An experiment was conducted to study the effect of temperature and pH on in vitro nutrient degradability, volatile fatty acid profile and methane production. The fermenter used was the semi-continuous system, known as the rumen simulation technique (RUSITEC). Sixteen cylinders were used at one time with a volume of 800 ml, the dilution rate was set at 3.5%/hour, the infused buffer being McDougall's artificial saliva. Basal diet (9.6 g DM) used in RUSITEC consisted of (DM) 6.40 g Timothy hay, 1.86 g crushed corn and 1.34 g soybean meal. The food for the fermentation vessel was provided in nylon bags, which were gently agitated in the liquid phase. The experiment lasted for 17 d with all the samples taken during the last 5 d. Treatments were allocated at random to four vessels each and were (1) two temperature levels of $39^{\circ}C$ and $41^{\circ}C$ (2) two pH levels of 6.0 and 7.0. The total diet contained ($g\;kg^{-1}$ DM) 957 OM, 115 CP and $167MJ\;kg^{-1}$ (DM) GE. Although increase in temperature from $39^{\circ}C$ to $41^{\circ}C$ reduced degradation of major nutrients in vitro, it was non-significant. Interaction effect of temperature with pH also reflected a similar trend. However, pH showed a significant (p<0.05) negative effect on the degradability of all the nutrients in vitro. Altering the in vitro pH from 7 to 6 caused marked reduction in DMD from 60.2 to 41.8, CPD from 76.3 to 55.3 and GED from 55.3 to 35.1, respectively. Low pH (6) depressed total VFA production (61.9 vs. 34.9 mM) as well as acetate to propionate ratio in vitro (from 2.0 to 1.5) when compared to pH 7. Compared to pH 7, total gas production decreased from 1,841 ml to 1,148 ml at pH 6, $CO_2$ and $CH_4$ production also reduced from 639 to 260 ml and 138 to 45 ml, respectively. This study supported the premise that pH is one of the principal factors affecting the microbial production of volatile fatty acids and gas. Regulating the ruminal pH to increase bacterial activity may be one of the methods to optimize VFA production, reduce methane and, possibly, improve animal performance.
연구배경: 암세포는 빠른 증식 속도로 인하여 상대적인 저산소증에 노출되면서 비정상적인 종양 혈관을 형성하여 치명적인 병인을 형성한다. EGCG는 녹차의 추출물로 간세포암주 및 전립선암주에서 HIF-1$\alpha$의 발현을 억제하는 것으로 알려져 있다. 그러나 EGCG의 비혈관 증식성 효과에 대해서는 아직 정확히 규명되어 있지 않다. 본 연구에서는 EGCG가 비소세포폐암주에서 HIF-1$\alpha$ 및 VEGF의 발현에 대한 억제 가능성을 확인하여 보고자 하였다. 방 법: 비소세포폐암주인 A549를 RPMI배지에서 계대 배양하였다. 저산소 유사 상태는 Modular Incubator Chamber (MIC-101)을 이용하였고 5% 이산화탄소와 95% 질소 혼합 가스를 5분 동안 공급하여 저산소 상태를 만들었으며 세포 배양액을 채취하여 혈액가스분석기(Blood Gas Analyzer ABL725)로 세포 배양 상태를 측정하였다. 세포의 증식 상태는 MTT 방법을 실시하였다. EGCG는 0, 12.5, 25, 50,100 ${\mu}mol/L$로 농도 변화를 주어 실험을 시행하였으며 16시간 동안 저산소 상태를 만든 뒤 HIF-1$\alpha$, VEGF, $\beta$-actin mRNA에 대해 Real time PCR을 시행하였다. 결 과: 48시간과 72시간에서 저산소 상태에 놓인 A549 세포의 증식능력은 대조군에 비하여 억제되었다. EGCG 는 저산소화에 의해 유도된 HIF-1$\alpha$의 mRNA의 전사를 유의하게 억제하였다. 그러나 이러한 억제 효과는 VRGF mRNA 발현에는 미치지 못하였다. 결 론: EGCG는 HIF-1$\alpha$의 발현을 억제함으로써 비소세포암주에서의 예방적 항암요법이나 항암 치료요법 시의 주요 작용 목표로 사용될 수 있을 것으로 보인다.
