Molecular Phylogenetic Study of Korean Tilia L. (한국산 피나무속(Tilia L.) 식물의 분자 계통학적 연구)

  • The genus Tilia is characterized by linear form bracts of which the lower part is attached to the peduncle of a cyme. This character is distinguished from the others genus of Malvaceae. The purpose of this study is verifying the phylogenetic relationship of genus Tilia. Phylogenetic analyses were conducted to evaluate relationships of 10 taxa of Tilia in Korea and Japan including one outgroup (Gossypium hirsutum). The molecular phylogenetic analyses were conducted with sequences based on ITS, trnL-F and rpl32-trnL region. The combined data result of ITS, trnL-F and rpl32-trnL was formed by 6 clades. T. kiusiana situated as the most basal clade. T. amurensis, T. taquetii and T. rufa are composed a clade. T. koreana, T. insularis and T. japonica was formed independent clade. T. insularis has the closest relationship with T. japonica. T. miqueliana, T. mandshurica, and T. megaphylla are composed a clade and showed a sister relationship than other species.

Taxonomic study on viola albida var. albida and its related taxa (태백제비꽃과 근연분류군의 분류학적 연구)

  • Taxonomic studies were conducted to evaluate the interspecific relationships in Viola albida var. albida and its related taxa using seven populations for morphology, palynology and anatomy. Molecular phylogenetic studies were also examined in 28 populations including 19 Korean, four Chinese, two Japanese, one American population and two outgroups using nrITS, and 27 populations except V pinnata for trnL-F region of chloroplast DNA. Morphological differences was observed among seven populations of three species in leaf shape, but characters such as serrate number of leaf margins, petal size, pistil shape were showed overlap between populations. Pollen shape of seven populations was monad and grain shape on the polar axis was semi-angular. Morphology of aperture was tri-colporate, and the surface sculpturing was scabrate in rugulate. The grain shape of equatorial view of five populations was prolate whereas V albida var. taknhashii type 1 and V albida var. chaerophylloides type 3 were subprolate. The anatomical characters of rnidvein of leaf, petiole, peduncle, root were also described for the species. The stomatal apparatus of the leaves was observed only in abaxial surface, and the number of stoma per unit ($mm^2$) were abundant in incised than lobate or cleft margin of the leaves. The nrITS analysis shows that V. pinnata and V. dissecta was monophyletic and occupied a basal position in the V. albida var. albida and its related taxa. The other clade including infraspecific populations of V. albida, and V. eizanensis was paraphyletic. The trnL-F noncoding region analysis was similar to the ITS tree. According to the above observations in morphology, palynology, anatomy, and molecular phylogenetic analysis, the significant differences were not found except for leaf shape in Viola albida var. albida and its related taxa, therefore V. albida var. takanhashii and V. albida var. chaerophylloides were considered to be an infraspecific taxa of V. albida var. albida rather than an independent species, subvariety or variety of V. pinnata and V. dissecta.

Biochemical and cultural characteristics of mineral-solubilizing Acinetobacter sp. DDP346 (미네랄 가용화능을 갖는 Acinetobacter sp. DDP346의 생화학적 및 배양학적 특성)

  • In this study, to select strains suitable as microbial agent from among rhizosphere microorganisms present in rhizosphere soil and roots, the mineral solubilization ability, antifungal activity against 10 types of plant pathogenic fungi, and plant growth-promoting activity of rhizosphere microorganisms were evaluated. As a result, DDP346 was selected because it has solubilization ability of phosphoric acid, calcium carbonate, silicon, and zinc; nitrogen fixing ability; production ability of siderophore, indole-3-acetic acid, and aminocyclopropane-1-carboxylate deaminase; and antifungal activity against seven types of plant pathogenic fungi. DDP346 showed a 99.9% homology with Acinetobacter pittii DSM 21653 (NR_117621.1); phylogenetic analysis also revealed a close relationship with Acinetobacter pittii based on the 16S rRNA base sequence. The growth conditions of DDP346 were identified as temperatures in the range of 10-40 ℃, pH in the range of 5-11, and salt concentrations in the range of 0-5%. In addition, a negative correlation coefficient (r2 = -0.913, p <0.01) was shown between pH change and the solubilized phosphoric acid content of Acinetobacter sp. DDP346, and this is assumed to be due to the organic acid generated during culture. Consequently, through the evaluation of its mineral solubilization ability, antifungal activity against plant pathogenic fungi, and plant growth-promoting activity, the potential for the utilization of Acinetobacter sp. DDP346 as a multi-purpose microbial agent is presented.

Molecular phylogeny and divergence of photosynthetic pathways of Korean Cypereae (Cyperaceae) (한국산 방동사니족(사초과) 식물의 분자계통과 광합성경로의 분화)

  • Multiple changes of the photosynthesis pathway are independent evolutionary events occurring in the phylogeny of flowering plants, and such changes have occurred more than five times in Cyperaceae. In the tribe Cypereae, the C4 photosynthetic pathway appeared only once and is regarded as a synapomorphy of the C4 plants within this tribe. The morphological delimitation of genera within Cypereae does not correspond to their molecular phylogenetic relationships. In this study, the molecular phylogeny was compared with the photosynthetic pathways of Korean Cypereae (18 species of Cyperus, 1 species of Kyllinga, and 1 species of Lipocarpha). The photosynthetic pathways were determined by observing the leaf anatomy. The phylogenetic analysis was performed using three DNA regions (nrITS, rbcL, and trnL-F). According to the position of the photosynthetic tissue, 4 species (C. difformis, C. flaccidus, C. haspan, and C. tenuispica) and 16 species (14 Cyperus species, K. brevifolia var. leiolepis, and L. microcephala) were confirmed as C3 and C4 plants, respectively. Tribe Cypereae was divided into the CYPERUS and FICINIA clades, and all species of Korean Cypereae plants belonged to the CYPERUS clade in the phylogenetic analysis. Within the CYPERUS clade, C4 plants were monophyletic but their phylogenetic relationships were unclear. The genera Kyllinga and Lipocarpha were not supported as an independent genus in either case because they were nested by the Cyperus species in the molecular phylogenetic trees in the present and in previous studies. To determine the classification within the CYPERUS clade, a detailed morphological study and a molecular phylogenetic analysis at a high resolution will be necessary.
