Application of Image Analysis System for Red Tide Organisms

  • Relative DNA contents in some harmful algae were measured using DAPI staining and image analysis system. This method was useful to identify some morphologically similar species and isolates from harmful algal blooms (HABs). In exponential phase, Prorocentrum micans had higher relative DNA content (RD) of $1.83\pm0.52$ than any other isolates, followed by Cochlodinium polykrikoides $(1.10\pm0.46)$ Alexandrium tamarense $(0.93\pm0.32)$ Gyrodinium impudicum $(0.56\pm0.17)$, Scrippsiella trochoidea $(0.41\pm0.26)$ and P. minimum$(0.05\pm0.01)$. When they were fixed with Lugol's solution, it was difficult to d,iscern C. polykrikoides from G. impudicum under the light microscope, but the DNA contents were quite different in two species. C. polykrikoides contained about twice as much RD as G. impudicum under the same culture conditions and exponential phase. DAPI­stained DNA feature in C. polykrikodes showed concentrated in the peripheral part of the cell, but in G. impudicum showed a compact structure in the central part. Although A. tamarense and S. trochoidea were morphologically similar under the light microscope, nuclear DNA content of A. tamarense was twice as much as that of S. trochoidea.

Cytogenetic Study of Pleuronectes obscurus, Konosirus punctatus and Pseudoblennius percoides

  • Our objective was to clarify the cytogenetic characteristics, including karyotypes, cellular DNA content, and nuclear size of erythrocytes, of black plaice Pleuronectes obscurus, dotted gizzard shad Konosirus punctatus, and perch sculpin Pseudoblennius percoides, collected from the coastal areas of Jo Island, Busan, Korea. Karyotypes of P. obscurus and K. punctatus both had a diploid number of 48 and a fundamental number (FN) of 48, with a chromosome formula of 48T. The karyotype of p. percoides had a diploid number of 46 and FN of 56, with a chromosome formula of 10SM +36T. No sex-associated heteromorphic pairs were detected for any species. The variation in DNA values (P. obscurus=1.15 pg/nucleus, K. punctatus=1.56pg/nucleus, P. percoides=1.11 pg/nucleus) was positively related to variation in chromosome FN.

  • Glucosylated polyethylenimine (GPEI) was synthesized as a tumor-targeting gene carrier through facilitative glucose metabolism by tumor glucose transporter. Particle sizes of GPEI/DNA complex increased in proportion to glucose content of GPEI, whereas surface charge of the complex was not dependent on glucosylation, partially due to inefficient shielding of the short hydrophilic group introduced. GPEI with higher glucosylation (36 mol-$\%$) had no cytotoxic effect on cells even at polymer concentrations higher than 200 $\mu$g/mL. Compared to unglucosylated PEl. glucosylation induced less than one-order decrease of transfection efficiency. Transfection of GPEI/DNA complex into tumor cells possibly occurred through specific interaction between glucose-related cell receptors and glucose moiety of GPEI. Gamma imaging technique revealed GPEI/DNA complex was distributed in liver. spleen. and tumors.

ITS 및 rbcL 염기서열에 근거한 한국 자생 옻나무속의 계통분류 (Phylogeny of Korean Rhus spp. Based on ITS and rbcL Sequences)

  • 한국산 옻나무속 6종에 대하여 분자식물학적 방법으로 계통유연관계를 확인하기 위하여, nrDNA의 ITS 구간과 cpDNA rbcL 염기서열을 사용하여 계통분석한 결과 ITS 1의 길이는 $246{\sim}253\;bp$이었고, ITS 2는 $234{\sim}244\;bp$이었다. ITS 1의 길이는 Rhus sylvestris와 R. succedanea에서 246 bp로 가장 작았으며, R. verniciflua에서 253 bp로 가장 긴 것으로 나타났다. ITS 2의 길이는 R. verniciflua가 234 bp로 가장 짧았으며, R. trichocarpr가 244 bp로 가장 길게 나타났다. 이들 분류군의 G+C Content는 ITS 1에서는 $58.0{\sim}68.13%$의 범위를 나타냈고, ITS 2에서는 $59.75{\sim}68.46%$로 나타나 두 구간이 비슷한 비율을 보이고 있었다. ITS 1에서의 G+C content는 R. sylvestris가 58.0%로 가장 낮았으며, 가장 높은 값은 R. ambigua가 68.13%로 확인되었다. ITS 2에서는 외군인 Cotinus coggygria가 59.75%로 가장 낮았으며, R. ambigua가 68.46%로 가장 높게 나타났다. 한국산 옻나무 속에서 ITS 염기서열은 일반적으로 피자식물이 갖는 G+C content 범위 안에 포함되는 것으로 확인되었다. 한편, rbcL의 길이는 1,428 bp로 모든 종에서 동일하였다. 또한 rbcL의 G+C content는 $43.56%{\sim}43.77%$로 나타나 종간에 거의 차이가 없음을 확인하였다. 연구결과 rbcL gene은 옻나무속의 종간 계통유연관계를 해석하는데 유용하지 않았으며, ITS 1 구간의 염기서열 변이는 향후 옻나무속을 분류할 때 신속하게 분류할 수 있는 분류 marker로 이용할 수 있다고 판단되었다.