본 연구에서는 초음속 유동장 내 연료 분사시 연소기의 공간적인 제한 조건을 고려하여 복수의 분사기간 배치 간격을 변화시키고 그에 따른 유동 구조, 연료의 침투 거리 및 연료-공기의 혼합 특성을 비교 분석하였다. 이를 위하여 널리 알려진 단일 분사구를 이용한 실험 조건을 모사하여 적용된 수치 모델을 검토하였으며, 동일한 분사 조건을 갖는 복수의 분사기를 이용하여 비반응 유동 해석을 수행하였다. 해석 결과를 바탕으로 분사구 간 거리에 따라 전압력 손실, 침투 거리, 및 혼합 성능 등을 정량적으로 비교하였다. 해석 결과 분사구 간 배치 거리가 매우 짧은 경우 분사 연료가 서로 융합되면서 유동장이 2차원 특성을 나타내었고 전반적으로 낮은 혼합 효율 특성과 높은 전압력 손실을 발생하였다. 분사구 간 거리가 멀어짐에 따라 분사 가스간의 상호작용이 감소하면서 혼합 효율이 증가하고 전압력 손실이 낮아지는 것이 관찰되었다.
Serious environmental problems have been caused by the greenhouse effect due to carbon dioxide($CO_2$) or nitrogen oxides($NO_x$) generated by the use of fossil fuels, including oil and liquefied natural gas. Many countries, including our own, the United States, those of the European Union and other developed countries around the world; have shown growing interest in clean energy, and have been concentrating on the development of new energy-saving materials and devices. Typical non-fossil-fuel sources include solar cells, wind power, tidal power, nuclear power, and fuel cells. In particular, organic solar cells(OSCs) have relatively low power-conversion efficiency(PCE) in comparison with inorganic(silicon) based solar cells, compound semiconductor solar cells and the CIGS [$Cu(In_{1-x}Ga_x)Se_2$] thin film solar cells. Recently, organic cell efficiencies greater than 10 % have been obtained by means of the development of new organic semiconducting materials, which feature improvements in crystalline properties, as well as in the quantum-dot nano-structure of the active layers. In this paper, a brief overview of solar cells in general is presented. In particular, the current development status of the next-generation OSCs including their operation principle, device-manufacturing processes, and improvements in the PCE are described.
An oxygenator is a very important artificial organ and widely used for patients with lung failure or during open heart surgery. Although an oxygenator has been widely studied worldwide to enhance its efficiency, studies on oxygenators, in particular when using a pulsatile blood flow, are domestically limited. Therefore, a new oxygenator was developed in the lab and animal experimental results are described in the paper. The oxygenator is composed of polycarbonate housing and polypropylene hollow fibers. It has a total length of 400 mm and a surface area of $1.7 m^2$. The animal experiment lasted for 4 hours. The blood flow rate was set to 2 L/min and a pulsatile blood pump, T-PLS (Twin-Pulse Life Support), was used. Samples were drawn at the oxygenator's inlet and outlet. The total hemoglobin (Hb), saturation oxygen ($sO_2$), and partial oxygen pressure ($pO_2$), partial $CO_2$ pressure ($pCO_2$), and plasma bicarbonate ion concentration ($HCO_3^-$) were measured. The oxygen and carbon dioxide transfer rates were also calculated based on the experimental data in order to estimate the oxygenator's gas transfer efficiency. The oxygen and carbon dioxide transfer rates were $16.4{\pm}1.58$ and $165.7{\pm}10.96 mL/min$, respectively. The results showed a higher carbon dioxide transfer rate was achieved with the oxygenator. Also, the mean inlet and outlet blood pressures were 162.79 and 137.92 mmHg, respectively. The oxygenator has a low pressure drop between its inlet and outlet. The aim of own preliminary study was to make a new oxygenator and review its performance when applying a pulsatile blood pump thus, confirming the possibility of a new oxygenator suitable for pulsatile flow.
