant has lost functions in photosynthesis, such as pigmentation.
The good growth of Populus alba ${\times}$ P. glandulosa is shown at the foothill of the mountain where good soil moisture is available, but its growth is poor at the upper part of the mountain where soil moisture is limitted. Stomatal frequency, stomatal size and transpiration rates that affect soil moisture demand were determined in order to know how this character is inherited from its parents. Number of stomata per $mm^2$, length and width of stomata from the abaxial leaf and transpiration rate ($g/cm^2$) per day in P. alba ${\times}$ P. glandulosa showed intermediate values between those of their parents, while P. alba showed the highest values and P. davidiana the lowest values. The values of stomatal frequency of P. alba ${\times}$ P. glandulosa were close to the expected mean values of parents in the $x^2$ test, and in the regression analysis the transpiration rates for the hybrid and its midparent had strong relationship with 0.97 of regression coefficient. The fact that these intermediate characteristics were observed in the hybrid suggests incomplete dominance in the hybrid. It is concluded that soil moisture demand in P. alba ${\times}$ P. glandulosa and its parents is closed related to stomatal frequency, stomatal size and transpiration rate.