수종 의치상 레진의 세포반응에 관한 연구 (AN IN VITRO STUDY ON CELLULAR RESPONSE OF SEVERAL DENTURE BASE RESINS)

  • The present study quantitates the in vitro cytotoxicity of a variety of denture base acrylic resins using cell culture techniques combined with image analysis to measure nuclear area and DNA contents. In this study, a comparison was made among direct curing, heat curing and microwave curing resins. The results obtained from this study were as follows : 1. Morphologically, cell process and nucleus became prominent but macroscopic difference according to the resins were nit observed. In addition, increased cellular density around the specimen were observed. 2. In DNA contents measurements, $S-G_2M$ phase cell was 15.47%, 14.58% in control and heat curing resin on 1st day and the others group $21.39\sim33.36%$ were measured. 3. Nuclear area and DNA contents were increased on 3rd day except DNA content of the microwave curing resin group. These results suggest that denture base acrylic resins stimulate gingival fibroblasts in vitro, especially stimulation of direct curing resin is larger and longer than the others.

Hyperoside Protects Cells against Gamma Ray Radiation-Induced Apoptosis in Hamster Lung Fibroblast

  • Ionizing radiation, including that evoked by gamma (${\gamma}$)-rays, induces oxidative stress through the generation of reactive oxygen species, resulting in apoptosis, or programmed cell death. This study aimed to elucidate the radioprotective effects of hyperoside (quercetin-3-O-galactoside) against ${\gamma}$-ray radiation-induced apoptosis in Chinese hamster lung fibroblasts, V79-4 and demonstrated that the compound reduced levels of intracellular reactive oxygen species in ${\gamma}$-ray-irradiated cells. Hyperoside also protected irradiated cells against DNA damage (evidenced by pronounced DNA tails and elevated phospho-histone H2AX and 8-oxoguanine content) and membrane lipid peroxidation. Furthermore, hyperoside prevented the ${\gamma}$-ray-provoked reduction in cell viability via the inhibition of apoptosis through the increased levels of Bcl-2, the decreased levels of Bax and cytosolic cytochrome c, and the decrease of the active caspase 9 and caspase 3 expression. Taken together, these results suggest that hyperoside defend cells against ${\gamma}$-ray radiation-induced apoptosis by inhibiting oxidative stress.

  • Astaxanthin을 생산하는 효모 Phaffia rhodozyma로부터 제조된 상보적 돌연변이 균주간의 세포융합을 통하여 astaxanthin을 대량 생산하는 균주를 얻고자 시도하였다. 이들의 원형질체 융합빈도는 $1.3$\times$10^-^5-6.0$\times$10^-^5$이었고 DNA함량, 핵염색, UV조사에 대한 생존력 비교 그리고 형질분리분석등으로 핵융합을 확인하였다. 융합체 중 F1은 야생형의 모 균주와 비교했을때 astaxanthin생성량이 약 3배 증가하였다.

Transcriptional Regulation of the Drosophila Proliferating Cell Nuclear Antigen Gene and raf Proto-oncogene by Ursolic Acid in Drosophila Cultured Kc Cells