본 연구에서는 바이오매스 및 플라스틱에 가스화 효율을 높이기 위한 탄산염 촉매 또는 Ni based 촉매를 혼합한 시료에 대하여, 고정층 반응기를 이용하여 급속 등온 열분해 실험을 수행하여, 생성된 가스의 온도, 시료 및 촉매의 영향에 관한 분석을 통하여, 최적의 수소 생성 수율을 얻고자 한다. 고위발열량 측정 결과, 바이오매스보다 플라스틱 폐기물의 발열량이 높아짐을 알 수 있었다. 원소분석 결과로부터 수소 함량은 플라스틱 시료가 더 높았다. 계산된 활성화 에너지는 촉매 적용에 의해 감소하였고, 5 wt% 이상의 경우에는 큰 변화를 보이지 않았다. 수소수율은 플라스틱 폐기물이 포함된 시료, 온도에 대해서는 대부분 높은 온도 범위에서 최대값이 얻어졌다. 또한 대부분의 시료에서 높은 혼합비를 갖는 조건의 수소수율이 가장 높은 결과를 보였으나, 5 wt% 이상의 조건에서는 촉매 혼합비 증가의 영향은 미비하여, 활성화 에너지의 결과와 잘 일치함을 확인하였다. 전체적으로 촉매 반응이 무촉매 반응에 비하여 높은 수소수율이 얻어졌다. 촉매 종류에 대하여, 탄산염 촉매인 $Na_2CO_3$ 및 $K_2CO_3$보다 Ni-$ZrO_2$ 촉매가 수소 생산을 위한 목적에 더 적합한 촉매임을 확인하였고, 본 연구로부터의 최대 수소수율을 위한 조건은 $900^{\circ}C$, 20 wt%의 Ni-$ZrO_2$(1:9) 촉매가 혼합된 Pitch Pine, Polyethylene 시료에 대하여 65.9 vol%의 높은 수소수율의 결과를 얻었다.
최근 구제역과 같은 가축질병으로 인한 살 처분된 가축매몰지의 적절한 모니터링 시스템의 필요성이 대두되고 있다. 그러나 현재 사용되는 모니터링 시스템은 살 처분된 매몰지 현장에서의 시료를 채취한 후 1~2주간의 모니터링 기간을 필요로 함으로 실시간 감시가 필요로 하는 매몰지에 대한 연속측정이 불가능하다. 이에 본 논문에서는 이런 실시간 모니터링을 가능하게 하는 우선 센서네트워크 기반의 가축매몰지 환경정보 모니터링 시스템 설계 및 구현을 제안한다. 제안된 시스템의 무선 센서노드는 환경센서(Dust, Co2, NH3, H2s, Temperature, Humidity)와 위치확인을 위한 GPS센서로 구성된다. 제안된 시스템은 게이트웨이를 통해 원격지 서버에 전송된 매몰지 환경정보를 분석함으로 언제 어디에서나 유해환경 모니터링이 기능하고 위치확인을 통한 상황에 따른 능동적인 대응을 가능케한다. 따라서 제안된 시스템의 유효성 검증을 위해 규정된 가스 배출관에 센서를 설치하였고, 통합 모니터링이 가능한 테스트 베드에서 실시간으로 수신된 데이터를 수집, 분석하여 시간의 변화에 따른 매몰지 위치의 악취환경변화를 모니터링 하였다. 이를 통한 상황에 따른 실시간 대응을 함으로 유사시 환경오염의 사전예방이 가능 한 것을 확인할 수 있었다.
메탄은 온실효과가 이산화탄소 보다 20배 이상인 대표적인 non-$CO_2$ 온실가스이다. 매립지는 주요 인위적 메탄 발생원으로, 매립지의 메탄 발생량은 연간 35~73 Tg(tera gram)으로 추정된다. 바이오커버(개방형 시스템)과 바이오필터(폐쇄형 시스템)을 이용하는 생물학적 방법은 메탄을 회수하여 자원화하기에는 메탄 농도가 너무 낮거나 가스 포집정이 설치되어 있지 않는 노후화된 매립지나 소규모 매립지로부터 메탄 배출을 저감할 수 있는 유용한 방법이다. 메탄을 유일탄소원과 에너지원으로 활용하는 메탄산화세균은 이러한 생물학적 방법에 있어 메탄을 산화시켜 제거하는데 매우 중요한 역할을 담당한다. 토양, compost, 지렁이 분변토 등과 같은 다양한 충전재를 이용하여 실험실 규모의 바이오커버/바이오필터의 메탄산화효율에 관한 많은 연구가 진행되었다. 이 중에서 compost는 가장 많이 이용되고 있는 충전재이고, compost를 이용한 바이오커버/바이오필터의 메탄산화속도는 50에서 $700\;g-CH_4\;m^{-2}\;d^{-1}$로 보고되고 있다. 또한, 실제 매립지에 파일럿 규모의 바이오커버/바이오필터를 설치하여 메탄 배출 저감 효과에 관한 연구도 진행되고 있다. 매립지의 메탄 배출 저감은 탄소배출권 거래와 연관될 수 있으므로, 바이오커버/바이오필터에 의한 메탄 저감량을 정확하게 평가하는 것이 매우 중요하다. 그러므로, 매립지 현장에 설치된 바이오커버/바이오필터의 성능을 평가하는 방법은 표준화되어야 하며, 메탄 저감량을 정확하게 정량화할 수 있는 방법 개발이 필요하다.
분유 분말의 C. sakazakii와 양파 분말의 B. cereus의 포자를 저해하는데 있어 가장 효과적인 플라즈마 형성 가스는 각각 He-$O_2$와 He이었다. 분유 분말에 대한 콜드 플라즈마 처리시간이 길어질수록 그리고 형성 전력이 높아질수록 C. sakazakii의 저해도가 커졌으며, 900W, 40분 처리시 가장 큰 저해율을 나타내었다. 양파 분말에 접종된 B. cereus 포자는 콜드 플라즈마 단독 처리와 마이크로파와 병합된 콜드 플라즈마 처리에 대하여 강한 저항력을 가지고 있었으나 열처리($90^{\circ}C$, 1분)를 두 형태의 플라즈마 처리에 선병합시켰을 때 약 90%가 저해되었다. 병합 처리한 결과, 각각의 방법을 단독으로 처리한 것보다 병합 처리시 B. cereus 포자의 저해율(약 1.0 log spores/g)이 높아졌다. 본 연구는 분체 식품에 존재하는 식중독 균의 효율적인 살균 방법을 제시하였고, 분체 식품의 새로운 살균 공정으로서의 콜드 플라즈마의 적용 가능성을 보여주었다.
본 연구에서는 GC-FID를 이용하여 식품첨가물공전에 사용량이 정해져 있는 빵류, 캡슐류, 건조과실류 채소류 및 과실류 채소류 중 유동파라핀의 함량을 측정하였다. 시료에서 n-hexane으로 유동파라핀을 추출하고 $KMnO_4$ 산화반응을 통해 추출물 중 불포화탄화수소의 극성을 증대시킨 다음 aluminium oxide를 충진한 SPE 카트리지로 정제하여 GC-FID로 측정하였다. 검출한계와 정량한계는 각각 10 mg/kg과 20 mg/kg으로 개별 n-alkane으로 정량하는 방법이 아닌 넓은 봉우리로 나타나는 유동파라핀의 면적의 합으로 정량하는 문헌들(15,17)의 값과 유사하게 나타났다. 유동파라핀은 빵류, 건조과실류 채소류 및 캡슐류에서 이형제 등으로 사용되고 있으나 최종 완제품에서 사용기준 이하로 검출되었고, 과실류 채소류에서 모두 불검출로 나타남에 따라 안전한 수준으로 관리되고 있음을 알 수 있었다.