  • Promoter of the Drosophila proliferating cell nuclear antigen (PCNA) gene contains DRE (Drosophila DNA replication-related element) required for the high level expression of replication-related genes. Recently, we found that promoter region of the D-raf (a Drosophila homolog of the human c-raf-1) contains two sequences homologous to the DRE and demonstrated the DRE/DREF (DRE-binding factor) involvement in regulation of the D-raf gene. In this study, using ursolic acid (UA), a pentacyclic triterpene acid reported to possess antitumor activities, we examined effects of UA on proliferation of the Drosophila cultured Kc cells and on expression of the PCNA and D-raf genes. UA showed an inhibitory effect on proliferation of the Kc cells in a concentration-dependent manner in DNA content assays and [3H]thymidine incorporation assays. The IC50 value of anti-proliferative effects of UA in DNA content assays was about 7.5uM. UA showed inhibitory effects on expression of the PCNA as well as on that of the D-raf, which were examined with the reporter plasmic p5'-168DPCNACAT or p5'-878DrafCAT, respectively. The results obtained in the present study suggest that expression of the PCNA and D-raf genes is coordinately regulated in at least UA-treated Kc cells and that down-regulation of expression of the PCNA and D-raf genes might be related with the antitumor activities of UA.

핵전이에 의한 Aspergillus niger와 Penicillium chrysogenum의 잡종에서의 Lipase 고생산균주의 개발 (Development of Lipase Hyper-producing Strain from Hybrids between Aspergillus niger and Penicillium chrysogenum by Nuclear Transfer)

  • Aspergillus niger와 Penicillium chrysogenum간의 lipase 우수 생성 종간 형질전환체를 획득하고, 유전분석을 통하여 다음과 같은 결과를 얻었다. (1) P. chrysogenum의 영양요구물을 확인하여 본 결과 Tyrosine 요구성임을 알 수 있었다. (2) 핵전이를 위한 원형질체 형성 및 재생 조건에서는 Novozym 234의 농도 1%, 삼투안정제는 0.6M KCl, 효소의 처리 시간은 180분 그리고 최적 pH는 5.8로 나타났다. (3) 핵전이에 의한 형질전환의 빈도는 $1.3{\times}$10^{-3}$$ $-3.8{\times}$10^{-3}$$으로 비교적 낮은 편이었다. (4) 유전적 안정성, conidia의 크기, DNA 함량의 측정 그리고 핵염색의 결과 형질전환체의 핵형은 aneuploid로 추정되었다. (5) 형질전환체의 lipase활성은 모균주와 비교하여 1.4-2.2배 증가하였다.

  • 근년(近年) 고등동물세포(高等動物細胞)에 있어서 유전자(遺傳子)의 본체(本體)인 DNA에서 RNA를 경과(經過)해서 특이적(特異的)인 단백질(蛋白質)의 생합성(生合成)에 도달(到達)하는 경로(經路)에 대(對)해서는 많은 연구(硏究)에 의해서 확립(確立)되어졌으나 그 조절기구(調節機構)에 대(對)해서는 불명(不明)한 점(點)이 많다. 개체(個體), 기관(器管), 세포내구조(細胞內構造) 급(及) DNA의 준위(準位)에서의 방사선(放射線)의 장해(障害)에 대(對)해서도 연구(硏究)되고 있으나 소위(所謂) 방사선감수성(放射線感受性) 급(及) 비감수성(非感受性)의 각장기(各臟器)에서 분리(分離)한 Chromatin (DNA-Histone-잔여단백(殘餘蛋白)의 고차구조결합체(高次構造結合體)에 대(對)한 DNA, RNA, 전단백질(全蛋白質)과 유전수식체(遺傳修飾體)라고 생각되는 Histon-단백(蛋白)의 화학조성(化學組成)을 검출(檢出)했으며 겸(兼)해서 chromatin의 생물활성(生物活性)인 RNA 합성능(合成能)(priming activity)에 대(對)한 방사선(放射線)의 영향(影響)을 조사(調査)하는데 의의(意義)가 있다. 전리방사선(電離放射線) 조사(照射)에 의해서 생체(生體)의 DNA의 합성조해(合成阻害)가 잘 알려진 사실(事實)이나 분화(分化)한 생체조직(生體組織)에서의 DNA의 합성(合成)보다도 일반대사(一般代謝)에 중요(重要)한 역할(役割)을 한다는 것도 생각된다. 세포(細胞)의 대사(代謝)는 내분비계등(內分泌系等)의 "Effector-DNA-RNA-단백합성(蛋白合成)이라는 정보유전기구(情報遺傳機構)에 의해서 제어(制禦)되어 있다. 이 연구(硏究)는 방사선생물학상(放射線生物學上) 중요(重要)한 것은 논할(論) 필요(必要)도 없으며 방사선동위원소표지화합물(放射線同位元素標識化合物)을 사용(使用)하여 생화학적(生化學的)으로 추구(推究)하였다.
